Effect of Electron-beam Irradiation on Development and Reproduction of Bemisia tabaci, Myzus persicae, Plutella xylostella and Tetranychus urticae (전자빔 조사가 담배가루이, 복숭아혹진딧물, 배추좀나방과 점박이응애의 발육과 생식에 미치는 영향)

  • Four major agricultural insect pests, Bemisia tabaci, Myzus persicae, Plutella xylostella and Tetranychus urticae, were irradiated with 30, 50, 70, or 100 Gy electron-beam. Longevity, egg hatching, emergence, and fecundity of the test insects were measured. Hatchability of B. tabaci, P. xylostella and T. urticae declined with increasing irradiation doses, and all B. tabaci and T. urticae eggs were dead at 100 Gy. When eggs of B. tabaci, P. xylostella and T. urticae were irradiated, hatch was inhibited. B. tabaci adults grown from 70 Gy irradiated eggs did not lay eggs. Fecundity of P. xylostella from the 100 Gy irradiated eggs decreased. When B. tabaci, P. xylostella, M. persicae and T. urticae nymphs/larvae were irradiated, the results were similar as those of the hatched eggs. When P. xylostella pupae were irradiated with 100 Gy, fecundity of emerged adults decreased and no eggs hatched. When B. tabaci, P. xylostella, M. persicae and T. urticae adults were irradiated with 70 and 100 Gy, fecundity decreased and egg hatch of B. tabaci was inhibited. However, the longevity of adults did not change and electron-beam irradiation of all developmental stages had no effect on the longevity of adults.

Quality Changes of Salted Chinese Cabbages with Electrolyzed Water Washing and a Low Storage Temperature (전해수 세척 및 저장 온도에 따른 절임배추의 품질변화)

  • To extend the shelf-life of salted Chinese cabbages, electrolyzed water (EW) was used to wash raw Chinese cabbages before the salting process (to control microbial growth), and different storage temperatures evaluated (0, 4, and $10^{\circ}C$). A tap water washing group (TW) was used for comparison. The initial total bacterial population was 5.36 log CFU/g in the TW treatment and 3.50 log CFU/g in the EW treatment. The EW treatment decreased bacterial numbers by approximately 2 log CFU/g compared to the TW treatment and kept this initial population number for 32 days at $0^{\circ}C$. The salinity had no difference during storage. In general, several factors (pH, reducing sugars, hardness, and transmittance) decreased over time, and decreased slowly with EW treatment and $0^{\circ}C$ storage. Overall, the salted Chinese cabbages with EW treatment showed lower bacterial populations compared to TW treatment, and when stored at $0^{\circ}C$, delayed decreases in quality.

Control Efficacy of Ethaboxam on Chinese Cabbage Clubroot Caused by Plasmodiophora brassicae (Ethaboxam의 배추 뿌리혹병 방제효과)

  • Ethaboxam[(RS)-N-(a-cyano-2-thenyl)-4-ethyl-2-(ethylamino)-1,3-thiazole-5-carboximide] is a novel fungicide with high level of activity against Oomycetes fungi. The control effects of ethaboxam technical and various ethaboxam formulations were investigated against P. brassicae, the causal agent of clubroot disease in Chinese cabbage. When ethaboxam was applied to infested soil, club formation caused by P. brassicae was strongly inhibited at 8.33 mg/L soil and $EC_{50}$ of ethaboxam was 2.65 mg/L soil. Five ethaboxam formulations [10% suspension concentrate (SC), 15% SC, 2% granule (GR), 5% GR, 25% wettable powder] and mixture formulation of ethaboxam and metalaxyl (3%+1% GR) exhibited good efficacy against the pathogen. 10% SC, 15% SC, and 2% GR formulations of ethaboxam showed better disease controlling efficacy on Chinese cabbage clubroot than the other formulations. The $EC_{50}$ values of 10% SC, 15% SC, and 2% GR formulations of ethaboxam were 3.72 mg AI/L soil, 1.1 mg AI/L soil, and 4.95 mg AI/L soil, respectively. Among them, soil drenching application by 15% SC formulation of ethaboxam exhibited the most in vivo antifungal activity on P. brassicae. These results indicate that ethaboxam has a high potential for the control of clubroot disease.

Characteristics of Waste Brine from the Salting Process of Chinese Cabbage (배추의 절임공정 중 폐염수의 특성)

  • Major characteristics of waste brine from the repeated salting and rinsing processes of Chinese cabbage for Kimchi were investigated. Salt concentration of brine was increased with the number of successive salting steps from 12% after 1st salting step to 14% after 5th step. Total waste brine which is the representative of wastewater produced from salting process of cabbage showed 6-8% salt concentration. The pH values in all samples decreased with the number of reuse of brine and showed pH 5.9 and pH 5.2 for the mixture of brine for quarter-cut and small-cut cabbage, respectively, after 5th salting process. Soluble solid contents increased with repetition and resulted in 14-16 $^{\circ}Brix$. Total brine showed increased COD values with the number of reuse of brine and 40-50 ppm of COD values for both quarter-cut and small-cut cabbages.

In Vivo Antifungal Activities of Various Fungicides against Plamodiophora brassicae (다양한 살균제의 배추 뿌리혹병 방제효과)

  • In vivo antifungal activity of 44 fungicides consisting of 3 clubroot fungicides, 7 Oomycetes fungicides, 7 botriticides, 7 blasticides, 9 sterol biosynthesis inhibitors, and 11 broad spectrum fungicides were investigated against Plamodiophora brassicae, the causal agent of clubroot disease in Chinese cabbage. When fluazinam, flusulfamide and cyazofamid, commercial fungicide to control clubroot of Chinese cabbage in Korea, were applied to infested soil, club formations by P. brassicae were strongly inhibited at pot (35 $cm^2$) per 0.63 mg. Ethaboxam and cymoxanil, Oomycetes fungicides, completely controlled Chinese cabbage clubroot at 5 mg/pot, but cymoxanil represented sever phytotoxicity. Besides, dichlofluanid and procymidone of botriticides effectively controlled the development of Chinese cabbage clubroot at 2.5 mg/pot. Chlorothalonil, quintozene and trichlamide, broad spectrum fungicides, showed disease-control efficacy of 85%, 100% and 100% at 2.5 mg/pot, respectively. Most of sterol biosynthesis inhibitors displayed the strong antifungal activity against P. brassicae on cabbage seedlings and plant growth -retarding activity. From these results, 7 fungicides were selected and further tested in vivo antifungal activity against P. brassicae in glasshouse. Among them, ethaboxam showed the most antifungal activity against P. brassicae on cabbage seedlings, followed by fenarimol, procymidone, nuarimol and chlorothalonil.

Effects of Wasabi (Wasabia japonica Matsum) on the Physicochemical Characteristics of Baechu Kimchi during Fermentation (고추냉이가 배추김치의 발효 중 이화학적 특성에 미치는 영향)

  • In this study, we explored the application of Wasabi addition to baechu kimchi with regard to quality improvement, by reviewing the optimum Wasabi level and its effects on the product physicochemical characteristics during fermentation. The final weight percentages of Wasabi in the baechu kimchi were adjusted to 0, 2, 3, 4, and 5% per weight of baechu, respectively, and the samples were fermented for 35 days at $10^{\circ}C$. Following fermentation, the treatments over 3% Wasabi contained the highest pH and lowest total acidity values. Total vitamin C and reducing sugar content increased initially, up to certain fermentation times depending on the level of Wasabi contents within the treatments over 3% Wasabi, and decreased afterwards. The water soluble pectin content increased as the fermentation period increased; however, it decreased with increasing levels of Wasabi. In conclusion, the optimum levels of added Wasabi in the baechu kimchi were 3% and 5% per weight of baechu, to have a fermentation-retarding effect on the product.

  • 식물체가 건조 스트레스를 받으면 각 기관 물질 생산의 변이, 분화 및 발달 억제를 통해서 식물의 생산활동을 현저히 저하시켜, 식물의 생장, 형태, 개체발생 및 대사생리에 영향을 미치는 것으로 연구되어 왔다. 최근 기후 온난화로 인해 온도가 점진적으로 상승하고 가뭄과 같은 이상기상이 빈번하게 발생함에 따라 많은 노지 작물의 농업 생산성이 약화되고 있다. 배추는 우리나라의 대표적인 식품 중 하나인 김치의 주재료로 연중 안정적인 생산과 공급이 필요시 되지만, 배추의 경우 건조 조건에서 엽육조직의 붕괴와 같은 생리장해가 발생되기 때문에 최근 발생되고 있는 이상 기후의 영향으로 안정적인 생산이 어려워지고 있는 실정이다. 하지만, 배추에서는 이상 기후에 대응이 가능한 내건성 품종의 육성을 위한 연구는 미흡한 실정이다. 따라서 본 연구에서는 내건성이 높은 배추 개발을 위한 기초소재를 찾는 것을 목적으로 농촌진흥청 유전자원센터에서 보존하고 있는 재래종 배추 30계통을 분양받아 강릉원주대학교 생명과학대학 내의 조직배양실을 이용하여 연구를 수행하였다. 배지는 Tissue Culture Square Dish(125×125×20mm)에 Agar를 녹여 40ml씩 분주하여 고체배지를 조성한 후, 건조 처리구 Polyrthylene glycol 6000(PEG) 0%(Control), 20%(Mild Stress), 30%(Severe Stress)를 설정하여 60ml씩 추가 분주하여 배양기 28℃에서 15시간 처리를 하였다. 분양받은 각 계통의 종자는 1% 차아염소산 나트륨으로 10분간 종자표면을 살균한 후, 5번 정도 멸균수로 헹군 후, 표면 살균한 재래종 종자를 고체배지 시험관에 6립씩 치상하였다. 식물체 생육은 각 처리구별 3반복으로 하였으며 주간 12시간 주기, 광도 2,400Lux, 온도 20℃의 조직배양실에서 치상 후 7일간 생육하였다. 치상 후 7일간 생육시킨 식물체를 채취하여 지상부생체중, 지하부 생체중, 뿌리 길이를 측정하였으며, 지상부와 지하부로 나누어 50℃에서 72시간 건조시킨 후, 건물 생산량을 조사하였다. 본 실험 결과 건조 처리(PEG-6000)는 배추의 생장을 저해하였지만, 생육 저해 정도는 계통간의 차이가 있는 것을 관찰할 수 있었으며 무처리구와 스트레스 처리구간의 생장량 변화 정도를 기반으로 군집분석을 수행한 결과 'IT110483'과 'IT104903' 계통이 실험에 공시된 계통들 중 상대적으로 강한 건조 내성을 가진 것을 확인할 수 있었다. 따라서, 재래종 배추 계통 중 일부는 건조내성이 강한 새로운 배추 품종을 육성하는데 있어 유용하게 이용될 수 있을 것으로 사료된다.

Effect of Nutrient Solution Concentration on the Growth and Mineral Uptake of Various Wrap-up Vegetables and Herbs Grown with Mixed Planting in DFT Hydroponics (담액 수경재배 시 양액농도 처리가 혼식한 쌈 채소류와 허브류의 생육과 무기양분 흡수에 미치는 영향)

  • The twenty seven wrap-up vegetables (13 Compositae, 14 Brassicaceae) and seven herbs (6 Labiatae, 1 Umbelliferae) were cultivated with a deep flow technique (DFT) hydroponic beds and treated with 3 levels of nutrient solution concentrations of 1.2, 2.4, and $3.6dS{\cdot}m^{-1}$ in summer and autumn season. The pH and electrical conductivity (EC) change of nutrient solution, fresh weight, and mineral contents of plants were investigated. The pH was maintained lower in high electrical conductivity (EC) treatment and in summer than autumn. EC of nutrient solution in EC $3.6dS{\cdot}m^{-1}$ treatment increased up to $4.8dS{\cdot}m^{-1}$ during the growing period in summer season. The growth of tested plants showed high variations by plant species and nutrient solution concentrations. The coefficient variation (CV) of the shoot fresh weight of plants was higher in summer than autumn. The growth of Compostiae and herbs was better at EC $1.2dS{\cdot}m^{-1}$, and 14 Brassicaceae was better at EC $2.4dS{\cdot}m^{-1}$ in summer. In autumn, the growth was better at EC $2.4dS{\cdot}m^{-1}$ in all plants except kale 'TBC F1' and red rape 'honchaetae'. In mineral contents, total nitrogen and potassium were higher in autumn than summer. Total nitrogen, potassium, calcium, magnesium were higher in Brassicaceae than others. Iron and manganese, however, were higher in Compositae. As the results, this study suggests that mixed planting of 27 wrap-up vegetables and 7 herbs in DFT hydroponics in two seasons was possible and EC $1.2dS{\cdot}m^{-1}$ in summer and EC $2.4dS{\cdot}m^{-1}$ in autumn be recommended as for the nutrient solution concentration to produce them safely year round.
