AN IMMUNOHISTOCHEMICAL STUDY ON THE EXPRESS10N OF GROWTH FACTOR IN PERIODONTAL TISSUE DURING THE EXPERIMENTAL MOVEMENT OF RAT INCISORS (백서의 실험적 치아 이동시 치주조직내 성장인자 발현에 관한 면역조직화학적 연구)

  • This study was designed to evaluate the expression of growth factor in periodontal tissue during the experimental movement of rat incisors by LSAB(Labelled streptavidine Biotin) immunohistochemical staining for EGF(Epidermal growth factor). 23 Sprague-Dawley rats were divided into a control group(3rats) and experimental groups(20rats), where a force(75g) from helical springs across the maxillary incisors was applied. Experimental groups were sacrificed at 12 hours, 1, 4, 7 and 14 days, after force application, respectively. And Tissue slides of control and experimental groups were studied immunohistochemically and histologically. The results were as follows : 1. In 14days after force application, periodontal fibers were strectched on the tension side, and compressed In pressure side of all experimental groups, and the arrangement of periodontal fibers was not recovered yet. 2. The degree of EGF expression in control group was strongly positive in the oral epithelium, predentin, capillaries in pulp and periodontal spaces. But osteoblasts and osteoclasts were stained mildly positive. 3. EGF expression was mild and diffuse in 12 hours, 1, 4 and 7 days of experimental groups and was not significantly different between the tension and pressure sides. 4. The degree of EGF expression in the 14-day experimental group was higher than any other group. And the tension side showed a more positive EGF expression than the pressure side. The apical area revealed a more positive EGF expression than the cervical area.

Effects of Dietary Supplementation of Illite on Humoral Immunity against Salmonella typhimurium Flagella Antigen in Laying Hens (산란계에 Illite 급여가 Salmonella typhimurium 편모항원에 대한 체액성 면역 반응에 미치는 영향)

  • This study was carried out to investigate dietary effects of illite on humoral immune response against Salmonella typhimurium flagella in Hyline Brown laying hens. Total of twenty eight 36-week-old hens were divided into two groups; The first was fed commercial diet as control and the other was fed diet containing 2 % illite. Each group was divided into 2 sub-groups and then performed immunization of Salmonella flagella with different adjuvants which were Freund's adjuvant and croton oil. The rates of egg-production in all groups were normal range but no difference was found between illite-treated and untreated groups. The serum antibody titers of Freund's adjuvant-used subgroup in illite fed groups were significantly increased, especially from 6-to 9-week after $1^{st}$ immunization compared to those of control groups (p<0.05). And the antibody titers of croton oil-used subgroup in illite-fed groups were also significantly increased, especially at 4-, 6- and 7-week (p<0.05). The results demonstrated that the feeding illite stimulated the immune response against S. typhimurium flagella antigen in laying hens and suggest that the supplementation of illite to the poultry diets may support protective effects against bacterial infections such as Salmonellosis.

Effects of Fermented Milk with Hot Water Extract from Acanthopanax senticosus and Codonopsis lanceolata on the Immune Status of Mouse (가시오가피와 더덕 추출물을 첨가한 발효유가 마우스의 면역기능에 미치는 영향)

  • We investigated the immunomodulatory actions of water extract from Acanthopanax senticosus in male ICR mice. The mice were treated with fermented milk containing three added doses of freeze dried extract: 3 mg/kg (A), 9 mg/kg (B), and 27 mg/kg (C) of body weight with Acanthopanax senticosus: Codonopsis lanceolata (8:2) for 7 and 10 weeks, respectively. Organ weights, plaque-forming cell tests, agglutination tests, IgG tests, differential white cell counts, and histological tests were performed at the 7th and 10th weeks of dietary treatment. There were no significant differences in body weight and organ weight. The spleen indices of group B at 7 weeks and group C at 10 weeks were significantly higher than those of the control group (p<0.05). For the plaque-forming cell test, groups B and C at 7 weeks, and group C at 10 weeks, showed significant increases over the control group (p<0.05). The agglutination test decreased with an extended experimental period. Groups A, B, and C at 7 weeks, and groups B and C at 10 weeks, had greater antibody responses to sheep red blood cells (SRBC) than the control group. The IgG antibody production of group C at 7 weeks and groups B and C at 10 weeks were significantly higher than the control group (p<0.05). In groups B and C, lymphocyte percentage was higher than the control group, and their spleen and thymus tissues showed active immune reactions.

Effects of aquatic drugs and immune response in color carp, Cyprinus carpio, to Aeromonas hydrophila (Aeromonas hydrophila에 대한 비단잉어의 면역반응과 수산약제의 영향)

  • This study was carried out in order to investigate the immunosuppressive factor and immune response of color carp. The protection and serum antibody production of juvenile color carp aganist Aeromonas hydrophila were investigated on the effect of temperature differences and injected several aquatic drugs, i.e. Hydrocortisone, Oxytetracycline, Chloramphenicol and Ascorbic acid. The fish were injected intraperitoneally with 1mg/fish of HKC and FKC at three different temperature conditions as $16^{\circ}C$, $24^{\circ}C$, and $32^{\circ}C$ respectively. There were induced better protection and serum antibody production in the fish which had been kept at $24^{\circ}C$ than in the fish which had been kept at $16^{\circ}C$ and $32^{\circ}C$. The FKC immunized fish were followed 24 hrs later with intraperitoneal injection of 40mg/kg body weight of Hydrocortisone, 60mg/kg body weight of Oxytetracycline. 60mg/kg body weight of Chloramphenicol and 30mg/kg body weight of Ascorbic acid, respectively. The control fish were injected PBS only. The fish given the above aquatic drugs reduced serum antibody production level and protection rate when compared to control fish. As the results, immune response of juvenile color carp immunized FKC at $24^{\circ}C$ was more effective than $16^{\circ}C$ or $32^{\circ}C$ and immune response of juvenile color carp injected several aquatic drugs which was seemed to be immunosuppressive factor.

Characterization and Immunomodulation Activity of Lactobacillus sakei L2 and L8 Isolated from Chicken Cecum (닭의 맹장으로부터 분리한 Lactobacillus sakei L2와 L8의 특성 및 면역활성)

  • The aim of this study was to investigate the potential of lactic acid bacteria (LAB) strains as probiotics. Two strains were isolated from healthy chicken cecum and their acid and bile tolerance, residual organic acids, antibacterial activity against pathogenic bacteria, and immunomodulation activity were measured. Identification of the isolated strains was performed using the API 50CHL system and phylogenetic analysis using 16S rDNA sequencing. The isolates were determined to be Lactobacillus sakei strains. The acid tolerance of strains L2 and L8 was high enough that 75% of the inoculum survived in pH 2 for 2 h. The bile tolerance of both strains was observed at a 1% Oxgall concentration in MRS broth. The production of organic acids (lactic acid and acetic acid) and pH changes during growth were monitored and the maximum concentrations were obtained after 48 h of incubation. Culture supernatants of the two LAB strains showed strong antibacterial activity against pathogenic bacteria. The heat-killed LAB cells also induced high levels of immune cell proliferation compared with the control, and stimulated IL-6 and TNF-α production in mouse macrophages. Therefore, L. sakei strains L2 and L8 can be considered suitable probiotic bacteria.

Inhibitory Effect of Chlorine Dioxide on Phenoloxidase Activation of the Indianmeal Moth, Plodia interpunctella (화랑곡나방(Plodia interpunctella)의 페놀옥시데이즈 활성화에 대한 이산화염소의 억제 효과)

  • Phenoloxidase (PO) is an oxidizing enzyme and plays crucial roles in insect immunity and cuticle sclerotization. High oxidizing activity of chlorine dioxide gives effective control activities against microbes and insect pests. These allowed us to assess any inhibitory activity of chlorine dioxide against PO with respect to insect immunity. PO activities of the Indeanmeal moth, Plodia interpunctella, was detected in both hemocytes and plasma. Upon bacterial challenge, PO activity was significantly increased especially in plasma. However, the immune challenge coupled with chlorine dioxide treatment did not enhance PO activity. When different chlorine dioxide concentrations were incubated with activated PO by immune challenge, they did not inhibit the activated PO. These results indicate that chlorine dioxide suppresses PO activity by inhibiting PO activation.

Enzyme-linked Immunosorbent Assay Strip Sensor for Rapid Detection of Staphylococcus aureus (Staphylococcus aureus 신속 검출을 위한 효소면역측정 스트립 센서)

  • In this study, an established enzyme-linked immunosorbent assay and immuno-chromatography technique are combined to fabricate an immuno-strip sensor for the detection of S. aureus. The immuno-strip is manufactured by using four different functional membranes. The capture antibody is immobilized on the nitrocellulose membrane due to the high affinity and the capillary action through porous membranes induces a flow of sample. A colorimetric signal is appeared according to the enzyme reaction and is analyzed by the digital camera (qualitative analysis) and home-made image analysis software (quantitative analysis). Under the optimal conditions, samples with S. aureus in the range of $2.7{\times}10^4{\sim}2.7{\times}10^7CFU/mL$ can be detected by the colorimetric method within 30 min.

Control efficacy of BtPlus against two mosquitoes, Aedes koreicus and Culex vagans (한국숲모기와 줄다리집모기에 대한 비티플러스 방제 효과)

  • Two mosquito species were collected in still-water near farming area in Andong, Korea. Based on morphological characters, these two mosquitoes were identified as Aedes koreicus and Culex vagans, respectively. DNA barcode analyses supported the identification. An entomopathogenic bacterium, Bacillus thuringiensis subsp. israelensis (BtI), exhibited insecticidal activities against the two mosquito species and its virulence was more potent than that of B. thuringiensis subsp. kurstaki. It has been known that the bacterial metabolites of Xenorhabdus spp. suppress insect immunity and enhance pathogenicity of B. thuringiensis. This study tested the effect of the bacterial culture broth of Xenorhabdus spp. on enhancing BtI pathogenicity. Among three Xenorhabdus spp., culture broth of X. ehlersii (Xe) was relatively effective to enhance BtI pathogenicity against both mosquito species. Indeed, organic extracts of Xe culture broth suppressed the hemocyte-spreading behavior, suggesting the presence of immunosuppressant in the culture broth. These results suggest a formulation of BtPlus by mixing BtI spore and Xe culture broth to be applied to control the two mosquito species.

Antimicrobial Activity of Specific IgY against Salmonella gallinarum (Salmonella gallinarum에 대한 specific IgY의 항균력)

  • In this study, we investigated the antimicrobial effects of anti-Salmonella gallinarum-specific IgY separated from egg yolk obtained from layers immunized by S. gallinarum. The comparison tests of vaccination, content of IgY and innoculation number were examined by microscopic observation, turbidity, and pH. The results show that the ratio of anti-S. gallinarum IgY in the total IgY was 23%. Also, the anti-S. gallinarum IgY had selectivity only to S, gallinarum. The 0.3% addition of anti-S. gallinarum-specific IgY resulted in agglutinating clusters of S, gallinarum cells, but the agglomeration didn#t occur in IgY from layers not immunized nor in the control group. Microscopic observation indicated agglomerative cells when IgY was added at 0.2% or higher, and the pH and turbidity examinations revealed that a suppression effect was remarkable in IgY at more than 0.1%. These results suggest the possibility that IgY extracted from eggs and obtained from layers immunized by S. gallinarum can be used to prevent fowl typhoid.

Effects of the Duration of Highly Intensive Exercise on Lymphocyte Cell Death in Rats (고강도운동 지속시간이 rat의 림프구 세포사에 미치는 영향)

  • The time-dependent effects of highly intensive exercise on the hematological properties of leukocytes, as well as $CD4^+$ and $CD8^+$ level changes as T-lymphocyte activation subsets and the cell death of lymphocytes in rats were studied in this research. Twenty, 60, and 120 min of highly intensive exercise was performed daily for 8 weeks. Total leukocyte counts in the blood of rats exercising for 20 min were elevated; they then decreased to less than the level of the control group up to 120 min. The patterns of lymphocyte level changes were directly influenced by exercise duration and the extents of alteration were similar to the total leukocytes counts. The levels of $CD4^+$ and $CD8^+$ in the blood of the exercising rats were not statistically different even when the exercise was continued for 120 min; thus, the exercise did not affect T-lymphocyte activation. Early- and late-stage lymphocyte apoptosis was not affected by the length of exercise, except that late-phase apoptosis was slightly increased at 120 min, suggesting that aging processes for lymphocyte apoptosis might be stimulated at that time. As the exercise time became longer, stimulated necrosis of lymphocytes was observed, so damage in lymphocytes and a potential loss of immunity might be presumed. The current observation suggests that long-term, highly intensive exercise might result in a loss of immunity that could be due to the damage of lymphocytes in terms of both their numbers and inflammation-related functions. The results suggest that under highly intensive exercise conditions, more than 20 min of exercise should not be suggested for health care purposes.
