Hypoxia and NF-${\kappa}B$; The Relation to Chemoresistance (저산소증과 NF-${\kappa}B$의 항암제내성과의 연관성 고찰)

  • 항암치료는 현재 암환자의 주요한 치료임에도 불구하고 항암제내성과 같은 문제점을 가지고 있다. 약물내성은 다양한 기전에 의해 발생하는데 수송단백질의 과발현, 비독성화발현, 손상유전자의 복구, 세포사멸신호의 변화, STAT-3와 NF-${\kappa}B$의 발현 등이 포함된다. 암세포는 저산소환경에서 발생하며 일반세포에 비해 무산소해당에 상대적 의존도가 높고 이는 암세포의 성장과 전이를 촉진하는 인자가 된다. 항암제가 효과를 내기 위해서는 산소가 필요한데 저산소환경은 이를 방해하며 또한 유전자의 불안정화로 인해 약물내성이 유도된다. NF-${\kappa}B$는 주요 전사인자 중 하나로서 각종 염증과 암에서 지속적으로 활성화되며 암세포의 변화, 증식, 침윤, 전이에 관여한다. 환경적 스트레스 등과 대부분의 항암약제들이 NF-${\kappa}B$를 활성화시키며 임상적으로도 암환자의 생존과 연관된 중요한 예후인자이다. NF-${\kappa}B$의 발현은 항암제로 인한 암세포의 자멸을 회피하게 만들고 수송단백질을 활성화시켜 항암제내성을 유도한다. 강황, 적포도, 고추, 건칠 등 다양한 천연물에서 NF-${\kappa}B$를 억제시키는 효능이 발견되었으며 이는 항암제내성을 억제시키고 항암제의 효과를 증대시킨다. 저산소환경의 개선과 NF-${\kappa}B$의 억제는 상호연관성을 가지고 있으며 항암제내성의 개선뿐만 아니라 암치료제 개발의 새로운 연구목표가 될 수 있다.

The geochemical properties of phosphorus from sediments of Lake Shihwa (시화호 퇴적물에서 인의 지화학적 특성)

  • We have studied the phosphorus geochemistry in sediments from anoxic environments of Lake Shihwa. The dominant sedimentary phosphorus forms were detrital apatite P and Al-bound p, whereas the amount of Fe-bound P was low because of anoxic condition. Based on the correlation between TP(Total Phosphorus) and OC(Organic Carbon), the behavior of sedimentary phosphorus was influenced by organic matters. It shows that dissolved and solid phosphorus concentrations, the flux of phosphate and the correlation coefficients between sedimentary phosphorus and organic matter In St. Cl were higher than those in St. C2. The results indicate that the concentrations and distributions of phosphorus In sediments were controlled by organic matters.

Sediment Oxygen Consumption Rate and Hydrogen Sulfide Release by Dissolved Oxygen Depletion in Hypoxic Area of the Gamak Bay, Korea (가막만 빈산소 해역의 퇴적물 산소소모율과 용존산소 고갈에 의한 황화수소 용출)

  • This study investigated sediment oxygen consumption rates and geochemical characteristics of sediment in hypoxic area of the Gamak Bay based on the chamber experiments and geochemical analyses. The organic carbon contents of surface sediment in the Gamak Bay showed that the inner bay area has higher organic carbon content than those of the outer bay. They toward the outer bay, contents dropped off. The vertical profiles of calcium carbonate ($CaCO_3$) content at piston core sediment assumed that the hypoxia have been frequently occurred during past century in the northern inner bay. The benthic chamber experiments were conducted in February, May, August and November 2010, 2011 in the hypoxic area of the Gamak Bay. In the sediment incubation experiment with chamber at site C3 in the northern inner bay and site C17 in the southern outer bay, the sediment oxygen consumption rate ranged from $3.98mmol\;m^{-2}d^{-1}$ to $12.43mmol\;m^{-2}d^{-1}$ and $3.28mmol\;m^{-2}d^{-1}$ to $8.18mmol\;m^{-2}d^{-1}$, respectively. When the oxygen was completely depleted, the toxic hydrogen sulfide was released with $1.38mmol\;m^{-2}d^{-1}$ and $1.3mmol\;m^{-2}d^{-1}$, respectively.

Analysis of the Substrate Removal Characteristics of TPA Using OUR and NUR Tests, and Simulation with ASM1 (호흡률과 탈질률 실험과 ASM1을 이용한 전산모사를 통한 TPA의 기질 분해 특성 평가)

  • In this study, nitrate uptake rate(NUR) and oxygen uptake rate(OUR) tests were conducted for the assessment of application of Terephtalic acid(TPA) by-product as an alternative external carbon source for sewage treatment plant(STP). With the ASM1 installed in GPS-X the substrate removal characteristic was investigated with simulation by obtained data from NUR and OUR test. As a result, the fraction of RBDCOD(readily biodegradable COD) was mort than 90% and specific denitrification rate was observed about 8.00 mg $NO_3^-$-N/g VSS/hr that was similar to conventional external carbon source. In the next step, sensitivity analysis for heterotrophic biomass in ASM1 was conducted. Optimized parameters of ${\mu}_{max,H}$, $K_s$, ${\eta}_g$, and $b_H$ were 6.60/day, 23.3 mg/L, 0.88, and 0.54/day, respectively. Then, relative mean squared error(RMSE) was reduced to about 40%. Optimized parameters value were well corresponded with the substrate removal characteristics of high maximum and final endogenous specific OUR and high specific NUR.

Phosphorus Removal (Characteristics by Anoxic Oxic Process) by Anoxic and Oxic Processed Combined with Iron Electrolysis (철 석출장치가 결합된 무산소.호기공정에 의한 인 제거 특성)

  • In this study, the (phosphorous removal) the characteristics of phosphorous removal due to (the iron compound precipitated) iron compound precipitation by iron electrolysis in (the anoxic. oxic process) anoxic and oxic processes (equipped with the) in an iron precipitation device were analyzed. During the device operation period, the average concentration of BOD, T-N, and T-P were 219.9 mg/l, 54.6 mg/l and 6.71 mg/l, respectively. The BOD/$COD_{Cr}$ ratio was 0.74, and the BOD/T-N and BOD/T-P ratios were 4.0 and 32.8, respectively. The removal rate of (the organic matters) organic matter (BOD and $COD_{Cr}$) was very high at 91.6% or higher, and that of nitrogen was 80.5%. The phosphorous concentration (of the final) in the treated water was 0.43 mg/l (0.05-0.74 mg/l) on average, and the removal efficiency was high at 90.8%. The soluble T-P concentrations in (an) the anoxic reactor, oxic reactor (II) and final treated water were 1.99 mg/l, 0.79 mg/l and 0.43 mg/l, respectively, which indicated that the phosphorous concentration in the treated water was very low. Regardless of the changes in the concentrations of (organic matters) organic matter, nitrogen and phosphorous in the influent, the quality of the treated water was relatively stable and high. The removal rate of T-P somewhat increased with the increase in the F/M ratio in the influent, and it also linearly increased in proportion to the T-P loading rate in the influent. In the treatment process used in this study, phosphorous was removed (using) by the precipitated iron oxide. Therefore, the consumption of organic (matters) matter for biological phosphorus removal was minimized and (most of the organic matters were) was mostly used as the organic carbon source for the denitrification in the anoxic reactor. This (can be an economic) treatment process (without the need for the supply of additional organic matters) is economic and does not require the supply of additional organic matter.

Effects of MK-801, CNQX, Cycloheximide and BAPTA-AM on Anoxic Injury of Hippocampal Organotypic Slice Culture (해마 조직 절편 배양을 이용한 무산소 손상에 대한 MK-801, CNQX, Cycloheximide 및 BAPTA-AM의 효과)

  • Objective : Glutamate induced excitotoxicity is one of the leading causes of cell death under pathologic condition. However, there is controversy whether excitotoxicity may also participate in the neuronal death under low intensity insult such as simple hypoxia or hypoglycemia. To investigate the role of NMDA receptor in low intensity insult, we chose anoxia as the method of injury and used organotypically cultured hippocampal slice as the material of experiment. Materials & Methods : The hippocampal slices cultured for 2-3 weeks were exposed to 60 minutes of complete oxygen deprivation(anoxia). Neuronal death was assessed with Sytox stain. Corrected optical density of fluorescence in gray scale, used as cellular death indicator, was obtained from pictures taken at 24 and 48 hours following the insult. The well-known in vivo phenomenon of regional difference in susceptibility of hippocampal sub-fields to ischemic insult was reproduced in HOSC(hippocampal organotypic slice culture) by complete oxygen deprivation injury. Results : $CA_1$ was the most vulnerable to complete oxygen deprivation in hippocampus while $CA_3$ was resistant. Oxygen deprivation for 10 and 20 minutes with glucose(6.5g/l) present was insufficient to induce neuronal death in the cultured hippocampal slice. However, after 30 minutes exposure under anoxic condition, neuronal death was able to be detected in the center of $CA_1$ area. The intensity and area of fluorescence indicating cell death correlated with the duration of oxygen deprivation. NMDA receptor and non-NMDA receptor blocking with MK-801(30 & $60{\mu}M$) and CNQX($100{\mu}M$) did not provide cellular protection to HOSC against damage induced by oxygen deprivation, but increased intracellular calcium buffering capacity with BAPTA-AM($10{\mu}M$) was effective in preventing neuronal death (p=0.01, Student's t-test). Cycloheximide($1{\mu}g/ml$, $10{\mu}g/ml$) provided no protection to HOSC against insult of complete oxygen deprivation for 60 minutes and combined therapy of MK-801(30 & $60{\mu}M$) and cycloheximide(1 & $10{\mu}g/ml$) was also ineffective in preventing neuronal death. Conclusion : The results of this study show that the another mechanism not associated with glutamate receptor(NMDA & non NMDA) may play major role in cell death mechanisms induced by complete oxygen deprivation and increased intracellular calcium during anoxia may participate in the neuronal death mechanism of oxygen deprivation. Further investigation of the calcium entry channel activated during oxygen deprivation is necessary to understand the neuronal death of anoxia.

Removal Characteristics of Nitrogen and Phosphorus in swine wastewater by Using Acetic acid on the SBR Process (SBR에서 아세트산을 이용한 양돈폐수의 질소·인 제거 특성)

  • This study was performed : 1) to find the suitable HRT(hydralic retention time), 2) to evaluate the effects of the ratio of mixing/aeration time and injection time of external carbon source, for the removal of organics, nitrogen and phosphorus in swine wastewater by SBR(sequencing batch reactor process), which is one of the biological treatment process. The result of this study were summarized as follows : (1) As the ratio of mixing/aeration time was higher, $NH_4{^+}-N$ removal efficiency was increased and it was increased with increasing injection time of external carbon source because nitrification was affected by denitrification microbes propagation when injection time of external carbon soruce was shorted. T-N removal efficiency was increased with increasing the ratio of mixing/aeration time and injection time of external carbon source. (2) The T-P removal efficiency showed a great difference in each operating condition, and it was increased with increasing the ratio of mixing/aeration time increased and when the injection time of external carbon source was shorted because denitrification was done with effect by denitrification microbes propagation. (3) The highest removal efficiency of organic and nitrogen were obtained by the operating condition of Run 4-1(the ratio of mixing/aeration time : 16.5/5.5, injection time of external carbon source : 15hours) and T-P were obtained by the operation condition of Run 4-2(the ratio of mixing/aeration time : 16.5/5.5, injection time of external carbon source : 3hours), and efficiency(effluent concentration)of $BOD_5$, $COD_{Mn}$, $COD_{Cr}$, T-N and T-P in the treated water was 96.1%, 87.7%, 90.6%, 86.6% and 84.5%, respectively.

  • Behavior of trace metals in Masan Bay, Korea was studied in August 1998 when the oxygen deficient condition occurred. Dissolved Cd and Zn concentration in the surface waters decreased with increasing distance from Masan Harbor. Dissolved concentrations of Cd, Cu Pb and Zn in the surface waters were higher than bottom waters. Particulate(acid-teachable fraction) concentrations of Cu, Cd and Pb in the surface waters of Masan Bay decreased with increasing distance from Masan Harbor. Bottom waters contained higher concentrations of particulate Cd, Cu, Pb and Zn than surface waters. Distribution coefficients between dissolved and particulate phase (K/sub d/) of Cu and Cd decreased with the increasing distance from the Harbor, possibly due to reaction of these elements with sewage-derived particulate matter Al, Zn, Cu, Cd, and Pb in the surface sediment showed relatively high concentration in the inner Masan Bay.

The Practical Study for the Treatment of Fish Processing Saline Wastewater Using Immersed MBR (iMBR 공정을 이용한 수산물가공폐수 처리에 관한 실증적 고찰)

  • The study is the result of an practical operation analysis for the full scale fishery product wastewater treatment plant with immersed MBR (iMBR) process. Since fishery product industries show a wide range of wastewater generation by the season, design and operation of the equalization basin are very important factor. The aeration system for the equalization basin mixing can save the chemical consumption for followed system through the restriction of acid fermentation. The concentrations of wastewater primary DAF process treated were BOD 2,291 mg/L, $COD_{Mn}$ 530 mg/L, SS 256.8 mg/L, T-N 38 mg/L, T-P 13.5 mg/L respectively. It was considered that iMBR is the most efficient biological process for high salinity content wastewater since It is irrelevant to the capability of the sludge precipitation. SADp and SADm were 0.31, $26.5m^3/hr{\cdot}m^3$ respectively. In iMBR process, the critical F/M ratio was derived at 0.08~0.10 gBOD/gMLSS by analysing the correlations between MLSS, normalized TMP and temperature. The effluent concentrations were BOD 1.8 mg/L, $COD_{Mn}$ 12.4 mg/L, SS 1.0 mg/L, T-N 7.85 mg/L, T-P 0.1 mg/L and removal efficiencies were 99.9%, 97.6%, 96.3%, 95.7%, 97.8% respectively.

Design and Environmental/Economic Performance Evaluation of Wastewater Treatment Plants Using Modeling Methodology (모델링 기법을 이용한 하수처리 공정 설계와 환경성 및 경제성 평가)

  • It is not easy to compare the treatment processes and find an optimum operating condition by the experiments due to influent conditions, treatment processes, various operational conditions and complex factors in real wastewater treatment system and also need a lot of time and costs. In this paper, the activated sludge models are applied to four principal biological wastewater treatment processes, $A_2O$(anaerobic/anoxic/oxic process), Bardenpho(4 steps), VIP(Virginia Initiative Plant) and UCT(University of Cape Town), and are used to compare their environmental and economic assessment for four key processes. In order to evaluate each processes, a new assessment index which can compare the efficiency of treatment performances in various processes is proposed, which considers both environmental and economic cost. It shows that the proposed index can be used to select the optimum processes among the candidate treatment processes as well as to find the optimum condition in each process. And it can find the change of economic and environmental index under the changes of influent flowrate and aerobic reaction size and predict the optimum index under various operation conditions.
