A monoterpenoid compound, benzylideneacetone (BZA), is a metabolite of an entomopathogenic bacterium, Xenorhabdus nematophila. Its primary biological activity is an inhibitor of phospholipase $A_2$, which catalyzes the committed step of biosynthesis of various eicosanoids that are critically important to mediate insect immune responses. When BZA was applied to two-spotted spider mite, Tetranychus urticae, it exhibited a dose-dependent mortality in leaf-disc assay. Subsequently BZA was tested against T. urticae infesting apples in a field orchard, in which it showed a significant control efficacy, which was not statistically different with that of a commercial acaricide. BZA also had significant antibacterial activities against three species of plant pathogenic bacteria when it was added to the bacterial cultures, in which it showed the highest inhibitory activity against a bacterial wilt-causing pathogen, Ralstonia solanacearum. The bacterial pathogen caused significant disease symptom to young potato plants. However, BZA significantly suppressed the disease occurrence. This study suggests that BZA can be used to develop a novel crop protectant to control mite and bacterial pathogen.
The sprouting characteristics of Scirpus planiculmis and storage methods of corm for the herbicide screening were investigated under both laboratory and greenhouse conditions. Sprouting and elongation of S. planiculmis corms were continued during storage at low temperature ($5^{\circ}C$) and wet condition. However, sprouting of corm was failed when the surface water was removed from the corm, and elongation was reduced by the treatment of 0.5-1.0% sodium chloride(NaCl) solution. Alternate temperature was more effective on the sprouting of S. planiculmis corm than constant temperature. Optimum temperature for the sprouting of S. planiculmis corm was alternation between $25^{\circ}C$ to $35^{\circ}C$. Storage at $-20^{\circ}C$ for 10 min. or 14.7% dehydration made the corm of S. planiculmis to fail for sprouting. S. planiculmis corm was more tolerant to sodium chloride than Echinochloa crus-galli or Oryza sativa(Donginbyeo) seeds. Sprouting rate was decreased by cutting of the corm. Sprouting percent by vertical cuttings, horizontal cuttings, and vertical plus horizontal cuttings were 72, 56, and 28%, respectively.
These surveys were conducted for the purpose of fundamental data of environmental ecology to save the damage of the fieldmice at the National Park of Mt. Jiri, shown especially in the 5 areas of Jang-dang, U-pyung, Jung-san-ri, Sea-seok and Dae-sung area from October, 1983 to September, 1984 during which the fieldmice were very active. The results were summarized as follows; 1) Fieldmice observed were 3 species: Clethrionomys rufocanus regulus, Apodemus speciosus peninsulae and Apodemus agrarius coreae. 2) Types of fieldmice hole were of 5 types; around the roots of trees, at fallen trees, between rocks and the ground, on the surface of the slope ground, on the surface of the ground. The holes were found mostly around the trees (33.2%). 3) Inhabitant ratio was the highest at Dae-sung area (22.6%). Breeding was the most active in July. 4) Females found in July at Sea-seok area were the most in the number (6.6%). Males found at U-pyung area were the most in July in the number, at Sea-seok area in May, at Dae-sung area in July (5.4%). 5) The weight ranged between 20.3g and 57.4g. Clethrionomys rufocanus regulus was the heaviest (37.8g), Apodemus speciosus peninsulae the next and Apodemus agrarius coreae the highest. The heaviest were those found at Dae-sung area.
An endoparasitoid wasp, Cotesia plutellae, has been used for a biological control agent against the diamondback moth, Plutellae xylostella. It has a symbiotic polydnavirus in their reproductive tract, which is required for its successful parasitization. Here, we measured a specific replication time of the polydnavirus during female development of C. plutellae. We, also, analyzed the reproductive potentials of female C. plutellae under mating or different host conditions. At $25^{\circ}C$, pupal C. plutellae began to develop adult tissues such as compound eyes and wings since day 2. At day 5, all adult tissues including antennae were developed and were ready to emerge. With polyclonal antibody raised against C. plutellae polydnavirus, an immunobloting could confirm virus replication at day 4 during pupal stage. Virus particles could be visualized by transmission electron microscope in the oviduct lumen of day 5 pupae. After adult eclosion, venom gland and ovarian calyx increased in size, though ovarioles did not. Mated females layed large number of eggs (over $60\%$) at first 4 days during their mean longevity of ca. 8 days at $25^{\circ}C$. Unmated females showed less active ovipositional behavior, where all the eggs developed into males. C. Plutellae parasitized both P. xylostella and fall webworm, Hyphantria cunea. However, C. Plutellae developed faster and showed higher successful paarasitization in P. xylostella than in H. cunea.
Purpose of this study was a development of a sprayer arm auto control system that could be operated according to distance from pear trees for automation of pest control. Auto control system included two parts, hardware and software. First, controller was made with an MCU and relay switches. Two types of ultra-sonic sensors were installed to measure distance from pear trees: one on/off type that detect up to 3 m, and the other continuous type providing 0~5 V output corresponding to distance of 0~3 m. Second, an auto control algorithm was developed to control. Each spraying arm was controlled according to the sensor-based distance from the pear trees. And it could dodge obstacles to protect itself. Max and min signal values were eliminated, when five sensor signals was collected, and then signals were averaged to reduce sensor's noises. According to results of field experiment, auto control test result was better than non auto control test result. Spraying rates were 69.25% (left line) and 98.09% (right line) under non auto control mode, because pear trees were not planted uniformly. But, auto control test's results were 92.66% (left line) and 94.64% (right line). Spraying rate was increased by maintaining distance from tree.
This study was carried out to investigate effect of $CaCO_3$ treatment on cultivation of oyster mushroom for suppression of green mold disease and for promotion of mycelial growth to stabilize mushroom production in field and laboratory experiment. Treatment of $CaCO_3$ in PDA media promoted mycelial growth of mushroom and suppressed that of green mold. Addition of $CaCO_3$ in rice straw substrate increased mushroom mycelial growth compared with control. In that case, growth of green mold increased up to treated 0.6% $CaCO_3$ but decreased in treatment beyond 0.8% $CaCO_3$. There were some differences on effect of $CaCO_3$ treatment according to green mold species. Trichoderma longibrachiatum was effected but T. virens was not effected by treated $CaCO_3$. Differences among mushroom strains by treated $CaCO_3$ were not shown. It is confirmed that treatment of $CaCO_3$ can promote mushroom mycelial growth but it's not clear in the field. In the result of field test, treatment of $CaCO_3$ in rice straw substrates tended to increase yield and decrease incidence of disease compared with non-treatment. These results suggest that $CaCO_3$ treatment on cultivation of oyster mushroom can be applied to take preventive steps against of green mold disease.
The differential susceptibility of the two spotted spider mite, Tetranychus urticae and its predator, Phytoseiulus persimilis against extracts of Gleditsia japonica var. koraiensis, Camellia sinensis, Cinnamomum cassia, Lantana camara and their mixtures was evaluated. The plant extracts tested were much less toxic to P. persimilis than to T. urticae. The plant extracts tested had little effect on the survival of P. persimilis adult females. Moreover, reproduction of P. persimilis adult females and eclosion of eggs deposited by treated predators were not seriously affected. The treatments of the plant extracts tested showed no serious toxic effect on P. persimilis eggs and exhibited 84~100% hatchability. The treatments with mixtures 3 and 1 were effective against the adult females of T. urticae and yielded 76.0% and 72.7% adulticidal activity at 7 days after treatment, respectively. The treatment of mixture 2 revealed 63.3% adulticidal activity. Generally, the acaricidal activity of the individual treatment of each plant extract was lower than the mixtures. The adult females of T. urticae treated with mixtures 3 and 1 produced only 29.5~31.3% as many eggs as the control females did. All the plant extracts tested exhibited no noticeable toxic effect to the eggs of T. urticae. These results suggest that mixtures 3 and 1 might be used for the control of T. urticae, and expected to be promising candidates for use in integrated mite management program with P. persimilis.
Two Psyllidae species of Cacopsylla pyricola (Foerster) and C. pyrisuga (Foerster)damaging pear trees have been reported in Korea. However, their ecological characteristics and damagepatterns have not been evaluated yet. To establish basic control measures of C. pyricola, field phenology,overwintering ecology, seasonal fluctuation and temperature-dependent development of C. pyricola wereexamined. C. pyricola overwintered under the bark scale of pear trees as winter form adults and theymoved to fruiting twigs from mid-February. Honeydew produced by C. pyricola nymphs and adults asthey feed caused serious black sooty mold on leaves and fruits. The seasonal occurrence of C. pyricolawas different every year. In 1993, characterized by cold temperature and heavy precipitation, C. pyricolapopulation was maintained highly during growing season. However, the population was decreased rapidlyfrom early July in 1994, year of hot and dry weather condition. In 1995, year of average temperature, thedensity of C. pyricola population was decreased during hot months of July and August, and rebuilt up inSeptember and October. The development periods of C. pyricola eggs were 13.33 days at 15"C, 9.32 daysat 20$^{\circ}$C, 7.82 days at 25"C, 6.60 days at 30$^{\circ}$C, and 7.75 days at 35$^{\circ}$C. The development periods ofnymphs were 33.75 days at 15OC, 23.77 days at 20$^{\circ}$C, 15.21 days at 25"C, and 17.40 days at 30$^{\circ}$C. Theirdevelopment periods and mortalities were increased in higher temperatures. The parameters of nonlineardevelopment model, Weibull and linear development models of Cacopsylla pyricola were estimated.models of Cacopsylla pyricola were estimated.
The Rhizoctonia blight causing by Rhizoctonia spp. is an important disease of turfgrass, requiring fungicide application to maintain acceptable conditions for turfgrass good qualities in the golf course. The experiment was conducted to determine the mean 50% effective concentration inhibiting mycelial growth ($EC_{50}$) value of to flutolanil, pyraclostrobine and hexaconazole to Rhizoctonia solani AG-1 IB, Rhizoctonia cerealis and Rhizoctonia solani AG2-2IV isolated from Gyeongbuk province of Korea in vitro. Five discriminatory concentrations of each fungicide were used to detect in vitro sensitivity. The mean of $EC_{50}$ values to three systemic fungicides was the lowest isolate of R. solani AG-1 IB. However, the sensitivity of fungicides to Rhizoctonia solani AG2-2IV were higher mean $EC_{50}$ value of 0.026 ${\mu}g\;a.i.\;ml^{-1}$ of pyraclostrobine and 0.044 ${\mu}g\;a.i.\;ml^{-1}$ of flutolanil. R. cerealis was the lowest sensitivity to hexaconazole which was an average $EC_{50}$ value of 0.022 ${\mu}g\;a.i.\;ml^{-1}$. Inhibition of mycelial growth rate (%) by three combine fungicides using the $EC_{50}$ value of each fungicide was the highest R. solani AG2-2IV. Results of this study were may confirmed in vitro response fungicide of three Rhizoctonia species for control of Rhizoctonia blight in the field.
Overwintering adults of sycamore lace bug (Corythucha ciliata) infected by an unidentified pathogenic fungus were found on the stems of street trees of sycamore in Cheongju city. The objective of this study was to describe this entomopathogenic fungus infecting overwintering sycamore lace bug adults. This unidentified fungus colonized the insect adult body and formed white colony with subglobose clusters of conidiocarps. The size of conidiocarps was 300 to $400{\mu}m$ and each conidium was 15 to $20{\mu}m$. The conidiospore was globus and 2.5 to $3.0{\mu}m$ in diameter, and the hyphae were 1 to $5{\mu}m$ thick. This fungus was successfully isolated and cultivated on potato dextrose agar medium (PDA). The fungal colony was white and then became light yellow. When conidia from this pure culture were inoculated into the overwintering adults, the fungus formed conidiocarps with the same morphology on the insect body and the lethal rate by the fungus was $88{\pm}16%$. This fungus has over 99% homology with Cordyceps bassiana (imperfect fungal name is Beauveria bassiana) in ITS-5.8s rDNA base sequence. The fungal ecology and the infection process of the fungus into its host need to be clarified before using this fungus as a biological control agent.
