Physicochemical properties, phytochemicals, and biological activities of heat-treated Elaeagnus multiflora juice and vinegar (열처리 보리수 과즙과 식초의 이화학적 특성, phytochemicals 및 생리활성)

  • In this study, vineger was produced after heat treatment of Elaeagnus multiflora juice and its fermentative characteristics were investigated. The heat-treated juice and vinegar of E. multiflora were similar in fruit color, with b values (redness) of 39.48 (juice) and 37.56 (vinegar). After 10 days of fermentation of E. multiflora fruit, the acetic acid bacteria viable cell number, pH, acidity, reducing sugar content, and alcohol content were 4.59-4.62 log CFU/mL, 3.14-3.45, 0.2-2.12%, 0.69-35.24 mg/mL, and 0.2%, respectively. The heat-treated juice and vinegar showed significantly higher radical scavenging and digestive enzyme inhibitory activities than untreated samples, and the levels of soluble phenolics, soluble flavonoids, flavan-3-ol derivatives, and phenolic and derivatives were increased. Additioinally, the heat-treated vinegar contained major organic acids, such as acetic acid (21.82 mg/mL), and major flavan-3-ols and phenolic acids, such as catechin ($72.24{\mu}g/mL$), catechin gallate ($273.36{\mu}g/mL$), epigallocatechin gallate ($68.35{\mu}g/mL$), protocatechuic acid ($12.84{\mu}g/mL$), and salicylic acid ($42.29{\mu}g/mL$). At $25{\mu}L/mL$ treatment, DPPH and ABTS radical scavenging activities and ${\alpha}$-glucosidase and pancreatic lipase inhibitory activities were 79.66%, 93.99%, 90.12%, and 64.85%, respectively. This result suggested that it is possible to produce new types of vinegar and beverages, using heat-treated E. multiflora juice.

Optimization of Combined Process of Enzymatic Hydrolysis and Solvent Extraction for Production of Lycopene from Elaeagnus umbellata (보리수 나무 열매로부터 라이코펜 생산을 위한 효소 분해 및 유기용매 추출 복합 공정의 최적화)

  • This study was undertaken to optimize combining the processes of enzymatic hydrolysis and extraction for lycopene production from autumn olive berry. The autumn olive berry was pulverized and suspended in water, followed by treatment with various hydrolytic enzymes including Ceremix, Celluclast, AMG, Viscozyme, Pectinex, Promozyme, Ultraflo and Tunicase. Reaction solutions were subjected to extraction by applying different organic solvents including acetone, ethyl acetate, hexane and chloroform. Highest yields of lycopene extraction were obtained with the Ceremix (hydrolysis enzyme) and chloroform (extraction solvent) combination. Subsequently, using this ideal combination, enzymatic hydrolysis conditions, including enzyme concentration, pH and temperature, were statistically optimized to 0.58%, 5.5 and 54.4℃, respectively, by applying the response surface method. The lycopene extraction yield increased 2.3-fold (22.6 mg/100g) by using the selected combined process. We propose that these results could be used for the future development of bioactive materials required for bio-health care products.

Antioxidative Activities of Various Solvent Fractions from Fruit and Leaf of Pinkpop Borisu (핑크팝 보리수 열매와 잎 용매 분획물의 항산화 활성)

  • To get basic data for the utilization of Pinkpop Borisu fruit and leaf as a raw material in functional food, antioxidative activities were investigated. The radical scavenging activity of various solvent fraction from fruit and leaf against the DPPH and ABTS radicals increased with the increasing amount of various solvent fractions and butanol fraction of leaf showed the highest DPPH radical scavenging activities. Among the various solvent fractions from fruit and leaf of Pinkpop Borisu, butanol fraction of leaf showed the strongest scavenging effect on ABTS radicals. Reducing power and ferric reducing/antioxidant power (FRAP) of the various solvent fractions from fruit and leaf increased with the increasing concentration of fractions and butanol fraction of leaf showed the highest reducing power. The butanol fraction of fruit and leaf showed strong antioxidative activity on linoleic acid system. Total phenolics contents of butanol fraction from fruit and leaf were 106.34 and 252.46 mg/g, respectively. The antioxidative activities of the butanol fraction from fruit and leaf are attributed to phenolic compounds. The present results suggest that butanol fraction of Pinkpop Borisu leaf would have the protective potential from oxidative stress induced by free radicals.

Component Analysis and Anti-Proliferative Effects of Ethanol Extracts of Fruits, Leaves, and Stems from Elaeagnus umbellata in HepG2 Cells (보리수나무 열매, 잎 및 줄기 에탄올 추출물의 함유성분 분석과 간암 세포 증식억제 효과)

  • The aim of this study was to evaluate the physicochemical properties and antioxidant and anti-proliferative activities of different plant parts of Elaeagnus umbellata Thunb. extracted with ethanol (EtOH). EtOH extract of stems presenting the highest content of polyphenols showed the strongest 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity ($EC_{50}=54.04{\mu}g/mL$). The total content of free amino acids decreased in the order of leaves (6,179.12 mg/100 g)> stems (1,211.69 mg/100 g)> fruits EtOH extract (378.88 mg/100 g), and asparagine (1,907.57 mg/100 g), ${\gamma}-aminobutyric$ acid (300.17 mg/100 g), and proline (233.48 mg/100 g) were the major free amino acid in leaves, stems, and fruits, respectively. Five phenolic compounds in each extract were measured by using liquid chromatography- tandem mass spectrometry, and gallic acid (98.95 mg/100 g) and (+)-catechin (1,575.99 mg/100 g) were present as major components in leaves and stems, respectively. EtOH extract of leaves showed the highest anti-proliferative activity against HepG2 cells as measured by 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyltetrazoliumbromide and lactate dehydrogenase assay but had no effects on Chang liver cells.

Analysis on the Components of the Emit of Elaeagnus multiflora Thumb (뜰보리수 열매의 유용성분 분석)

  • This study was conducted to analyze components of the fruit of Etaeagnus multiflora to form a part of studies on the nutritional and functional materials of fruits and leaves of E. multiflora, and the development of a functional food. The moisture content of the fruit was 82.34%, and the content of carbohydrate, crude protein, lipid and ash was 15.4, 1.29, 0.79 and 0.54%, respectively. The pH of the fruit extract was 3.29, acidity 0.64%, and brix 14.0. Total sugar was contained 1.03/100g and reducing sugar was 3.27 g/100g. The content of soluble protein was 0.48 g/100g, and polyphenol was 0.28 g/100g. The glucose and fructose as a free sugar was 401.96 and 370.34 mg/100g, and account for a great portion of the components. Total content of free sugary was 0 mg/100g. The total amount of organic acid was 294.44 mg/100g, and the content of citric acid was 265.01 mg/100g, malic acid 18.16 mg/100g, and succinic acid 8.50 mg/100g. In the free amino acid, the concentration of serine was the highest (13.93mg/100g), and alanine, aspartic acid, cystine and methionine were high in the above order. The content of ascorbic acid was 131.35 mg/100g and that of dehydroascorbic acid was 431.37 mg/100g. Potassium, magnesium and sodium content were 627.44, 140.28, and 56.70 mg/100g, respectively.
