Effect of feeding diet containing Cordyceps with fly pupa on blood lipid, fatty acid and TBARS in broiler chickens (브로일러의 혈액지질, 닭고기의 지방산 및 지방산패도에 관한 파리번데기 동충하초 급여효과)

  • The objective of this study was to determine the feeding effect of a diet containing Cordyceps with fly pupa on the changes in blood lipid profile from broiler chickens, fatty acids and thiobarbituric acid reactive substances (TBARS) in chicken meat. A total of 360 chicks (Ross strain 308) were divided on hatch day into four treated groups with respect to a 35-day feeding regimen: T1 (control group), T2 (2.0% Cordyceps with fly pupa), T3 (3.5% Corceps with fly pupa) and T3 (5.0% Cordyceps with fly pupa). Blood triacylglyceride and total cholesterol level was significantly decreased by 5.32-10.63% and 9.23-12.62%, respectively, in groups T2, T3 and T4 when compared to T1 (p<0.05). Water holding capacity was significantly highest in T2 (p<0.05), while there were no significant differences among groups T2, T3 and T. In chicken meat, the ratio of saturated fatty acid to unsaturated fatty acid was high in the T3 and T4 groups, the ratio of n-6 to n-3 fatty acid was low in the T2, T3 and T4 groups and oleic acid (18:1n-9) was high in the T2, T3 and T4 groups, when compared to T1 (p<0.05). TBARS tended to increase according to the storage time (in days), and was significantly lower in the chicken thigh muscles with skin in groups T2, T3 and T4 as compared to T1 (p<0.05). These results suggest that a diet containing 2.0-3.5% of Cordyceps with fly pupa may decrease blood lipid and improve both the shelf-life and quality of broiler chicken meat.

Effect of Cordyceps Militaris on Lipid Metabolism in Rats Fed Cholesterol Diet (동충하초(Cordyceps Militaris)가 고콜레스테롤혈증 흰쥐의 지질대사에 미치는 영향)

  • The effects of Cordyceps militaris on the weight gain, the food intake, the food efficient ratio, and the lipid concentrations of serum, liver, pancreas, heart and aorta in male rats fed cholesterol diet were studied. Male Sprague-Dawley rats were received 4 types of diets for 4 weeks, respectively : a normal diet(without cholesterol), a control diet(hypercholesterolemic diet), a hypercholesterolemic diet supplemented with 3% fruiting body of Cordyceps militaris(CF), a hypercholesterolemic diet supplemented with 3% mycelium of Cordyceps militaris(CM). The body weight gain of rats fed the control diet were significantly increased compared to those of the rats fed diets with CF or CM. But, the hepatic and pancreatic weights of rats fed diets with CF or CM were heavier than those of rats fed the normal diet. In serum of rats fed the CM diet, the concentrations of total cholesterol, triglyceride, LDL-cholesterol and the atherogenic index were significantly lower than those of rats fed the control diet. Also, serum HDL-cholesterol concentration and HDL-cholesterol/total cholesterol ratio were significantly lower in rats fed the control, CF or CM diets than those in rats fed the normal diet. Whereas, the hepatic concentrations of the total lipid, the total cholesterol and the triglyceride were significantly higher in rats fed 3 hypercholesterolemic diets than those in the rats fed normal diet. The heart and pancreatic concentrations of total cholesterol were the highest in the rats fed diet with the CR among rats fed experimental diets. In conclusion, the CM feeding decreased the concentrations of the cholesterol, triglyceride, and LDL-cholesterol and the atherogenic index in the serum of the rats. But the CF or CM feeding could not decrease the hepatic concentrations of the total lipid, cholesterol, triglyceride in the rats fed hypercholesterolemic diets.   • Objective : The aim of this study was to investigate the effect of CMM oral administration and herbal acupuncture at BL13 LU4 on Asthma induced by ovalbumin in rats Methods : Several experimental items were measured and compared each other ; that is body weight change, oxygen consumption rate, Albumin change, white blood corpuscle change, Erythrocyte change, IgE change. Results : 1. Body weight was significantly increased in herbal acupuncture group after its administration while there was no such a response in oral administration group. 2. Oxygen Consumption rate was significantly increased in oral administration group and herbal acupuncture group. 3. Albumin in serum was significantly increased in oral administration group after its administration while there was no such a response in herbal acupuncture group. 4. White blood corpuscle in blood was significantly increased in oral administration group after its administration while there was no such a response in herbal acupuncture group. 5. Erythrocyte in blood was significantly increased in oral administration group after its administration while there was no such a response in herbal acupuncture group. 6. IgE in blood was decreased in oral administration group and herbal acupuncture group but there was no significance in oral administration group and herbal acupuncture group. Conclusion : Based on the above results it is assumed that CMM oral administration and CMM herbal acupuncture can help the treatment of Asthma induced by ovalbumin in rats.

Morphological Changes by Submerged Culture Conditions for the Mycelial Optimal Growth of Cordyceps sinensis and Immunological Properties of Hot Water Extract of Mycelium (동충하초 균사체 최적 성장을 위한 심부배양 조건에 따른 형태학적 변화 및 균사체 열수 추출물의 면역학적 특성)

  • The mycelial dispersed growth of Cordyceps sinensis was optimized in submerged batch culture at initial pH of 5.0, 150 rpm, and $25^{\circ}C$. The morphological data showed much more dispersed growth of C. sinenesis at initial pH of 5.0. Also, projected area, main hyphal length and number of tips for the mycelial growth of initial pH 5.0 were higher than those of other initial pHs. The industrial medium for mycelial production of C. sinensis was determined to be molasses of 100 g and crushed brewery yeast of 10 g per liter as carbon and nitrogen sources, respectively. With these culture conditions, the maximum production of mycelia was approximately 30.0 g per liter by batch culture in 5-liter jar fermenter with no controlled pH. This result suggests that large-scale mycelia production of C. sinensis may be possible in submerged batch culture. The hot water extract of mycelia from C. sinensis was mainly composed of 83.0% carbohydrate, 11.8% protein, 1.9% lipid, and 2.4% ash and there were present glucose, mannose, galactose, and arabinose as molar ratio of 8.79 : 2.59 : 1.34 : 1.0 in the carbohydrate, respectively. In the experiment using spleen cell and macrophage, the extract showed potent mitogenic and immuno-stimulating activities and among various components, an important factor that contribute to the immunological activities was turned out to be carbohydrate moiety.

Effects of Cordyceps militaris on Key Enzymes of Carbohydrate Metabolism (Cordyceps militaris 성분이 당대사 관련 효소 활성에 미치는 영향)

  • The present study was designed to investigate the antidiabetic effect of Cordyceps militaris on activities of hepatic glucose-regulating enzymes, such as glucokinase (GK), pyruvate dehydrogenase (PDH) and acetyI-CoA carboxylase (ACC). We have isolated the active compounds, CM-A and CM-B from C. militaris and the extracts are under investigation to determine their mechanism of action. Hepatic GK, PDH and ACC activities were significantly (p < 0.05) increased compared with the control. Treatment with CM-A led to a rise in percentage of enzymes by 380$\%$,396$\%$ and 286$\%$, respectively, relative to control levels. CM-B were 329$\%$, 312$\%$ and 239$\%$, respectively. The increase in GK and ACC activities was linearly proportional with increased ratio of CM-A. Our findings suggest that C. militaris exerted antihyperglycemic potency, which is thought to be mediated through activation of GK, PDH and ACC enzymes related to glucose metabolism directly or indirectly and therefore C. militaris is promising as antidiabetic functional foods.

Genotoxicity Study of Water Extract of Cordyceps militaris Grown Upon Protuetja dreujtarsis (굼벵이 유래 밀리타리스 동충하초 열수 추출물의 유전독성평가)

  • Water extract of Cordyceps militaris grown upon Protuetja dreujtarsis (CMPD) was examined for the genetic toxicity-bacterial mutagenicity, chromosome aberration, and micronucleus formation. For mutagenicity assay, bacterial reversion test with Salmonella typhimurium TA98, TA100, TA1535, TA 1537, and E. coli WP2uvrA were performed. The extract at the concentrations of $50{\sim}5,000{\mu}g/plate$ did not induce mutagenicity at all. Chromosome aberration test was performed by using Chinese lung (CHL) cells. There was no significant chromosome aberration in CHL cells with S-9 mixture at the concentrations of $312.5{\sim}1,250{\mu}g/ml$ of the extract and without S-9 mixture at the concentrations of $1.2{\sim}19.5{\mu}g/ml$ of the extract. For micronucleus test, ICR mice were treated with the extract at the dose of 0.5, 1, and 2g/Kg. The frequencies of the micronucleated polychromatic erythrocytes (MNPCE) in bone marrow preparations of the extract-treated group were not increased compared to the untreated control group. Taken together, our results show that water extract of CMPD did not induce any harmful genotoxicity.

Effects of Extracts from Mixed Culture with Tricholoma Matsutake Mycelium and Cordyceps Militaris Mycelium on Blood Glucose in Streptozotocin-Induced Diabetic Rats (송이버섯과 동충하초 균사체 혼합배양 추출물의 투여가 Streptozotocin으로 유발한 당뇨 쥐의 혈당에 미치는 영향)

  • This study was performed to investigate the influence of extracts from mixed culture with Tricholoma matsutake mycelium and Cordyceps militaris mycelium on hypoglycemia in streptozotocin-induced diabetic rats. Experimental animals were divided into 7 groups : normal control group(NC), diabetes control group (DC), positive control group(PC), non-fermented OCM(oriental medicine & cereal medium) extracts group (UM), crude polysaccharide of non-fermented OCM extracts group (UME), fermented OCM extracts group (UF), crude polysaccharide of fermented OCM extracts group (UFE). NC, DC groups were orally administered saline, PC group was orally administered acarbose. UM, UME, UF, UFE groups were orally administered each extract once a day for 14days. Blood glucose level was lower in the all administering OCM extract groups (UM, UME, UF, UFE) than in the diabetes group(p<0.05), and specially UF, UFE groups were similar to tendency of PC group. ALT, ALP activity in OCM groups were not significantly lowered than PC group(p<0.05). AST activity was not different with PC group. The results of this study show that extracts from mixed culture with Tricholoma matsutake mycelium and Cordyceps militaris mycelium may have a beneficial effect on the hypoglycemia.

  • The fruiting bodies of Isaria japonica have been traditionally used in Korea to treat cancer. An apoptosis-inducing compound, 4-Acetyl-12, 13-epoxyl -9-trichothecene-3, I5-diol (AETD), was isolated from the methanol extract of fruiting bodies of Isaria japonica Yasuda by bioassay -guided fractionation. The apoptosis of murine bladder cancer cell line (NBT-Ⅱ) by the compound was accessed by propidium iodide staining flow cytometric analysis, and apoptosis-inducing activity at $IC_{50}$ concentration (5 nmol/L) was further confirmed by a nuclear morphological change, a ladder pattern of DNA fragmentation, and an activation of caspase-3. These results indicate that AETD induces apoptosis of NBT-Ⅱ cells via expression of caspase-3.

Physiological Activity of Extracts from Mixed Culture of Medical Herbs and Mycelia of Tricholoma matsutake and Cordyceps militaris by Fermentation (송이버섯과 동충하초 균사체를 혼합 배양한 한방추출물의 발효에 의한 생리활성)

  • Objectives : The purpose of this study was to investigate extracts from mixed culture of Oriental medicines and cereal medium and mycelia of Trichloma matsutake and Cordyceps militaris by fermentation to develop new material for pharmaceutical products and medicinal food, Methods : To evaluate physiological activities of OCM extracts, we examined antioxidant activity(total polyphenol contents, electronic donating ability, SOD-like activity), ${\beta}$-glucan contents, nitric oxide production and cytotoxicity by MTT assay. Results : Total polyphenol contents of fermented OCM(UF) and non-fermented OCM(UM) extracts were more than 40% UM and UF of DPPH radical scavenging activity was 25.67%, 23.43% respectively. Total polyphenol content of non-fermented extract (UME) was 12.57%, while that of fermented extract(UFE) was 7.05%. SOD like activity showed UM 85.35%, UF 76.18%, UME 58.42%, UFE 72.21%. UME, and UFE 31.43%, ${\beta}$-glucan contents of UME and UFE were more than 40%. NO productions of UME, and UFE showed a LPS dose dependent tendency. Cytotoxicity on Raw 264.7 cell showed more than 90% viability. Inhibitory effect of UFE on HT1080 cell growth was higher than UME. Conclusions : These results showed that extracts from mixed culture of mushroom mycelium and OCM have physiological activities which can be used in pharmaceutical products and medicinal food.

A Study on the Quality Characteristics of Doenjang Prepared with Paecilomyces japonica, from Silkworm (누에 동충하초(Paecilomyces japonica)를 첨가하여 제조한 된장의 품질특성 변화에 관한 연구)

  • Change in Quality Properties of Doenjang Prepared with the Powder of Paecilomyces japonica and extract of P. japonica by different solvents were investigated during 90 days of fermentation at 20$^{\circ}C$. The moisture content was not significantly different, pH of Doenjang made with P. japonica was lower than control group and decreased continuously according to the fermentation time. Amino nitrogen continuously increased till 60 days and decreased slightly on 90 days. Value of L, a, b in Color decreased with proportion to fermentation period and L, a, b value of Doenjang made with P. japonica was lower and in Particular that of Doenjang made with P. japonica powder was lowest. From the result of sensory evaluation test, the color of control group was similar to “yellow”but that of Doenjang made with powder of P. japonica was close to“dark brown”and that of Doenjang made with extract of P. japonica was darker than that of control group and the preference of dark color was low. Texture was“glossy and smooth”in all and preference was high. In salty taste, Doenjang by P. japonica addition was stronger and Doenjang made with extract was stronger than that made with powder. Doenjang made with powder of P. japonica was weaker than other groups in sweet taste. In flavor and overall Preference, Doenjang made with P. japonica was scored lower slightly in than control group and Doenjang made with powder of P. japonica was the lowest in score.
