유류오염토양처리를 위한 미생물활성화방안 연구

  • 유류로 오염된 토양에 있어서의 미생물의 분포 및 활성도에 관한 연구는 오염토양의 생물학적 복원기술을 적용시키고자 하는 지역에서는 매우 중요한 연구이다. 따라서 본 연구에서는 유류 오염된 지역에서의 미생물 활성도를 ETS(Electron Transport System) 및 그 이외의 몇 가지 방법을 통하여 고찰해 보았다. 본 연구결과에 따르면 순화된 토양에서는 이미 유류를 산화시킬 수 있는 Burkholderia spp.가 이미 우점을 이루고 있었고, 생물학적 복원기술 중 Biostimulation법을 적용할 경우 이미 존재하는 미생물(Indigeonous microorganism)에 의한 유류오염물질의 산화를 가속화시킬 수 있는 것으로 나타났다.

육포의 제품 성분조성에 따라 존재하는 위해 미생물의 균수 측정 및 안전성 평가

  • 육포의 제조과정에 따른 미생물 분포를 조사하였다. 실험결과 초기 원료육에서는 $10^2{\sim}10^4\;cfu/g$ 정도의 미생물 분포를 보였으며, 양념은 $10{\sim}10^2\;cfu/g$ 정도의 미생물 분포를 나타내었다. 육포를 제조하기 전 단계인 양념육의 미생물 분포는 $10^2{\sim}10^4\;cfu/g$정도였다. 그러나 육포를 제조한 후에는 미생물의 수가 급격히 줄어들었다. 이는 육포의 건조를 위해 $70^{\circ}C$의 열로 가열하였기 때문이라고 생각되며, 육포의 제조 전 과정에서 미생물의 오염은 없었던 것으로 생각된다. 또한 식품에서 문제시되는 포자생성균이나 대장균군은 모든 검체에서 검출되지 않았다.

  • 시중의 정육점 및 백화점 등에서 유통 중인 10종의 우육 원료에 대한 일반 세균수, 저온균수, 고온균, 혐기성균 및 진균류, 대장균군에 대한 미생물학적 분포와 병원성 미생물에 대한분리${\cdot}$동정을 실시하였다. 실험결과 원료 우육에서 중온균은 $3.8{\times}\;10^3{\sim}1.4{\times}\;10^5\;cfu/g$으로 높은 분포를 보였다. 저온균은 $9.2{\times}\;10^3{\sim}1.0{\times}\;10^5\;cfu/g$으로 지표세균 중에서 가장 높은 분포를 나타내었고, 혐기성균은 중온균, 저온균과 유사한 분포를 보였으나 상대적으로 적게 검출되었고, 고온균은 모든 검체에서 검출되지 않았다. 대장균군 또한 모든 시료에 대해서 검출되지 않았다. 효모와 곰팡이류는 $2.2{\times}\;10^1{\sim}7.8{\times}\;10^2\;cfu/g$으로 검출되었다. 병원성 미생물은 우육 sample B, G, H에서 B. cereus 만이 검출되었고, 동정결과 99.8%의 상동성을 보였다.

The Soil Properties and Microbial Numbers of soil Samples Collected from Polluted and Unpolluted Areas in Korea (오염지역과 비오염지역의 토양의 특성과 토양 미생물의 분포)

  • A total of 112 soil samples collected from polluted and unpolluted areas in Korea were investigated for physical properities (such as soil moisture, organic matter and soil pH) and biological properties (such as microbial numbers). The results of organic matter and soil pH showed a great variation(p=0.01) in the four areas, whereas soil moisture and organic matter were similar among three plant vegetations. There was a significant relationship(p=0.01 or 0.05) between soil pH and microbial number These results imply some variations in soil environment and may lead to unfavorable changes of plant vegetation in soil. Presumably, the above results appear to be resulted from soil acidification caused by an acidic rain.

Comparison of Microbial Community Compositions between Doenjang and Cheonggukjang Using Next Generation Sequencing (차세대 염기서열 분석법을 이용한 전통 된장과 청국장의 미생물 분포 분석)

  • To profile the microbial compositions of Korean traditional fermented paste made from whole soybeans, Doenjang and Cheonggukjang, and compare their taxonomic differences, we analyzed the V3-V4 region of 16S rRNA of naturally fermented foods by using next generation sequencing. α-Diversity results showed that values indicating bacterial community abundances (OTUs) and richness (ACE, Chao1) were statistically significant (p=0.0001) in Doenjang and Cheonggukjang. Firmicutes was the most common phylum in both groups, representing 97.02% and 99.67% in the Doenjang and Cheonggukjang groups, respectively. Bacillus was the most dominant genus, accounting for 71.70% and 59.87% in both groups. Linear discriminant (LDA) effect size (LEfSe) analysis was performed to reveal the significant ranking of abundant taxa in different fermented foods. A size-effect threshold of 2.0 on the logarithmic LDA score was used for discriminative functional biomarkers. On the species level, Bacillus subtilis, Tetragenococcus halophilus, and Clostridium arbusti were significantly more abundant in Doenjang than in Cheonggukjang, whereas Bacillus thermoamylovorans, Enterococcus faecium, and Lactobacillus sakei were significantly more abundant in Cheonggukjang than in Doenjang. Permutational multivariate analysis of variance (PERMANOVA) showed that the statistical difference in microbial clusters between the two groups was significant at the confidence level of p=0.001. This research could be used as basic research to identify the correlation between the biochemical characteristics of Korean fermented foods and the distribution of microbial communities.

Microbial Community in Various Conditions of Soil Microcosm (벤젠과 톨루엔 분해에 적합한 미소환경과 토착미생물군의 분포변화)

  • Biological treatment of benzene and toluene contaminated soil was investigated in laboratory microcosm of 16 different types for degrading benzene and toluene by indigenous bacteria. At the experimental conditions of the microcosms fast degrading benzene and toluene, moisture contents were 30% and 60% in a soil gap and content of powdered-activated carbon(PCA) for adhesion of benzene and toluene-degrading bacteria was 1% in total soil mass. At the conclusion of the shifted bacteria community, Case 6 and case 7 were operated until 10 days, and then the total cell number and the number of benzene and toluene degrading bacteria were investigated. The total cell number of Case 6 and Case 7 increased 488 fold and 308 fold of total indigenous cell, respectively. The number of benzene and toluene degrading bacteria increased and maintained the percentages occupied in pre-operating microcosm. Species of benzene and toluene degrading bacteria in microcosm changed from species of Gram negative bacteria to Gram positive bacterial species after soil exposed to benzene and toluene.

Bioindicator in Advanced Wastewater Plants (고도처리장의 Bioindicator)

  • The occurrence and abundance of protozoa at advanced wastewater treatment plant were compared with operating parameters and effluent quality using statistical procedures. It seemed that plant operating conditions influenced the distribution of protozoa in the mixed liquor. In statistical analysis, the distribution of protozoa showed the operating condition of plant and predicted effluent quality. Once enough data concerning protozoa, operating parameters and effluent has been gathered, the operator has a valuable tool for predicting plant performance and near-future effluent quality based on microscopic examination. Plant operator manipulates operating conditions if he knows near-future effluent quality is deteriorating. Perhaps more importantly it can be used to actually control the plant to adjust the operating conditions to obtain the protozoal populations that have been shown to provide the best effluent quality.

Application of the Fluctuating Microbial Counts Using Probability Approaches in Food Industries (식품산업체에서 확률분포 모델을 이용한 불규칙적인 미생물 수 분포 활용)

  • Sequences of industrial microbial counts of foods shows irregular fluctuating patterns as adeinition of fluctuating microbial counts(FMC). Recently, it beame clear that the FMC was considered as having a lognormal distribution as a first order approximation. Application of lognormal distribution to the industrial microbial counts could produce useful information in practice. This study is intended to verift the application method of lognormal idstribution in FMC. The one year's records for microbial counts of frozen dumplings from two companies were obtained, and the statistical analysis was carried out to estimate the frequencies of future events where counts exceeding selected levels and to compare the sanitation level of the two companies. The results showed that this spplication method enable translation of irregular recourds of microbial counts into an useful information such as te actual probalities of outburst of a given level and the quantitative predictions of potential hazards in the processing.

  • The physico-chemical and biological factors of coastal sea water were measured bimonthly from 1976 to 1979 for elucidating the relationship between microbial distribution and environmental factors at Masan and Jinhae bay. The experimental results are summarized as followings : 1) The polulation size of bactriz in sea water were increasing as the water temperature increased, and that was higher at station 2 and 3 than at station 1. The number of fungi showed the highest value on July on bottom. The population size of yeast showed no seasonal variation and also showed a relation with the geographic distance. 2) The correlationship between microbial distribution and environmental factors showed little coefficiency in surface water. And the other hand, at bottom water, between general bacteria and water temperature and dissolved oxygen, and between yeast and salinity, there were relatively high coefficiecy.

The Characteristic Study of the Microbial Habitat in the Muwisa Museum, Gangjin (강진 무위사 성보박물관 전시실의 미생물 서식환경 특성 연구)

  • The exhibition hall of museum has been damaged by abundant dust and mold of the air in the room. Occurrence of microorganisms on the surface of the cultural heritage will cause damages such as coloration, materials degradation, deterioration, etc. In this study, we investigated the temperature/humidity and microbes distribution in exhibition hall, display showcase, and on surface of wall paintings in the Muwisa museum. From some samples collected in exhibition hall and display showcase, we identified so many varieties of micro-organisms such as Aspergillus sp., Penicillium sp., Cladosporium sp.. In particular, we could see a large number of microorganisms in the number 1, 2 exhibition hall compared with the number 3 exhibition hall. The results examining the distribution of the temperature and humidity were showed as $28{\pm}4^{\circ}C$ and 70~80%, respectively. The number 1 exhibition showcase was higher humidity environment than the number 2, 3 exhibition hall compared to the distribution ratio, which considered as correlation with the distribution of microorganism concentration. As these results, we understood that high temperature and humidity in the exhibition hall and display showcase of the Muwisa museum were characteristics caused by environmental management and hermetically-sealed rooms.
