Role of Retinoic Acid in the Gut Mucosal Immunity (장관면역에 있어서 레티노인산의 역할)

  • 장관은 우리 몸에서 가장 넓은 표면적을 가지고 있으며, 외부로부터 침입하는 이물에 대항하여 면역세포의 배치가 필수적이다. 항원과 만난 적이 없는 naive lymphocyte는 임파절 등 2차 임파기관의 수지세포(dendritic cell)가 임파구에 항원을 제시할 때 레티노인산(retinoic acid)을 생산하여 제공함으로써, 소장 조직에 특이적으로 임파구가 호밍(homing)하는 능력을 부여한다. 한편, 장에 있어서는 식품항원에 대한 면역반응을 억제할 필요가 있는데, RA는 T세포의 기능분화를 억제하여 면역관용의 성립에도 관여함이 밝혀졌다.

  • 실험실에서 면역세포가 종양세포를 죽일 수 있다든가 혹은 드물지만 암 환자에서 암이 자연적으로 소실되었다는 임상적 보고 그리고 AIDS 환자와 같은 면역계의 장애를 가진 환자에서 Kaposi 육종, 특정 형태의 림프종이 더 잘 생긴다는 것 등으로 미루어 보면 동물이나 인간에 발생하는 암에 대해 어느 정도의 면역반응이 일어나는 것은 분명하다. 인체는 규칙적으로 암세포를 생산하지만 면역계에 의해 제거되는 것으로 추측된다. 그러나 아직도 면역계가 어떻게 암을 억제하는지 그리고 면역계가 암발생을 억제하는데 왜 실패하는지에 대해서는 정확히 모르고 있다. 이러한 의문점들은 앞으로 종양 면역학이 규명해야 할 과제이다.

Immunohistochemical study of the endocrine cells of the gastrointestinal mucosa of the African clawed toad, Xenopus laevis (아프리카 발톱두꺼비의 소화관내분비세포에 관한 면역조직화학적 연구)

  • The gastrointestinal endocrine cells of the African clawed toad, Xenopus laevis have been investigated immunohistochemically using the avidin-biotin method. Seven antisera were tested and three endocrine cell types immunoreacted with antisera to neurotensin, GRP and substance P. A moderate number of neurotensin-immunoreactive cells were weakly reacted in the small intestine. GRP-immunoreactive cells were mainly situated among the upper portion in the fundic glands, and the basal portion in the pyloric glands. These cells were oval and round in shape. On the other hand, in the intestine they were thin spindly cells with the epithelium. Substance P-immunoreactive cells were observed in among intestinal epithelium. However, no secretin-, motilin-, M-Enk- and PYY-immunoreactive cells were found in the GIT of the African clawed toads.

  • The regional distribution and relative frequency of neurohormonal peptides-producing cells were demonstrated in the gut of the stomachless teleost, the Prussian carp, Carassius auratus Linnaeus, using 10 types of specific antisera raised against mammalian regulatory peptides. The gut of the Prussian carp was divided into five portions from proximal to distal (Segments I~V). Most of immunoreactive cells in the epithelial lining portion, between epithelial cells, were generally spherical or spindle shape having long cytoplasmic process that reached to the lumen (open typed cell) while cells showing round in shape (close typed cell) were found in the basal portions of epithelial lining occasionally. Somatostatin-, cholecystokinin (CCK)-8- and pancreatic polypeptide (PP)- immunoreactive cells were observed in this study. However, no serotonin-, glucagon-, chromogranin A-, secretin-, vasoactive intestinal peptide (VIP)-, substance P- and bombesin-immunoreactive cells were found. Somatostatin-immunoreactive cells were restricted to most proximal segments of the gut (Segment I) with rare frequency and CCK-8-immunoreactive cells were demonstrated in the proximal segments of the gut (Segments I and II) with a few to rare frequencies. In addition, pancreatic polypeptide-immunoreactive cells were demonstrated in the proximal to middle segments (Segments I~III) with moderated to rare frequencies. In conclusion, the distribution and relative frequency of these immunoreactive cells are well corresponded to the previous reports in stomachless teleost but somewhat peculiar patterns are also detected.

Immunomodulating Effects of Red Ginseng on the Regulation of Cytokine Release in vivo (홍삼의 생체 내 사이토카인 분비에 대한 면역조절효과)

  • Cytokines released from innate immune cells play key roles in the regulation of the immune response. Red Ginseng (RG, steamed and dried root of Panax ginseng C.A. Meyer) is known to show different pharmacological effects by changed composition of saponins compared with Panax Ginseng. In this study, we examined the immunomodulatory effects of RG on the regulation of cytokine release in mice. RG was injected i.p at doses of 0.5, 5 and 50 mg/kg for 6 weeks. We assessed that the weight index of immune organs such as thymus, and spleen, and the mitogen blastogenesis of splenocytes. We also determined the levels of circulating cytokines in serum from RG-treated mice using ELISA assay. The weight index of thymus and spleen, and proliferation of mitogen response of splenocytes have increased in the RG-injected groups. In addition, the levels of IFN-${\gamma}$, TNF-${\alpha}$, IL-6, IL-12 and IL-2 concentrations have significantly increased in the serum of RG-treated mice, but that of IL-10 has not. These results suggest that RG has immune stimulating effects and could be useful as a immunoregulator of circulating cytokine release in vivo.

An histological and immunohistochemical study of endocrine cells in the gastrointestinal tract of the Amur lizard (Takydromus amurensis) (아무르장지뱀의 위장관 내분비세포에 관한 조직화학적 및 면역조직화학적 연구)

  • The distribution and the frequency of endocrine cells in the GIT of the Amur lizard(Takydromus amurensis) were investigated using silver techniques and immunohistochemical method. Only argyrophil cells stained by the Grimelius technique were found numerous in the pyloric glands and moderate in the duodenum. Two types of immunoreactive endocrine cells were identified by immunohistochemical method. Bovine CG-immunoreactive cells were demonstrated the entire GIT. BPP-immunoreactive cells were restricted in the duodenum and the ileum. The results showed that: the number of argyrophil cells was lower than the number of cells stained with bovine CG antiserum. Therefore, bovine CG-immunostaining and the Grimelius silver technique did not correspond with various endocrine cells in the Amur lizard.

  • 기능성 다당체는 자연계에 두루 존재하며 특히 많은 천연소재 유래 다당체의 경우 거의 부작용없이 광범위한 면역조절 기능을 나타내는 것으로 보고되고 있다. 이들 다당체의 면역증진 기능성은 다양한 경로와 타켓을 대상으로 나타나고 있으며 각 다당체의 기능성 차이는 그들에 구조적 차이에 기인한 것으로 판단되고 있다. BRM의 일종으로 천연소재 유래 다당체는 면역기관이나 T와 B 림프구, 대식세포, NK cell, LAK 세포 및 수지상 세포 등 면역세포의 활성화, 사이토카인 및 항체의 생성, 적혈구의 면역기능에 영향을 미쳐 생체방어 기능, 즉 면역기능을 증진시켜 주는 것으로 보고 되었다. 또한 lentinan, PSK, PSP, PUPS 등과 같이 이미 잘 알려진 다당체의 경우, 이들 다당체가 지닌 품질 안정성, 정확한 효능, 낮은 독성 및 부작용 등을 근간으로 다양한 항암치료제의 보조제로서 활용가능성도 높아지고 있다. 반면 다당체를 이용한 기능성 구명 연구에서는 몇 가지 제한이 있는 것도 사실이다. 첫째로 비교가능한 적절한 기능성 다당체의 비교 표준이 부재하다는 것이며 두 번째로는 대부분의 다당체 관련 연구가 보다 철저한 구조적, 정성적 연구결과 없이 주로 추출물을 이용한 연구로 진행되어 정확한 분자수준에서의 면역조절 기전을 구명하기 어렵다는 점이다. 마지막으로 정확한 기능 구명을 위한 신호전달경로 관련 연구, 수용체 관련연구 및 임상연구 등이 부족하다는 점이다. 따라서 향후 다당체를 이용한 면역력 증진 등 기능성 구명연구의 활성화를 위해서는 보다 정확한 기능성 다당 분획의 구조분석 및 동정 연구가 수행되어야 할 것이며 이는 분자 수준의 기능성을 이해하는데 주요한 요인이 될 것으로 판단되었다.

Tyrosine Hydroxylase - Immunoreactive Dopaminergic Neurons in the OX Retina (Ox retina내 tyrosine - hydroxylase 면역 반응되는 dopaminergic neuron에 대하여)

  • The morphology of dopaminergic neurons in the adult ox retina was studied. The dopaminergic neurons were identified using antibody immunocytochemistry. The great majority of tyrosine hydroxylase - immunoreactive neurons were located at the innermost border of the inner nuclear layer. The processes were monostratified and ran laterally within layer 1 of the inner plexiform layer. The second major population of tyrosine hydroxylase - immunoreactive neurons was displaced amacrine cells. The processes of displaced tyrosine hydroxylase - immunoreactive amacrine cells were also located within layer 1 of the inner plexiform layer. Some processes of a few neurons were located in the outer plexiform layer. A very low density of neurons had additional bands of tyrosine hydroxylase - immunoreactive processes in the middle and deep layers of the inner plexiform layer. The processes of dopaminergic neurons widely extended radially and formed large, moderately branched dendritic fields. These processes occasionally had varicosities but did not have "dendritic rings". These results indicate that dopaminergic cells make up specific neuronal cell types in the ox retina.

  • 최근 컴퓨터와 인터넷의 급속한 발전과 더불어 컴퓨터의 데이터를 파괴하는 바이러스나 정보를 빼내기 위한 해킹 등이 만연하고 있다. 이에 컴퓨터의 데이터를 보호하기 위한 방법들이 연구중에 있는데 이 중 외부의 침입물질에 대해 자체적인 보호와 제거기능을 가지는 생체면역시스템을 이용한 컴퓨터면역시스템 구축에 대해 활발히 연구가 진행되고 있다. 생체 면역시스템은 바이러스나 병원균 등의 낮선 외부 침입자로부터 자신을 보호하고 침입자를 제거한다. 본 논문에서는 생체면역시스템의 면역세포 중 하나인 세포독성 T세포의 자기(Self)와 비자기(Nonself)를 구분하는 기능을 이용해 자기변경 검사 알고리즘을 구현하였다. 구현한 알고리즘은 자기로 인식하는 자기파일에서 자기를 구분하는 MHC 인식부를 구성한다. 이렇게 구성한 MHC 인식부는 자기파일을 대표하는 값을 이용하여 변경된 파일을 구분한다. 이 알고리즘을 변경된 자기파일에 적용함으로써 컴퓨터 해킹이나 바이러스에 의한 자기파일의 변경 검사의 유효성을 검증한다.

Immunohistochemical study of the pancreatic endocrine cells in the ICR mice (ICR 마우스 췌장 내분비세포에 대한 면역조직화학적 연구)

  • The regional distribution and relative frequency of the pancreatic endocrine cells in the ICR mouse were studied by immunohistochemical (PAP) method using four types of specific antisera against insulin, glucagon, somatostatin and human pancreatic polypeptide (PP). The pancreas of mice could be divided into three portions; pancreatic islets, exocrine and pancreatic ducts. Pancreatic islets, furthermore, were subdivided into three regions (central, mantle and peripheral region) according to their located types of immunoreactive cells. In the pancreatic islet portions, insulin-immunoreactive cells were located in the central and mantle regions but most of somatostatin-, glucagon- and PP-immunoreactive cells were detected in the mantle and peripheral regions with various frequencies. In addition, PP-immunoreactive cells were also found in the central regions of pancreatic islets of ICR mouse. In the exocrine portions, all four types of immunoreactive cells were demonstrated in the ICR mouse. In the pancreatic duct portions, insulin- and glucagon-immunoreactive cells were situated in the epithelial lining of ICR mouse with a few and rare frequencies, respectively. In addition, rare PP-immunoreactive cells were also demonstrated in the subepithelial regions of the pancreatic duct. However, no somatostatin-immunoreactive cells were demonstrated.
