본 연구는 관상가치가 높아 조경 및 관상소재로 개발이 가능한 큰개관중의 전엽체 증식 및 포자체 형성에 적합한 배양조건을 구명하고자 수행되었다. 실험재료는 무가온 온실에서 채집한 포자를 기내에서 발아시켜 전엽체를 획득한 후 8주 간격으로 계대배양하여 실험에 사용하였다. 배지종류에 따른 전엽체의 기내 증식 및 형태형성의 영향을 알아보고자 배양된 전엽체 300mg을 메스로 다진 다음 농도를 1/4, 1/2, 1, 2배로 조절한 MS와 Knop배지에 8주간 배양하였다. 그 결과, 1MS배지에서 전엽체의 생체중이 5.5g으로 가장 많이 증가하였다. 1MS를 제외한 타 처리구는 생체중의 증가수준이 1.1-3.0g에 머물러 1MS배지보다 저조한 수준을 보였다. 현미경을 이용한 전엽체의 관찰결과, 1MS배지는 엽육의 색이 녹색으로 생육이 양호하였다. 전엽체의 증식이 가장 저조하였던, 2MS와 1/4MS는 생육의 저조뿐만 아니라 노화현상도 관찰되었다. 포자체 형성을 위한 최적의 토양조건을 알아보고자, 원예상토, 피트모스, 펄라이트 및 마사토의 비율을 5종류로 달리하여 배양토를 혼합하였다. 혼합된 토양은 사각분($7.5{\times}7.5{\times}7.5cm$)에 충진하여 기내배양된 전엽체 1g을 증류수와 함께 10초간 분쇄한 다음 토양표면에 분주 후 10주간 재배하였다. 그 결과, 원예상토가 높은 비율로 첨가된 혼합조건에서 포자체의 형성이 우수하였다. 그중 원예상토와 마사토를 2:1(v:v)로 혼합한 토양, 원예상토와 펄라이트를 2:1(v:v)로 혼합한 토양에서 포트 당 각 357.0, 339.8개의 포자체가 형성되었다. 또한 형성된 포자체의 생육을 조사한 결과 원예상토와 마사토를 2:1(v:v)로 혼합한 토양에서 생체중, 엽수, 엽장, 엽폭, 근수, 근장 및 SPAD value 등의 생육수치가 우수하였다. 따라서 큰개관중의 전엽체 증식에 적합한 배지는 1MS로 판단되었으며, 포자체 대량생산을 위해서는 원예상토와 마사토를 2:1(v:v)로 혼합한 토양이 적합하다고 판단된다.

Effect of BA concentrations and culture methods on in vitro plant multiplication from shoot-tip cultures of Wasabia japonica was studied. Shoot-tips with leaf primordia and apical meristem were cultured on MS basal medium for all the experiments. Liquid medium for 2 weeks followed by semi-solid medium for 4 weeks containing 1.0 mg/L BA was the best to number of shoots (22.8) and shoot length (3.5 cm). Shoots proliferated could be divided into ca. 5 to 11 of cultures for the multiplication of plantlets. Divided plantlets showed root formation (90%) well onto MS basal medium without growth regulators like IBA and NAA. After rooting, all the plantlets transferred into the pots containing composed soil (bio-media Co., peatmoss $8{\sim}10%$, coir dust $66{\sim}70%$, zeolite $13{\sim}17%$, vermiculite $3{\sim}7%$, perlite $2{\sim}4%$) and grown well into whole plants with multiple shoots.
This experiment was carried out to obtain the information on the crossability, variation of chromosome number in pollen mother cell (PMC) and somatic cell of the progeny from the cross between hexaploid triticale cv. Sinkihomil and two diploid rye varieties. Seed set was 39.3 to 41.6% (averaged 40.5%) in the cross between triticale (P$_1$) and rye(P$_2$), which resulted in 0.33% in F$_2$(selfed F$_1$), 2.69% in F$_1$/P$_1$ 5.47% in F$_1$/P$_2$ respectively. However, seed set was extremely low in both reciprocal crosses when triticale was used as male. Germination rate of the crossed seed was 94.0% in F$_1$ 40.8% in F$_2$(selfed F$_1$), 59.5% in F$_1$/P$_1$ and 65.9% in F$_1$/P$_2$ from the cross between triticale and rye, respectively. Pollen fertility of F$_1$ plant was averaged 18.7% in the cross between triticale and rye. Number of Uni-, Bi-, and Trivalent in PMC was 12. 6, 6.94, and 0.53, respectively, in the F$_1$ between the triticale and rye. There were 28 chromosomes in F$_1$, 21 to 34 in F$_2$, 34 to 38 in F$_1$/P$_1$ and 19 to 23 in F$_1$/P$_2$ from the cross between the triticale and rye, respectively.
Anthracnose fungus was most pathogenic on soybean sprout, of the fungi and bacteria isolated from rotten sprout on market. Bacterial strains associated were not virulent. Dry heat (DHT) applied even as high as $65^{\circ}C$ for 30min. was not effective enough to eliminate the artificially inoculated Colletotrichum gloeosporioides propagules from seedllots. Hot water immersion treatment (HWT), at elevated temperature of $55^{\circ}C$ for 20 min, did eliminate the pathogen but reduced seed germinating and retarded sprout growth: Seed germination was practically acceptable when the seedlots were exposed to at $55^{\circ}C$ for 5 min, but about 20% anthracnose propagules survived. Accordingly, we have optimized the HWT scheme for 5 min at $60^{\circ}C$. This scheme was validated, at small to large scale production system, that surely rule out the possible carry over of the bacterial contaminant from seedlots. This result should improve the shelf-life of soybean sprout on the market.
Antifungal activity on major seedborne diasease of crops was screened by the treatment of the extracts from 50 medicinal plants in vitro and in vivo. The extracts of garlic and taxus, Rheum undulatum, Achiranthes japonica, Glycyrrhiza uralensis, Oenothera lamar kiana treated with the blotting filter paper and water agar methods inhibited the growth of Pyricularia oryzae, Alternaria sesamicola, Colletotrichum gloeosporioides, and Alternaria brassicicola among the tested plants. Antifungal activities on infected seeds by soaking methods were shown even at the dilution of the extracts by 10 times. The activity was the highest in soaking seeds at $25^{\circ}C$ for 24 hours. The effect of plant extract on seed germination was not significant as compared with untreated seed. However, early growth of seedling was increased by the treatment of extracts. The extract of taxus slightly inhibited the seed germination of radish and chinese cabbage but those of Achirunthes japonica, Glycyrrhiza uralensis and Oenthfera lamarkiana showed severe damage on the seed germination and early growth of seedling.
This study was carried out to investigate propagation characteristics of seedling and rhizome in Epimedium koreanum NAKAI. Seed germination essentially needed after-ripening duration after harvesting for 250days and more by stratification. At $20^{\circ}C$ seed emergence days and root length were 8 and 0.46cm respectively. The above-ground part of E. koreanum grew until 60days after root cutting and the underground part of it grew after that time. The optimum amount of organic fertilizer was fermented cow-manure with husk 2,000kg/10a that showed $57.5cm^2$ in leaf area per plant, 6.2cm in new rhizome length, and 0.43% in icariin content, an index component of E. koreanum, while inorganic N-fertilizer made E. koreanum worse as it increased. In nutriculture extruded rice hull media was better for growth than perlite media at 1/2 NHRI solution.
The most remarkable aspect in the hormesis law is that dose of harmful agents can produce effect that are diametrically opposite to the effect found with high doses of the same agent. Minute quantities of a harmful agent bring about very small change in the organism and control mechanisms appear to subjugate normal processes to place the organism in a state of albert and repair. The stimulated organism in more responsive to changes in environmental factors than it did before being alerted. Routine functions, including repair and defense, have priority for available energy and matetial. The alerted organism utilizes nutrients more efficiently, grows faster, shows improved defense, and lives longer. Accelerated germination, sprouting, growth, development, blooming and ripening, and increased crop yield and resistance to disease are found in plants. Another concept supported by the data in that low doses of ionizing radiation provide increased resistance to subsequent high doses of radiation. The hormesis varies with subject plant, variety, state of seed, environmental and cultural conditions, physiologic function measured, dose rate and total exposure. The results of hormesis are less consistently found, probably due to the great number of uncontrolled variables in the experiments. The general dosage for radiation homlesis in about 100 (10 to 1,000) times ambient or 100 (10 to 1,000) times less than a definitely harmful dose, but these must be modified to the occasion. Although little is known about most mechanisms of homzesis reaction, overcompensation of repair mechanism is offered as one mechanism.
This study was conducted to investigate in vitro mass propagation methods suitable for each growth stage of A. aristata (G. Forst.) Tindale, from spore germination to sporophyte formation. Among spores germinated in $1/8-1{\times}MS$ medium and Knop medium, Knop medium yielded the highest germination percentage (87.1%). We cultured prothalli obtained from germinating spores for 8 weeks on media with different concentrations of sucrose and active carbon, as well as different concentrations and ratios of nitrogen, to select a suitable growth medium. A. aristata (G. Forst.) Tindale prothalli grew most actively in MS medium with 3% sucrose and 20 : 40 mM of $NH_4Cl$ and $KNO_3$ (total concentration of 60 mM). We investigated sporophyte formation according to soil type, finding that bedding soil mixed with perlite at a 2 : 1(v / v) ratio yielded the highest number of sporophytes per pot ($73.8/7.5{\times}7.5cm\;pot$). By contrast, when peat moss was used alone or mixed with other substrates, prothallus development and sporophyte formation were suppressed. Therefore, the most effective propagation method for A. aristata (G. Forst.) Tindale is to grow prothalli in MS medium and to induce sporophyte formation in a mixture of bedding soil and perlite (v / v = 2 : 1).
The plantlet was regenerated on MS medium containing BAP (2 mg/I) and 1M (1 mg/I) from leaf discs of pepper after 3 weeks of culture. And then, we investigated the activity of peroxidase and esterase and the pattern of their isozymes from leaf, stem and root in order to observe physiological and biochemical changes on the developemental stage, respectively. The peroxidase was expressed with tissue specificity because peroxidase activity according to the developemental stage of the tissue was not only highest in the leaf of the pepper at 10 days after it germinated but also 2 new bands of its isozyme were found in pI 7.2 and pI 5.2. However, a new pI 3.4 band was found in the leaf and root of the pepper after 14 days of germination, and in the stem was found out pI 5.2 band. As regeneration of leaf dises was progressed, its peroxiase activity was increased about 80% more than that of control after 14 days of culture and new pI 3.2 and 6.5 bands of it isozyme were found. The results suggested that peroxidase would be connected with regeneration of pepper. Also, esterase activity was increased about 50% more than that of control after 14 days of culture, the pattern of esterase isozyme was shown to be 3 cathodic bands and 1 anodic band after 7 days of culture.ulture.
Herbicide options for weed control in sorghum is very limited, hence there is a need for exploring potential herbicides. Sorghum herbicide tolerance field trails were conducted at two locations, Yaechoun, Gyeongsangbuk-do, and Miryang, Gyeongsangnam-do, in 2013. Tolerance of sorghum was evaluated following the pre-emergence application of methabenzthiazuron 70% (WP), simazine 50% (WP), oxadiargyl 1.7% (EC), and dimethenamid-P 5% + pendimethalin 20% (EC) at the standard rate 157.5 g, 75 g, 5.1 g, and 75 g a.i. $10a^{-1}$, respectively. As well as double the standard rate. On a phytotoxicity scale of 0 to 9, methabenzthiazuron (WP) induced injury to sorghum up to level 1 at the standard rate and to 3 at double the rate, but did not significantly affect the yield any statistical difference from the untreated. Simazine (WP) induced phytotoxicity up to levels 2 and 4 at single and double rates, respectively. Simazine (WP) did not significantly affect yield: however, the values were numerically lower than those in the methabenzthiazuron (WP) treatment. Oxidiargyl (EC) and dimethenamid + pendimethalin (EC) induced no or slight phytotoxicity; however they failed to provide effective weed control at the standard rate (32 and 68% control, respectively). Out of the tested, methabenzthiazuron (WP) was found to have potential for use in sorghum whereas the other herbicides caused unacceptable levels of injury.
