Development of Reagent Solution for Identifying the Chloride-based Packaging Materials (염소계 합성수지 포장재 식별용 진단시약 개발)

  • In respect of environmental hazard or human safety, the use of the chloride-based packaging materials like polyvinyl chloride (PVC) blister and film, Polyvinylidene chloride (PVDC) laminated film has been restricted with national regulations. In this study, I intended to develope the reagent solution for identifying the chloride-based packaging materials, so that it was made by mixing methanol ($CH_3OH$) saturated with sodium hydroxide (NaOH) and pyridine as solvent with same ratio. The efficacy of this reagent solution was verified by the analysis of chemical mechanism and experiments that showed the color change (brown) of the specimens. This analysis methode using reagent solution is so simple, convenient, inexpensive and immediate that I expect it will be useful in the field of packaging.

Anesthetic-Induced Acid-Base Equilibrium and Its Electric Dichroism on the Bacteriorhodopsin (Bacteriorhodopsin에 관한 마취제 유도 산-염기 평형 및 Electric Dichroism)

  • When anesthetics were added to a bacteriorhodopsin (bR) in the purple membranes, the 570 nm absorption band shifts to 480 nm. This anesthetic-induced spectroscopic change is reversible. The apparent pKa (6.3) of this equilibrium depends on the nature of the anesthetics in which bR is dispersed. The electric orientation measurements showed that the native bR is easily achieved by relatively small electric field which is oriented at $60^\circ$, while anesthetic-treated bR is not the case. These results demonstrate that the subtle changes in the chromophore and the protein structure surrounding the chromophore by anesthetics influence the spatial orientation of the charged residues in the protein matrix surrounding chromophore.

Coloring Analysis of Digital Textile Printing According to the Type of Fiber (섬유 종류에 따른 디지털 텍스타일 프린팅의 발색성 비교 분석)

  • Whole process of textile printing is made by computer and it remarkably improves environment problem. This digital textile printing is becoming next step environment friendly textile printing method. But, still now range of textile possible for digital textile printing is limited, and also color analysis according to fiber types is not completed. The ink printed on the fabrics through DTP printer can be absorbed and fixed into textile without any blots by pre-treatment using suitable media solution for fabrics types. The chemical formulation of media solution used in the pre-treatment process varies according to the types of textiles and inks for DTP products. First, I studied reference books or articles about color analysis of digital textile printing. Second, I recorded pre-process, printing, post-process and coloring of silk, wool, nylon at same condition. After that, I analyzed $L^*\;a^*\;b^*$, Total K/S, ${\Delta}E$ and studied color intensity and coloring. According to this study, I suggested particular textile special for coloring and manual for affective coloring control. It showed that the performance of the digital printing on the Silk, Wool and Nylon blend fabrics treated by the media solution developed in this study was better than the one treated by the previous media solution for each single inks.

  • Methyl mercaptan oxidase was isolated and purified from Thiobacillus thioparus TK-m for the detection of mercaptans. The procedure of purification involved DEAE-Sephacel and Superose 12 column chromatographies with recovery yields of 47.5 and 48.5 %, and specific activity of 374 and 1240.8 units/mg-protein, respectively. The molecular weight of purified methyl mercaptan oxidase was determined to be 66.1 kDa by SDS-PAGE. Optimum temperature for activity was observed at $55\;^{circ}C$. This enzyme was activated by $(NH_4)_2SO_4$ and NaCl and inhibited by $NH_4Cl$.

  • To develop a home-version assay system for plasma lipoprotein cholesterol, variables that can control the assay performance were optimized. The system was constructcd by using two major components: nitrocellulose membrane strip with immobilized enzymes (cholesterol esterase, cholesterol oxidase, and horseradish peroxidase); and sample carrier solution containing non-ionic detergent (Triton X-100) and chromogen (3,3'-diaminobenzidine). Once a sample combined with the carrier was absorbed from the bottom of the strip, cholesterol was delivered by capillary action to the immobilized enzymes and a sequential reactions took place. In the final reaction, the chromogen was oxidized and then generated a color as signal that was proportional to the concentration of cholesterol. The signal intensity was enhanced by optimizing conditions for the immobilization of enzymes and the chemical composition of carriel. Under these conditions, a dose-response curve was obtained and revealed a high sensitivity enough to measure the cholesterol in blood.

Effect of Color Developing by Heat Treatment on Cotton Fabrics Dyed with Catechu Extract (아선약염색 면직물의 열발색 공정에 의한 발색효과)

  • In order to clarify the availability of heat treatment for catechu dyeing, effect of color developing by heating process on cotton fabrics dyed with catechu extract was investigated. Dyed fabrics were heated various time(min) at $150^{\circ}C$. The cases of non-mordanting and mordanting were compared, and examined the effect of sodium acetate as an agent to promote the color developing to fabrics. Experiments with after-mordanting method showed that various colors can be obtained using catechu. K/S values of dyed fabrics with Al, Cu, Fe mordant were higher than the case of no mordant. The fabric by Cu mordanting showed 3 times to the case of no mordant. Fabrics dyed with catechu by mordanting lowered $L^*$ value while there was little change in $a^*$, $b^*$ value as the heating process progressed. Therefore, K/S values were increased due to heating process. The effects of adding sodium acetate in dyeing bath on dyed fabrics were shown higher K/S values, and the $L^*$ values were lower than those without sodium acetate according to heating time increase. The result show sodium acetate had a promoting effect on the color developing to the fabrics by heating process, it was very effective.

  • In the ternary system of $CaF_2-CaO-P_2O_5$. Apatite$(Ca_{10}(PO_4)_6F_2)$ single crystal having a congruent point was grown by FZ process. The atmospheric condition was kept by oxygen blowing. Adjusting the growth parameters of rotation rate, growth rate and gas amount, we tried to find the optimum growth condition. By partly substituting Ca as Co element, the absorption of infrared is increased and the color effect was observed. Using the Laue back reflection, XRD and FTIR analysis, the characterization of the crystal was carried out.

  • The PPP-MO method has been used to calculate the $\pi$ electronic excitation energy of fluorane dyes. A good correlation between the absorbed and calculated values for the ${\pi}{\rightarrow}\pi*$ transition was found for fluorane dyes. The effect of substituents on the color of fluorane dye was studied on the basis of these calculated results, i.e. HOMO-LUMO energy levels and $\pi-electron-density$ changes accompanying the first excitation.

Color Enhancement of Titanium with Nitrogen ion Implantation (질소이온주입을 이용한 티타늄 발색 향상)

  • We implanted $N^+ion$ into TiO$_2$/Ti substrates with 70 keV by varying dose of 0, 2, 5, and $10{\times}10^{17}/cm$^2$$. In addition, $N^+ion$implanted TiO$_2$ specimens were annealed at $600^{\circ}C$ for 2 hours in Atmosphere. We investigated the color evolution, surface roughness, and hardness of specimens with doses. We report that the color changed from white into dark-yellow as dose increased. ion implanted surfaces became smooth when they were annealed. Moreover, hardness increased up to 10% when we annealed ion implanted TiO$_2$. Our results imply that we may enhanced titanium color and surface hardness.

The Distribution of ATPase and Porin in the Bovine Heart Mitochondrial Cristae (소(牛) 심근 미토콘드리아의 ATPase와 porin의 분포)

  • ATP is the energy source synthesized at the electron transferase that consist of complex I, II, III, IV and V in mitochondrial cristae. The complex V functions as ATPase which composed of sub-complex $F_0$ and $F_1$. Porin or VDAC (voltagedependent anion-selective channel), is a family of small pore-forming proteins of the mitochondrial outer membrane, and play important roles in the regulated flux of anion, proton and metabolites between the cytosolic and mitochondrial compartments. The channel allows the diffusion of negatively charged solutes such as succinate, malate, and ATP in the fully open state, but of positively charged ions in subconducting state. In this study, in order to investigate the relationship of the function and localization between porin and ATPase we observed the distribution of porin and ATPase in the mitochondria of the bovine heart. Monoclonal antibodies against porin and ATPase ${\beta}$-subunit were used to detect porin and ATPase using light microscope with immunohistochemistry and immunofluorescence, and using electron microscope with immunogold-labeling. ATPase were stained in longitudinal section region in cardiac muscle, porin were stained in longitudinal section region in cardiac muscle. We viewed more specific pattern of localization and distribution of these proteins using immunofluorescence method. There were some region which were labeled with porin or ATPase respectively, and others which were labeled both proteins in cardiac muscle. The electron microscope results showed that immunogold labeled porin were labeled locally at mitochondrial outer membrane and ATPase were labeled evenly at mitochondrial cristae. But ATPase was not labeled at mitochondria cristae. These results confirmed the subcellular localizations of porin and ATPase in mitochondrial outer membrane and cristae. Also, we assumed that ATP synthesis always does not activation in all mitochondria exist in the bovine cardiac muscle.
