Morphological Study on Differentiation of Hepatocytic Stem Cell by Intrasplenic Transplantation after Partial Hepatectomy (간부분 절제술 후 비장내 이식한 간세포화 줄기세포의 분화에 관한 형태학적 연구)

  • This study was performed to evaluate regenerative effects of intrasplenic stem cell transplantation after partial hepatectomy. To evaluate the regenerative effects, Sprague Dawley rats were used. In vivo the embryonic stem cells of blastocysts were collected from superovulated rats on day 3.5 after the vaginal plug checked. The embryonic stem cells were cocultured with hepatocytes for 8 days, they were transplanted into the spleen. After the intrasplenic transplantation of cultured stem cells, they were initially distributed near the periarterial lymphatic sheath after transplantation in the hematoxylin-eosin staining. Their number were formely increased and their size enlarged at forming small lobules. The embryonic stem cells in the culture proliferated and initially proliferated around the periarterial lymphatic sheath and later they around the trabecula with blood vessels. After the transplantation of stem cells, their cell organelles were well developed rough endoplasmic reticulum at the 20th with prominent epidermal growth factor reaction, developed smooth endoplasmic reticulum at the 30th day, well differentiated bile canaliculi with increased transforming growth factor-$\beta$ and apoptosis reactions.

Studies on the Embryonic Development of the Oriental Tobacco Budworm, Heliothis assulta Guenee(Lepidoptera: Noctuidae) (담배나방의 배자발생(胚子發生)에 관한 연구)

  • Embryonic developmental rates of the Oriental tobacco budworm, Heliothis assulta Guenee, were compared at various constant temperatures and 16 hours of light, and detailed embryogenesis was also studied at $25^{\circ}C$. The egg was nearly globular in form and had an average equatorial diameter of 0.53mm. A single micropyle was in the center of the circular area at the anterior pole of the egg. Durations of embryogenesis at 20, 25, 30, and $35^{\circ}C$ were 147, 81, 61, and 67hrs, respectively. Embryonic death was especially higher at $35^{\circ}C$ than any other temperatures investigated. Embryogenesis progressed with changes in color and pattern, which are quite characteristic at each developmental stage of the embryo. At $25^{\circ}C$, organogenesis began in 14hrs after oviposition, formation of gut completed in 44hrs and eclosion occurred in 80hrs. The embryo formed along the long axis at early developmental stage, moved towards the equatorial plane in 24hrs, and made a half-turn on the plane in 36hrs. In 40hrs, head was oriented to the anterior pole of the egg until eclosion.

Effect of tebufenozide on embryonic and postembryonic development of sycamore lace bug, Corythucha ciliata(Hemiptera: Tingidae) (버즘나무방패벌레(Corythucha ciliata)의 배자발육과 후배자발육에 미치는 tebufenozide의 영향)

  • These studies were carried out to investigate the effects of nonsteroidal ecdysone agonist, tebufenozide on embryonic and postembryonic development of sycamore lace bug, Corythucha ciliata. Tebufenozide exhibited ovicidal activity($LC_{50}=4.0$ ppm). Susceptibility of each instar nymphs to tebufenozide was nearly same($LC_{50}=2.3{\sim}6.0$ ppm). The tebufenozide treatment in the final instar affected the emergence, longevity and fecundity of surving adults significantly. Tebufenozide applied at high concentration (10 ppm) to younger adults delayed prioviposition period, and decreased adult longevity, the number of eggs laid per female and hatchability. The treatment also inhibited the ovarian development of the female adults.

Development of Traditional Baeja Design Applied Jogakbo and Knot (조각보와 매듭을 활용한 전통 배자 디자인 개발)

  • In this study, Creative motifs using face composition of Jogakbo and Knot Symbol were developed, and applied to traditional Baeja of Joseon Dynasty to develop design contents of traditional clothes. As for study method, 7 motifs of new formative image that integrates traditional beauty and contemporary sense were developed by applying Knot Symbols and face compositions of Jogakbo with the use of Adobe Illustrator CS6 and Adobe Photoshop CS6 vector graphic software. The motifs were designed in contemporary image in face compositions like rectangle pattern, triangle pattern, dual rectangle pattern, vertical and horizontal pattern, pinwheel pattern, gojunmun pattern and free pattern by involving various changes like repetition, rotation, reduction, expansion and decomposition and using the colors used in the Jogakbo. It is desired that through this study, traditional Baeja may develop to bear traditional and contemporary image so that our traditional clothes design may become global. Also it is anticipated that this study will contribute to development of culture products of Hanbok like Jeogori, pants and skirt that require change of design in the global era while maintaining traditional beauty to appeal to the emotions of world citizens.

A Study of the Liposome-Mediated Transgenic Chicken Production (리포좀을 이용한 형질전환 닭 생산에 대한 연구)

  • Microinjection of DNA is a general method for generating transgenic animals, but the rate of transgenesis in chickens is very low. So it was carried out to investigate the efficiency of liposome-mediated gene transfer in stage one cell of chicken embryo with GFP expression vector. In order to determine efficiency and duration of the introduced foreign gene, it was microinjected DNA with liposome or naked DNA into the germinal disc of stage one cell or stage-X chicken embryo. Analysis of reporter gene expression in day-4 embryos showed that GFP expression was observed only in the liposome-mediate embryo groups and detectable up to day-8 embryos. The results suggest that stable integration of the introduced gene using liposome is a rare event. Nevertheless the liposome-mediated gene transfer may be a useful method to transfer a foreign gene into the stage one cell of chicken embryos.

Proteome Analysis of Chicken Embryonic Gonads: Identification of Major Proteins from Cultured Gonadal Primordial Germ Cells

  • The domestic chicken (Gallus gallus) is an important model for research in developmental biology because its embryonic development occurs in ovo. To examine the mechanism of embryonic germ cell development, we constructed proteome map of gonadal primordial germ cells (gPGC) from chicken embryonic gonads. Embryonic gonads were collected from 500 embryos at 6 day of incubation, and the gPGC were cultured in vitro until colony formed. After 7-10 days in cultured gPGC colonies were separated from gonadal stroma cells (GSCs). Soluble extracts of cultured gPGCs were then fractionated by two-dimensional gel electrophoresis (pH 4-7). A number of protein spots, including those that displayed significant expression levels, were then identified by use of matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry and LC-MS/MS. Of the 89 gPGC spots examined, 50 yielded mass spectra that matched avian proteins found in on-line databases. Proteome map of thistype will serve as an important reference for germ cell biology and transgenic research.

The Effects of Breed, Laying Age and Egg Storage Period on the Hatchability of Korean Native Chickens (한국토종닭의 품종, 산란 연령 및 종란의 보관 기간이 부화 능력에 미치는 영향)

  • This study investigated the effects of breed, laying age, and egg storage period on hatching performance. Eggs were collected from early laying period (27 to 29 weeks old) and late laying period (50 to 52 weeks old) in Hwanggalsaek Jaeraejong, Korean Rhode Island Red, and Korean White Leghorn chicken breeds. Eggs laid for each period were stored for 3, 7 and 14 days and then hatched. Fertility, hatchability, and embryonic mortality were observed to determine the hatching performance. The results showed that hatching performance was affected by breed, laying age, and egg storage period with independently or interactively. The Korean Rhode Island Red breed had the lowest hatching performance. Hatchability and early embryonic mortality rate increased in early laying period eggs compared to the late laying period, but the late embryonic mortality rate increased in late laying period eggs. Additionally, hatching performance decreased as the egg storage period increased. Using breed-specific hatching management techniques, avoiding late laying period eggs, and shortening the egg storage period to seven days after laying may improve the hatching performance.

A Study on Developing Women's Vest Designs through the Application of Traditional Bojaki Technique (전통보자기 기법을 응용한 배자 디자인 개발에 관한 연구)

  • The purpose of this paper is to develop waist coat designs that can be worn in the present day by researching design techniques and traditional waist coat forms shown on Korean patchworks called 'Jokakbo', which was commonly used in our traditional culture. By selecting patchwork as the primary motive in particular. The Bojaki, also known as deungguhli or baeguhli(背巨里), gives off a natural and vibrant color sensation. A new design that could be practically worn with modern clothing was developed by sufficiently reflecting this color sensation as well as the aesthetic quality of its geometric shape. On the basis of theoretical consideration of traditional cloth and vest, total of 7 points were designed and produced. These works contain diverse application of the traditional Bojaki and vest. The design of the Bojaki grants rhythmic sense to the side where diverse colors and formative patterns shown on the cloth can be seen as monotonous. The entire side opening of the traditional waistcoat can cover multiple sizes. Depending on the movement, the front and back naturally spreads. The vest can also be worn in layers or used as a cotton quilt depending on the season. And after minimal design alterations, it will be possible to wear the vest with diverse modern clothing due to its straight line and form.
