Suppression of Nitrate Accumulation in Vegetables by Foliar Application of Micronutrients (미량원소 엽면 처리에 의한 엽채류의 질산태 질소 축적 억제)

  • Suppression of nitrate accumulation in vegetables through foliar application of micronutrients was investigated. Spinach and lettuce were grown in pots under greenhouse condition. Micronutrient solutions containing Cu, Mn, Mo, and Zn were used; chitosan was added into one and the other contained chitosan oligomers. The micronutrient solutions were sprayed on the leaves at 3 and 4 weeks after transplanting of 20-day-old seedlings. Plants were harvested at 5-weeks after transplanting. Yield, contents of chlorophyll, Brix value, micronutrient, and nitrate, and nitrate reductase activity were measured. Fresh weights of lettuce and spinach were significantly increased by the foliar application of micronutrients. Contents of chlorophyll and micronutrients were higher in micronutrient-treated plants, while those of nitrate were reduced by about 10 and 14-23% in lettuce and spinach, respectively. Compared to the control plants, nitrate reductase activity was higher in plants treated with micronutrients. Results of this study indicate the effect of micronutrients on the suppression of nitrate accumulation was relatively small in comparison to the contents of nitrate in leaves of spinach and lettuce. To maximize the effect, nutrient composition in solution, application time, and frequency should be further examined, taking into consideration nitrogen level in soil and other environmental factors including light condition.

Modified Atmosphere Packaging of Fresh Strawberries by Antimicrobial Plastic Films (항균성 플라스틱 필름을 이용한 딸기의 환경기체조절포장)

  • Low density polyethylene (LDPE) films incorporated with 1% antimicrobial agents of Rheum palmatum extract, Coptis chinensis extract, and Ag-substituted inorganic zirconium matrix, were applied to modified atmosphere packaging of 200 g fresh strawberries. Plain LDPE film package, PVC wrap and perforative pinhole package of the film impregnated with 1% Rheum palmatum extract were also constructed for comparative purpose. All the packages were stored for 13 days at $5^{\circ}C$ and measured in package atmosphere, microbial count and quality attributes of the strawberry fruits. The antimicrobial LDPE films retarded the growth in total aerobic bacteria, lactic acid bacteria and yeast on the fruits, and resulted in significantly lower decay. The degrees of reduced microbial growth and fruit decay in the antimicrobial film packages were more pronounced, when applied by hermetical sealing to produce the modified atmospheres of low $O_2$ (<4.0%) and $CO_2$ concentrations with $6.3{\sim}9.0%$. The hermetically sealed packages of antimicrobial LDPE films also showed better retention of fruit firmness and did not give any negative effect on the physical and chemical qualities of strawberries.

Bio-Green' Functional Water Supply Influences Mineral Uptake and Fruit Quality In Tsugaru Apples (‘바이오 그린’ 기능수 처리가 사과 쓰가루 품종의 무기성분 흡수와 과실품질에 미치는 영향)

  • Commercial Bio- Green(B.G.) functional water was manufactured through a series of processes : water - ultra-purification - adding catalysts - energy imprinting fermenting with energized water + zeolite and others + photosynthetic bacteria in fermenter longrightarrow filtering. Control(0), 5 or 10 liters per plant of B.G. functional water were supplied to the orchard soil under canopy of 10 year- old ‘Tsugaru’/M26 apple trees on March 20, May 20 and June 20, 1995, respectively. pH and content of Ca and Mg of orchard soil were increased by supply with B.G. functional water. However, P$_2$ $O_{5}$, K, and B contents were not influenced by the treatment. At harvest time soluble solid content of flesh tissue and anthocyanin of fruit skin were increased by the treatment. B.G functional water treatment showed higher root activities, and photosynthesis of leaves than that of control. Also B.G. functional water treatment enhanced Ca content in fruit skin and flesh tissues, whereas not affected N, K, and Mg contents. During storage at 4$^{\circ}C$ cold room, the more volume of B.G. functional water supply showed lower bitter pit symptom. Respiration and ethylene evolution in fruit decreased, while fruit firmness increased by the treatment during storage.

  • There are several purpose to introduce microorganism into the Soil. The major purpose is to promote plant growth and inhibit plant pathogens. The model example is to put in nitrogen fixing symbiotic bacteria, Pythium and Rhizobium. In order to achieve the intended goal, the introduced microorganism should survive and colonize with sufficient density. The survival of introduced microorganism depend upon biotic and abiotic factors. Predation and competition are important among biotic factor. Water tension, organic carbon, inorganic nutrients(N, P), pH are important factor among abiootic factor. Soil texture and distribution of soil pore are also important in the survival and colonization of introduced microorganism. Selection by soil ecosystem for inoculant is a crucial factor for colonization. Good example are control of autochtonous microorganism and the introduction of surfactant biodegrading Pseudomonas. Sometimes, carriers such as peat and montmorillonite can be added to help colonization. Carriers can protect introduced microorganism by supplying protective microhabitat. Organic polymer is also used as a carrier to immobilize bacteria or industrial enzymes. Examples of these carrier are calcium alginate, agarose and k-carrageenan. The function of these carrier is to provide microhabitat and help colonization for introduced microorganism.

Changes in Quality Attributes of Sigumjang with Fermentation (시금장 발효기간에 따른 품질 변화)

  • The various components of sigumjang were analyzed according to fermentation time. Aerobic bacteria were increased with fermentation time. Yeasts and molds were increased until 3 days after fermentation and then not changed. Six free sugars, 3 volatile organic acids and 6 non-volatile organic acids were detected. The content of free amino acids was $1,407.9{\sim}3,053.9\;mg%$. Glutamic acid was most abundant component among the amino acids, followed by phenylalanine, arginine and valine. The ratio of essential amino acid was $34.4{\sim}37.0\;mg%$. The content of mineral was $12,966.7{\sim}13,864.5\;mg%$. Potassium was the most abundant in quantity among the minerals in sigumjang except sodium which was added artificially. The principal fatty acids were linoleic acid, palmitic acid and oleic acid. The ratio of unsaturated fatty acids was $76.3{\pm}1.1%$. As a result of sensory evaluation, the highest score was obtained 3 to 5 days after fermentation.

  • In the traditional medicine, the basidiocarps of Poria cocos (Fr.) Wolf naturally collec­ted have been widely used as a traditional remedy for dropsy, diarrhea, gonorrhea and nervous disease more than 3500 years ago. It was recently found to be composed of the particular compounds having antitumor effects. Therefore, its demand has been gradually increased, whereas it is still dependent upon the natural harvest. The artificial cultivation of P. cocos was carried out to observe the morphological characteristics and to investigate the cultural characteristics of P. cocos isolates collected. The morphological characteristics of its basidiocarps was observed. The physiological aspects of its isolates were also investigated.

A Study of Heavy Metal-Contaminated Soil Remediation with a EDTA and Boric acid Composite(I): Pb (EDTA와 붕산 혼합용출제를 이용한 중금속으로 오염된 토양의 처리에 관한 연구(I): 납)

  • To choose a organic acid and in-organic acid composite which is the most effective in soil-flushing process cleaning lead-contaminated sites, lead removal rates were investigated in the experiments with some organic acids; 0.01M of EDTA showed the highest lead-extraction rate ($69.4\%$) compared to the other organic acids. Furthermore, the lead removal rates were measured with 0.01M of EDIA and 0.1M of in-organic acid ; a EDTA and boric acid composite showed the highest lead-extraction rate ($68.8\%$) at pH5 compared to the other composites. As the concentration of boric acid was increased from 0.1M to 0.4M in a 0.01M of EDTA and boric acid composite, lead removal rate was decreased from $68\%\;to\;45\%$. But as the concentration of EDTA was increased from 0.01M to 0.04M in a EDTA and 0.1M of boric acid composite, permeability was decreased from $6.98{\times}10^{-4}cm/sec$ (0.01M of EDTA) to $5.99{\times}10^{-4}cm/sec$ (0.04M of EDTA). However, permeability was increased from $4.41{\times}10^{-4}cm/sec$ (0.03M of EDTA) to $6.26{\times}10^{-4}cm/sec$ (0.03M of EDTA and 0.1M of boric acid composite). indicating EDTA could increase lead dissolution/extraction rate and decrease permeability. In this system, lead remediation rate is the function of lead dissolution rate from soils and permeability of the composite into soils, and the optimized [EDTA]/[Boric acid] ratio is [0.01M]/[0.1M].

Effect of Medium and Growth Regulators on Tuber Propagation by in vitro Culture of Yam (Dioscorea japonica TUNBERG) (산약(山藥)의 기내증식(器內增殖)에 미치는 배지(培地)와 생장조정제(生長調整濟)의 영향(影響))

  • The low seed propagation is one of the problem needed a lot of seed tuber for the propagation in yam. Therefore this experiment was carried out to understand the possiblility of seed tuber propagation by tissue culture of yam. In-vitro stem node of yam was cultured by concentration treatments of 1/2, 1/4 and 1/8 with MS medium additted with each concentration levels of IAA, NAA, IBA, kinetin and BA. Acorrding to the Iower concentration than MS medium, length of shoots was promoted, leaf emergence shoots and rooting shoots were increased at 1/8 MS medium during the culturing period of stem node in yam. Fixed IBA and kinetin under the concentration of MS mdeium was inhibited severely by the heigh concentration additted with lAA $1mg\;/\;{\ell}\;and\;NAA\;4mg\;/\;{\ell}$. But fixed IBA $5mg\;/\;{\el}l\;and\;kinetin\;2mg\;/\;{\ell}$ with concentration of 1/8MS medium was remarkably promoted leaf emergence shoots and rooting shoots by $1mg\;/\;{\ell}$ of additted lAA and NAA. Percentage of induced shoots was increased by combination treatments of lAA. $1.5mg\;/\;{\ell}\;and\;kinetin\;2mg\;/\;{\ell}$, also leaf emergence shoots and rooting shoots were promoted by combination treatments of lAA $1.5mg\;/\;{\ell}\;and\;kinetin\;2mg\;/\;{\ell}$.

Effects of $GeO_2$ and Citric Acid on Germanium Content of Callus and Plant in Angelica koreana MAX (강활(羌活)의 캘러스 및 식물체(植物體) 중(中) Ge함량(含量)에 미치는 $GeO_2$와 Citric Acid의 영향(影響))

  • This study was conducted to investigate the effect of growth regulators and $GeO_2$ on the induction and proliferation of callus and the effect of $GeO_2$ and citric acid on the Ge content of callus from explants and plant, Angelica koreana Max. The results obtained were summarized as follows. The callus induction was most effective on MS (Murashige and Skoog) medium containing 1. 0ppm 2, 4 - D with petiolule. The proliferation of callus was most effective at 2. 0ppm 2, 4 - D on the medium, at 2. 5ppm $GeO_2$ on the medium containing 2. 0ppm 2, 4 - D, and at $0.\;1{\sim}1mM$citric acid on the medium at pH6 containing 2. 0ppm 2, 4 - D and 2. 5ppm $GeO_2.$ The more $GeO_2$ in MS medium up to 20ppm, the more Ge content in callus. Ge content in callus was highest when the medium was supplemented with 0. 1mM citric acid and the pH of medium was low. The Ge content in plant was high in order of leaf > root > stem. Application $GeO_2$ to the soil increased Ge content in plant and application of 1mM citric acid with $GeO_2$ resulted in increasing Ge content highest in plant, but application more than l0mM citric acid resulted in Ge content decreased. Application of $GeO_2$ increased Ge content in callus and plant but had a tendency to decrease some mineral content, on the other hand application of $0.\;1{\sim}1mM$ citric acid with $GeO_2$ had a tendency to increase mineral content.

Degradation of Triclosan by the Photolysis, the Fenton, and the Hybrid Reaction with Fe$^{2+}$ and UV : A Comparative Study (광반응, 펜톤, 그리고 Fe$^{2+}$와 UV의 조합반응을 이용한 Triclosan의 분해 : 공정 비교 연구)

  • The degradation mechanism of Triclosan(TCS), which is a potent broad-spectrum antimicrobial agent and has been considered as an emerging pollutant, was investigated in the Fenton and the hybrid reaction with Fe$^{2+}$ and UV-C. The results show that the Fe$^{2+}$ is oxidized to 30% by $H_2O_2$, 28% by UV-C, and 15% by UV-A for 10 min. The degradation rate of TCS for beginning time(10 min) was higher in UV-C only reaction than that in hybrid reaction, which of the order was inverted according to the lapse of reaction time. The effect of methanol was the greatest in Fenton reaction, in which the degradation rate of TCS decreased from 90% to 5% by the addition of methanol. Chloride, ionic intermediate, was produced to 77% for 150 min of hybrid reaction(Fe$^{2+}$ + UV-C), which was the greatest. In case with methanol, the generation rate of chloride for 15 min was ignorable in all reactions($\leq$2%) but the hybrid reaction with Fe$^{2+}$ and UV-C(12%). Additionally, the removal rate of TOC in each reaction was estimated as the followed orders; Fe$^{2+}$ + UV-C > Fe$^{2+}$ + $H_2O_2$ > Fe$^{2+}$ + UV-A > UV-C > UV-A. However, the Fenton reaction was almost stopped after 90 min because the reaction between Fe$^{2+}$ and $H_2O_2$ cannot be kept on without adding the oxidant. The phenomena was not observed in the hybrid reaction. In view of generating chloride, the reductive degradation of TCS may be in the hybrid reaction with Fe$^{2+}$ and UV-C, which is favorable to mineralize halogenated organic compounds such as TCS. Consequently, the hybrid process with Fe$^{2+}$ and UV-C may be considered as the alternative treatment method for TCS.
