Microbial Synthesis of Physiologically Active Compounds (미생물반응을 이용한 생리활성 물질의 합성)

  • 지구상에서 생합성된 천연물은 완급의 차이는 있겠으나 궁극적으로는 CO$_{2}$ H$_{2}$O, NH$_{3}$ 등으로 다시 산화분해된다. 이 분해과정은 미생물의 효소반응으로 이루어지며 생물권의 항상성도 일차적으로는 미생물의 이 산화분해작용으로 유지된다고 볼 수 있다. 미생물이 영위하는 이러한 효소반응을 생리활성물질의 공업적합성에 이용한 초기의 예로는 부현피질호르몬의 합성과정에서 Rhizopus nigricans를 사용한 수산화반응과 ascorbic acid 합성과정에서 Acetobacter xylinum을 사용한 탈수반응이 유명하며 이러한 반응은 아직도 이용되고있다. 특히 지난 십수년간의 응용미생물학의 발전은 대단한 것이며 여러가지 항생물질, 당질, amino산, 핵산관련물질들이 생리활성물질과 함께 발효법으로 생산되고 있다. 그러나 이글에서 다루는 미생반응은 발효과정에서 생산되는 미생물자체의 일차적 대사산물 (primary metabolite)이나 이차적대사산물 (secondary metabolite)를 대상으로 한것이 아니며 어디까지나 외부에서 공급되는 화학물질에 대한 균체의 효소반응산물을 목적물로 하고있다. 또한 근래 활발히 이용되는 균체고정및 효소고정법을 이용한 효소공학적수법도 제외하고 배양과정의 균체 또는 배양후의 군체를 이용한 미생물반응에 한정코저 한다.

Brief Review on the Microbial Biodegradation of Asphaltenes (아스팔텐의 미생물 분해 연구동향)

  • It was known that crude oil can be mainly divided into saturates, aromatics, resins, and asphaltenes. If microbial biodegradation of asphaltenes is effectively viable, additional oil production will be expected from depleted oil reservoir. Meanwhile, biodegradation can be applied to other aspects, such as the bioremediation of spilled oil. In this case, the biodegradation of asphaltenes also plays an important role. It has been already reported that asphaltenes are decomposed by bacterial consortia. However, the biodegradation mechanism of asphaltenes has not been clearly presented. The major reason is that the molecular structure of asphaltenes is complicated and is mainly in a aggregated form. In this paper, it was presumed that the biodegradation process of asphaltenes may follow the microbial oxidation mechanism of saturates and aromatics which are easier biodegradable than asphaltenes among the crude oil components. In other words, the biodegradation process was explained by serial stages; the contact between asphaltenes and bacteria in the presence of biosurfactants, and the decomposition of alkyl groups and fused-rings within the asphaltene structure.

Effects of Low Powered Ultrasonic Wave Exposure on Microcystis sp. (Cyanobacteria) (저출력 초음파가 Microcystis sp. (Cyanobacteria)에 미치는 영향)

  • This study aimed to investigate the effects of ultrasonic wave on colonial changes of Microcystis sp. causing water-bloom. The effects of frequency change and exposure time of ultrasonic wave on Microcystis sp. were also perceived by measuring the concentration of chlorophyll-a. The concentrations of chlorophyll-a showed a decreasing tendency in all experimental groups exposed to ultrasonic waves, especially the highest decrease at 70 kHz. Before exposed to ultrasonic wave, single cells of Microcystis sp. were observed in a form of colonies which were densely accumulated onto their mucilage layer. However, after one hour exposure to 70 kHz ultrasonic wave, the bonds of colonies were weakened and single cells were separated from the colonies. Moreover, the single cells of the colony were destroyed with the 5 hour exposure time at 70 kHz. As a result, we proposed that the frequency strength and the exposure time of ultrasonic wave might have effects in decrease of biomass of blue-green algae by reducing the concentration of chlorophyll-a, thereby changing and destroying forms of Microcystis sp. colonies.

Microbiological Characteristics of Heavy Metal Ion-Tolerant Microorganisms. (중금속 내성균주의 미생물학적 성질)

  • Cadmium ion-tolerant microorganisms were isolated from the sludge and soil of a cadmium ion-polluted area, a zinc mineralized area, in Kyung Sang Pook Do, Korea. A strain, C-7, which showed tile highest tolerance to cadmium ion was selected by screening from 18 cadmium tolerant microorganisms. By the taxonomical characteristics of this strain, it was identified as a variant of Erwinia sp.. The strain grew in a medium cadmium ion up to a concentration of 2, 800 $\mu\textrm{g}$/ml and the maximum intercellular accumulation of Cd$^{2+}$ was measured to be 28.60 mg/g dried cells (57.2%) during incubation in medium containing 50 $\mu\textrm{g}$/ml under aerobic condition at 28$^{\circ}C$ for 24 hour.r.

Transformation of Trichoderma koningii Using Isolated Nuclei (분리 핵을 이용한 Trichoderma koningii의 형질전환)

  • When protoplasts from auxotrophie mutant of Trkhoderma koningii CUT121(Lys-, Met-) were mixed with isolated nuclei of wild type T. koningii ATCC 261 13 and treated with PEG solution, protrophic colonies were produced with frequency of more than 30 percent. One of segregants from prototrophic colonies showed increased xylanase activity and other polysaccharide-hydrolyzing activities comparable to those of wild type strain. Through measurement of DNA contents, induced segregation, and analysis of isozyme patterns, it was revealed that the prototrophic colonies were transformants resulted from exchange of genetic materials between the two kinds of nuclei used. These results suggest that nuclei transfer technique is more efficient than conventional protoplast fusion technique for strain improvement of Trichoderma species.

Effects of crude Saponin on growth and Aflatoxin production by Aspergillus parasiticus (Saponin이 Aspergillus parasiticus의 발육과 Aflatoxin생합성에 미치는 효과)

  • The research was carried out for the purpose of finding effects of gerbal saponins on aflatoxin synthesis by Aspergillus parasitics NRRL 2999. A. parasiticus with $10^6$ conidia were grown at $30^{\circ}C$ for 9 days on the enriched medium that is optimum for the frowth and aflatoxins production by the mold. The inhibitory effect on the growth and aflatoxins produced by the mold occurred in the presence of 0.36% of crude red-ginseng saponin showing both the growth and aflatoxins production come to 62.3% (growth), 38.7% (aflatoxin $B_1$) and 22.9% (aflatoxin $G_1$) of the control. Thd next effective saponin to inhibit the growth and aflatoxins production was from burdock seeds. However, saponin extracted from honeysuckle flowers had no inhibitory effect. The mold caused no changes in the pH of the medium when it contained red-ginseng saponin. Red-ginseng saponin was more effective than the white-ginseng in inhibiting both the growth and aflatoxin production.

  • The Silleuksa temple in Yeoju city is adjacent to the South Han River and often has high humidity and fog days. In addition, its proximity to forests renders it prone to damage by insects and microorganisms. In order to obtain the basic data necessary for the establishment of a cultural property preservation management plan, the biological distribution of the wooden buildings and the termite distribution in the surrounding forests were monitored for three years. The wood-boring bee, an insect known to inflict damage to heritage structures, was confirmed, and the distribution of insects varied annually and seasonally, presumably due to the location characteristics of the site and the climate change during the year. In the forest behind, termite colonies were identified as a whole and their distribution pattern was different every year. As per the results of the microorganism survey, the species distribution and pollution degree were different at the entrance of the building. Therefore, it needs continuous biological distribution and conservation environment monitoring to mitigate the effects of wood-damaging organisms.were different according to the entrance of the building.

Conical Diffuser Design and Hydraulic Performance Characteristics in Bioreactor Using Empirical and Numerical Methods (원뿔형 산기관 설계와 생물반응조에서 수력학적 운전특성에 관한 실험 및 해석)

  • In this study, we develop a highly efficient conical-air diffuser that generates fine bubble. By inserting a sufficient number of aerotropic microorganisms with dissolved oxygen from an air diffuser and minimizing the air-channel blockages within the air diffuser, we expect to improve the efficiency and durability of the decomposition process for organic waste. To upgrade the conventional air diffuser, we perform experiments and numerical analysis to develop a conical-type that generates fine bubble, and which is free from nozzle blockage. We complement the air-diffuser design by numerically analyzing the internal air-flow pattern within the diffuser. Then, by applying the diffuser to a mockup bioreactor, we experimentally and numerically study the bubble behavior observed in the diffuser and the 2-phase fluid flow in the bioreactor. The results obtained include statistics of the cord length and increased velocity, and we investigate the mechanisms of the fluid-flow characteristics including bubble clouds. Throughout the study, we systemize the design procedures for the design of efficient air diffusers, and we visualize the fluid-flow patterns caused by bubble generation within the mockup bioreactor. These results will provide a meaningful basis for further study as well as the detection of oxygen transfer and fluid-flow characteristics in real-scale bio-reactors using sets of air diffusers.

  • 소형화된 기계가공시스템은 사용재료의 다양화와 에너지 및 공간의 감소와 같은 장점을 가지고 작고 정밀한 부품을 가공할 수 있는 시스템으로 주목받고 있다. 이러한 시스템이 비록 그 크기가 일반적인 가공시스템에 비해 작지만 정렬 및 조립공정, 기계요소의 불완정성에 의한 기하학적 오차는 여전히 존재한다. 기하학적 오차 평가는 기계시스템의 정밀도를 효과적으로 적은 비용으로 향상시킬 수 있는 오차보정기술을 적용할 수 있는 토대가 된다. 일반적으로, 3 축의 직선축으로 이루어진 공작기계는 21 개의 오차요소를 가진다. 레이져간섭계는 이러한 오차요소를 평가하는데 널리 사용되고 있지만 광학계를 정렬하고 설치하는 데 많은 어려움이 있으며 한번의 설치로 한 개의 오차요소만이 측정 가능하다. 또한, 소형공작기계의 경우, 그 크기로 인해 기존의 레이져 간섭계를 직접적으로 적용할 수 없다. 따라서, 본 연구에서는 소형공작기계를 포함한 소형가공시스템의 기하학적 오차 평가를 위한 새로운 다자유도 측정시스템을 제안하였다. 5 개의 정전용량변위센서를 사용하는 이 시스템을 통해 한 축의 움직임에 따른 5 개의 오차요소를 동시에 측정 가능하다. 균질 변환행렬을 이용한 측정알고리듬을 구성하고 이를 모의시험을 통해 평가하였다. 수학적 모델링을 통해 각 센서의 출력값을 유도하고 이를 이용하여 각 오차요소를 계산하기 위한 식을 유도하였다. 여기서, 단순화된 식을 적용한 경우, 임의의 오차에 대한 측정 알고리듬의 정확도를 평가하였다. 또한, 측정 시스템의 설치시 발생하는 셋업오차에 대한 측정 알고리듬의 민감도 분석을 행하였다. 제안하는 측정 시스템은 구조가 간단하고 고가의 부가장비가 필요치 않다. 또한, 적은 비용으로 구성할 수 있으며 높은 측정 정밀도를 가지고 소형가공시스템에 필요한 오차 평가를 행할 수 있다.가 함유된 계란을 생산하고 섭취하였을 때 특정항체들의 결합을 통해 병원성 미생물의 성장이나 군체를 형성하는 것을 무력화시켜 결과적으로 병원균을 감소시키거나 억제시킨다는 점이다. 오늘날 약물에 내성을 지닌 박테리아의 출현으로 질병감염을 막는데 항생제의 사용효과가 점차 감소하고 있기 때문에 이러한 항생제를 대체할 수 있는 방안으로 계란항체를 이용할 수 있다.한 중공 플랜지 형상의 단조 방법 중 보다 적절한 단조방법인 압조 단조에 있어서 일반적으로 사용되고 있는 SM10C에 대한 유한요소 해석을 수행하였으며, 제품의 형상비에 따라 폴딩 결함의 발생 유무를 검토하고, 폴딩 결함 없이 단조하기 위한 중공 플랜지의 형상한계 비를 제시하였다.도 경미하게 나타났으나, 경련이 나타난 쥐에서는 KA만을 투여한 흰쥐와 구별되지 않았다. 이상의 APT의 항산화 효과는 KA로 인한 뇌세포 변성 개선에 중요한 인자로 작용할 것으로 사료되나, 보다 명확한 APT의 기전을 검색하고 직접 임상에 응응하기 위하여는 보다 다양한 실험 조건이 보완되어야 찰 것으로 생각된다. 항우울약들의 항혈소판작용은 PKC-기질인 41-43 kD와 20 kD의 인산화를 억제함에 기인되는 것으로 사료된다.다. 것으로 사료된다.다.바와 같이 MCl에서 작은 Dv 값을 갖는데, 이것은 CdCl$_{4}$$^{2-}$ 착이온을 형성하거나 ZnCl$_{4}$$^{2-}$ , ZnCl$_{3}$$^{-}$같은 이온과 MgCl$^{+}$, MgCl$_{2}$같은 이온종을 형성하기 때문인것 같다. 한편 어떠한 용리액에서던지 NH$_{4}$$^{+}$의 경

Egg Antibody Farming and IgY Technology for Food and Biomedical Applications (식품과 생의학을 위한 계란 항체생산과 IgY 기술의 활용)

  • It has been recognized that the hen. like its mammalian counterparts. provides young chicks with antibodies as protection against hostile invaders. This system facilitates the transfer of specific antibodies from serum to egg yolk. and provides a supply of antibodies called immunoglobulin Y(IgY) to the developing embryo and the hatched chick. The protection against pathogens that the relatively immuno-incompetent newly hatched chick has. is through transmission of antibodies from the mother via the egg. Egg yolk. therefore. can be loaded with a large amount of IgY against pathogens which can immobilize the existing or invading pathogens during the embryo development or in day-old chicks. Thus. the immunization of laying hens to various pathogens results in production of different antigen-specific IgY in eggs. Egg yolk contains 8~20 mg of immunoglobulins (IgY) per $m\ell$ or 136~340 mg per yolk suggesting that more than 30 g of IgY can be obtained from one immunized hen in a year. By immunizing laying hens with antigens and collecting IgY from egg yolk. low cost antibodies at less than $10 per g compared to more than $20.000 per g of mammalian IgG can be obtained. This IgY technology opens new potential market applications in medicine. public health veterinary medicine and food safety. A broader use of IgY technology could be applied as biological or diagnostic tool. nut-raceutical or functional food development. oral-supplementation for prophylaxis. and as pathogen-specific antimicrobial agents for infectious disease control. This paper has emphasized that when IgY-loaded chicken eggs are produced and consumed. the specific antibody binds. immobilizes and consequently reduces or inhibits the growth or colony forming abilities of microbial pathogens. This concept could serve as an alternative agent to replace the use of antibiotics. since today. more and more antibiotics are less effective in the treatment of infections. due to the emergence of drug-resistant bacteria.
