Sensitivity study of the Yfiler® PLUS PCR Amplification Kit in forensic casework samples (법과학 현장시료에서 Yfiler® PLUS PCR Amplification Kit의 민감도 연구)

  • A variety of Y-STR analysis kits have been developed and used in the forensic field. Prior to the forensic application of a new kit, laboratory validation and sensitivity tests are essential processes in selecting suitable alternatives and for assuring that standard operating procedures are followed. In this paper, we have performed a sensitivity study of a new commercial kit, the Yfiler® PLUS PCR Amplification Kit (Yfiler plus kit, released in 2014) by comparing it with the AmpF/STR® YfilerTM PCR Amplification Kit (Yfiler kit, released in 2004). The Yfiler plus kit includes the 17 Y-STR loci of the Yfiler kit and has been supplemented with 10 new Y-STR loci. First, we analyzed the sensitivity difference between the two kits using commercial control DNA 2800M and 007. In addition, we compared the detection rate between the two kits from the 16 selected forensic casework samples of less than 0.5 ng concentrations. The results show that the sensitivity and detection rate of the Yfiler plus kit are higher than the corresponding rates of the Yfiler kit. In addition, we were able to obtain more Y-STR profiles with the use of the new kit. Thus, we suggest that Yfiler plus kit is a more effective forensic tool to detect Y-STR profiles from forensic casework samples of low concentrations.

  • Vacuum metal deposition(VMD) using gold and zinc is known as a very sensitive technique of developing latent fingerprints on nonporous surfaces by excellent malleability and ductility of gold. However, VMD produces empty print which cannot be identified. There is only presumption about the cause of occurrence of empty print, the exact cause is not known. In this study, we experiment on the freshness of fingerprints and sensitivity of techniques that are estimated as the factors causing empty print, based on this, we suggest cyanoacrylate fuming is effective to redevelop empty print.

  • 평균조도 계산법 중에서 국내에서 주로 사용되는 3배광법과 ZCM의 정확도를 비교하고, 그 적용한계를 파악하였다. 비교방법은 전반확산형 조명기구와 직접식 조명기구에 대하여 3배광법과 ZCM에 의한 조명률을 계산하고, 이 값들을 이용하여 여러 상황하에서의 평균조도를 계산하였다. 몬테카를로 시뮬레이션으로 동일상황에서의 조도값을 구하고, 이 값과 3배광법과 ZCM의 조도값을 각각 비교하였다. MCS법의 정확도는 Moon위 해석적인 방법과 비교하여 입증하였다. 연구결과는 다음과 같다. 1. 방의 크기에 따른 평균조도 비교에서 전반확 형 조명기구와 직접식 조명기구의 경우 3배광법 보다 ZCM이 평균조도 오차가 적었다. 2. 광원의 수 변화에 따른 평균조도 비교에서 직접식 조명기구의 경우 3배광법은 광원의 수가 증가할수록 평균조도 오차가 감소하다가 다시 증가하였고 ZCM 은 점차적으로 감소하였다. 3. 실내면 반사율 변화에 따른 비교에서 직접식 조명기구의 경우 오차 범의가 3배광법에서 크고 ZCM에서 적게 나타냈다. 4. 방의 형태 변화에 따른 평균조도 비교에서 직접식 조명기구의 경우 방의 폭이 좁고 길이가 긴 공간에서 3배광법과 ZCM모두 오차가 크게 낱났다. 또 정방형에서 가까울수록 오차는 작아지면서 3배광법 보다 ZCM의 오차가 적었다. 그러므로 우리나라에서 혼용되고 있는 3배광법과 ZCM중에서 정확도가 높은 ZCM선택이 바람직하다.

  • There are so many study on Heumheum Sinseo as a lawbook. But the study on Heumheum Sinseo as investigation and forensic manual is uncommon. So this study was performed to find the value of Heumheum Sinseo as investigation and forensic manual. In Heumheum Sinseo, we could find the basic ideology of a sense of justice on humanitarianism and rationalism, and democratic thoughts on criminal law such as the presumption of innocence, speedy investigation clause and Limiting the admissibility of the confession. And so many scientific method on crime investigation are described in Heumheum Sinseo. We expect that this study serve as moment for us to be able to find it have 'practical' value to us.

Natural isotopes and trace element analyses in glass samples (판유리 시료에서 동위원소 및 미량원소 분석법)

  • Glass is frequently encountered as types of materials that are submitted to forensic science laboratories as a result of trace evidence transfers. The repeatability and the reproducibility of trace element analysis were presented. An analysis of variance (ANOVA) was performed on laser ablation inductively coupled plasma spectrometric analyses of the fragments to identify the source. Pairwise comparisons were completed for all samples. In a pairwise comparison, each sample was compared to each other for a possible [n(n-1)/2] (n : numbers of the samples) total comparison to associate/discriminate samples using Tukey's HSD method. The aim of this study was to determine the utility of LA-ICP-MS for multi-element analysis of forensic samples. The 12 glass fragments from two manufacturers were collected and analyzed to identify the source. An analysis of variance (ANOVA) was performed on 31 elements in NIST 612 Trace elements in Glass. Elements were classified into four categories defined by the combination of precision and variation of inter-samples. We selected 11 elements, 209Bi, 90Zr, 121Sb, 178Hf, 59Co, 238U, 208Pb, 140Ce, 118Sn, 49Ti and 137Ba. 6 pairs out of 66 possible pairs were not distinguished when compared by 137Ba (p<0.05). However, all samples were distinguished using both 49Ti and 137Ba (p<0.05). In conclusion, multi-elemental analysis with LA-ICP-MS is a potential tecnique for the discrimination of forensic samples.

  • 전력계통의 과도 안정도 해석의 접근방법에는 SI(Simultaneous Implicit)법과 PE(Partitioned Explicit)법 두 가지방법을 사용해오고 있다. SI법에는 Trapezoidal법 등이 있고, PE법에는 Runge-Kutta법, Euler법등이 사용되고 있다. SI법인 Trapezoidal법은 PE법의 Runge-Kutta법 또는 Euler법에 비해 시간간격을 크게 해서 계산속도를 줄일 수 있다는 장점이 있지만, 정화도면에서는 신뢰한 수 없는 단점이 있다. 이 논문에서는 포물선 사법을 이용하여 Trapezoidal법의 정확도를 개선학 수 있는 방법을 제시하고 명확한 수학적 증명을 통해 타당성을 보여준다. 연속함수와 불연속함수에 대해서 Runge-Kutta법과 Trapezoidal법과 제안한 방법을 적용시켜서 제안한 방법의 정화함을 보여준다.
