Preparation of Molecularly Imprinted Poly(methacrylic acid) and Its HPLC Separation Characteristics of Retinoids (분자각인 Poly(methacrylic acid)의 제조 및 레티노이드 화합물의 HPLC 분리 특성)

  • Molecularly imprinted polymers were prepared in particle forms by crosslinking methacrylic acid (MAA)) using all trans-retinoic acid as a template. The HPLC column packed with the prepared molecular imprinted polymers showed high capability in separation of retinoid derivatives. The column capacity factor and selectivity increased with increasing MAA to template ratio when the incorporated template amount was fixed, as it statistically generated more binding sites between host molecules and template. Molecularly imprinted polymer particles prepared via an emulsion polymerization method were round-shaped and their sizes were more uniformly distributed, but their separation capability was inferior to those obtained by solution polymerization method. It was presumably because the loss of interaction strength between MAA and the template due to hydrogen bonding either between MAA and water or between template and water during the synthesis of molecularly imprinted polymers.

  • 본 연구의 목적은 $SPC/F_2$ water model에 대하여 정해진 여러 온도(250K, 280K, 300K, 350K)에서 경수(light water)와 중수(heavy water)의 일정부피 열용량($C_V$)을 계산하는 것이다. 모의실험은 path integral molecular dynamics (PIMD) 방법을 바탕으로, 계산상에서 물 분자 64개에 대해서 실제 물의 밀도에 맞는 일정한 부피를 설정한 후, 이 때 ring-polymer의 bead수는 양자효과를 보일 수 있는 24개와, 양자효과가 없는1개로 실험을 진행했다. 그 결과로 system의 $C_V$를 얻었고, 수소 동위원소의 변화, 온도의 변화, 양자조건의 여부에 따라 나타나는 차이를 각각 비교했다. 모의실험 결과로 온도가 낮을수록, 양자효과가 클수록, 수소의 질량이 작을수록 열용량이 증가하는 결과를 보였다.

Cholesterol-lowering Effects of Unripe Black Raspberry Water Extract (복분자 미숙과 물추출물의 콜레스테롤 개선 효과)

  • We investigated the effects of unripe black raspberry water extract (UBR-W) and oxidation-LDL treatment on cholesterol levels. Experiments using an established human hepatocellular carcinoma cell line (HepG2) showed a time-dependent increase in expression of LDL receptor after UBR-W treatment. Expression of LDL receptor-related genes, such as SREBP1 and 2, increased upon UBR-W treatment. However, expression of HDL-related genes was unaffected by UBR-W. HMG-CoA reductase activity was reduced by UBR-W treatment, whereas HMG-CoA mRNA expression significantly increased. In addition, the ApoB/ApoA1 mRNA level, which is a predictor of cardiovascular risk, was reduced in a time-dependent manner by UBR-W treatment. Macrophage-like cells (RAW 264.7) showed increased expression of ox-LDL-related genes, such as CD36, scavenger receptor-A, adipophilin, and PPAR-gamma, upon ox-LDL treatment compared to untreated control cells, and quantitative lipid analysis indicated a dramatic increase in lipid accumulation. However, UBR-W treatment significantly reduced expression of ox-LDL-related genes and largely prevented lipid accumulation. The results indicate that UBR-W mediates a cholesterol-lowering effect via inhibition of cholesterol synthesis and induction of LDL uptake through SREBP.

Effects of Unripe Black Raspberry Extracts on Prostate Cancer Cell Line and Rat Model of Benign Prostatic Hyperplasia (복분자 미숙과 추출물이 전립선암 세포주와 전립선비대 백서모델에 미치는 영향)

  • Benign prostatic hyperplasia (BPH), which is commonly found in aging men, is characterized by hyperplasia of prostatic stromal and epithelial cells beginning in the periurethral zone of the prostate. The prevalence of BPH increases in an age-dependent manner. Here, we investigated the protective effects of unripe Rubus occidentalis extracts (UROE) on BPH development using a prostate cancer cell line and testosterone-induced BPH rat model. Experiments using an established hormone-dependent prostate cancer cell line (LNCaP) showed that UROE treatment significantly decreased expression of androgen-related genes, including androgen receptor (AR), prostate specific antigen (PSA), and 5-alpha reductase 2, but not 5-alpha reductase 1, which was also observed in flutamide-treated cells. Further, AR and PSA gene expression was reduced by UROE treatment under androgen-stimulated conditions using dihydrotestosterone (DHT). BPH animals displayed elevated prostate weights. However, UROE as well as finasteride treatment significantly reduced prostate weights and DHT levels compared to testosterone-induced BPH animals. Histopathological analysis also showed that UROE treatment suppressed testosterone-induced prostatic hyperplasia. Taken together, the results suggest that UROE may effectively inhibit the development of BPH and thus may be a useful agent in BPH treatment.

Extraction Characteristics of Rubi Fructus in Relation to Drying Methods and Extraction Solutions (복분자의 포제방법에 따른 추출물 특성)

  • This study was observed the effects of drying methods on the extraction characteristics of Rubi fructus (fruits of Rubus coreanus). Extraction yields of soluble solids and total sugar were high in the Rubi fructus dried by freeze drying, followed by infrared drying and sun drying. Extraction yield of phenolic compounds and DPPH radical-scavenging activity of extracts were in the following order; the Rubi fructus dried by freeze drying, the Rubi fructus dried by infrared drying, the Rubi fructus dried by sun drying. L value was the highest in the Rubi fructus dried by freeze drying, and a and b value were lowest in the fruit dried by freeze drying. These results suggest that freeze drying has an beneficial effect to enhance the quality of Rubi fructus. Water and ethanol extractions was more effective in the extraction of soluble solids and the antioxidative components.

Thermo-Degradation Kinetics of Polyethylene (폴리에틸렌의 열분해 Kinetics)

  • Pyrolysis of polyethylene was carried out in the stainless steel reactor of internal volume of $10cm^3$. Pyrolysis reactions were performed at temperature $390{\sim}450^{\circ}C$ and the pyrolysis products were collected separately as reaction products and gas products. The molecular weight distributions(MWDs) of each product were determined by HPLC-GPC and GC analysis. Distribution balance equation for MWDs of random and specific products were proposed to account for initiation-termination and propagation-depropagation, such as hydrogen abstraction, chain cleavage, coupling of polymer and radical. A separate chain-end scission process produces low molecular weight noncondensable gases(C1 through C5) of average molecular weight 38. Activation energies of the random-chain scission and chain-end scission rate parameters, respectively, were determined to be 35, 17 kcal/mole.

  • The intramolecular formal iron-catalyzed [4+2] cycloaddition reaction of acetylenic dienes to produce bicyclic products has been investigated. These reactions proceeded, competing via intramolecular carbocyclization and bimolecular cycloaddition. However, the iron-catalyzed bicyclic carbocyclization of 4,4-dimethyl-3-(tert-butyldimethylsiloxy)-1-phenyl-6,8-decadien-1-yne(11) yielded exclusively bicyclic diene product(yield 78.1%). From this result, we found that the substrate containing a dimethyl substitutent at the 4,4-position helped to promote the intramolecular reaction, which is stable to the iron catalyzed conditions.

  • 천연가스를 대체하며 21세기 신 에너지원으로 기대되고 있는 메탄 하이드레이트가 주목을 받게된 것은 1930년대 시베리아의 화학 플랜트에서 고압의 천연가스 수송용 파이프라인이 막히는 사고가 빈번하게 발생하여 그 원인을 조사한 결과, 파이프 내에서 가스와 물이 결합하여 하이드레이트를 형성하고, 그것이 파이프의 내벽에 부착되어 파이프를 막고 있다는 것으로 밝혀지면서 천연가스 하이드레이트가 주목을 받게 되었다. 또한 메탄 하이드레이트의 경우 46개의 물분자에 8개의 메탄가스 분자가 포획된 구조로, 그 분자식은 $CH_4{\cdot}5.75H_2O$이다. 따라서 메탄가스와 물의 이론적 용량비가 216:1로써, 표준상태에서 $1m^3$의 메탄 하이드레이트는 $172m^3$의 메탄가스와 $0.8m^3$의 물로 분해된다. 만약 이와 같은 특징을 역으로 이용할 경우 메탄을 주성분으로 하는 천연가스를 물에 포집시켜 인공적으로 하이드레이트를 제조할 수 있기 때문에 천연가스 수송 및 저장의 수단으로써 그 중요성이 커지고 있으며, 액화수송보다 18-24%의 비용절감이 이루어진다고 보고하였다. 그러나 인공적으로 메탄 하이드레이트를 제조할 경우 가스 포집율의 예측이 매우 어려운 것으로 알려져 있다. 따라서 본 연구에서는 동일한 조건에서 메탄 하이드레이트 형성의 반복성 실험을 10회 수행한 결과 과냉도가 클수록 최대최소차이가 줄었고 또한 교반을 시킬 경우도 최대최소차이가 줄어 들었다.

Studies on Fibrinolytic Activity of Jujube (Zizyphus mauritiana) Extract (대추 추출물의 fibrinolytic activity에 관한 연구)

  • Crude extract prepared from jujube (Zizyphus mauritiana) possseses fibrinolytic activity hydrolyzing fibrin. A clear transparent region is observed where fibrin is hydrolyzed, and its diameter increased as the added jujube extract increased. The fibrinolytic activity of jujube extract seems to be heat and acid stable since its activity was retained after heat treatment at $100^{\circ}C$ for 10 min or an acid treatment by incubating at pH 2.0, 3.0, and 4.0 for 3 hours. But the jujube extract dialyzed with a cellulose membrane with molecular weight cutoff of 12,000 and 2,000 lost its activity, which suggests the fibrinolytic activity might be compounds of low molecular weight.

Hot-water Extract of Rubus Coreanus Miquel Suppresses VEGF-induced Angiogenesis (복분자 온수추출물의 VEGF-유도성 혈관신생 억제효과)

  • The interruption of angiogenesis using herbal extracts is now recognized as a useful approach for treating many solid tumors. To date, the best-validated antitumor approach is to target the vascular endothelial growth factor (VEGF)-induced angiogenic pathway. In the present study, we first identified the antiangiogenic activity of a hot-water extract of Rubus coreanus Miquel (RCMHE) in vitro and ex vivo. This extract suppressed VEGF-induced angiogenesis, the phosphorylation of extracellular regulated kinase (ERK), p38 and the activation of matrix metalloproteinases (MMPs). RCMHE also inhibited the VEGF-responsive phosphorylation of VEGFR2. These results clearly show that RCMHE may have potential therapeutic value for angiogenesis-associated human diseases through the suppression of angiogenesis and the interruption of the phosphorylation of VEGFR2.
