Analysis of Physical Status on COVID-19: Based on Impacts of Physical Activity (COVID-19에 대한 운동중재효과 분석)

  • The purpose of this perspective research is to discuss the potential role of exercise-interventions in COVID-19, terms of prevention and prognosis in the periods of the COVID-19 vaccine. SARCO-CoV-2. COVID-19 was detected as a new virus causing severe cardiovascular and respiratory complications. It emerged as a global public health emergency and national pandemic. It caused more than 1 million deaths in the first 6 months of the pandemic and resulted in huge social and economic fluctuations internationally. Unprecedented stressful situations, such as COVID-19 blue and COVID-19 red impact on many health problems. In healthy individuals, COVID-19 infection may induced no symptoms (i.e., asymptomatic), whereas others may experience flu-like symptoms, such as ARDS, pneumonia, and death. Poor health status, such as obesity and cardiovascular and respiratory complications, are high risk factors for COVID-19 prevention, occurrence, and prognosis. Several COVID-19 vaccines are currently in human trials. However, the efficacy and safety of COVID-19 vaccines, including potential side effects, such as anaphylaxis (a life-threatening allergic reaction) and rare blood clots, still need to be investigated. On the basis of direct and indirect evidence, it seems that regular and moderate physical exercise can be recommended as a nonpharmacological, efficient, and safe way to cope with COVID-19. Physical inactivity and metabolic abnormalities are directly associated with reduced immune responses, including reduced innate, CMI, and AMI responses. Due to prolonged viral shedding, quarantine in inactive, obese and disease people should likely be longer than physical active people. Multicomponent and systemic exercise should be considered for the obese, disease, and elderly people. More mechanism research is needed in this area.

Oxidized LDL is a Chemoattractant for the Eosinophils and Neutrophils (산화 저비중 리포 단백이 호산구와 호중구에 대한 화학주성)

  • Background : Rhinovirus infection of the airways results in increased permeability of the airway vascular endothelium with the influx of plasma proteins, including lipids such as LDL. In vitro studies on the effect of oxLDL on leukocytes has shown many pro inflammatory effects on multiple leukocytes. We hypothesized that oxLDL is one mechanism for recruiting granulocytes to the airways during a RV infection. Therefore, chemotaxis and transendothelial migration, in response to nLDL, was determined for these granulocytes. Methods : nLDL was oxidized with 5mM Cu2S04 for 20-24 hours. 3-5 105 cells were loaded into the Transwell filter while the chemotatic agonists were placed in the lower well for chemotaxis. Confluent monolayers on HPMEC were grown on Transwell filters for transendothelial migration. The filters were washed and eosinophils and neutrophils loaded on to the filter with the chemotatic agonist was were placed in the lower well. The wells were incubated for 3 hours. The number of migrating cells was counted on a hemocytometer. Results : OxLDL, but not nLDL, is chemotatic for eosinophils and neutrophils. The level of granulocytes chemotaxis was dependent on both the concentration of LDL and its degree of oxidation. OxLDL stimulates eosinophil and neutrophils migration across HPMEC monolayers (+/-IL-$1{\beta}$ preactivation) in a dose dependent manner. Conclusion : Increased vascular permeability during a RV infection may lead to the influx and oxidation of LDL. The resulting oxLDL. is one possible mechanism for the recruitment of neutrophils and eosinophils to the airway interstitial matrix. Once in the airways, granulocytes can further interact with oxLDL to promote airway inflammation.

Investigation of Viruliferous Insect Rate of Planthoppers Captured by Smart Sky Net Trap (SSNT) in Korea during 2015-2017 (2015-2017년 국내 스마트 공중 포집기에 포획된 벼 주요 멸구류의 밀도 변동 및 보독충률 조사)

  • Major viruses infecting rice are transmitted by planthoppers such as small brown planthopper (SBPH), brown planthopper (BPH) and white-backed planthopper (WBPH). In this study, we investigated planthoppers captured during 2015 to 2017 by a smart sky net trap (SSNT) system installed in 40 areas in Korea, which is an automatic, rapid and real-time insect surveillance system. The average rates of captured migration plnathoppers was 27.5%, 17.2%, 15.3% and 10.9% in Chungcheongnamdo, Jeollanamdo, Jeollabukdo and Gyeonggido, orderly. The highly migrated month was July for SBPH, July to August for WBPH and August for BPH. To investigate the viruliferous rates of planthoppers of rice during 2015 to 2017, we performed RT-PCR using specific primers for each rice virus. RBSDV was detected from 0.4% in SBPH, while no viruses were detected in BPH and SBPH. Rice planthoppers exist all around in Asia. They can move long distance by wind from southern countries to Korea. Monitoring the migration of rice planthoppers and their viruliferous rates is important to prevent the outbreaks of rice virus diseases.

  • Plant viruses of tobacco including tobacco mosaic virus (TMV) and potato virus Y (PVY) cause severe economic losses in leaf-tobacco production. Cultural practices do not provide sufficient control against the viruses. Use of valuable resistant cultivars is most recommendable for the control of the viruses. However, conventional breeding programs are not always proper for the development of virus-resistant plants mostly owing to the frequent lack of genetic sources and introduction of their unwanted properties. Therefore, we tried to develop virus-resistant tobacco plants by transforming commercial tobacco cultivars, NC 82 and Burley 21, with coat protein (CP) or replicase (Nlb) genes of TMV and PVY necrosis strain (PVY-VN) with or without untranslated region (UTR) and with or without mutation. Each cDNA was cloned and inserted in plant expression vectors with 1 or 2 CaMV 35S promotors, and introduced into tobacco leaf tissues by Agrobacterium tumefaciens LBA 4404. Plants were regenerated in kanamycin-containing MS media. Regenerated plants were tested for resistance to TMV and PVY In these studies, we could obtain a TMV-resistant transgenic line transformed with TMV CP and 6 genetic lines with PVY-VN cDNAs out of 8 CP and replicase genes. In this presentation, resistance rates, verification of gene introduction in resistant plants, stability of resistance through generations, characteristics of viral multiplication and translocation in resistant plants, and resistance responses relative to inoculum potential and to various PVY strains will be shown. Yield and quality of leaf tobacco of a promising resistant tobacco line will be presented.

Phospholipase D Activity is Elevated in Hepatitis C Virus Core Protein-Transformed NIH 3T3 Mouse Fibroblast Cells (C형 간염바이러스의 core 단백질에 의해 암화된 쥐의 섬유아세포에서 phospholipase D 효소활성의 증가)

  • Hepatitis C Virus (HCV) is associated with a severe liver disease and increased frequency in the development of hepatocellular carcinoma. Overexpression of HCV core protein is known to transform fibroblast cells. Phospholipase D (PLD) activity is commonly elevated in response to mitogenic signals, and PLD has been also reported to be overexpressed and hyperactivated in some human cancer. The aim of this study was to understand how PLD can be regulated in HCV core protein-transformed NIH3T3 mouse fibroblast cells. We observed that in unstimulated state, basal PLD activity was higher in NIH3T3 cells overexpressing HCV core protein than in vector-transfected cells. Although expression of PLD and protein kinase C (PKC) in core protein-transformed cells was similar with that of control cells, phorbol 12-myristate 13-acetate (PMA), which is known to activate PKC, stimulated significantly PLD activity in core protein-transformed cells, compared with that of the control cells. PLD activity assay using PKC isozyme-specific inhibitor, and PKC translocation experiment showed that PKC-$\delta$ was mainly involved in the PMA-induced PLD activation in the core-transformed cells. Taken together, these results suggest that PLD might be implicated in core protein-induced transformation.

Epidemiologic and clinical features in children with acute lower respiratory tract infection caused by human metapneumovirus in 2006-2007 (2006-2007년 소아 급성 하기도 감염증에서 유행한 메타뉴모바이러스의 유행 및 임상 양상)

  • Purpose : The causes of acute lower respiratory tract infection (ALRTI) are mostly attributable to viral infection, including respiratory syncytial virus (RSV), parainfluenza virus (PIV), influenza virus A/B (IFV A/B), or adenovirus (ADV). Several Korean studies reported human metapneumovirus (hMPV) as a common pathogen of ALRTI. However, studies on seasonal distribution and clinical differences relative to other viruses are insufficient, prompting us to perform this study. Methods : From November 2006 to October 2007, we tested nasopharyngeal aspiration specimens in children hospitalized with ALRTI with the multiplex reverse transcriptase-polymerase chain reaction to identify 6 kinds of common pathogen (hMPV, RSV, PIV, IFV A/B, and ADV). We analyzed positive rates and clinical features by respiratory chart review. Results : We detected 38 (8.4%) hMPV-positive cases out of 193 (41.8%) virus-positive specimens among 462 patients. HMPV infection prevailed from March to June with incidence peaking in April. HMPV-positive patients were aged 15 years (76.3%), and the ratio of boys to girls was 1.2:1. The median age was 27 months. HMPV primarily caused pneumonia (76.3 %) (P=0.018). Average hospitalization of HMPV-associated ALRTI patients was 5.8 days. In addition, they showed parahilar peribronchial infiltration (100%) on chest X-ray, normal white blood cell count (73.7%), and negative C-reactive protein (86.8 %) (P>0.05). All hMPV-positive patients recovered without complication. Conclusion : HMPV is a common pathogen of ALRTI in Korean children, especially in 1-5 year olds, from March to May. Immunocompetent children diagnosed with hMPV-associated ALRTI may have a good prognosis.

리보솜 불활성화 단백질의 삼차원 구조 규명과 그 구조의 신약개발에의 응용

  • 본 연구는 보리 씨앗에 존재하는 리보솜 불활성화 단백질(RIP) 의 삼차원 구조를 X-선 결정학 방법을 이용하여 밝히고, 그 결과로 분자 차원에서 기능을 이해하는 것을 목적으로 하고있다. 리보솜불활성화 단백질은 N-glucosidase 반응을 통하여 단백질 합성을 저해하기 때문에 세포를 죽일 수 있다. 따라서 암세포만을 특정적으로 인식하는 다른 물질과 결합시키면 암세포만을 특정적으로 죽일 수 있는 면역독소로 이용될 수 있다. 또, 최근에는 항바이러스의 작용을 함이 밝혀져 많은 연구가 진행되고 있다. 단백질 삼차원 구조 규명을 위해서는 여러가지 단계가 있는데 지난 번 과제까지 성공적으로 리보솜 불활성화 단백질의 대량 분리와 X-선 결정학의 필수 요건 좋은 결정을 길렀고, 이번에는 구조 해석을 위해 꼭 해결해야하는 위상문제를 극복하기 위하여 여러가지 실험을 진행하였다. 우선, 비슷한 구조인 피마자씨에서 분리한 Ricin의 A-체인과 미국자리공 잎에서 분리한 Pokeweed antiviral protein의 삼차원 분자좌표를 이용하여 분자치환법으로 위상문제 해결을 시도하였다. Ricin 의 A-체인을 이용하였을 때 분자의 위치가 정확히 찾아지지 않았고, 다른 모델인 Pokeweed antiviral protein을 이용하여 X-PLOR 프로그램내의 PC refinement법으로 분자치환을 시도하였다. Euler각도로 (187.37, 22.5, 311.94) 의 회전해 (Rotation solution) 를 가지고 있었고, 이러한 해에 맞추어서 분자를 돌려둔 후, 이동해 (Transaltion solution) 을 구해서 그 위치 (Orientation) 로 분자를 이동하였다. 이 때 R값은 53.9 % (8.0 - 3.5$\AA$) 이였고, 부분좌표 (Fractional coorcdinate) 에서는 0.102, 0.000, 0.261 이고, 직교좌표 (Orthogonal coorclinate) 에서는 4.616, 0.000, 13.167 의 결과를 얻었다.

Study on the Small Sized Robots Actuator using Piezoelectric Ceramic Bender (압전세라믹 벤더를 이용한 소형로봇용 구동원에 관한 연구)

  • This study proposed piezoelectric ceramic bender actuators for application to small walking robots. As the space where human access has recently become increasingly restricted (e.g., highly concentrated radioactive storage areas, viral contaminated areas, terrorist zones, etc.), the scope of using robots is becoming more diverse, and many actions that were possible only in the past have been attempted to be replaced by small robots. This robotic concept has the advantage of being simple in structure, making it compact and producing a large size work force. The dynamic modeling, using finite element analysis, maximized the robot's mobility performance by optimizing the shape of the actuator, and the results were verified through fabrication and experimentation. The actuator moved at a maximum speed of 236 mm/s under no load conditions, and it could move at a speed of 156 mm/s under load conditions of 5g. The proposed actuator has the advantage of modular additions depending on the mission and required performance, which ensured that they are competitive against similar drive sources previously created.

  • Rapid development of the IT technology and mobile communications has increasingly improved many kinds of digital devices arise, as well as the mobile technology. However, the attacks (virus, hacking and Ip spoofing etc) have also increasingly grown dogged on any region including the society security. As the visual data is prone to copy, delete and move etc, it is necessary that attesting to the integrity of forensics evidence is crucial, as well as data transmission security. This paper presents a framework model using digital forensics method and the results of its performance evaluation for mobile security. The results show that the integrity of the visual data can be obtain with high security and make a proposal refer to prevention of Mobile IP Spoofing attack using our hashing data.

Analytical Performance of Sensitivity and Specificity for Rapid Multiplex High Risk Human Papillomavirus Detection Kit: HPV ViroCheck (고위험군 HPV 검출을 위한 분석적 민감도와 특이도 성능평가)

  • Human papillomaviruses (HPVs) are major causes of cervical cancer. Sixteen high risk HPVs, including HPV 16, HPV 18, HPV31, HPV 33, HPV 35, HPV 39, HPV 45, HPV 51, HPV 52, HPV 53, HPV 56, HPV 58, HPV 59, HPV 66, HPV 68, and HPV 69 are found in cervical cancer. HPVs 16 and 18 are mainly presented in 70% of cervical cancer. Therefore, identifying the presence of these high-risk HPVs is crucial. The objective of this study is to establish the HPV ViroCheck for detecting 16 HR-HPVs and genotypes of HPVs 16 and 18, as well as to analyze the analytical performance of HPV ViroCheck. We performed the analytical sensitivity of HPV E6 / E7 genes of 16 high risk HPVs to confirm the limit of detection. Then, a cross reactivity of HPV ViroCheck with microorganisms and viruses related to the cervix were analyzed for analytical specificity. Analytical sensitivity of high risk HPV genotypes ranged from 1 to 100 copies when using cloned DNAs. The limit of detection was 10 cells for both SiHa and HeLa cells. Cervical-related microorganisms and viruses did not show cross-reactivity to HPV DNA. Moreover, the intra- and inter-assay coefficient variations (CVs) were below 5%. In conclusion, HPV Virocheck will be useful for the detection of 16 HR HPVs, as well as HPV 16 and HPV 18 genotypes rapidly.
