  • Forty eight fungal isolates were isolated from the maeius collected through the whole nation. Out of them, nine isolate were observed to be zygomyceteous fungi(4 genera and 9 species): Mucor hiemalis, M. circinelloides f. griseo-cyanus, M. circinelloides, M. jansseni, M. racemosus f. racemosus, Mortierella isabellina, Rhizopus stolonifer, R. oryzae, and Absidia corymbifera. Four species(M. circinelloides f. griseo-cyanus, M. hiemalis, M. jansseni, and R. oryzae) identified here were known as species involved in maeiu but not described in detail. The others were first reported as a species inhabiting in the maeiu. Two species of M. hiemalis f. hiemalis. and M. circinelloides f. griseo-cyanus were determined to play a role in biochemical changes of soybean in the fermentation of maeiu. However, the others seldom inhabited but considered to be comtaminated fungi in maeiu.

Various 14 species of Penicillium were isolated from maejus, but all of them were not considered to be involved in the maeju fermentation. Otherwise, S. brevicaulis was especially collected from the maejus of various located areas, but were also related to the good quality of maejus under labratory conditions. A. flavus, A. oryzae P. tulolense, and P. funiculosum were also observed to be involved in the process of maeju.

  • This study was conducted to investigate flavor components of sigumjang meju and barley bran. Among 52 kinds of flavor components identified in barley bran, the content of hexanal was the most followed by butanoic acid, 2-pentylfuran and hexanoic acid. Among 66 kinds of flavor components identified in 12 sigumjang mejus, the contents of 2-furancarboxaldehyde, 1-(3-methoxyphenyl)-ethanone and tetramethylpyrazine were more than those of any other components. Twenty two kinds of flavor components were detected in both barley bran and sigumjang meju. The contents of butanoic acid, hexanal and 2-pentylfuran were higher in barley bran than sigumjang meju. The contents of 2-furancarboxaldehyde, 1-(3-methoxyphenyl)-ethanone and tetramethylpyrazine were higher in sigumjang meju than barley bran. Thirty kinds of volatile components were detected only in barley bran and 44 kinds were detected only in sigumjang meju.

  • The radical scavenging activity of the methanol extracts was in the order of doenjang〉 meju〉 soybean. The metha-nol extracts were further fractionated with dichloromethane, ethyl acetate, butanol and water in sequence. Among the solvent fractions of the methanol extracts, the ethyl acetate fraction demonstrated high levels of radical scave-nging activity. On the other hands, the nitrite scavenging activity of the methanol extrats was in the order of doen-jang〉soybean〉meju. All fractions from doenjang were exhibited the strong nitrite scavenging effect. Especially, $H_2O$, EtOAc and BuOH fractions with 6gmg/10$m\ell$ concentration exhibited much stronger nitrite scavenging effect than that of L-ascorbic acid. The ethyl acetate fraction showed remarkable antioxidant and nitrite scavenging effects compared with the other fractions obtained from soybean, meju and doenjang. The ethyl acetate fraction of doenjang was fur-ther purified by repeated chromoatograpic methods and isolated three compounds. There are identified as genistin, dai-dzein and genistein on the basis of spectral data.

  • Based on the previous studies, R. stolonifer was selected for the model system of Korean soy sauce preparation process, using the fermented soybean. The system of meju fermentation was refined; the optimal conditions of temperature and relative humidity were $25^{\circ}C$ and 90%, respectively. The optimal temperature for soy sauce fermentation was evaluated to the $25^{\circ}C$ and, in the latter half of the fermentation, some aeration was observed to positively affect the sensory evaluation of soy sauce. In the sensory evaluation, the soy sauce fermented with B. subtilis and A. sojae was showed to be better than one fermented with R. stolonifer. Conditions of the pasteurization and ultrafiltration process were also established for industrialization of the soy sauce. Finally, the standard systems using the soybean fermented was, by the industrial scales, proposed for Korean traditional soy sauces.

  • In order to evaluate the migration of fungi into doenjang from its materials, meju and solar salt, microbial communities were analyzed using pyrosequencing. Dominant fungi of meju were Botrytis spp. (57.94%) and Dothiorella samentorum (24.08%). Unidentified fungal species (37.53%), unassigned species (32.60%) and several fungal species of small portion were identified in solar salt. In doenjang, Candida versatilis were predominantly detected (92.62%). Non-halophilic mold were dominantly identified from meju (low-salt fermented soybean), while halophilic bacteria and archaea for solar salt and salt-tolerance fungi such as C. versatilis for doenjang (high-salt fermented soybean) were frequently detected. These results implied that most predominant fungal species might not be migrated from meju and/or solar salt into doenjang.

  • This study was carried out to examine the quality characteristics of Doenjang manufactured with or without soaking Meju in brine according to salt concentrations (8 and 12%) during fermentation for 6 months. The moisture content and salinity of Doenjang fermented for 6 months were 54.9-60.3% and 7.8-12.5%, respectively. Doenjang using soaking Meju in brine had higher pH and lower titratable acidity than that using non-soaking Meju. The reducing sugar content in all samples was increased until 2 months and then decreased regardless of soaking Meju, especially that of non-soaking 8% Doenjang was the highest. The 8% low-salt Doenjang was shown the highest amino-type nitrogen content, especially the soaking Doenjang was higher than the non-soaking Doenjang. ${\alpha}$-amylase activity of all samples during fermentation were continuously decreased from 0.91-0.94 Unit/g to 0.01-0.06 Unit/g, especially the 8% soaking Doenjang was shown the highest activity after 2 months. Total bacterial count of the soaking Doenjang at the 6 months was in range of 7.8-8.0 log CFU/g and that of the non-soaking Doenjang was in range of 7.2-7.5 log CFU/g. By taste analysis, the 8% soaking Doenjang was shown the similar taste pattern with commercial Doenjang. In conclusion, the 8% low-salt Doenjang manufactured with soaking Meju in brine was a suitable concentration in order to reduce salt intake.

  • Whole soybean cereal was fermented with four fungal strains (Mucor and Rhizopus) in pilot meju fermentation system. The pH range of the fermented soybean cereal was $7.16{\sim}8.38$, the contents of reducing sugar and amino-nitrogen were $0.54{\sim}2.64%,\;93{\sim}312mg%$, respectively, and that of free fatty acid ranged $0.06{\sim}2.00%$. The components of the amino acid, organic acid, free sugars and fatty acid showed distinctive patterns among four groups of fermented soybean cereals. Amylase activity and carbohydrate degradation rate of R. oryzae, protease and protein degadation rate of R. stolonifer was higher than other strains. But lipase and lipid degradation rates of four strains were similar. The odor concentrates of the soybean cereals fermented with Mucor strains were similar to Aspergillus strains, but Rhizopus were possessed of the flavor components of Bacillus and Aspergillus. Soysauce, made from M. hiemalis and R. stolonifer fermented soybean cereal showed excellent sensory evaluation and it was proposed that the two strains will be useful in Korean soysauce process.
