Properties of Promoters from Alkali-tolerant Bacillus sp. (알카리 내성 Bacillus속 Promoter의 특성)

  • The promoters of alkali-tolerant Bacillus sp. had been cloned in the promoter probe vector pPL703 and recombinant plasmid p-12 had been constructed. As a result of subcloning, two different promoters were found to exist in the cloned 2.9 kb promoter fragment and two recombinant plasmids p-l2B1 and p-l2B2, each harboring different promoter, were constructed. The promoter activity, which was expressed in the CAT specific activity, of p-l2B1 was 7 times higher than that of p-l2B2. The promoter activity as a function of growth revealed that both promoters of p-l2B1 and p-l2B2 were expressed after the late logarithmic growth phase and repressed in the presence of 1.0% (w/v) glucose.

In-vitro Anti-thrombosis Activity of Sphagnum palustre (수태의 항혈전 활성)

  • Sphagnum palustre (SP), a species of moss belong to the Sphagnaceae family, is used as a dwarfed potted plant, in diapers, bandages, and soil additives. Although, SP can be found all over the world and is very cheap, the study of SP components and bioactivities are still at a rudimentary stage. In this study, the hot-water extract of SP (HWSP) and its subsequent organic solvent fractions were prepared, and their in-vitro anti-thrombosis activities were evaluated. The results showed that the water residue of HWSP has a strong anti-coagulation activity with significant extensions of thrombin time, and platelet aggregation activity. Our results suggest that the SP has the potential to be a novel resource for anti-thrombosis agents. This report provides the first evidence of the anti-thrombosis activity of SP.

Properties of Extracellular Cytosine Deaminase from Arthrobacter sp. JH-13 (Arthrobacter sp.JH-13이 생산하는 세포외 Cytosine Deaminase의 성질)

  • Some properties of an extracellular cytosine deaminase produced from Arthrobacter sp.JH-13 were examined after 20-80% of ammonium sulfate fractionation. Among some substrates, this enzyme utilized cytosine and 5-fluorocytosine as a substrate. The optimum pH and temperature for the activity of this enzyme were found to be near 8.0 and $40^{\circ}C$, respectively. The ensyme was more stable in 0.2M of Tris-HCl buffer than 0.2M of potassium phosphate buffer. The enzyme was generally stable below $50^{\circ}C$, but inactivated completely at $70^{\circ}C$. 1mM of $Fe^{3+},\;K^+\;and\;Na^+$ increased the enzyme activity, but 0.01mM of $Co^{2+},\;Cu^{2+},\;Ni^{2+},\;Hg^{2+},\;Ag^{2+},\;Zn^{2+},\;Ba^{2+},\;and\;Mg^{2+}$ markedly inactivated the enzyme activity. 0.1mM of p-chloromercuribenzoate, trichloroacetic acid, and N-ethylmaleimide compleyely inhibited the enzyme activity, but 0.1mM of 2-mercaptoethanol slightly increased the enzyme activity.

Studies on Physiological Activity of Bacillus subtilis JM-3 Isolated from Anchovy Sauce (멸치액젓으로부터 분리한 Bacillus subtilis JM-3의 생리활성기능에 관한 연구)

  • In previous paper, we isolated the bacteria, Bacillus subtilis JM-3, with proteolytic and fibrinolytic activity for candidate microorganisms that have rapid fermenting and physiological functions from anchovy sauce. This study was carried out to search physiological functions of Bacillus subtilis JM-3, such as antimicrobial, antioxidative, antimutagenic, angiotensin-converting enzyme inhibition, and anticarcinogenic activity in vitro. The cell free culture of Bacillus subtilis JM-3 showed strong antibacterial activity against Listeria monocytogenes, antioxidative activity with 87% of inhibition rate against linoleic acid, 50% of antimutagenic activity against N-nitrosodimethylamine and N-nitrosomorpholine, and 88.9% of growth inhibition rate against SNU-1 cell line (stomach cancer cell of human). However, Bacillus subtilis JM-3 did not show angiotensin-converting enzyme inhibition activity.

Growth and Succession of Protozoa Population in the Activated Sludge Plant. (하수처리장의 원생동물 천이)

  • This study concerns the succession process of protozoa populations in two activated sludge pilot plants. Protozoa populations with similar structures developed and succeeded each other in two plants. It was observed that initial sludge inoculum did not influence the protozoan composition but shorten the period of the plant stabilization. It seemed the protozoan in aeration tank was come from the influent. The composition of the influent sewage influenced the structure of protozoa in the pilot plants. The flagellates was very abundant during the initial starting phase. These rapidly decreased after growth peak. Then sarcodina, ciliates were increased in succession. In ciliates, free-swimming forms appeared at first and crawling forms and stalked forms were followed. After 36 days from the start, the sludge stabilization phase was characterized by a populations of ciliates dominated by stalked and crawling type and low value of effluent BOD was achieved.

Killer Characteristics of Candida dattila KI09 and Kl12 Strains (Candida dattila K109와 K112 균주의 Killer 특성)

  • Candida dattila K109 and K112 isolated from grapes in Korea showed killer activity toward Saccharomyces cereuisiae 5 x 47, S. cereuisiae 1368, Hamenula, Torulopsis, Kluyueromyces, Debaryomyces, and Brettanomyces, and showed the most effective killer activity at 22-26$^{\circ}C$ and at pH 3.9-4.1. The killer actvity of both toxins were remarkably decreased at higher temperature than $25^{\circ}C$ and higher pH than pH 4.0. And the toxins were suggested to be glycoproteins inactivated by pronase E and pepsin. The killer activity was not cured by incubation at elevated temperature of 30-37"C, but cured by treatment with 0.0105-0.3 ppm cycloheximie.imie.

  • Melhylovorus sp. strain SS1 grown on methanol was found to show activities of key enzymes of the linear route, $NAD^+$-linked formaldehyde and formate dehydrogenases, and the cyclic route, hexulose-6-phosphate synthase, glucose-6-phosphate isomerase, glucose-6-phosphate dehydrogenase, and 6-phosphogluconate dehydrogenase, for formaldehyde oxidation. The activities of the cyclic route enzymes were higher than those of the linear route enzymes. The bacterium also exhibited activities of the key enzymes of the ribulose monophosphate and Entner-Doudoroff pathways and transaldolase involved in the formaldehyde assimilation and the enzymes involved in the biosynthesis of extracellular polysaccharide. Cells grown in the presence of 2.3 mM ammonium sulfate were higher in the productivity of extracellular polysaccharide, but lower in the growth yield, than those grown in the presence 7.6 mM ammonium sulfate. The activities of 6-phosphogluconate dehydrogenase, phosphoglucomutase, and UDP-pyrophosphorylase in cells grown under nitrogen-limited condition were higher, but that of 6-phosphogluconate dehydratase/2-keto-3-deoxy-6-phosphogluconate aldolase was lower, than those in cells grown in the presence of sufficient amount of nitrogen source.

  • Lipase (EC 3.1.1.3) in the culture filtrate of Streptomyces coelicolor A3(2) was active on ${\alpha}$-naphthyl-butyrate as well as on various triacylglycerols with different lengths of acyl chains. The extracellular lipase was purified 15-fold by ammonium sulfate fractionation, Sephadex G-100, DEAE-Cellulose and Phenyl-Sepharose CL4B column chromatography with overall yield of 16%. It showed an molecular weight of 34.7 kDa by SDS-polyacrylamide gel electrophoresis. The enzyme activity with tributyrin as substrate was optimal at pH 8.0~9.0 and at $37^{\circ}C$. The enzyme activity decreased when the chain length of acyl group of triacyglycerol increased. A-factor, a hormone-like regulator of Streptomyces differentiation inhibited the lipase activity, which might corelate with the low enzyme activity in early exponential growth phase.

Analysis of Amino Acid Residues Involved in Activities of Chitin Deacetylase of Aspergillus nidulans (Aspergillus nidulans에서 분리된 키틴 탈아세틸화 효소활성에 영향을 미치는 아미노산 잔기 분석)

  • Native chitin deacetylase of Aspergillus nidulans was purified to apparent homogeneity by a combination of phenyl-Sepharose and Q-Sepharose column chromatography. In order to analyze the amino acid residues involved in the enzyme activity, the enzyme was chemically modified with chemical agent, which selectively reacted with the specific amino acid residue on the protein. When the enzyme was chemically modified with diethylpyrocarbonate, which specifically reacted with histidine residues on the protein, the activity was eliminated. The chitin deacetylase, chemically modified with 100 ${\mu}M$ modifier at the residue of arginine or tyrosine, has shown to have decreased activities. It was shown that the modification at aspartic acid or glutamic acid did not affect the enzyme activity to a greater extent, which would not implicate that acid amino residues were directly involved in catalytic reaction and would affect on the global structures of the proteins. This results demonstrated that histidine and tyrosine residues of enzyme would participate in an important function of the chitin deacetylase activity.

Anti-thrombosis Activities of the Root Extract of Moringa oleifera Lam (모링가 지하부의 항혈전 활성)

  • Moringa oleifera Lam (MOL) has been used as a traditional medicine to treat various cancers and inflammation. Whereas the bioactivities of the MOL leaf and seed are well reported, the study of the root is still rudimentary. In this study, the ethanol extract of MOL (EEMOL) and its subsequent organic solvent fractions were prepared and their anticoagulation activity in vitro and platelet aggregation inhibitory activity were evaluated. The EEMOL had negligible anticoagulation and strong platelet aggregation activities. However, the hexane and ethyl acetate fractions of EEMOL showed significant inhibition against thrombin, prothrombin, coagulation factors, and platelet aggregation, without hemolytic activity up to 1.0 mg/ml. Our results suggest that the active fractions of MOL root have potential as new anti-thrombosis agents.
