Cucurbitacin-I, a natural triterpenoid derived from Cucurbitaceae family plants, exhibits a number of potentially useful pharmacological and biological activities. Indeed, the previous study demonstrated that cucurbitacin-I reduced the proliferation of colon cancer cells by enhancing apoptosis and causing cell cycle arrest at the G2/M phase. CD44, a type I transmembrane protein with the function of adhering to cells, mediates between the extracellular matrix and other cells through hyaluronic acid. Recent studies have demonstrated that an overexpression of the CD44 membrane receptor results in tumor initiation and growth, specific behaviors of cancer stem cells, the development of drug resistance, and metastasis. The aim was to examine the effect of cucurbitacin-I on CD44 expression human ovarian cancer cells because the effect of cucurbitacin-I on CD44 expression has not been reported. The expressions of CD44 mRNA and protein were detected using a quantitative real-time reverse-transcription polymerase chain reaction and a Western blot analysis, respectively. Treatment with cucurbitacin-I inhibited the expression of CD44 mRNA and protein. A subsequent analysis revealed that cucurbitacin-I blocked the phosphorylation of activator protein-1 (AP-1) and nuclear factor kappa-B ($NF-{\kappa}B$), which are key regulators of CD44 expression. Taken together, the data demonstrate that cucurbitacin-I regulates the AP-1 and $NF-{\kappa}B$ signaling pathways, leading to decreased CD44 expression.

The elution behaviors of Ni(II), Cu(II), Co(II), and Cr(II) in 1-phenyl-3-methyl-4(2-hydroxy-5-X-phenylazo)-5-pyrazolone, [Pm(2-OH)(5-X)PaPz](X=H, $CH_3$, $NO_2$, Cl) chelates have been studied by reversed phase HPLC. Thirteen metal-[Pm(2-OH)(5-X)PaPz] chelates were prepared and characterized by UV, IR, MS, and ICP spectroscopic methods. These metal-2-hydroxyarylazopyrazolone chelates were successfully separated on Novapak-$C_{18}$ column using methanol/water mixtures as a mobile phases. It was found that the chelates were eluted properly in an acceptable range of the capacity factor value($0{\leq}log\;k^{\prime}{\leq}1$). The dependence of the capacity factor(log k') on the volume fraction of water in the binary mobile phase showed a good linearity. Also, there was a good linear dependence of the capacity factor on the liquid-liquid extraction distribution ratio($D_c$) in methanol-water/n-pentadecane extraction system by the batch method. It suggested that the retention of the chelates in the reverse phase liquid chromatographic system be largely due to the solvophobic effect.
This review investigated theoretical principals and practical application examples on recirculation system of soil washing-wastewater treatment-treated water recycling. As for technologies which have attempted to remediating metals-contaminated soil in and around country, there are reactive barriers, encapsulation, solidification/stabilization, soil washing, and phytoremediation. Among those, in particular, this review covers soil washing technology which physicochemically removes contaminants from soils. The major drawbacks of this technology are to generate a large amount of wastewater which contains contaminants complexed with ligands of washing solution and needs additional treatment process. To solve these problems, many chemical treatment methods have been developed as follows: precipitation/coprecipitation, membrane filtration, adsorption treatment, ion exchange, and electrokinetic treatment. In the last part of the review, recent research and field application cases on soil washing wastewater treatment and recycling were introduced. Based on these integrated technologies, it could be achieved to solve the problem of soil washing wastewater and to enhance cost effective process by reducing total water resources use in soil washing process.
The hypothalamo-pituitary-gonadal hormone axis is centrally controlled by a complex regulatory network of excitatory and inhibitory signals, that is dormant during infantile and juvenile periods and activated at puberty. The kisspeptins are the peptide products of the KiSS-1 gene and the endogenous agonists for the G protein-coupled receptor 54(GPR54). Although KiSS-1 was initially discovered as a metastasis suppressor gene, a recent evidence suggests the KiSS-1/GPR54 system is a key regulator of the reproductive system. Yet the actual role of the KiSS-1/GPR54 system in the neuroendocrine control of gonadotropin secretion remains largely unexplored, the system could be the first missing link in the reproductive hormonal axis. Central or peripheral administration of kisspeptin stimulates the hypothalamic-pituitary-gonadal axis, increasing circulating gonadotropin levels in rodents, sheep, monkey and human models. These effects appear likely to be mediated via the hypothalamic GnRH neuron system, although kisspeptins may have direct effects on the anterior pituitary gland. The loss of function mutations of the GPR54(GPR54-/-) have been associated with lack of puberty onset and idiopathic hypogonadotropic hypogonadism(IHH). So kisspeptin infusion may provide a novel mechanism for HPG axis manipulation in disorders of the reproductive system.
Honokiol is a small molecular weight ligand originally isolated from the Chinese medicinal herb Magnolia officinalis, a plant used in traditional Chinese and Japanese medicine [9]. In a previous study, the effects of honokiol were shown to have anti-angiogenic, anti-invasive and anti-proliferative activities in a variety of cancers [1,3,4,11,13,17,24,29,30]. We showed previously that direct production of nitric oxide (NO) by treatment of NO donor, sodium nitroprusside (SNP), led to apoptosis in rabbit articular chondrocytes [15,16]. This study confirmed that NO-induced apoptosis was suppressed by honokiol treatment in a dose-dependent manner as determined by cell phenotype, MTT assay, Western blot analysis and FACS analysis in articular chondrocytes. Treatment of honokiol inhibited SNP-induced expression of p53 as well as DNA fragmentation in articular chondrocytes, but increased expressionof pro-caspase-3. Inhibition of SNP-induced apoptosis by honokiol treatment was rescued by LY294002, the specific inhibitors of phosphoinositide 3-kinase (PI-3K) in articular chondrocytes. Our results indicate that honokiol inhibits NO-induced apoptosis via PI-3K/AKT pathway in rabbit articular chondrocytes.

본 고에서는 한국통신(Korea Telecom) IMT-2000 시험시스템(이하: Trial system 라고 함) 단말기용 전력증폭단으로 적용하기 위한 다단구동증폭기 및 전력증폭기를 GaAs MMIC로 설계 구현하는 기술을 제시하였다. 설계된 구동증폭기는 3단으로구성하여 RF(Radia Frequency) 송신신호(1955$\pm$70MHz)대역에서 2단 (중간단)의 이득 조정범위가 40 dB이상이 될 수 있도록 능동부품인 MESFET를 Cascade 형으로 구성하고 MESFET의 게이트(gate)에 조정전압을 인가하는 증폭기를 설계하여 GaAs MMIC화 1 침(크기4$\times$5 mm)으로 제작하였다. 아울러, 본 논문에서는 제시한 구동증폭기는 동작주파수 대역폭 범위기 3.5배이고 출력전력은 15dBmm 이며, 출력전력이득이 25~27dB이고 반사계수는 -15~20dB이며 이득평탄도 3dB(동작주파수 대역폭내)로써 Trial system용 단말기의 최종단인 전력증폭단의 출력단 특성을 효과적으로 나타내었다. 그리고, 전력 증폭기는 2개의 입력단에서 출력되는 신호를 분배하는 전력분배기와 병렬구조인 4개의 증폭단에서 출력되는 출력신호를 외부에서 접속하는 전력결합기를 접소하여 구성하였으며 RF(Radio Frequency) 주파수(1955 $\pm$70NHz)에서 대역폭을 4배로 설계하여 광대역인 대역폭을 구현하였고 출력전력은 570mW이며, 출력부가효율(PAE; Power Added Efficency)가 -15$\pm$20dB이고, 이득 평탄도(Gain flatness)는 동작주파수 대역내에서 0.5dB이며 입출력 전압정재파비(Input & Output VSWR)가 13이하인 고출력 전력증포기를 GaAs MMIC화 1칩 (크기; 3$\times$4mm)으로 제작하였다.의 다양성이나 편리성으로 변화하는 것이 국적을 바꾸는 것보다 어려운 시 대가 멀지 않은 미래에 도래할 것이다. 신세기 통신 과 SK 텔레콤에는 현재 1,300만명이 넘 는 고객이 있으며. 이들 고객은 어 이상 음성통화 중심의 이동전화 고객이 아니라 신세기 통신과 SK텔레콤이 함께 구축해 나갈 거대란 무선 네트워크 사회에서 정보화 시대를 살아 갈 회원들이다. This study has been carried out to examine the feasibility of washing technique for reducing the heavy metal contamination level of tailing wastes and agricultural soil surrounding abandoned metal mines. Some organic acids with low molecular weight were used as washing solution. Initial contamination levels of copper and lead for some soil samples were found to exceed the standard levels of countermeasure and concern, and those of cadmium to approach the standard level of countermeasure. Experimental results using sequential extraction method revealed that more than half of copper and lead existing in tailing wastes are adsorbed forms available for plants. There are some proportional relationships between metal concentrations determined by using 0.1N HCI solution and those determined by sequential extractions. Citric acid was turned out to be superior to oxalic acid and acetic acid with low molecular weight in washing above three metals. When citric acid is used for washing heavy metals from soil, it is desirable to operate at pH less than 5.5 for better washing effect. Metal removal effect by citric acid solution has been proved to depend upon solution concentration and the mass ratio of solution to soil. Addition of SDS(Sodium Dodecyl Sulfate) to citric acid improved the washing effect of cadmium among three metal most significantly. while copper removal did not change. Washing technique using citric acid for removal of heavy metals from agricultural soil or tailing wastes is recognized to be an effective remediation method.
For the development of basic genetic materials for specific and effective therapeutic approach to suppress multiplication of hepatitis C virus (HCV), HCV internal ribosome entry site (IRES)-targeting hammerhead ribozyme which activity is allosterically regulated by HCV regulatory protein, NS5B RNA replicase, was developed. The ribozyme targeted most effectively to +382 nucleotide (nt) site of HCV IRES RNA. The allosteric ribozyme was designed to be composed of sequence of RNA aptamer to HCV NS5B, communication module sequence which can transfer structural transition for inducing ribozyme activity upon binding NS5B to the aptamer, and sequence of ribozyme targeting +382 nt of HCV IRES. Noticeably, we employed in vitro selection technology to identify the most appropriate communication module sequence which can induce ribozyme activity depending on the US5B protein. We demonstrated that the ribozyme was nonfunctional either in the absence of any proteins or in the presence of control bovine serum albumin. In sharp contrast, the allosteric ribozyme can induce activity of cleavage reaction with HCV IRES RNA in the presence of the HCV NS5B protein. This allosteric ribozyme can be used as lead compound for specific and effective anti-HCV agent, tool for highthroughput screening to isolate lead chemicals for HCV therapeutics, and ligand for biosensor system for HCV diagnosis.

A new analytical luminescence method for the Eu+3 and Tb+3 ions was studied using the luminescence enhancement by the treatment of the o-phenanthroline on the nylon membrane. Compared to the specific emission intensities of the ions in aqueous(or ethanol) solution, if the aqueous ion is spotted on the nylon membrane, the luminescence intensities were extremely enhanced. There was additional enhancement effect of the luminescence intensities of the ions on the nylon membrane, if the ion on the nylon membrane is treated with o-phenanthroline. Based on the luminescence enhancement, the detection limits were lowered by more than 7 order of magnitude compared to that of solution sample, and also lowered by about 1 order of magnitude compared to that of previous TLC method. The dynamic ranges and correlation coefficients of the calibration curves near the detection limit were 2∼3 order and ∼0.99, respectively. It was also shown that the luminescence intensity was in its maximum when the ion on the nylon is treated with ∼4 mole ratio of o-phenanthroline. The energy-transfer mechanism was explained for the theoretical background of the luminescence enhancement.
Screen printed carbon electrodes (SPCEs) with immobilized osmium-based hydrogel redox polymer, uricase and PEGDGE can be used to apply uric acid electrochemical detecting. The osmium redox complexes were synthesized by the coordinating pyridine group having different functional group at 4-position with osmium compounds. The synthesized poly-osmium hydrogel complexes are described as PAA-PVI-$[Os(dCl-bpy)_2Cl]^{+/2+}$, PAA-PVI-$[Os(dme-bpy)_2Cl]^{+/2+}$, PAA-PVI-$[Os(dmo-bpy)_2Cl]^{+/2+}$. The different concentrations of uric acid were measured by cyclic voltammetry technique using enzyme-immobilized SPCEs. The prepared SPCEs using PAA-PVI-$[Os(dme-bpy)_2Cl]^{+/2+}$ showed no interference from common physiologic interferents such as ascorbic acid (AA) or glucose. The resulting electrical currents at 0.33 V vs. Ag/AgCl displayed a good linear response with uric acid concentrations from 1.0 to 5.0 mM. Therefore, this approach allowed the development of a simple, point of care in the medical field, disposable electrochemical uric acid biosensor.
