Indoor Rearing Method of Diving Beetles: Cybister japonicus, Cybister tripunctatus orientalis, Cybister brevis (물방개류 실내 사육법)

  • The aim of this study is to develop indoor-rearing methods of the diving beetles. In nature, both the adult diving beetle and its larvae are voracious aquatic predators. The larvae beetles hunt relatively bigger size of tadpoles and small fish for food source. However, due to difficulties of the food supplement for rearing diving beetles at indoor-condition further motivated us to develop new artificial food. Three separate experiments were performed. In the first experiment, adult beetles were provided with one of the several food choice treatments to self-compose their preferred foods that are affordable on the market at lower price. The second experiment was also to develop artificial diet that is possible for rearing larvae beetle under indoor condition. The larvae beetles were restricted to raw squid, artificial food source and mosquito larvae as a control at the first stadium and small fish and raw squid during second to third stadium duration. According to our result, adult beetles selected a food that made of boiled squid and dead small fish while, the young larvae consumed small fish, mosquito larvae and raw squid. Although, the larval food restriction on law squid caused noticeable decrease in survival, the result still supported the possible survival rate of keeping larvae at indoor condition. Moreover, pupation rate experiments, in which groups of larvae were placed at different mats, natural soil and fermented sawdust, showed that 80% of diving beetles pupated on the sawdust. This result indicates that female beetle preferentially selected to oviposit along soft and moist area.

  • There has been a successful re-entry in the form of infant foods and as a product concept of "well-being" milk by feeding goat natural medicinal plants in high mountain lands and goats eating natural feeds. Typical composition of cow's milk and goat's milk are not significantly differ in major nutritional constituents. However, the noticeable differences between milks of the bovine and caprine species concern in the dimensions of the micelles, in casein composition, in size of the micelles and in the mineral charge of the micelle, but the ratio Ca/Pi in the micelle is very close for the two species The potential market in Korea could be expected to expand by keeping its freshness and nutritional benefits. The supply of goat milk products all year around is also an important to the consumers. In order to increase its market scale of goat milk, product manufacturers need extensive advertising promotion. Domestically, goat milk is currently manufactured at small scale dairy goat milk companies and consumed mainly in the form of fresh or fermented goat milk, while imported goat milk powder is used to produce infant goat milk formula by major dairy companies. Decreasing the unpleasant goaty flavour for the Korean consumers would be essential for the researchers who work for dairy science and technology.

  • 하천내에서 작은 물고기의 국지적인 회유 또는 이동 습성은 먹이를 구하고, 산란 및 성장과 집단에서 분산하고, 위험으로부터 도피하는 것에 밀접한 연관이 있다. 하천의 합류부와 연결부에 제방(levee), 도로(road) 등이 설치되는 경우 일반적으로 암거형태의 인공수로로 상 하류를 연결되는데, 암거내 수리특성상 평수시 저유량, 고유속, 저수심의 수리특성을 보인다. 이러한 수리특성은 물고기가 이동하는데 장애(barrier)가 되어 생태적으로 하천내 종다양성 감소와 일부 어종의 멸종을 초래한다. 어도 설치시 설계기준에서 가장 주목할 부분은 어도를 이용할 대상어종의 범위와 유영능력이다. 하천설계기준(2005, 한국수자원학회)에서 제시된 물고기의 유영능력은 대부분 국외 어종을 대상으로 외국에서 실험한 결과를 이용하고 있다. 그러므로 이러한 기준이 국내 어종에 대해 적합한지에 대한 검토가 필요하다. 본 실험의 목적은 국지회유성인 피라미와 각시붕어가 인공수로를 통과할 때의 유영행동 중 소상성공율과 유영속력을 평가하고자 함이다. 유영행동의 실험방법은 Brett(1967)에 의해 제안된 2가지 방법인 증진유속방법과 고정유속 방법중에서 물고기의 이동경로와 소상능력을 확인하기에 용이한 고정유속방법을 사용하였다. 또한, 효과적인 실험을 위해 실험어종(피라미 100마리, 각시붕어 100마리)이 실험 중에 최적의 건강상태를 유지할 수 있도록 물고기의 적응 기간(24시간)과 실험 중 휴식기간을 두었다. 실험결과 체형이 방추형(fujiform)인 피라미의 경우 평균유속 $0.7{\sim}0.8m/s$구간에서 소상성공율이 61%에서 26%로 급격하게 하락하는 현상이 나타났다. 그리고 측편형(compressiform)의 체형을 가진 각시붕어의 경우 평균유속 0.4m/s에서 소상성공율이 25%로 나타났으며 그 이상의 유속에서는 10%이하로 나타났다. 실험어종의 이동경로를 분석한 결과 대부분 수로바닥에 밀착하여 최저유속구간을 이용하여 이동하는 것으로 나타났으며, 이동경로의 지점유속은 평균 유속의 70%정도로 나타났다. 실제 하천에 설치된 인공수로가 생태통로의 기능을 유지하기 위해서는 평상시 수리특성이 대상어종의 유영능력을 초과하지 않도록 설계되어야 한다. 특히, 하천에 설치된 인공수로의 길이가 $5{\sim}10m$정도일 경우라면 피라미의 소상 가능성을 50% 이상 확보하기 위해서는 평수시 설계유속이 0.7m/s를 초과하지 않아야 할 것으로 판단된다.

무당개구리에서 내분비계장애물질에 의한 발생독성 평가

  • 내분비계 장애물질 (Endocrine Disruptors: EDs)이란 ‘내분비 기능에 변화를 일으켜, 생체 또는 그 자손의 건강에 위해한 영향을 미치는 외인성 물질’로서 세계야생생물보호기금 (World Wildife Fund: WWF)의 목록에는 67 여종, 일본 후생성에서는 142 여종의 물질을 내분비계 장애물질로 분류하고 있다. 이렇게 분류된 내분비계장애물질 가운데에는 많은 종류의 농약이 포함되어 있으며, 이들이 자연상태계에 미치는 영향에 대한 많은 보고가 있다. 양서류는 먹이연쇄의 중위포식자로 내분비계 장애물질의 순환 및 생채축적 회로에서 중요한 위치를 갖는다. 또한 농경지나 계류 등에 서식하며 산란하는 습성이 있다. 이러한 서식지는 농약 등에 노출받기 쉬우므로 환경오염평가 대상동물로 중요하다. 본 연구에서는 국내에서 사용되고 있는 제초제, 살충제 등이 양서류 발생에 미치는 영향을 평가하고, 이들 물질에 의한 노출을 평가할 수 있는 biomarker 유전자를 발굴하고자 하였다. 우리나라 전역에 많이 분포하는 무당개구리(Bombina orientalis)의 초기 배아발생 과정에서 이들 농약을 처리하였을 때, 물질의 종류에 따라 정도의 차이는 있으나 농도 의존적으로 배아 및 올챙이의 치사율이 높게 나타났다. 또한 농약은 배아 및 올챙이의 기형을 유발하였다. 기형의 종류는 농약에 따라 서로 상이한 형태로도 나타났으나, 특히 몸통휨 또는 꼬리휨등의 척추골 기형이 많이 나타났다. 환경 위해성 평가 biomarker 유전자로서 골격계 형성에 관여하는 Sox9 유전자를 선택하였다. 무당개구리에서 처음으로 Sox9 유전자를 동정하였으며, 농약을 처리한 실험군에서의 Sox9 유전자의 발현 양상은 대조군에 비하여 높게 나타났다. 본 연구결과 생태계에서 내분비계 장애물질의 위해성 평가를 위해 양서류 초기배아의 치사 및 기형의 정도를 관찰하는 방법이 유효할 것으로 사료된다. 또한 야생동물의 biomarker 유전자를 발굴과 이를 이용한 위해성 평가는 더욱 정밀한 평가법으로 유용할 것이다.

Studies on the Feeding Activity and Environmental Tolerance of Geoduck Clam, Panope japonica (코끼리조개의 섭식활동 환경내성에 관한 연구)

  • Experiments on the feeding activity and environmental tolerance of geoduck clam, Panope japonica caught at the coastal water of Kangwon province located in the East Sea of Korea were performed from January to December, 1994. Twenty genera and 33 species of plankton were found in the digestive tract, and most of them are phytoplankton. In spring, 21 species of planktonic foods were indentified, but in sumer, only 11 species were identified. Nitzschia longissima and Rizosolenia alata were present evey month. Weight of the digestive tract was fluctuate seasonally : higher winter and spring, and lower summer. Adult geoduck clams could be tolerated within the range of 5 to $20^{\circ}C$ water temperature, and they could tolerated considerably well temperature than higher temperature within the range.

Ecological Characteristics of Termite(Reticulitermes speratus kyushuensis) for Preservation of Wooden Cultural Heritage (목조문화재의 보존을 위한 한국산 흰개미의 생태적 특성 연구)

  • In this study, after analyzing several local climate characteristics of South Korea, I validated distribution, invasion, foraging, underground activities, attack season as ecological characteristics and also temperature, relative humidity, and tree species as preference characteristics of Korean termites (Reticulitermes speratus kyushuensis Morimoto). Especially, southern part of the Korean peninsula is a suitable area for inhabitation and motion of termites holding same ecological characteristic like R. speratus kyushuensis. Busan is a neighboring district at field distribution north limiting temperature of Coptotermes formosanus Shiraki and Chuncheon is a passing area through the Korean Peninsula of field distribution north limiting temperature of Reticulitermes speratus Kolbe. The termite attack of wood devices was about 34.5% for 3 years in the forest of Jongmyo. Although the attack rate of termite increased each year, the detection rate decreased and the missing rate was high by degrees. I confirmed a foraging habits which is a part of termite colony was a role of continuous decomposition and another was a role of new food hunt as experimental results. The foraging termites were found under ground at Jongmyo in Seoul from April to November in the 2001 and the most active period was on July and August. The termite invasion rate of bait station increased in every monitoring. Through the increasing attack rate of bait station during 2nd monitoring (November, 2000) and 3rd monitoring(March, 2001), I confirmed that termites moved into the deep underground in winter, and were working continuously to forage. R. speratus kyushuensis inhabiting at the Korean Peninsula is a species which has food consumption rate with higher temperature. The termite revealed the greatest amount of food(filter paper) at $30^{\circ}C$(90% RH), but showed increasing death rate at over $32^{\circ}C$. Also, survival rate of this termite was 97% at 84% RH($30^{\circ}C$), but killed 100% at 52% RH($30^{\circ}C$) and 70% RH($30^{\circ}C$). For wood feeding, this was observed the preference in a pine tree(Pinus densiflora) above all others. Survival of termites was high(87%) at a pine tree, but low(13.5%) at a paulownia tree(Paulownia coreana). In this study, I presented the biological characteristic of termite(R. speratus kyushuensis Morimoto) and confirmed the deterioration degree of termite on wooden cultural heritage in Korea. Depending on climate and soil temperature, each area in the southern part of the Korea Peninsula, has some different active period and different distribution of R. speratus kyushensis. With these results, I expect that this report helps to prepare the integrated pest management(IPM) of the termite on wooden cultural heritage in Korea, and it may help to reduce the economical loss from termite damage in Korea.

Early Life History and Spawning Behavior of Chinese Minnow, Rhynchocypris oxycephalus Reared in the Laboratory (실험실에서 사육한 버들치의 산란습성(産卵習性) 및 초기생활사(初期生活史))

  • This study was conducted to observe the spawning behavior and early life history of chinese minnow, Rhynchocypris oxycephalus reared in the laboratory. The spawning period of Rhynchocypris oxycephalus is from May to June in nature. The eggs of Rhynchocypris oxycephalus were spawned on the sand and surface of the gravel. The fertilization eggs were demersal in shape and adhesive, released as a clump forming a thin layer and their diameter were 1.70~1.90mm (mean 1.80 mm, n=20). Hatching of eggs was started in 88 hour 45 minute after fertilization at water temperature $19{\pm}0.5^{\circ}C$ and finished in 90 hour. Newly-hatched larvae were measured 4.87~5.02 mm in total length (TL, mean 4.94 mm), mouth and anus were not opened. 6 days old larvae were 6.32~6.56 mm in TL (mean 6.44 mm). Yolk sac was almost absorbed, mouth and anus was began to open. 13 days old larvae were 6.74~6.91 mm in TL (mean 6.82 mm). Part of Dorsal fin was began to rising and myomere number was 15+23=38. 25 days after hatching, total length of larvae was 8.45~8.60 mm (mean 8.52 mm). Dorsal and anal fin rays became differentiated, and also caudal part of the notocord flexion was achieved at $45^{\circ}C$. In the time, growth rate was higher than the other stage. Aggregate numbers of all fin rays were completed at 16.39~16.57 mm in TL (60 days after hatching), at which time the larvae reached the juvenile stage, but fin-fold on ventral was remained yet. External features of adult specimens were almost completed at 80 day old juveniles (18.69~18.87 mm in TL).

Analysis of Damages and Rice Consumption by Golden Apple Snails(Pomacea canaliculata: Ampullariidae) at Growth Stages of Rice (벼 생육시기별 왕우렁이의 벼 섭식량 및 피해해석)

  • The golden apple snail was introduced to the Korea from Japan as a potential food for people in 1983. It is one of the pests of direct-seeded rice in korea. This study was conducted to investigate rice(Oriza sativa) consumption and the extent of damage by the golden apple snail(Pomacea canaliculata) in direct-sown paddy fields. Food consumption of the golden apple snail was highest at $30^{\circ}C$. When the emergence of the 1st, 3rd, and 5th leave of rice supplied food, the consumption of young(shell height 15mm) and adult golden apple snail(over shell height 30mm) at $30^{\circ}C$ were 19.0/79.8, 11.0/54.5, 5.5/18.5, and 0.0/2.0 individuals, respectively. Young rice seedings(before 5th leaves of rice) are very susceptible to golden apple snail damage because the young, tender leaves and stems favor the snail's feeding habits. The field experiment with four treatments (0, 2, 5 and 10 individuals respectively per 10 $m^2$) was carried out. The most serious yield loss was 16% from the plot with 10 adults released at emergency stage of rice.

The Effect of Cone Protective Net and the Morphological Variation of Cone and Seed of Korea Rare Endemic Pinus pumila Regel (희귀수종 눈잣나무의 구과보호망 효과 및 구과와 종자의 형태적 변이)

  • This study was conducted to investigate the effect of cone protective net and the variation of cone and seed characteristics of a unique and rare dwarf stone pine (Pinus pumila Regel) population in Mt. Seorak which is in the southern peripheral of South Korea. Iron protective net (50 mesh, 25 ${\times}$ 25 cm) was effective in reducing birds and rodents damage to conelet, thereby it was possible to safely collect seed genetic resources. Early July was observed to be the most appropriate season to cover conelet with Iron protective net. P. pumila have ovoid-triangle shaped, wingless seeds and indehiscent cones, seeds dissemination is mainly by the hoarding behavior of nutcrackers and rodents. Statistical analysis showed that there were significant differences among individuals within population, and the mean characteristics of the species were 35.3 mm, 25.6 mm, 1.38, 6.6 g and 39.3 ea in the cone length, cone width, cone index, cone weight and in number of seeds per cone, respectively. Coefficients of variations in seed weight and number of seeds per cone were relatively high (21.7%, 21.5%, respectively) compared to other traits. Based on the correlation analysis between cone characteristics and seed characteristics, the length of cone and seed showed a significant positive correlation with the seed size and the seed weight, but the width and thickness of seed showed a negative correlation with the number of seeds per cone.

Feeding Habits of the Glass Eel Anguilla japonica Determined by C and N Stable Isotopes in the Nakdong River Estuary of the Korean Peninsula (안정동위원소를 이용한 낙동강 하구 실뱀장어의 먹이 습성 분석)

  • Wild glass eels found in the Korean peninsula are thought to migrate along the North Equatorial Current and undergo the processes of spawning and incubation in the Mariana Trench. Juveniles of the wild glass eels are collected from the southern and western coasts of the Korean peninsula and used as seeds for aquaculture. To investigate the feeding behavior of wild glass eels, we collected glass eels from the Nakdong River estuary during March and April 2014 and analyzed the total length, wet weight, dry weight, carbon and nitrogen contents, and stable isotope ratio of eels as well as water temperature and salinity. Water temperature in the Nakdong River estuary was $13.2{\pm}1.0$ (mean ${\pm}$ 1SE; range, $10.1{\sim}15.7)^{\circ}C$ and salinity was $24.8{\pm}2.4$ (13.2~34.0), showed a lower range from 13.2 to 30.0 (a mean of 21.2) when the floodgates were opened. The total length of glass eels was $56.5{\pm}0.2$ (51.0~63.6) mm, wet weight was $70.9{\pm}1.4$ (33.6~133.2) mg, and dry weight was $16.5{\pm}0.3$ (10.1~29.1) mg. Carbon and nitrogen contents of glass eels were $51.0{\pm}0.8%$ and $13.9{\pm}0.1%$, respectively. Mean ${\delta}^{13}C$ and ${\delta}^{15}N$ values of glass eels collected from the Nakdong River estuary were -20.9±0.2‰ and 6.1±0.1‰, displaying similar values to those of leptocephalus, glass eel larvae collected from the North Equatorial Current. Therefore, this result suggest that the glass eels collected from the Nakdong River estuary do not feed on prey after metamorphosis from eel larvae to glass eels their migration.
