Optimization of the process conditions for the emulsification of rice bran oil using response surface methodology (반응표면분석법을 이용한 현미유 유화 제조공정 최적화)

  • In this study, the optimization of the manufacturing process for the oil-in-water emulsification of rice bran oil was performed by response surface methodology (RSM) using varying amounts of the emulsifier (0.05-0.25%), varying rotation speeds of the homomixer (4,000-8,000 rpm), and varying water to oil ratios (0.8-1.6%) as independent variables, and the emulsion stability index (ESI) as the dependent variable. The optimization conditions predicted by the RSM model were 0.2%g of the total amount of the rice bran oil emulsion, emulsified at the homomixer rotation speed of 6,700 rpm using a water to oil ratio of 1:3. The ESI of the rice bran oil emulsion prepared under the optimized conditions was 95.7%, which was similar to the predicted value of 94.4% obtained by the RSM model. The transmission stability and the backscattering values were found to agree with each other over time and the turbiscan stability index was less than 0.7, indicating that the aggregation and upper floatation were less while the dispersion stability was maintained.

Cytotoxic Effect of Isolated Protein-bound Polysaccharides from Hypsizigus marmoreus Extracts by Response Surface Methodology (반응표면분석에 의한 해송이버섯(Hypsizigus marmoreus) 추출물 중 단백다당체의 암세포 성장억제효과)

  • This study used response surface methodology (RSM) in an effort to optimize the water extraction conditions of Hypsizigus marmoreus in order to increase cytotoxicity activity of the extract. A central composite design was applied to investigate the effects of independent variables, which included the extraction temperature ($X_1$), extraction time ($X_2$), the ratio of solvent to sample ($X_3$) on dependent variables of the extracts, including extraction yield ($Y_1$) and protein content ($Y_2$). The estimated optimal conditions were as follows: $51.3^{\circ}C$ extraction temperature, 8.2 hrs extraction time, and 46.7 mL/g of solvent per sample. The extract (CE) was extracted at optimal condition and crude polysaccharides (CPS) were obtained from CE by ethanol precipitation, dialysis, and freeze drying. Neutral (NPS) and acidic (APS) fraction of polysaccharides were seperated from CPS by ion chromatography. The growth inhibitory effects of the APS (0.5 mg/mL) on AGS human cancer cells were 73.97%. CPS showed the highest growth inhibitory effects on HepG2 human cancer cell at 0.5 mg/mL. However all fraction polysaccharides from Hypsizigus marmoreus showed lower than 20% growth inhibition on SW480 human cancer cell.

Development of an Onion Vinegar Beverage Containing Yuza (Citrus junos Sieb ex Tanaka) and Its Biological Activity (유자 첨가 양파발효음료의 제조 및 생리활성)

  • Onion vinegar has an undesirable flavor and taste that results from alcohol and acetic acid production from fermentation. In this study, we have used onion vinegar to develop an onion vinegar beverage with better sensory quality. The objective of this study was to determine the optimum blending ratio by using response surface methods to produce an onion vinegar beverage containing Yuza (Citrus junos Sieb ex Tanaka). The optimal formula for a fermented onion beverage was determined using a central composite design by the response surface methodology. The independent variables were obtained by regression analysis of the reaction surface of brown sugar, apple extracts and Yuza extracts. The optimum mixing ratio for onion vinegar:water:brown sugar:apple extracts:Yuza extracts was 6.0:77.6:4.9:9.2:2.3 (w/w). The actual overall acceptance was 7.08 under optimum conditions, which was close to the maximum predicted value of 6.96. The concentration of phenolic compounds, total flavonoids, and quercetin present in the onion vinegar beverage was 14.8 mg/100 g, 2.6 mg/100 g and 1.4 mg/100 g, respectively. The onion vinegar beverage showed antimicrobial activity against Staphylococcus aureus, Escherichia coli, Salmonella typhimurium and Enterobacter aerogenes. It also showed antioxidant effects, with a DPPH radical inhibition rate of 18.2% and superoxide dismutase (SOD)-like activity of 11.5%. In conclusion, the onion vinegar beverage described here seems to have nutritional value and potential biological activity.

Preparation and characterization of the primary gas standards for isoprene (아이소프렌 일차표준가스의 제조 및 특성 평가)

  • Isoprene is a one of the biogenic volatile organic compounds (BVOCs) and it is known as a source of the tropospheric ozone and formaldehyde. In addition, isoprene is a trace component of the exhaled breath and it is a potential biomarker for the diagnosis of diseases such as lung cancer. In these regards, isoprene gas standards are required for the accurate measurement of isoprene in air samples. To establish a standard for isoprene gas, gravimetric preparation and characterization of primary gas standards were studied. The primary gas standards were produced independently in 4 aluminum cylinders and concentrations were examined by GC-FID. As a result, the uncertainty of the gravimetric preparations including purity of the raw material was 0.01% and reproducibility of the preparation of independent 4 cylinders was 0.08%. The primary gas standards for isoprene showed 14 months of long-term stability. The relative expended uncertainty of 2.8% (95% of confidence level, k=1.96) was assigned to the certified value of 10 ${\mu}mol$/mol level of isoprene based on the quantitative evaluation of the purity, weighing, reproducibility, adsorption and long-term stability.

Processing Optimization and Antioxidant Activity of Chocolate Added with Mulberry (오디 초콜릿의 제조 최적화 및 항산화 활성)

  • The purpose of this study was to determine the optimal mixing conditions for two different amounts of added mulberry powder and fresh cream to prepare functional chocolate with added mulberry powder. The experiment was designed according to the central composite response surface design, which showed 10 experimental points, and included two replicates for mulberry powder and fresh cream. The physiochemical, mechanical, and sensory properties of the test were measured, and these values were applied to the mathematical models. The results of the physiochemical and mechanical analyses of each sample, including pH, moisture content, total phenolic content, DPPH free radical scavenging activity, color L, color b, hardness, gumminess, and cohesiveness showed significant differences. The sensory characteristics of the samples tested were significantly different in flavor, texture, sourness, bitterness, and overall acceptability. The optimum formulation calculated by numerical and graphical methods was 25.76 g mulberry powder and 72.21 g fresh cream.

Optimization of the Preparation Conditions and Quality Characteristics of Sweet Pumpkin-Doenjang Sauce (단호박된장소스 제조조건의 최적화 및 품질 특성)

  • This study was conducted to develop a sauce prepared with sweet pumpkin and Korea Doenjang. The optimum conditions for manufacturing sweet pumpkin-doenjang sauce were investigated using the response surface methodology, based on the central composition design. The amount of stock added, the thickening agent, and doenjang were used as the independent variables, and the sensory characteristics (taste, flavor, color, and overall acceptability) were used as the dependent variables to evaluate the optimum conditions for the preparation of the sauce. The optimum conditions for the maximized-responses variables in the preparation of the sauce were 448.5 g of sweet pumpkin stock, 331.5 g of the thickening agent, and 20.0 g of doenjang. The quality characteristics of sweet pumpkin-doenjang sauce that was manufactured at optimum conditions were as follow: 89.55% moisture content, 0.70% crude protein, 0.10% crude lipids, and 0.71% crude ash. The pH of the sauce was 5.96; total acidity, 0.08%; and soluble solids, 6.80$^{\circ}Brix$. The total polyphenol content of the sauce was 5.70 mg/L. The electron-donating ability and reducing power of the sauce were, 14.24% and 1.64 OD, respectively.

  • For the conservation treatment of the Central Asia mural painting which is to be exhibited in the new museum in Yongsan, we analyzed the pigments used in this mural painting and examined to identify the species of the straw in the wall. We also analyzed the species of the wood of the wooden protective frame and the material of the paper in it, in order to review the material and technique of the conservation treatment performed before the mural painting had been brought to the National Museum of Korea in 1916. The results were as follows: the black pigments of Bon4075 and Bon4078 is carbon(C); the white pigment on the background is gypsum[Ca(SO)4(H2O)2]; the red pigment is lead oxide(Pb3O4) and hematite(Fe2O3) etc. The straw, which had been mixed into the wall to prevent the wall from cracking, was proved to be either wheat straw or oats straw. The wooden protective frame, which protects the mural painting now, was proved to be made of Salix, Populus, Cryptomeria japonica and pine. The paper discovered in the frame was proved to be made of the bark of a mulberry.

Roasting Conditions for Optimization of Citri Unshii Pericarpium Antioxidant Activity Using Response Surface Methodology (반응표면분석을 이용한 진피의 항산화 활성 최적화를 위한 로스팅 조건 확립)

  • This study was conducted to establish roasting conditions for optimization of Citri Unshii Pericarpium antioxidant activity using response surface methodology (RSM). A central composite design was applied to investigate the effects of two independent variables, namely roasting temperature ($40{\sim}100^{\circ}C$; $X_1$) and roasting time ($5{\sim}15min$; $X_2$), on responses such as electron donating ability ($Y_1$), total phenolic content ($Y_2$), total flavonoid content ($Y_3$), and hydroxyl radical scavenging activity ($Y_4$). The maximum electron donating ability was 72.38% at a roasting temperature of $71.12^{\circ}C$ and roasting time of 9.39 min. The maximum total phenolic content was 10.76 mg tannic acid equivalents/g at a roasting temperature of $69.71^{\circ}C$ and roasting time of 8.39 min. The maximum total flavonoid content was 105.99 mg quercetin equivalents/100 g at $72.54^{\circ}C$ and 8.64 min. The maximum hydroxyl radical scavenging activity was 60.33% at $68.97^{\circ}C$ and 9.84 min. Based on the superimposition of three dimensional RSM with respect to electron donating ability, total phenolic content, total flavonoid content, and hydroxyl radical scavenging activity under various conditions, optimum conditions were established as follows: roasting temperature of $70.90^{\circ}C$ and roasting time of 9.03 min.

Processing of Functional Porridge with Optimal Mixture Ratio of Mulberry Leaf Powder and Mulberry Fruit Powder (뽕잎분말과 오디분말의 최적 혼합비율을 이용한 기능성 죽 제조)

  • The purpose of this study was to develop a functional porridge prepared with mulberry leaf and mulberry fruit powder, which can ameliorate hypertension. The experiment was designed according to the central composite design. For optimization of the mixture ratio of mulberry leaf powder (MLP) and mulberry fruit powder (MFP), the independent variables were defined as MLP (X1) and MFP (X2) and the dependent variables were defined as K (Y1), Na (Y2), ${\gamma}$-aminobutyric acid (GABA) (Y3), cyanidin-3-glycoside (C3G) (Y4), rutin (Y5), and flavonoid (Y6). The optimal MLP to MFP mixture ratio according to the response surface method were 5.41 g of MLP and 2.65 g of MFP. The amounts of K, Na, GABA, C3G, rutin, and flavonoid in the optimal MLP and MFP mixture were 1,844.22 mg/100 g, 52.74 mg/100 g, 139.98 mg/100 g, 1,134.89 mg/100 g, 101.56 mg/100 g, and 201.28 mg/100 g, respectively. The amounts of Ca, K, Mg, and Na in the functional porridge at this optimal point were 27.66 mg/100 g, 131.32 mg/100 g, 19.57 mg/100 g, and 3.59 mg/100 g, respectively. Overall, this functional porridge can help reduce hypertension.

An Empirical Study on the Dual Burden of Married Working Women : Testifying the Adaptive Partnership, Dual Burden and Lagged Adaptation Hypotheses (근로기혼여성의 이중노동부담에 관한 실증연구: 가사노동분담에 관한 협조적 적응, 이중노동부담, 적응지체 가설의 검증)

  • The purpose of this article is to empirically testify three hypotheses on the relation between married women's employment and the allocation of unpaid domestic work within households - i.e., adaptive partnership (AP), dual burden (DB) and lagged adaptation (LA) models. The AP hypothesis assumes that, when wives are employed, husbands spend more time doing housework in order to compensate for their wives' increased responsibility. The DB model, by contrast, indicates that, even if married women are employed, their burden on domestic work does not decrease. In this case, therefore, the dual burden of married women can be expected. Between these two opposite views, the third, alternative hypothesis has been suggested recently. The LA model argues that the behaviours of households are adaptive to the changing environments but over a period of many years and even across generations. The article has analysed the total work time as well as unpaid domestic work time to testify these three hypotheses, utilising 1999 Time Use Survey data of the National Statistical Office. The research results can be summarised as follows. First, married working women worked 100 minutes more than their male spouses. Second, the average domestic work time of married men, 23-25 minutes per day, was no more than 5-10% of that of women. Third, the effects of age and women's employment were not statistically significant in multiple regression models, which means that the DB hypothesis explains the situation of married working women in Korea. Based on these findings, the article suggested the expansion of the public social service system to mitigate the dual burden of married working women, the introduction of compensatory credit for caring work, and the directions of further empirical research using the time use survey data.
