  • Interspecific crossing was conducted to obtain hybrid lilly with better quality of adapting to the unfavorable environment using oriental Lilium spp. as a female parent. Average interspecific crossing rate between oriental and asiatic lines by cut-style pollination was as low as 29%. Although the average germination rate of zygotic hybrid embryo between oriental 'Rodolfa' ${\times}$ asiatic 'Toronto' was increased from 76.6% to 78.3%. on 114 or 1/6 strength MS medium compared to the control, the rate of zygotic embryos formation between oriental 'Farolito' ${\times}$ asiatic 'Buff Pixie' was significantly enhanced up about 86~90% on 1/2 strength MS medium. Meanwhile, germination rate of interspecific hybrid embryo between oriental 'Snow Cristal' ${\times}$ asiatic 'Royal Trinity' showed up 100%. Germination rate of interspecific hybrids slightly increased by addition of $0.5-1.0mg{\cdot}L^{-1}$ GA. Germination rate of zygotic embryos between oriental 'Farolito' ${\times}$ asiatic 'Buff Pixie' and oriental 'Belcanto' ${\times}$ L. callosum was highest on the MS medium containing $0.01mg{\cdot}L^{-1}$ NAA and $0.1mg{\cdot}L^{-1}$ kinetin, as 66.6% and 45.2% respectively.

  • The experiment was conducted to elucidate the growth response of rice seedling to heavy metals, copper and zinc. The copper and zinc concentration in rice seedling was 50mg/l and 100mg/l, respectively. Germination. chlorophyll and organic compounds were investigated in seedling of rice with or without heavy metals. Root growth and germination were inhibited at 50mg/l Cu and at 100mg/l compared to control and such reduction in growth was more remarkable at 100mg/l Zn than at 50mg/l Cu. Chlorophyll content was slightly reduced at both 50mg/l Cu and 100mg/l Zn. Malic acid was remarkably increased at 50mg/l Cu compared with that at Zn 100mg/l regardless of rice cultivars. The content of organic acids such as succinic, citric, and malic acid and free proline was increased with treatment of copper and zinc.

  • This study was conducted to examine effects of concentrations of abscisic acid (ABA) or /kinds of osmotica on induction of somatic embryos (SEs), germination and plantlet regeneration in Japanese larch (Larix kaempferi). In comparison of duration of culture, concentrations of ABA and osmoticum, the highest induction number (191/g tissue) of the SE was showed in $60{\mu}M$ ABA+0.2 M sucrose for 4 weeks culture. However, the lowest number (3.5~23.5) of SEs was induced from $4{\mu}M$ ABA+0.1 M sucrose, regardless of culture duration for SEs induction. In comparison of germination efficiency of SEs, the highest induction frequencies of cotyledon (90.9%), hypocotyl (95.8%) and root (96.5%), respectively, were obtained from the SEs that cultured from the treatment of $60{\mu}M$ ABA+0.2 M sucrose with 5 weeks culture. In contrast, the lowest germination response was showed in SEs that induced from the treatment of $4{\mu}M$ ABA+0.1 M sucrose. In comparison of effect of different kinds/concentrations of osmotica for germination and plantlet regeneration, the best response was obtained from the treatment of 0.2 M sucrose with induction of cotyledon (98.3%), hypocotyl (78.4%), root (57.5%) and plantlet regeneration (54.8%), respectively.

  • Experiments were conducted to find out the method of reducing viviparous germination of rice seeds using growth regulators during grain filling period. Among the recommended varieties, early Japonica type showed higher rate of viviparous germination, while late Japonica type and Tongil type varieties did not show viviparous germination in field condition. Grain showed high germination ability from 30 days of grain filling period in Daeseongbyeo of Japonica type and 35 days in Taebackbyeo of Tongil type. Growth regulators, NAA, CMH and Reglone were effective for reducing viviparous germination to the two rice varieties and three treatment stages in this experiment. NAA and CMH were most effective at 100 ppm and Reglone was at 0.4-0.6% concentration. Germination rates of rice seed were not different from non-treatment when NAA CMH were treated, while Reglone lowered germination rate. NAA and CMH had no effects on grain filling rate for both varieties, which showed high grain filling rate as the non-treatment, but Reglone results low grain filling rate. Grain color was not changes when NAA was applied, while CMH and Reglone made a lot of grain to dark brown as the concentration became higher. When grain were hulled, NAA had no difference with the non-treatment whereas CMH and Reglone had lower rate of perfect brown rice when they were applied at 20-30 days after heading.

  • In order to investigate the stimulatory effect(hormesis) of low dose gamma-radiation on the seeding growth of old seeds with respect to antioxidant defense systems, various doses of gamma radiation to the 5-year-old dried seeds of Chinese cabbage (Brassica campestris L. cv Sulin eockaei). Compared to the new, 1-year- old seeds, old seeds irradiated by low does gamma radiation in the range of 1-2 Gy showed vigor growth as revealed by statistically significant increases both in the germination rates and the leaf size and fresh weight. Further, seedlings grown from seeds treated by low dose gamma radiation showed higher peroxidase and catalase activities than non-treated seedlings. These results suggest that the antioxidant defense systems could be closely related to the stimulatory effects of low dose radiation. • This experiment was conducted to elucidate the salt tolerance mechanism of crop plants. Plants used in this experiment were rye, barley, Italian ryegrass. Seeds were treated at saline condition and the saline conditions were controlled by NaCl. The results were summerized as follows. In rye, germination ratio and speed were above 90% and 80% at NaCl concentrations below 0.5% but germination ratio was seriously decreased at NaCl concentration above 0.6%. In barley germination ratio was not less than 80% at NaCl concentration below 0.8%. In italian rye grass germination ratio was decreased gradually by increasing NaCl concentration but germination speed was seriously decreased at NaCl concentration above 0.6% as not more than 7.5%. Water absorption rate was low in barley than rye and it was decreased by increasing NaCl concentration and duration of absorption. In barley, carbohydrate content was decreased slightly in response to increased NaCl concentration and content of reducing sugar was independent of NaCl concentration. In rye, reducing sugar content was increased until 0.3% of NaCl and increased seriously after 20 hours. $\beta-Amylase activity was high at control and 0.6% lot of NaCl, but it was not general tendency.

  • This study was conducted to evaluate the effect of optimal aeration time of livestock manure slurry with fruits pomace on germination index. Six co-digestates of livestock manure slurry with fruits was aerated with 0.1 ㎥ air/㎥·min for 54 days. The maturity of digestates was evaluated using the germination method. The germination index(GI) of co-digestate of SS + CS + MP was more than 70 at the 30th day of aeration. The GI of co-digestate of SS + CS was more than 70 at the 36th day of aeration. The GI of digestate of swine manure slurry alone was 70 at the 54th day of aeration. The co-digestate of SS + MP caused to shorten 24 days of aeration period to reach GI of 70, compared to swine manure digestate. These results suggest that the germination index of seed could be used to establish the optimal aeration time for co-digestate of liquid fertilizer.

  • During 10 days germination of perilla seeds for sprouts preparation, the changes of proximate composition and antioxidant activities were monitored, and the inhibitory effect of sprouts extracts on perilla oil oxidation was also studied. The moisture content in seeds(2.9%) was increased to 9.2% in sprouts at 10 days while crude ash content wasn't significantly. The crude fat and protein contents were reduced from 46.8% and 20.7% in seeds to 18.2% and 18.3% in sprouts, respectively, but reducing sugar and fiber contents increased from 2.2% and 14.8% to 12.8% and 22.4%, respectively. Compared with perilla leaf, sprouts at 10 days contained more fat, carbohydrate, reducing sugar, and fiber while less moisture, ash, and protein. Antioxidant activities during germination were increased and reached to maximum at 8 days in which Trolox equivalent antioxidant capacity(TEAC) based on DPPH and ABTS radical scavenging were 133.1 and 136.8 Trolox eq. mmol/kg, respectively, and ferric ion reducing power(FRAP) was 399.3 Fe(II) eq. mmol/kg. Polyphenol content(19.2 g/kg) was maximum at this stage, too. Perilla leaf showed similar TEAC but higher FRAP than the sprouts. When methanol extract of sprouts at 8 days was added to perilla oil, the oil oxidation was delayed in dose dependent manner. The induction time for oxidation was extended about 2.8 times by adding 2.5%(w/w) extract, that is, from 1.67 hr(control) to 4.62 hr. This induction time corresponded to 38% level of that of perilla oil containing 2.5% BHT.

  • This study was performed to evaluate changes in the chemical composition of germinated rough rice with high hydrostatic pressure treatment (HPT). Rough rice was germinated at $37^{\circ}C$ over 6 days (control), and then subjected to HPT at 30 MPa for 24 h. The highest crude protein content was 9.54% in the control sample after 6 days of germination. Crude lipid content increased from 2.04-2.74% (control) to 2.27-3.10% (HPT). HPT samples showed higher values of total free sugar and glucose content than those of the control. The total amino acid value was not significant, but the essential amino acid content increased from 0.45-5.09 mg/g in the control to 1.57-5.30 mg/g in the HPT sample. The major fatty acids were found to be palmitic, oleic, and linoleic acid. The content of oleic acid decreases with HPT, whereas that of linoleic and linolenic acid increased slightly during the initial stages of germination. These results suggest that HPT after germination efficiently depolymerizes chemical components and enhances the content of essential nutrients.
