Suppression of LH Concentration by Difluoromethylornithine in Gilts (Gilt에 있어서 Difluoromethylornithine에 의한 LH분비 억제)

  • While ornithine decarboxylase (ODC) is considered a key enzyme in the biosynthesis of polyamines, difluoromethylornithine(DFMO) acts as an inhibitor of polyamine synthesis. Cycling crossbred gilts were randomly assigned to one of two (treatment and control) groups (6/group). An indwelling silicone catheter was surgically implanted in the jugular vein of each animal. DFMO was dissolved in saline(200 mg/ml) and adminstered by i. m. injection at a dose of 80 mg/kg/day. The control group received an equivalent volume saline injection. DFMO was injected 3 times daily(08:00. 16:00. 24:00h) from day 16 of estrous cycle to 21 or until estrus. Once daily blood samples (10ml) were taken from day 14 until two days after the last DFMO treatment. Window blood samples were collected every 15 min for 8 h (from 08:00 to 16:00h) starting on day 16 and continuing until day 21 from one gilt per day. Serum progesterone (P$_4$), estradiol (E$_2$), LH and FSH were measured. Typical concentration profiles for P$_4$ and E$_2$ were seen during the follicular phase regardless of DFMO treatment. Injection of DFMO suppressed the preovulatory LH concentration in the serum(p<0.01) while having no effect on FSH profile. The present results indicate that DFMO had an inhibitory effect on LH secretion in the pig, but did not affect PI, E2 or FSH release.

Follicular Cysts of a Beagle Dog in Diestrous Stage (비글개에서 발정 휴지기중의 난포낭종)

  • A 2 years old female beagle dog was examined because of the vaginal discharge and vaginal swelling at 7th day after mating. Abdominal ultrasonography revealed multiple follicular cysts on both ovaries. Also, the ratio of superficial cells was above 90% based on vaginal cytology. She was still in standing heat, with the tail deflected up and flagging when a male sniffed the vulva and attempted to mount. The dog was diagnosed with multiple follicular cysts by vaginal cytology, clinical signs and ultrasonography. The dog was treated with GnRH $50{\mu}g$ injection by intramuscularly. The sizes of the cysts were decreased two weeks after GnRH treatment, and then the dog delivered an offspring at 68th day after last mating. To confirm the stage of estrous cycle, we performed hormone analyses, retrospectively $Estradiol-17{\beta}$ concentration was 78pg/ml at diagnosis. Progesterone concentration was 6.9ng/ml at the last mating, and 66.9ng/ml at diagnosis. Therefore, we confirmed that follicular cysts was observed during diestrous stage in a beagle dog.

Behavioural Estrous, Short Estrous Cycles and Serum Level of Estradiol-17$\beta$ durig Peri-Estrus following Superovulation Treatments in Korean Native Goats (산양의 과배란 처리에 따른 발정발현과 단발정주기의 발생 및 발정기의 혈중 estradiol-17$\beta$의 농도변화)

  • Behavioural estrus and short estrous cycles were observed and serum concentrations of estradiol-17$\beta$(E2) before and after of estrous were measured following superovulation treatments in 30 pluriparous Korean native goats. The goats were divided into 2 groups. Fifteen goats were injected IM with 1,000IU PMSG on Day 12 of the estrous cycle followed by 10mg PGF2$\alpha$ 48h later(P4+PMSG), and the other 15 goats were injected IM with 10mg progesterone(P4)in oil once daily for 10d beginning at any days of estrous cycle followed by 1,000IU PMSG and 10mg PGF2$\alpha$ at the 8th day of progesterone treatment(P4+PMSG group). After injection of PGF2$\alpha$, onset of standing estrus occurred in 12 of 15 goats(80.0%) at 50.0$\pm$7.7h and in 11 of 15 goats(73.3%) at 135.6$\pm$10.1h in PMSG and group and P4+PMSG group, respectively. The mean interval from PGF2$\alpha$ injection to first estrus was significantly(P<0.01) earlier in PMSG group than in P4+PMSG group. This result indicate that the delayed infusion of P4 in P4+PMSG group caused the later exhibition of their estrous behaviors. However, duration fo frist estrus(31.5$\pm$2.6h vs 26.2$\pm$2.3h), length of estrous cycle(14.1$\pm$3.3d vs 16.6$\pm$3.8d) and percentage of short estrous cycle(50.0% vs 45.5%) were not different between PMSG and P4+PMSG group. The mean concentration of serum E2 in 4 goats showing normal estrous cycle in P4+PMSG group(PP-NEC) was higher than in 6 goats showing normal(P-NEC) or in 6 goats showing short estrous cycle(P-SEC) in PMSG group. The peak level of serum E2 was observed at the time of onset of standing estrus in PP-NEC(67.6pg/ml), 6h earlier in P-NEC(53.1pg/ml) and 6h later in P-SEC(52.3pg/ml) than the onset of standing estrus. The profiles of serum concentration of E2 during the period of peri-estrus was similar in the goats of PMSG or P4+PMSG and also in the goats showing the subsequent estrous cycle of normal or short length.

Production of Transgenic Animals by the Testis-Mediated Gene Transfer II. Production of Transgenic Korean Native Goats (정소실질내 유전자 도입에 의한 형질전환동물의 생산 II. 형질전환 한국재래산양의 생산)

  • The totipotential spermatogonial stem cells of adult testis which give rise to mature sperm cells is well-known to incorporate foreign DNA as well as those of somatic cells. Also, the integration rates of foreign DNA after haploid stages are generally known to decrease and /or is simply bound foreign DNA into the sperm plasma membrane. To overcome these problems, liposome and DNA complexes were used to determine how direct injection of these complexes into testis were integrated into sperm genome and resulted in transgenic offspring. To study this purpose, cation liposome was gently mixed with WAF/hGH DNA (1 : 2) and the complexes were injected into testis. At 10, 20, 40, 60, and 80 days after direct injection into testis, mature sperm cells were recovered by using artificial virgin method from two goats and each semen except a part of semen used for DNA analysis such as PCR or Southern blotting was cryopreserved for the artificial insemination. The results obtained in this study are summarized as follows. 1. By PCR, the presence of exogenous DNA was confirmed up to 80 days after injection with liposome/DNA complexes. The highest integration rates were obtained at day 40 after direct injection. This results suggested that spermatogonial stem cells were integrated exogenous DNA into their genome. 2. Among 23 Korean Native Goats which were artificially inseminated, 4 goats resulted in pregnancy and produced 7 young goats. 3. Two young goats were confirmed as a transgenic by PCR and Southern blot analysis. Therefore, our results suggested that testis-mediated gene transfer can be used as a feasible tools for the production of transgenic livestock.

The Expression of Plasma Progesterone and Estradiol-17$\beta$ Level before Parturition in the Recipients Pregnant by Hanwoo SCNT Embryos (체세포 복제란 이식 한우의 분만 전 혈장 Progesterone과 Estradiol-l7$\beta$ 농도 변화)

  • This study was performed to analyze the characterization of plasma hormonal levels during pregnancy in the Hanwoo recipients pregnant by artificial insemination (AI) or somatic cell nuclear transfer (SCNT) embryos. The synchronized recipients pregnant by SCNT embryos produced by Hanwoo fetal fibroblast cells (n=8) and by AI (control, n=5) were used. The plasma hormonal levels were measured by RIA (P4 and E2) and ELISA (cortisol), respectively. In control, the increase of E2 and the decrease of P4 were occurred immediately before the initiation of parturition. The expression pattern of plasma P4 was similar in both groups from 50 to 10 days before parturition, however, it did not decrease even at the expected date of labor in the SCNT recipients. The plasma cortisol was expressed a lower level during pregnancy in the SCNT recipients. But, the cortisol was increased in the cow aborted around 100 days of pregnancy (n=1). Based on these results, it can be postulated that the failure of the hormonal changes immediately before parturition in the SCNT recipients may be one of the most important reasons for a delayed parturition in clone calving.

Changes of estrus signs and genital organs by hormomal induction of estrus in the bitch (인공발정유도견의 발정기 변화와 생식기의 변화)

  • To investigate changes of estrus signs and genital organs in the bitch by hormonal induction of estrus, fourteen bitches of nulliparous and multiparous(2nd-5th) were grouped into diestrus and anestrus according to their estrus cycle. The hormonal treatments were divided into four groups: group A($PGF_2{\alpha}+PMSG+hCG$) and group B(PMSG+hCG) in diestrus bitches and group C(GnRH+FSH+hCG) and group D(PMSG+hCG) in anestrus bitches. The external signs of proestrus and estrus as well as the vaginal smear findings and natural breeding as estrus detection were investigated in all the experimental groups. Also, genital organs were examined at two months after the hormone treatment. The bitches in anestrus showed 100% of male attraction, vaginal bleeding and vulvar swelling as proestrus signs after the hormonal treatment for estrus induction and they showed higher numerical value of signs than the bitches in diestrus. The group A showed the lowest value in proestrous signs of all the groups. The bitches in anestrus treated with GnRH+FSH showed 100% of positive estrus by vaginal smear findings and 75% of natural breeding as estrus detection index and these values were the highest of all the groups. Pregnancy was recognized in only group C and the conception rate was 7.14% in al the experimental animals. Of the side effects after the hormone treatment, external findings of continous male attraction, continous external swelling and purulent exudate were recognized in all the experimental groups and the bitches in diestrus showed higher value of the findings than the bitches in anestrus. Of the changes of genital organs after the hormone treatment, hypertrophy of uterine horn, sanguineous exudate and purulent exudate as uterine findings were recognized in all the groups and these findings were shown more in the bitches in diestrus than in those in anestrus. These results indicated that group C showed the highest value of all the experimental groups in external signs of estrus and estrus detection and also pregnancy was recognized only in that group, consequently, that the hormonal treatment of group C would be the most effective for estrus induction, and also indicated that bitches in anestrus were more suitable than bitches in diestrus for the induction of estrus. In addition, side effects in external genital organs and uteri after hormone treatment were shown more in the bitches in diestrus than in those in anestrus, indicating that bitches in anestrus would be of choice for estrus induction.

Studies on the Factors Affecting the IN-Vitro Maturation and Fertilization of Bovine Follicular Oocytes (소 난포란의 체외성숙과 체외수정에 영향을 미치는 요인에 관한 연구)

  • 본 연구는 소의 난포란의 체외성숙과 체외수정에 영향을 미치는 요인을 구명하기 위하여 미숙 난포란을 채취하여 형태적 분류를 통해 우수한 란을 공시한 후 난포의 크기, 정액의 형태, 수정능득법, 혈청, 호르몬, 난포액, 난구세포등을 첨가한 TCM-199 배양액에서 배양하면서 체외성숙 및 수정율을 조사하였는 바 그 결과는 다음과 같다. 1. 소 난포란을 채취하여 배양을 통해 형태적 분류를 했을때 A형란은 61.4%, B형란은 12.1%, C형란은 19.2%, D형란은 4.2%였으며 발생중지 또는 퇴화란은 3.0%였다. 또한 A, B, C형란을 배양액에 배양했을때 난포란의 체외성숙율은 각각 89.1%, 78.0%, 52.6%였으며, 수정율은 각각 78.1%, 66.1%, 33.3%였다. 2. 소 난포의 크기를 1-2mm, 3-5mm 및 5mm이상으로 분류하여 채취한 난포란의 수는 각각 67개, 98개, 63개 였으며, 이를 TCM-199 배양액에서 배양했을때의 체외성숙 및 수정율은 각각 56.7%와 44.8%, 82.5%, 72.4%와 46.0%와 28.6%였다. 3. 소 난포란의 체외수정에 있어서 정소상체 미부정자, 희택정액 및 동결정액을 이용하여 매정하였을때 체외수정율과 분할율은 각각 63.3%, 73.3%, 70.0%와 32.7%, 37.8%, 38.3%였다. 4. 소 난포란의 체외수정에 있어서 m-KRB 처리법, HIS처리법, Ca-IA처리법, BFF처리법 및 heparin처리법으로 각각 수정능획득을 유기하였을때 체외성숙 및 분할율은 각각 53.1%, 28.1%, 33.9%와 17.7%, 50.8%와 26.2%, 48.1%와 22.8% 및 58.8%와 32.8%로서 heparin 처리법이 가장 높았다. 5. 소 난포란의 체외성숙과 수정에 있어서 각 농도의 우태아혈청과 FSH, HCG, $\beta$-estradiol을 첨가한 TCM-199배양액에서 배양했을때의 체외성숙 및 수정율은 각각 76.0-82.3%와 26.2-70.0%로서 무첨가에 비해 첨가가 높았다. 6. 발정우혈청 및 우태아혈청 5-20%를 첨가한 배양액에서 배양했을때의 체외성숙율은 각각 71.7-76.9%, 74.0-80.6%였으며 체외수정율은 51.9-58%와 26.2-30.0%로서, 체외수정율의 경우 발정우혈청의 첨가가 우태아혈청의 첨가에 비해 높았다. 7. 난포액20-30%를 첨가한 배양액에서 배양했을때의 난포란의 체외성숙율은 각각 68.0%와 64.6%, 수정율은 각각59.6%와 60.4%로서 난포액10%, 50%를 첨가한 배양액에서 배양시의 체외성숙율과 수정율에 비해 높았다. 8. 1$\times$10 6 /ml의 난구세포를 첨가한 배양액에서 배양했을때의 난포란의 체외성숙율과 수정율은 각각 76.5%와 61.7%로서 FCS 10%와 1$\times$10 4 -10 5/ml 와 1 $\times$10 8/ml난구세포를 첨가한 배양액에서 배양시의 체외성숙율과 수정율에 비해 높았다.

Molecular biological studies on Heat-Shock Responses in Amoeba proteus: I. Detection of Heat-shock Proteins (아메바(Amoebaproteus)의 열충격 대응에 관한 분자생물학적 연구: 1 . 열충격 대응 단백질의 탐색)

  • 세균이 세포내 공생하는 xD strain과 모 세포주인 tD strain Amoeba proteus의 열충격 대응의 차이를 알아 보기 위하여 방사선 동위원소로 표지된 아미노산을 Ca2+_less Chalkley's 용액에서 음작용 경로를 통하여 90분 동안 흡수하게 하고, 저온 및 고온 스트레스에 대하여 새로 합성되는 스트레스 대응 단백질의 양상을 1, 2차원 전기영동 및 자기방사 사진법에 의해서 비교하였다 저온(10"C) 충격에 대응하여 아메바는 두 strain 모두 56.0 kDa, pl 6.0 단백질을 강하게 발현하였으며, xD strain에서는 tD strain과 달리 저온 충격 초기에 66 0 kDa, pl 5.5 단백질의 발현이 중단되었다. 한편 고온(33"C) 열충격에 대하여 두 strain 아메바에서 모두 10여종의 단백질이 새합성되는 것으로 확인되었으며, tD 아메바에는 이들 단백질의 새합성이 완만하게 이루어지는데 비하여 xD 아메바에서는 그중 66.0 kDa 단백질이 고온 대응 단백질로서 신속하게 새합성되는 것으로 나타났다. 이외에도 2차원 전기 영동 분석을 통하여 열충격에 의해서 발현이 촉진되는 다수의 단백질들을 탐지하였다 탐지된 아메바의 열충격 단백질은 분자량에 따라 hsp100군 2종, hsp90군, 3종, hsp70군 및 hsp60군 각 1종, 그리고 small csp군 4종으로 분류해 볼 수 있었다 두 분석의 결과를 종합해 보면 tD 아메바에는 저온 및 고온 충격에 대하여 열충격 단백질의 합성이 완만하게 상승하는 데 비하여 xD strain에서는 신속하게 이루어졌다. 이상의 결과로 보아 아메바의 세포내 공생 세균은 숙주의 열충격 대응기작에 변화를 야기한 것으로 판단된다한 것으로 판단된다. 10mg과 20mg의 estrogen 처리구 사이에 유두 직경, 길이 그리고 용적의 증가량에 있어서는 차이가 없었다. 10mg 및 20mg의 estrogen 처리는 초발정일령을 각각 20일 및 124일 단축시켰다. 전체적으로 이러한 결과는 송아지에 estradiol의 삽입은 성장과 유선 발달을 촉진시키고 초발정일령을 단축시킬수 있다는 것을 강력하게 지적한다. 일치하지 않으므로 더욱 정밀한 조사를 실시하여 분류학상의 위치를 정확히 밝혀 볼 필요가 있을 것으로 생각되었다.연한 도구이자 정신활동으로 보게함으로써, 주제 및 연구방법에서 획일성보다 다양성과 창조성이 강조되고 있다. 그리고 연구에 있어서 주제 의 다양성을 통해 보다 현실생활에 밀접하게 연결되어야 할 필요성은 학문이나 과학의 사회 성에 대한 새로운 인식을 가져다 주고 있다. 이러한 지리교육과정의 좌표의 변화된 측면들 을 고려하여, 지리교육과정의 새로운 방향은 다음의 세가지로 모색될 수 있다. 첫째, 爭點中 心 地理敎育課程이다. 사회쟁점에 대한 접근은 쟁점의 이해와 문제해결에의 지리적 관점의 활용을 통해 학습내용의 시사성과 사실성을 높힐 수 있다. 이때 문제해결능력을 통해 현대 시민의 자질 및 능력을 기를 수 있음은 물론, 다른 한편으로 실제세계 즉 학생의 실생활, 사 회, 국가, 세계에서 일어나는 일들과의 관련성을 갖게 함으로써, 내적 동기화와 외적인 자극 을 강력하게 결합할 수 있을 것이다. 이는 개인적 유관적합성과 사회적 유관적합성을 동시 에 확보하는 데 유리할 것이다. 둘째, 思考中心 地理敎育課程이다. 지리교육은 학생들을 지 식 및 기능의 숙달자가 되도록 할 것이 아니라 기본적 문장해독력의 수준을 넘어 능력있는 사고자로 길러내는 것을 목표로 하여야 한다.

Analyse the Winter Season Home Range of Felis catus resided in Gyeongju National Park of Korea (경주국립공원내 서식하는 들고양이 (Felis catus)의 동절기 행동권 분석)

  • The research was conducted in order to analyze the non-estrous season home range of feral cats which are resided in Gyeoneju National Park of Korea from October 2008 to January 2009. As a result of research, individual feral cats(♂) located outside the woods of national park was examined to 100% MPC $0.18\;km^2$, 95% KR $1.10\;km^2$, 50% KR $0.02\;km^2$, while individual feral cats(♀) located inside forest which near Baekunam temple was indicated to 100% MPC $0.31\;km^2$, 95% KR $0.28\;km^2$, 50% KR $0.03\;km^2$. This shows that feral cats located within forest has greater home range than outside the forest. The home range of feral cats is also analyzed during daytime and nighttime, and both individual feral cats located inside and outside the forest have a wide home range during nighttime than daytime and again feral cats located within forest has greater home range than outside the forest for both daytime and nighttime.

Studies on the Improvement of Performance and Reproductive Efficiency in Farm Animals IV. Assessment of Fertilizing Ability of Korean Native Bull by In Vitro Fertilization with Bovine Follicular Oocytes (가축의 개량 및 번식효율 증진에 관한 연구 IV. 우 난포란과의 체외수정에 의한 한우 종모우의 수정능력 평가에 관한 연구)

  • 본 연구는 종모우의 선발방법으로 난포란을 이용하여 실험실내 정자의 수정능력을 직접 검정하여 평가코자 시도되었다. 즉 본 실험은 후대검정중에 있는 한우 후보종모우 15두의 동결융해정자의 수정능력을 평가하기 위하여 정액을 고장액(HIS)에 처리한 후 DM에서 6시간 그리고 소 난포액이 20% 첨가된 DM에서 4시간 전배양하여 수정능을 획득시켜 정자의 활력과 첨체 반응율을 조사하였고 전배양된 정자의 체내(토끼 난관) 또는 체외수정능력을 조사하기 위하여 FCS 15%, 발정암소혈정(CSS) 10%가 첨가된 mKRB에서 체외성숙된 한우난포란과 수정시켜 수정능력을 평가하였으며 인공수정에 의한 개체별 수태율과도 비교 검토한 바 다음과 같은 결론을 얻었다. 1. 한우 난포란의 체외성숙율은 BSA 첨가구 에서 43.8%, FCS 15% 첨가구에서 67.4%, CSS10% 첨가구에서 69.9%이었다. 2. 토끼 난관에서 체외수정율은 BSA 참가구에서 43.8%, FCS 15% 참가구 41.2% 및 CSS 10% 참가구 35.0% 이었다. 3. 후보종무우 15두의 정액을 HIS-DM으로 처리후 6시간 전배양하였을 때 정자의 활력지수는 9-32%였고 첨체반응율은 19-44% 이었으며 20% 난포액을 첨가하여 4시간 전배양 하였을 때 정자의 활력지수는 9-13% 이었고 첨체반응율은 20-43%로 개체간에 차이가 있었다. 4. 체외수정율은 6.6-85.7%였으며, 발정암소혈청(CSS) 10%가 첨가된 mKRB에서 성숙시킨 난포란이 FCS 15% 첨가된 mKRB에서 성숙시킨 난포란보다 다소 높았으나, 정자수정능획득방법간에는 차이가 없었 다. 5. 체외수정율에 있어서 전배양후 정자활력지수와는 부의 상관이 었으며, 첨체반응율과는 낮은 정의 상 관을 나타냈다. 6. 종모우의 수태율은 체외수정율, 정자활력지수 및 첨체반응율과 낮은 정의 상관관계를 나타냈다. 7. 종모우의 개체간 수태율 우열순위에서는 수정율순위와의 사이에 더욱 낮은 부의 상관관계를 보였다. 8. 이상의 연구결과 비록 후대정검중의 제한된 자료로 인하여 종모우 수태율과 체외수정율간에 유의적 인 상관관계는 없었으나, 연결 한우 수정율 평가에 대한 실험실내의 검정가능성을 찾을 수 있었다.
