Gray hair is a representative sign of aging. Intrinsic aging, stress, and the external environment cause hair graying. Stress is known to be a major factor in the early onset of hair graying. We previously found that Pueraia lobata root extract (PLRE) can prevent hair graying by promoting melanin formation. However, it remains unknown whether PLRE can prevent hair graying induced by conditions of stress. In this study, we confirmed the effect of PLRE on stress-induced hair graying. A reporter cell line was newly constructed to confirm the expression of microphthalamia-associated transcription factor (MITF), the main transcription factor for melanin production. MITF expression and melanin pigmentation were reduced in human hair follicle tissue treated with the stress hormone cortisol or H2O2 to induce oxidative stress. PLRE treatment restored MITF expression and increased the amount of melanin pigment in the hair follicle. The expression of Tyrosinase related proteins-2 (TRP-2), a melanin synthesis enzyme in the hair follicle, also increased. In conclusion, PLRE can effectively prevent the inhibition of melanin synthesis by stress hormones and oxidative stress.
Juglans mandshurica belongs to the family Juglandaceae is known to contain a wide range of pharmacological activities including anti-cancer, anti-inflammation, astringent, and anti-human immunodeficiency virus-type 1 (HIV-1). Melanogenesis refers to the biosynthesis of melanin pigment in melanocytes. In this study, to investigate the whitening activity of the extracts from Juglans mandshurica, we measured effects on a tyrosinase activity, a melanogenesis, and a tyrosinase synthesis in the B16/BL6 melanoma cells and an antioxidant activity. The extracts significantly scavenged a 1,1-diphenyl-2-picrylhydrazyl (DPPH) and a superoxide anion radicals in a dose-dependent manner with a $SC_{50}$ value of $20\;{\mu}g/mL$ and $25\;{\mu}g/mL$, respectively. Also, the tyrosinase activity and melanogenesis were significantly inhibited by the extracts. Furthermore, the synthesis of tyrosinase protein was significantly decreased by the extracts in enzyme-linked immunosorbent assay. Double blind study on the clinical efficacy of a cream containing 2% of the extracts showed that the extracts have a significant skin whitening effect. Therefore, this study demonstrates that the extracts from Juglans mandshurica may be useful as a potential agent for skin whitening.
Melanin is a polymer substance that plays an important role in the determination of hair growth and skin color in vivo. However, melanin, which is over-produced by reactive oxygen species, is known to cause stains, freckles, and hypercholesterolemia, which are associated with aging. Previous studies have shown that polyphosphate, one of the components of Rhynchosia Nulubilis, inhibits skin aging induced by ultraviolet rays. The aim of this study is to investigate the direct effect of Rhynchosia Nulubilis ethanolic extract (RNEE) on melanin synthesis. In this study, RNEE showed no antioxidative effects on scavenging activity of DPPH radical in addition to reducing power. The cytotoxicity of RNEE was increased in a dose-dependent manner in an MTT assay. In addition, RNEE increased tyrosinase activity and melanin synthesis in DOPA-oxidation experiments. RNEE did not promote the conversion L-DOPA into melanin in live cells, but melanin production was promoted in the RNEE-treated group after H2O2 pretreatment compared to the control group in which melanin production was reduced by treatment with H2O2. In addition, RNEE increased the expression level of tyrosinase related protein-2 (TRP-2) and increased the expression level of tyrosinase related protein-1 (TRP-1) at a concentration of $16{\mu}g/ml$. In particular, it was found that RNEE increased the expression level of SOD-3, by which superoxide anion is converted to hydrogen peroxide, higher than the control and ${\alpha}$-MSH used as a positive control at a concentration of more than $16{\mu}g/ml$. The results suggest that RNEE can induce melanogenesis related to black hair.

It has been reported that the ethanolic extract of the root of Piper methysticum (P. methysticum) inhibits melanogenesis in melanocyte stimulating hormone (MSH)-activated B16 melanoma cells. Flavokawain B (FKB) and Flavokawain C (FKC) isolated from this extract have been found to inhibit melanin production based on anti-melanogenesis activity. This study was designed to find out the inhibition and its process of FKB and FKC on melanin synthesis in melan-a melanocytes. FKB and FKC inhibited melanogenesis at 10 μM, 5 μM respectively in melan-a melanocytes. However, they did not inhibit extracellular tyrosinase activity from melan-a melanocytes. FKB reduced the protein level of tyrosinase (Tyr), tyrosinase-related protein 1 (TRP-1), tyrosinase-related protein 2 (TRP-2), microphthalmia-associated transcription factor (MITF) and the mRNA level of Tyr and TRP-1. FKC reduced the protein level of TRP-2 and MITF and the mRNA level of TRP-1 and Tyr. The reduced expression of Tyr and TRP-1 might be resulted from the decreased MITF which regulates major melanogenic proteins. However, since the mRNA expression of MITF did not change by FKB and FKC treatment, the effects of FKB and FKC on extracellular signal regulating kinase (ERK)/AKT phosphorylation, known to regulate the degradation of MITF, were confirmed. FKB and FKC significantly increased the phosphorylation of ERK1/2, not in AKT. These results suggest that FKB and FKC may be helpful as a potential depigmenting agent for various hyper-pigmentary disorders.

Soybean (Glycine max), as one of the foods with high plant proteins, contains a large amount of bioactive compounds and known to be effective in cardiovascular disease and obesity as well as in improving skin condition. The purpose of this study was to investigate the anti-photoaging effects of soybean fermented with Lactobacillus Rhamnosus ($Bio-Peptone^{(R)}$) by assessment of cytotoxicity against UVB, collagen synthesis after UVB-irradiation, tyrosinase activity, and melanin synthesis. Results showed that $Bio-Peptone^{(R)}$ protected skin fibroblasts against UVB-induced cytotoxicity and increased type I collagen synthesis. Furthermore, $Bio-Peptone^{(R)}$ significantly inhibited tyrosinase activity and reduced melanin contents. This study suggests that $Bio-Peptone^{(R)}$ has protective effects against UVB-induced skin damage. Thus, it is concluded that $Bio-Peptone^{(R)}$ is able to prevent skin damage against UVB and thus acts as anti-photoaging materials by protecting skin forming wrinkles and skin pigments.

Melanin pigments are abundantly distributed in mammalian skin, hair, eyes, and nervous system. Under normal physiological conditions, melanin protects the skin against various environmental stresses and acts as a physiological redox buffer to maintain homeostasis. However, abnormal melanin accumulation results in various hyperpigmentation conditions, such as chloasma, freckles, senile lentigo, and inflammatory pigmentation. Tyrosinase, a copper-containing enzyme, plays an important role in the regulation of the melanin pigment biosynthetic pathway. Although several whitening agents based on tyrosinase inhibition have been developed, their side effects, such as allergies, DNA damage, mutagenesis, and cytotoxicity of melanocytes, limit their applications. In this study, we synthesized 4-chromanone derivatives (MHY compounds) and investigated their ability to inhibit tyrosinase activity. Of these compounds, (E)-3-(4-hydroxybenzylidene)chroman-4-one (MHY1294) more potently inhibited the enzymatic activity of tyrosinase (IC50 = 5.1±0.86 μM) than kojic acid (14.3±1.43 μM), a representative tyrosinase inhibitor. In addition, MHY1294 showed competitive inhibitory action at the catalytic site of tyrosinase and had greater binding affinity at this site than kojic acid. Furthermore, MHY1294 effectively inhibited α-melanocyte stimulating hormone (α-MSH)-induced melanin synthesis and intracellular tyrosinase activity in B16F10 melanoma cells. The results of the present study indicate that MHY1294 may be considered as a candidate pharmacological agent and cosmetic whitening ingredient.

This study was aimed to investigate skin whitening effects of Nomura's jellyfish (Nemopilema nomurai) hydrolyzed extracts (NHE). The extracts were prepared through the hydrolysis of N. nomurai using commercial proteolytic enzymes such as Protamex, Alcalase, Flavourzyme and Neutrase with optimum pHs (pH 5-8) at $55^{\circ}C$. Their whitening activities were examined from the inhibition of melanin synthesis using B16-F1 cell lines. Among the examined samples, Neutrase-treated extract (N-NHE) showed the most significant inhibition effect on melanin synthesis by 89.9% at a concentration of $100{\mu}g/mL$. This sample decreased the expression of tyrosinase and TRP-1 (tyrosinase related protein-1) proteins as well. These results suggested the potential application of NHE as whitening ingredients in cosmetic preparation.

Deficiency of collagen regeneration, denaturalization of fibers, increased melanin synthesis and reactive oxygen species are important factors inducing deterioration of healthy skin function. They induce freckles and decrease in skin elasticity. Vitamin C, vitamin A and their derivatives have been used as cosmetic ingredients for improvement of these problems but they have various problems. We have been developing the various derivatives of these ingredients. In this study, we investigated whitening effect of 3-o-cetyl-L-ascorbic acid (VCCE), a new vitamin C derivative. The VCCE inhibited melanogenesis in a dose dependent manner(44 % at $20\;{\mu}g/mL $) and tyrosinase expression. For 8 weeks, we also investigated skin brightening effect of VCCE on pigmented spots in UV-irradiated human skin. In results, VCCE showed a statistically significant skin whitening effect by mechanical and visual evaluation. Taken together, our findings suggest that the VCCE has potential benefits as an active ingredient for whitening cosmetics.

This study was performed to determine the whitening effect of organic solvent extracts from the centipede, Scolopendra subspinipes mutilans. We prepared different concentrations (50%, 70% and 100%) of ethanol, methanol, 100% ethyl acetate and water extracts. We tested melanin inhibitory effect and tyrosinase activity using B16/F10 melanoma cell. As a result, treatment of organic solvent extracts is decreased the biosynthesis of melanin and tyrosinase activity to 36 ~ 86%. Especially the 70% ethanol extracts was the most effective in B16/F10 melanoma cells. In the study on melanogenic protein expression, 70% ethanol extracts of Scolopendra subspinipes mutilans blocked glycosylation of tyrosinase. Therefore this result suggests that 70% ethanol extracts could be developed as skin whitening agents.

Ultraviolet radiation exposure is a major cause of extrinsic skin aging, which leads to skin hyperpigmentation. Loganin, a major iridoid glycoside obtained from Corni fructus, has anti-inflammatory, anti-diabetic, and neuroprotective effects. In this study, we investigated the mechanisms underlying the anti-melanogenic effects of loganin in B16F10 melanocytes treated with ${\alpha}$-melanocyte stimulating hormone (${\alpha}-MSH$) and 3-isobutyl-1-methylxanthine (IBMX). Anti-melanogenic activity was measured by treating cells with loganin at concentrations between 1 and $20{\mu}m$. Cell viability assays confirmed that doses of loganin up to $20{\mu}m$ were not cytotoxic. Loganin significantly and dose-dependently decreased intracellular melanin production. We also investigated potential molecular signaling pathways for the anti-melanogenesis effects of loganin. Western blotting showed that treatment with ${\alpha}-MSH$ and IBMX increased the phosphorylation of cAMP response element-binding protein (CREB) and the gene expressions of microphthalmia-associated transcription factor (MITF) and tyrosinase. Addition of loganin suppressed these increases, while promoting the phosphorylation of extracellular signal regulated kinase (ERK) and the anti-melanogenesis response. Our data therefore indicated that loganin could attenuate the increased melanin synthesis induced by ${\alpha}-MSH$ and IBMX treatment of B16F10 melanocytes. This attenuation appears to occur by downregulation of CREB phosphorylation and MITF and tyrosinase gene expression and upregulation of ERK phosphorylation. These finding suggests that loganin could be a valuable candidate for treatment of skin diseases related to hyperpigmentation.
