Sodium Hypochlorite Solution As a Chemical Wounding Agent for Improving Agrobacterium-mediated Chinese Cabbage Seed Transformation (Sodium hypochlorite처리에 따른 배추종자의 Agrobacterium이용 형질전환 증대)

  • Chinese cabbage (Brassica campestris ssp. napus var. pekinensis Makino) seeds/seedlings were transformed via vacuum-infiltration with recombinant Agrobacterium tumefaciens LBA4404 cells. The agroinfiltration method was determined to be unsuccessful for Chinese cabbage transformation during the analysis of hepatitis B surface antigen expression by ELISA. However, treatment of sodium hypochlorite solution, prior to agroinfiltration, to pregerminated or germinating 1 day- or 2 days-old seeds was proven effectively to enhance transformation efficiency, suggesting that chemical wounding caused by sodium hypochlorite reaction might facilitate Agrobacterium infection and, therefore, transient gene expression in Chinese cabbage sprouts.

Effect of Different Herbicides on Initial Rice and Weedy Rice Seedling Growth under Iron-coated Seeds in Flooded Direct Seeding (담수직파 파종 전 제초제 처리에 따른 철분코팅종자와 잡초성 벼의 출아, 입모 및 초기생육 영향)

  • This research was conducted to determine a pre-germinated herbicides with iron-coated seeds in water and wet hill seeded rice. Days of rice seedling emergence was the faster germination at the application of benzobicyclon > oxadiazon > thiobencarb > butachlor and the sealed iron-coated seeds with pre-germinated seeds> pre-germinated seeds> iron-coated seeds with pre-soaking. The seedling establishment was relatively high in untreated control and benzobicyclon among seed treatments by 63.7% and 75.7%. There was 100% seeds killed of pre-germinated seeds in terms of herbicide phytotoxicity and sealed iron-coated seeds with pre-germinated seeds in butachlor but benzobicyclon was of safe with 2% rice seeds killed. Infant rice seedling height was of 9.2-12.9cm in benzobicyclon and 11.9-16.3cm in untreated control and thus there was relatively normal development and growth at the initial rice seedling.

  • The intraspecific susceptibility of twelve Echinochloa colona(L.) Link ecotypes to herbicides was studied at three different stages of growth. At germination and post-germination stage, the Batangas ecotype was most susceptible to both butachlor (N-butoxymethyl-2-chloro-2', 6'-diethylacetanilide) and thiobencarb (S-4-dichlorobenzyldiethylthiocarbamate), whereas the Bukidnon ecotype was the most tolerant. The susceptibility of the ecotypes used was not associated with the seed weight and maturity. When the preemergence herbicides were applied at the 2-leaf stage of the ecotypes, the Nueva Ecija ecotype was least susceptible to butachlor at 1 ppm, but it was most susceptible to thiobencarb. The reverse was exhibited by the Cagayan ecotype. Differential susceptibility of the ecotypes to propanil (3', 4'-dichloropropionanilide) as determined by the necrotic length of the third leaf of 20-day-old seedlings was pronounced in the Camarines Sur, Iloilo, and IRRI (red) ecotypes. However, no necrotic tip appeared in the Cagayan and Batangas ecotypes. The differential responses varied with the type and the concentration of the herbicide studied and the growth stage of the ecotypes.

Selective Mechanism of Oxvfluorfen and Chlormethoxynil in Crops and Weed Species (Oxyfluorfen과 Chlormethoxynil의 선택작용성(選擇作用性))

  • The germination and the nutrient culture tests in the growth chamber and pot experiment under two types of soil conditions were conducted to determine the selectivity of oxyfluorfen and chlormethoxynil in crops and weeds and for characterization of selective mode of action, the absorption study was also conducted with different absorption methods and application time using $^{14}C$-oxyfluorfen. Oxyfluorfen showed more growth inhibitory effects than chlormethoxynil. In the nutrient culture test, rice growth was greatly inhibited at 2-leaf stage than at 4-leaf stage, and the shoot parts were more inhibited than the root parts. By preemergence application of both herbicides, higher growth inhibition was observed in sandy loam soil than in clay loam soil. Absorption and translocation of $^{14}C$-oxyfluorfen were higher by foliar application than by root treatment, and selectivity of crops and weed species may be explained partly by the amount of absorption.

Effect of Various Factors on Dormancy-Breaking of Digitaria sanguinalis Seeds (바랭이 종자(種子)의 휴면타파(休眠打破)에 관한 연구(硏究))

  • The effects of various factors on germination of dormant Digitaria sanguinalis seeds were studied to obtain the basic informations on establishing an effective control method in upland crops. Germination of dormant seeds was most significantly promoted by light treatment, about 88-89%, and followed by the treatment of alternating temperatures (15 or 20$^{\circ}C$ to 35$^{\circ}C$), showing the similar effects like light. Removal of seed coats also increased germination of dormant seeds by 72%, next to the treatments of light and altering temperature. Potassium nitrate ($KNO_3$) at $10^{-2}M$ and concentrated sulfuric acid ($H_2SO_4$) treatment at the duration of 8 minutes resulted in 20% and 37.8% of germination of dormant seeds, respectively, which were significantly higher than that of the untreated control. Under the conditions of altering temperature, gibberellic acid (GA) at $10^{-3}M$ to $10^{-6}M$, benzylandenin (BA) at $10^{-4}$ to $10^{-7}M$ and their combination had no effect on breaking of dormant weed seeds, and instead inhibited the promotive effects induced by the alternating temperature. However, under the constant temperature at 35$^{\circ}C$ in dark, all these treatments increased germination as much as 20% over the untreated control.

Antifungal activities of sulphamide and dicarboximide fungicides against Botrytis cinerea in several in vitro bioassays (여러 종류의 in vitro 생물검정에서 Botrytis cinerea에 대한 sulphamide계와 dicarboximide계 살균제의 활성 특성)

  • Two sulphamide (dichlofluanid and tolylfluanid) and three dicarboximide fungicides (iprodione, vinclozolin, procymidone) were used to investigate the correlation between in vitro antifungal activities and in vivo disease controlling activities against Botrytis cinerea, a causal agent of tomato gray mold and to develop efficient in vitro assays. They controlled effectively the development of tomato gray mold disease in vivo and their controlling activities were similar one another. However, several in vitro assays revealed that their in vitro antifungal activities were quite different between sulphamide and dicarboximide fungicides; the formers showed stronger inhibition activities for spore germination than the latters, whereas the formers inhibited mycelial growth less severely than the latters. The results indicate that the fungicides having different modes of action can show different in vitro antifungal activities according to in vitro assays, even if they have similar in vivo disease controlling activities. On the other hand, two rapid and efficient in vitro assays named Microtiter plate methods I (MPM I) and II (MPM II) were developed for the evaluation of fungicides for inhibitory activities against spore germination and mycelial growth of B. cinerea, respectively. The antifungal activities of five fungicides of two chemical groups in MPM I and II were correlated with the inhibitory activities against spore germination and mycelial growth using solid media, respectively.

Effects of Several Fungicides on the Spore Growth Period of Alternaria dauci, a Carrot Black Leaf Blight Fungus, Using a Rezasulin-based Spore Survival Assay (Rezasulin 기반 포자 생존 검정법을 이용한 당근검은잎마름병균 Alternaria dauci의 포자 생장 시기에 따른 몇 가지 살균제의 효과)

  • The effects of five fungicides on the spore growth phase of Alternaria dauci, which causes carrot leaf blight, were tested using the spore viability assay (SVA) and agar dilution method (ADM). The average EC50 values for chlorothalonil against seven isolates of A. dauci examined by SVA and ADM were 14.21 ㎍/ml and more than 100 ㎍/ml. Dithianon and folpet also had lower EC50 values in SVA than in ADM, while iminoctadine trisalbesilate had lower EC50 value in ADM. For fluazinam, the EC50 values of SVA and ADM were 1.63 and 2.40 ㎍/ml, respectively. As EC50 values of five fungicides according to the spore growth phase of A. dauci KACC 42997, the efficacy of each fungicide as chlorothalonil, dithianon, and folpet decreased when treated after spore germination rather than when treated with spores before germination. However, iminoctadine tris-albesilate was more effective when treated after spores germinated than when treated before treatment. The excellent effect of fluazinam on the pathogen was maintained until A. dauci KACC 42997 was cultured in potato dextrose broth for 6 hr and the germ tube grew beyond the size of the spore. However, when treated with iminoctadine tris-albesilate and fluazinam after culturing for 12 hr, as the EC50 values of the two fungicides increased to 8.87 and 20.65 ㎍/ml, their efficacies decreased. The results of this study show that the treatment time of the fungicide should be determined by considering the effect of the fungicide on the spore growth phase of pathogens.

Weed Control by Herbicides and Fertilizers Applied Separately or Combined on Kentucky Bluegrass Lawn (캔터키블루그래스 잔디에서의 제초제와 비료의 단독과 혼용처리에 의한 잡초방제)

  • Incorporating herbicides application into a fertilization program has several benefits including saving time and reducing traffics on the lawn. Premixed products of fertilizers and herbicides are commonly known as Weed & Feed in the lawn-care industry. To compare Weed & Feed with separate applications of fertilizers and herbicides on a Kentucky bluegrass (Poa pratensis L.) lawn, a Weed & Feed 28-3-3, containing 0.64% 2,4-D, 0.31% MCPP, and 0.03% dicamba of active ingredients, was used in this study. The first application was in May, with the second in June or Sept. Herbicides in forms of 2,4-D (LV-4, 4EC), MCPP (4EC), and dicamba (Clarity, 4EC) were applied at rates equal to the amounts in Weed & Feed or at half of the rates. The dominant weed in both locations was common dandelion (Taraxacum officinale Weber.) in 2005 and 2004. A secondary weed was Canada thistle (Cirsium arvense (L.) Scop.) in 2004 and broadleaf plantain (Plantago major L.) in 2005. When applied in May and June, fertilizer plus full rate of herbicides treatment achieved 112.3 and 83.7 days of acceptable turf quality in 2004 and 2005, respectively. During the same period, Weed & Feed resulted in 58.7 and 24.3 days of acceptable turf quality, respectively. Our study showed that Weed & Feed was generally as effective in weed control as the same amount of fertilizer plus half rates of herbicides sprayed although results may vary due to the timing of application. Fertilizer plus full rates of herbicides provided the same or better results of weed control than Weed & Feed.

  • This study was conducted to develop labor-saving cultural techniques for weed control using herbicides in Rehmannia glutinosa and Astragalus membranaceus, Napropamide, linuron(preemergence) and paraquat (postemergence) showed significant herbicidal effects. At the same time they did not turn out to be harmful to the growth of Rehmannia glutinosa. The yield of Rehmannia, glutinosa treated with paraquat was higher than that of the untreated. Napropamide showed significant herbicidal effect without harmful effects to the growth of Astragalus membranaceus. Etha1flura1in and metolachlor, however, were s1ightly harmful. The yield of Astragalus membranaceus was highest with napropamide.

  • Germinating ability of green seed in maturing period was investigated to develop the technics on generation shortening. Rapeseed did not germinate until 40 days after flowering (=DAF) in green seed and 50 DAF in dried seed. Effective germination to be utilized for generation shortening came on 55 DAF for dried seed by heating and on 75 DAF for green seed. The periodic time of seed dormancy in maturing period had two cycles and rest period was considered that it was different according to the ecological types of early, middle and late maturity.
