Comparative Studies of Invertase Isozymes Produced by Rhodotorula glutinis K-24 (Rhodotorula glutinis K-24가 생산하는 Invertase Isozymes군에 관한 비교 연구)

  • Rhodotorula glutinis K-24 was found to produce internal, cell wall bound, and external invertase. Internal invertase was purified by column chromatographies on DEAE-Sephadex A-50, Sp-sephadex C-50, gel filtration on Sephadex G-200 and isoelectric focusing. Cell wall bound invertase was partially purified by the following procedures; column chromatography on DEAE-Sephadex A-50 and gel filtration on Sephadex G-100. Optimum pH and temperature for enzymatic activities of internal and cell wall bound invertase were pH 3.0 and 6$0^{\circ}C$, respectively. Both enzymes were inhibited by HgC1$_2$, AgNO$_3$, MnSO$_4$, and sodium dodecylsulfate. The molecular weights of internal and cell wall bound invertases were estimated to be 310,000 and 61,000, respectively. Other physicochemical properties of the both enzymes were similar.

  • 동굴은 항상 폐쇄된 세계로 항온, 항습, 항암의 지하세계이므로 특수한 환경을 이루고 있다. 즉 동굴의 온도는 년중 거의가 일정하고 햇빛이 없는 캄캄한 암흑의 세계라는 것이 하나의 특색이다. 대체로 동굴의 대기는 기류의 움직임이 매우 느린 관계로 해구부근에는 대기온도와 동굴의 온도가 크게 달리 나타나지만 차차 안으로 들어가면서 동굴내의 대기온도는 동굴벽, 즉 동굴의 온도와는 거의가 비슷하게 나타난다. 이 때문에 굴속 깊은 지점의 온도는 석회암의 온도와 관계되고 있으며 그 온도는 대체로 지표의 년간 평균기온과 비슷하다.(중략)

  • Transient Analysis on heat transfer of rocket engine combustion chamber and wall temperature variation was carried out, especially, calculations of LOx/kerosene rocket engine with/without fuel film-cooling were conducted. Convective and radiative heat flux inside combustion chamber wall were calculated by the empirical equations for rocket engine combustion, and conduction of wall interior was calculated by numerical method with 2D axisymmetric grid. In this calculations the transient variations of wall temperature, the location changes of peak temperature and so on affected by film-cooling were analyzed.

  • 화재시뮬레이션용 소프트웨어인 FDS (Fire Dynamics Simulator)의 수직벽 화재 시뮬레이션에 있어서의 문제점을 파악하기 위해, 수직벽 프로필렌 화재에 대한 시뮬레이션을 수행하였다. 성능기반설계 등에 널리 사용되고 있는 이 전산유체역학 모델에 포함되어있는 주요 매개변수의 기본값을 사용한 경우, 수직벽 화재에서는 정확도가 매우 낮음을 확인하였다. 프로필렌 연소율 $10.08g/m^2$-s과 $29.29g/m^2$-s에 대한 주요 매개변수의 조사를 수행한 결과, 스마고린스키 상수(Smagorinsky constant)가 기본값인 0.2에서 수직벽에 형성된 경계층이 층류로 예측되었다. 스마고린스키 상수가 0.1일 때 온도분포가 실험과 비교적 잘 일치하였으나 벽면에서의 열유속에는 큰 오차가 있음을 확인하였다.

  • Instantaneous temperature probes were manufactured by pressing method. By using these probes, the instantaneous surface temperature and unsteady heat flux in the cylinder liner of DOHC engine were measured. The main results are as follows; ⅰ) the instantaneous surface temperature of cylinder liner are affected by the contact of piston ring as well as burning gas. ⅱ) the wall temperature of the siamese portion is much higher than other parts. ⅲ) it was shown that the rising trend of heat flux by burning gas are nearly limited to the 1/2-stroke distance from the top of cylinder liner.

  • Transient response of temperature distributions in a Pressurized Water Reactor (PWR) pressurizer vessel wall for the Inadvertent Auxiliary Spray transient has been analyzed with conservatism accounted for the resulting thermal stresses in the regions of the vessel wall which are wetted by the spray water droplets. In order to determine the forced convective heat transfer coefficient at the inner boundary surface of vessel wall where the droplets impinge on and flow down, the transient temperatures of spray droplets when they reach the inner surface of the vessel wall after travelling from the spray nozzle through the pressurizer interior space occupied with the saturated steam-noncondensable hydrogen gas mixture have been predicted. The transient temperature distributions in the vessel wall have been obtained by using the finite element method, and the typical results have been provided. It has been shown that the results of thermal analysis are consistent with representation of the input transient and have plausible physical meaning.

  • Objective : Polymethylmethacrylate(PMMA) is often used to reconstruct the spine after total corpectomy, but the exothermic curing of liquid PMMA poses a risk of thermal injury to the spinal cord. The purposes of this study are to analyze the heat blocking effect of pre-polymerized PMMA sheet in the corpectomy model and to establish the minimal thickness of PMMA sheet to protect the spinal cord from the thermal injury during PMMA cementation of vertebral body. Materials & Methods : An experimental fixture was fabricated with dimensions similar to those of a T12 corpectomy defect. Sixty milliliters of liquid PMMA were poured into the fixture, and temperature recordings were obtained at the center of the curing PMMA mass and on the undersurface(representing the spinal cord surface) of a prepolymerized PMMA sheet of variable thickness(group 1 : 0mm, group 2 : 5mm, or group 3 : 8mm). Six replicates were tested for each barrier thickness group. Results : Consistent temperatures($106.8{\pm}3.9^{\circ}C$) at center of the curing PMMA mass in eighteen experiments confirmed the reproducibility of the experimental fixture. Peak temperatures on the spinal cord surface were $47.3^{\circ}C$ in group 2, and $43.3^{\circ}C$ in group 3, compared with $60.0^{\circ}C$ in group 1(p<0.00005). So pre-polymerized PMMA provided statistically significant protection from heat transfer. The difference of peak temperature between theoretical and experimental value was less than 1%, while the predicted time was within 35% of experimental values. The data from the theoretical model indicate that a 10mm barrier of PMMA should protect the spinal cord from temperatures greater than $39^{\circ}C$(the threshold for thermal injury in the spinal cord). Conclusion : These results suggest that pre-polymerized PMMA sheet of 10mm thickness may protect the spinal cord from the thermal injury during PMMA reconstruction of vertebral body.

  • 균일하게 분포된 내부발열을 갖는 유체가 든 경사진(수평에서 45.deg.까지)정사각형 단면의 밀폐 공간 내에서의 2차원 자연대류 유동 및 열전달에 관한 수치적인 연구가 수행되었다. 4개 벽면온 도가 동일한 경계조건에서 내부발열로 인한 자연대류 유동이 Rayleigh수 1.5*$10^{5}$까지는 층 류유동 영역의 가정하에서 수치적으로 수렴되었다. 경사진 밀폐 공간에서의 유동형태 및 온도분 포는 수평인 경우에 비하여 윗쪽 벽면 근처에서 그 상이점이 많이 나타났다. 경사각도가 증가함 에 따라 평균 열전달율이 아랫쪽 벽과 오른쪽 벽에서 증가하였고 왼쪽 벽에서 감소하였으며 윗 쪽 벽에서는 거의 일정하였다.다.

  • Numerical simulation of natural convection along a vertical wall was carried out to evaluate the computational fluid dynamics simulator, which is to be utilized for study of vertical wall fires. The computed velocity and temperature profiles were compared with measurements over the turbulent boundary layer formed along the wall of 4m high and constant temperature. It fumed out that the simulator with default parameters failed to predict the turbulent natural convection showing the boundary layer flow laminar. The grid size $\Delta$x=5mm, ${\Delta}y={\Delta}z=10mm$ and Smagorinsky constant of the large eddy simulation $C_s$=0.1 were chosen through parametric investigations. Though turbulent mixing was not enough, the velocity distribution near wall, peak velocity, and temperature profile in the turbulent boundary layer agreed well with the measurements.

Growth of 3D TiO2 Nano-wall-like Structure with High Effective Surface Area (높은 유효 표면적을 갖는 3차원 TiO2 나노벽 유사구조의 성장)

  • Nano-materials with high effective surface areas have been applied to functional materials, such as high sensitive gas sensors and biosensors and high-efficiency catalytic materials. In this study, titanate sheets with a 3D nano-wall-like structure, high effective surface area, were synthesized vertically to the substrate by a chemical bath deposition (CBD) process using a Ti sheet and urea. The synthesis temperature and synthesis duration time were controlled to the optimal conditions of a 3D nano-wall-like structure in the CBD process. The synthesized ammonium titanate sheets with a 3D nano-wall-like structure were annealed in air to transform to TiO2 with a 3D nano-wall-like structure for various applications. As a result, the optimal temperature in the CBD process for the synthesis of a uniform ammonium titanate sheet with a 3D nano-wall-like structure was 90 ℃. TiO2 with a 3D nano-wall-like structure was obtained from the ammonium titanate sheet with a 3D nano-wall-like structure by annealing above 550 ℃ for three hours. In particular, TiO2 with a 3D nano-wall-like structure with a dominant rutile phase was obtained by post-annealing at 700 ℃. On the other hand, damage to the 3D nano-wall edge was observed after 700 ℃ post-annealing.
