Bioreactor Systems for the Cometabolic Biodegradation of Trichloroethlene (트리클로로에틸렌의 공동대사적 생분해를 위한 생물반응기 시스템)

  • Trichlooethylene (TCE) has become a widespread contaminant in air, soil, and underground water due to extensive industrial used and improper disposals. Since TCE is a suspected carcinogen and constitutes public health concerns, many treatment systems have been investigated to remove this hazardous waste. One of the most premising reactor systems for the treatment of TCE is trickling biofilter (TBF), in which monooxygenase (MO), the corresponding enzyme for initiating primary substrate oxidation, fortuitously degrades TCE via cometabolism. TCE, however, is not easily treated by simple TBF. This is mainly due to the toxicities of TCE and its degradation products to microbial film in TBF. In this paper, recent progresses on the development of bioreactor system for the treatment of TCE, especially gas-phase TCE, are reviewed. The potentials of novel biofilm reactor systems were also discussed for the long-term continuous treatment of TCE.

Probiotic Property of Lactobacillus pentosus Miny-148 Isolated from Human Feces (인체분변으로부터 분리한 유산균 Lactobacillus pentosus Miny-148의 생균제 특성 연구)

  • Three hundred lactic acid bacteria isolated from human feces were studied their probiotic characters to develop potential probiotics. The properties were tested on the basis of guideline for probiotic selection protocol such as tolerance for acid or bile salt, thermal stability, antimicrobial, anticancer cell, and antiviral activity. Strain Miny-148 was selected as a potential probiotic bacterium which showed resistance to low pH, bile salts and thermal stability. On the basis of fatty acid profiles and 16S rDNA sequences analysis, the strain was identified as Lactobacillus pentosus (similarity 99.9%). The strain, L. pentosus Miny-148, showed broad antimicrobial spectrum against E. coli O157:H7, Shigella flexneri, Bacillus anthracis, Staphylococcus aureus, E. coli, Vibrio cholerae, V. vulnificus, Salmonella typhimurium, and Methicillin-resistant S. aureus (MRSA). Cell-free culture supernatant of the strain also inhibited against the growth of HT-29 colon cancer cell and transmissible gastroenterits virus.

Synergistic Anticancer Activity of a Mixture of Anticancer Agent with Proteoglycan from Rhanella aquatilis against Human Colon Cancer Cell HT29 (Rhanella aquatilis 유래 당단백질과 항암제 혼합물에 의한 인체 대장암 HT29세포에 대한 항암상승효과)

  • In order to investigate the anticancer activity of an anti-yeast substance (AYS), a proteoglycan produced by Rhanella aquatilis AY2000, the cytotoxicity of the AYS against cancer cells was determined in vitro. The AYS was not cytotoxic to the human Jurkat T cell or the mouse sarcoma 180 cell, but was cytotoxic to the human colon cancer TH20 cell. The AYS was increasingly cytotoxic against human colon cancer cells in a dose-dependent manner at range from 62.5 to 500 ${\mu}g/ml$. Anticancer activity by combination of the AYS and an anticancer agent was also determined. The anticancer agent combined with the AYS was shown to possess greater synergistic anticancer activity against human colon cancer cells, as compared with the anticancer agent alone.

The Cytotoxic and Apoptotic Effect of Pseudomonas aeruginosa Exotoxin A on Human Leukemia K-562 Cells (인간 백혈병 세포에서 Psuedomonas aeruginosa exotoxin A에 대한 세포독성과 세포자멸사 효과)

  • After reports on regression of cancer in humans and animals infected with microbial pathogens date back more than 100 years, much effort has been spent over the years in developing a wild type or attenuated bacterial and purified bacterial proteins for the treatment of cancer. Pseudomonas aeruginosa exotoxin A (ETA) is known to inhibit cell growth and trigger significant cell death in various cancer cells. Although ETA induces apoptosis of cancer cells, its exact mechanism of action is not known yet. Four different assays were performed in this study: morphological assessment of apoptotic cells, cell cytotoxity, annexin-V binding assay, and cell cycle analysis. The proliferation and survival of the K-562 cells treated with ETA were decreased in a dose dependent manner. In addition, the apoptotic body of K-562 cells was induced by ETA treatment in a dose dependent manner. The ETA-induced apoptosis was confirmed by annexin-V binding assay. Flow cytometric analysis was examined to ascertain whether ETA could arrest the cell cycle at the sub-G1 phase. Our results suggest that P. aeruginosa ETA inhibits cell growth and induces apoptosis in human leukemia K-562 cells.

  • Rhodopseudomonas sp. KCTC 1437 evolved molecular hydrogen efficiently under light illuminated anaerobic culture condition in the presence of organic acids and various sugars. especially glucose when low concentration of NH$_4$ + or L-glutamate was added to cultures. It was revealed that hydrogen formation from Rhodopseudomonas sp. KCTC 1437 was mediated by two different enzyme systems. Under the nitrogen limiting condition, hydrogen evolution from glucose was catalyzed by nitrogenase. For the nitrogenase activation in vivo, the precultured cells drown on limiting concentration of NH$_4$$^{+}$ as a sole nitrogen source showed more capacity of hydrogen evolution from glucose in the presence of L-glutamate than any other cells .frown on sufficient concentration of NH$_4$$^{+}$, L-glutamate, NH$_4$$^{+}$, or both of L-glutamate and $N_2$. A significant volume of molecular hydrogen was evolved from glucose even in the presence of excess NH$_4$$^{+}$ either in the light or dark anaerobic condition, presumably due to the mediation of hydrogen evolution by fromic hydrogenlyase.enlyase.

Antioxidant Activity of Fermented Barley, Wormwood, Sea Tangle, and Soybean (발효 보리, 쑥, 다시마, 대두의 항산화효과)

  • Superoxide is involved in causing inflammation, cancer, and arteriosclerosis in many cases. Taking antioxidant material can be helpful in preventing the diseases. Natural food such as barley, wormwood, sea tangle, and soybean contain antioxidant ingredients. Antioxidant activity increase was determined by fermenting them with microorganism. To determine the activity, 1,1-diphenyl-2-picrylhydrazyl (DPPH) solution was used. When barley, wormwood, sea tangle, and soybean were fermented with Bacillus lichenifomis Bl, antioxidant activities of each fermented product increased 2.6, 1.6, 2.7, and 1.7 folds, respectively. Also, absorbance of fermented soybean was higher than that of soybean at the range of 250~290nm, which might be involved in differences of antioxidant activity of the two. Paraquat suppressed Esherichia coli DH5$\alpha$ growth by making superoxide inside the strain. However, when ethanol extract from fermented soybean was added into the GM (glucose-mineral) media containing the strain, its growth was recovered, suggesting that ethanol extract can move across E. coli, and can function as anti-oxidant material in vivo. Thus, it will be possible to develope antioxidant material from fermented soybean which can be taken orally.

Novel insight into the role of thiamine for the growth of a lichen-associated Arctic bacterium, Sphingomonas sp., in the light (Sphingomonas 속 세균의 명조건 생장에서 티아민의 필수적인 역할)

  • Bacteria in the polar region are under strong light and ultraviolet radiation. In this study, we investigated the effects of light on the growth of a psychrophilic bacterium, Sphingomonas sp. PAMC 26621, isolated from an Arctic lichen Cetraria sp. The growth of the strain in the light was lower than that in the dark. Surprisingly, thiamine increased the growth of Sphingomonas sp. PAMC 26621 in M9 minimal medium under light conditions. Thiamine increased the growth of the strain in a concentration-dependent manner along with ascorbic acid. N-acetylcysteine had no effect on the growth of the strain in the light. Thiamine and ascorbic acid also increased the activities of glucose-6-phosphate dehydrogenase and superoxide dismutase. The results of this study indicate that thiamine provided by the lichen symbiosis system plays an important role in light-induced oxidative stress in this Arctic bacterium as an antioxidant. Our study provide insight into the biochemistry and physiology of Arctic bacteria under strong light and ultraviolet radiation.

Effects of Dietary Acidifier($Lactacid^(R)$) and Essential Oil($Immunocin^(R)$) on the Performance, Nutrient Metabolizability, Small Intestinal Microflora and Immune Response in Broiler Chicks (유기산제제 ($Lactacid^(R)$)와 Essential Oil($Immunocin^(R)$)이 육계의 생산성과 영양소 이용율, 소장내 미생물 균총 및 면역 체계에 미치는 영향)

  • An experiment was conducted to investigate the effects of dietary acidifier($Lactacid^(R)$) and essential oil($Immunocin^(R)$) on the performance, nutrient metabolizability, small intestinal microflora, IgG level and leukocytes and erythrocytes in broiler chickens. Five hundred males and 500 females broiler chickens($Ross^(R)$) were divided into 20 pens of 50 chickens(25 birds in each sex). Five pens were assigned to each of four dietary treatments: control, diets containing antibiotics(Bacitracin methylene disalicilate), acidifier($Lactacid^(R)$) and essential oil($Immunocin^(R)$) dietary treatments. Birds were fed experimental diets ad libitum 5 wks. Weight gain, feed intake, and feed conversion rate were significantly affected by dietary treatment(P<0.05). Overall weight gain($0{\sim}5$ wks) of $Lactacid^(R)$ treatment was significantly lower than the others. Feed intake was highest(P<0.05) in the control followed by antibiotics, $Lactacid^(R)\;and\;Immunocin^(R)$ treatment. Feed conversion rate of $Immunocin^(R)$ treatment was lowest(P<0.05) followed by antibiotics, $Lactacid^(R)$ treatment and the control. Production indices of $Immunocin^(R)$ and antibiotics treatments were significantly higher than those of the control and $Lactacid^(R)$ treatment(P<0.05). $Immunocin^(R)$ treatment was the highest and antibiotics was lowest in serum IgG level. The number of leukocytes and stress index(neutrophil/lymphocytes) tended to be lower in $Immunocin^(R)$ treatment than others. There were no significant differences in erythrocytes among the treatments. The cfu of E. coli was significantly lower in $Immunocin^(R)$ and antibiotics treatments than $Lactacid^(R)$ treatment and the control. Metabolizability of crude protein was significantly lower in the control than $Lactacid^(R)\;and\;Immunocin^(R)$ treatment while that of NFE was significantly lower in $Immunocin^(R)\;than\;Lactacid^(R)$ and antibiotics treatments. It was concluded that essential oil product $Immunocin^(R)$ is as effective as antibiotics in improving feed conversion efficiency and production index while $Lactacid^(R)$ is not.

Curcumin-induced Cell Death of Human Lung Cancer Cells (Curcumin에 의해 유도되는 인간 폐암 세포주의 세포사멸)

  • Lung cancer is a type of cancer that has the highest mortality rate. It is mainly classified into small-cell lung cancer (SCLC) and non-small-cell lung cancer (NSCLC). Chemotherapy is used to treat lung cancer, but long-term treatment causes side effects and drug resistances. Curcumin is a bright yellow polyphenol extracted from the root of turmeric. It has biological activities, such as anti-oxidant, anti-cancer, and anti-inflammatory effects. In this study, we observed differential cell death in human lung cancer cells. Based on the results, curcumin at 10, 30, and 50 μM exhibited a dose-dependent inhibition on the cell survival of several lung cancer cells, with minor differential phenotypes. In addition, apoptosis, autophagy, and reactive oxygen species (ROS) regeneration were observed through flow cytometry. Curcumin dose-dependently increased these phenotypes in A549 (NSCLC) and DMS53 (SCLC), which were restored by corresponding inhibitors. Western blotting was performed to measure the level of expression of apoptosis- and autophagy-related proteins. The results indicate that Bax, PARP, pro-caspase-3, and Bcl-2 were dose-dependently regulated by curcumin, with seemingly higher Bax/Bcl-2 ratios in DMS53. In addition, autophagic proteins, p-AKT, p62, and LC3B, were dose-dependently regulated by curcumin. ROS inhibition by diphenyleneiodonium reduced the induction of apoptosis and autophagy generated by curcumin. Taken together, it is suggested that curcumin induces apoptosis and autophagy via ROS generation, leading to cell death, with minor differences between human lung cancer cells.

  • 인공토굴(人工土窟) 숙성식품의 장기가공보관 수단의 원조로 자리 잡아, 광천옹암리 토굴이 전통식품 산업의 지혜와 관광산업의 역동으로 변신하고, 인공 굴은 폐광토굴에서 농산물저장을 위한 인공제작 굴의 활용기술로 이어져 산업화사회에 기여하고 있다. 대표적인 인공토굴중의 하나는 광천토굴이며, 마을뒤편 야산에 활석암으로 된 암반을 굴착한 토굴이다. 폭과 높이가 각각 2m 정도, 200여 m의 토굴 속에 수많은 젓갈을 담은 드럼통을 저장하여 숙성, 발효시킨다. 젓갈은 우리나라의 대표적인 수산발효식품이며, 어패류 등에 비교적 다량의 식염을 가해 자가소화효소 및 미생물분해 작용으로 알맞게 숙성되는 원리를 이용한다. 젓갈은 일종의 균 식품으로 식품을 발효시킴으로써 독특한 맛과 향 영양을 갖게 되며, 빵, 요구르트, 장류(간장 된장 등), 김치, 막걸리, 동동주, 식혜뿐 아니라 심지어는 버섯조차도 균 식품에 해당한다. 특히 어패류를 염장 발효시켜서 독특한 감칠맛이 나도록 한 우리나라 특유의 저장식품으로 예로부터 기호식품, 조미료 및 김치의 재료로서 널리 식용되어 왔던 양질의 단백질인 동시에 칼슘과 지방질 공급원이기도 하다. 최근에는 상품화된 자연친화적 농산물저장 굴로 사용되는 인공토굴도 등장하였다. 그것은 타공판과 흙을 이용한 생태환경지중건축물(生態環境地中建築物)로서 우리조상들이 오래전부터 지열을 이용하여 주거와 농산물을 저장하였던 재래식 토굴을 현대화시킨 구조물의 지중저장토굴 공간이다.
