Parentage Testing for Thoroughbred Horse by Microsatellite DNA Typing (Microsatellite DNA형 분석을 이용한 더러브렛 말의 친자감정)

  • The objective of present study was to ascertain parentage of Thoroughbred(TB) horses in Korea. A total of 2,029 TB horse samples including 993 foal samples for parentage testing were genotyped for nine international minimum standard markers(AHT4, 5, ASB2, HMS3, 6, 7, HTG4, 10, and VHL20). This method consisted of multiplexing PCR procedure, and showed reasonable amplification of all PCR products. Genotyping was performed with an ABI 310 genetic analyzer. The number of alleles per locus varied from 5 to 11 with a mean value of 7.33 in TB. Expected heterozygosity was ranged from 0.544 to 0.837(mean 0.709) and the total exclusion probability of 9 microsatellites loci was 0.9978. Of the 9 markers, ASB2, HMS7 and HTG10 loci have relatively high PIC value(>0.7). All of the 993 foals were qualified by compatibility according to Mendelian fashion in the present DNA typing for parentage testing. These results suggest that the present DNA typing has high potential for parentage verification of TB horses.

Genetic Diversity Analysis of the Cheju Horse Using Random Amplified Polymorphic DNAs (PCR-RAPD를 이용한 제주말의 유전적 다양성분석)

  • This experiment was carried out to analyze genetic characteristics and to develop the breed specific DNA marker for Cheju-native horse. If this marker contains high repetitive sequences, it is possible to convert a RAPD marker of interest into a single-locus PCR marker called a sequence characterized amplified region(SCAR). Twenty six Cheju-native horse and Fifty thoroughbred genomic DNA were pooled and PCR. were accomplished using 800 random primers. Comparing the pooled DNA from Cheju-native horse and thoroughbred, we found 9 primers which identified markers present in the pooled DNA from breed but absent in the other breed. Among 9 random primers, 6 primers were thoroughbred specific and 3 primers were Cheju-native horse specific. Testing individual horse revealed that 5 marker showed the similar band pattern between Cheju-native horse and Thoroughbred. However, 4 marker were wholly absent in breed while present in the other breed. UBC $126_{3500bp}$, UBC $162_{500bp}$, and UBC $244_{1200bp}$ was detected only Thoroughbred and UBC $562_{560bp}$was detected Cheju-native horse, respectively. After determining of the cloned breed-specific fragment sequence, we designed the SCAR-primers and carried out PCR. Compared to random primer, RAPD-SCAR primer didn't show significantly higher specific band. However, RAPD analysis is useful for genetic characterization of Cheju-native horse.

An Approach to Identify Single Nucleotide Polymorphisms in the Period Circadian Clock 3 (PER3) Gene and Proposed Functional Associations with Exercise Training in a Thoroughbred Horse (국내산 경주마의 주기성 시계 유전자(PER3) SNP 및 운동에 따른 기능적 식별 접근 가능성 제안)

  • The period circadian clock gene 3 (PER3) plays a role in the mammalian circadian clocksystem. A regular exercise regime may affect the PER3 transcription in skeletal muscle. Although the effects of day length on circadian and circannual processes are well established in humans and mice, the influence of exercise on these processes in the horse has not been investigated. The present study investigated the expression of the PER3 gene following exercise in a thoroughbred breed of Korean horse. In addition, a comprehensive in silico nonsynonymous single nucleotide polymorphism (nsSNP) analysis of the horse PER3 gene and predicted effects of nsSNPs on proteins were examined. The expression of PER3 in skeletal muscle was significantly upregulated after exercise. Four nsSNPs were functionally annotated and analyzed by computational prediction. The total free energy and RMSD values of PER3 gene showed causative mutations. The results showed that nsSNP s395916798 (G72R) was associated with residues that have stabilizing effects on structure and function of PER3 gene. This study documented role of PER3 gene in phenotypic adaptation related to exercise in skeletal muscle. Further, the SNPs in PER3 could serve as useful biomarkers of early recovery after exercise in racehorses.

Eyelid Squamous Cell Carcinoma in a Horse in Jeju (제주도의 말에서 발생한 안검 중층편평상피 암종)

  • An 8-year-old, female, mixed Thoroughbred horse was presented for the evaluation of the left upper eyelid mass with ocular discharge during 4 months. After unilateral surgery, excised eyelid mass and left eye were submitted for diagnosis. Grossly, the removed mass was $6{\times}7{\times}2$ cm in size and dome shape with rough irregular surface. On the cut surface, mass was firm and yellowish white in color. Histopathologically, severely proliferated stratified squamous epithelia were heavily invaginated into dermis, and then formed rete ridge or numerous squamous islands. These neoplastic islands were composed of multiple layers of prickle cells with intercellular bridge and central laminated keratin pearls or necrotic cellular debris. Based on the above results, this case was diagnosed as squamous cell carcinoma in upper eyelid of horse.

Antimicrobial Susceptibility Patterns of Staphylococcus intermedius Isolated from Skin Lesions of Thoroughbred Horse. (더러브렛 말의 피부병변에서 분리한 Staphylococcus intermedius의 약제 감수성 양상)

  • Staphylococcus intermedius (S. intermedius) has been rarely isolated from horse. In this study, we investigated the antimicrobial susceptibility patterns of S. intermedius isolated from skin lesions of Thoroughbred horse in Daegu Equestrian Association. The skin lesions were showed with dehairing and slight purulent inflammation. Bacteria were isolated from skin lesions and identified as S. intermedius by biochemical tests and MicroLog (BIOLOG, California, USA). Antimicrobial susceptibility test of S. intemedius isolates was performed with 33 antimicrobial agents (BBL, Maryland, USA) by using the agar disk diffusion method. It showed a high sensibility in the amikacin, amoxycillin/clavuramic acid, ampicillin, cefoxitin, cephalothin, chloramphenicol, ciprofloxacin, enrofloxacin, florfenicol, kanamycin, neomycin, nitrofurantoin, norfloxacin, ofloxacin, oxacillin, penicillin G, tetracycline and vancomycin. The horse was treated with penicillin, and cured completely after two weeks. The present results showed valuable information for treatment and prevention of skin disease in horse.

Biochemical characteristics and antimicrobials susceptibility of pathogenic Streptoccus spp. isolated from respiratory tract of Thoroughbred horses in Jeju, Korea (제주지역에서 사육중인 더러브렛 말 호흡기로부터 분리된 병원성 Streptoccus spp.의 생화학성상 및 약제감수성 양상)

  • This study carried out to investigate the pathogenic Streptococcus spp. isolated from respiratory tract of Thoroughbred in Jeju province. The specimens were collected from nasal mucosa using a culture swab from 113 Thoroughbred horses. Suspected colonies were selected onto blood and MacConkey agar plate, and identified by standard biochemical properties using Vitek 2 system and PCR method. In this study, we isolated S. equi (n=6), S. zooepidemicus (n=31), S. equisimilis (n=5), S. dysgalactiae (n=2), S. agalctiae (n=1), non identified ${\beta}$-hemolytic Streptococcus spp. (n=1) from Thoroughbred horses. In antimicrobial susceptibility test, it showed a high sensibility in the most of antimicrobial except for neomycin, streptomycin, spectinomycin, erythromycin and clindamycin. These results will provide the basic information to establish control measures for the treatment and prevention of respiratory disease by pathogenic Streptococcus spp. in Thoroughbred horses in Korea.

  • A 11-year-old gelding Thoroughbred horse was presented with a history of unilateral nasal discharge of right side. The horse was diagnosed as maxillary sinusitis based on blood examination and radiography. The horse was treated with bone flap of right maxillary sinuses. Abscess of maxillary sinus was drained and irrigated with antibiotic-saline and povidone iodine solution. After surgical procedure, nebulization and medication applied by the result of antibiotic sensitivity test. After treatment for two weeks, the patient was recovered completely.

Outbreaks of Strangles due to Capnophilic Streptococcus equi subsp equi in South Korea (호이산화탄소성 Streptococcus equi subspecies equi에 의한 선역의 발생)

  • We reported an outbreak of clinical strangles in thoroughbred horses due to capnophilic Streptococcus equi subsp equi in South Korea. On three different farms, we isolated 17 S equi subsp equi isolates from 29 horses with or without abscesses in their lymph nodes. Of the 17 isolates, two isolates from clinical cases grew well in aerobic conditions, whereas 7/7 isolates from clinical cases and 8/22 isolates from the nasal discharges of horses did not. The latter 15 isolates were capnophilic, oxygen-sensitive, and $CO_2$-requiring S equi subsp equi, which could not grow in aerobic conditions, but which grew well in a $CO_2$ incubator with 5% $CO_2$, in anaerobic conditions using a GasPak, and with reduced oxygen tension in a candle jar. This study is the first report of a strangles outbreak caused by capnophilic S equi subsp equi in South Korea.

  • Antibodies to Anaplasma phagocytophilum and Ehrlichia chaffeensis were detected by the immunofluorescent antibody (IFA) test in sera collected from cats, thoroughbred horses and Holstein cattle in Gwangju, Jeonju and Jeju Island of Korea. Two hundred fifty four sera (33 feral and pet cats, 92 grazing horses and 129 grazing cattle) were obtained from Republic of Korea. Antibodies to A. phagocytophilum (titer $\geq$ 80) were detected in 6 of the 33 feral and pet cats (18.2%), and 1 seropositive cat (3.0%) also had antibodies to E. chaffeensis. Only 1 of 129 (0.8%) cattle and 2 of 92 (2.2%) horses had antibodies to A. phagocytophilum. Antibodies to E. chaffeensis were not detected in either of these animals. This is the first report of serological evidence of A. phagocytophilum and E. chaffeensis from cats, cattle and horses in Korea. These rickettsial agents could have an important impact on human health or impact animal health with economic losses among industrial grazing animals in Korea.

Peritonitis associated with Streptococcus equi subsp. zooepidemicus in a thoroughbred horse (더러브렛 말의 Streptococcus equi subsp. zooepidemicus 감염에 의한 복막염)

  • Fibrinopurulent peritonitis and abdominal abscesses associated with Streptococcus (S.) equi subsp. zooepidemicus is reported in a 1-year-old female thoroughbred horse. The horse died after showing 5-day history of the cold, severe abdominal distention, abdominal breathing, hyperthermia, anorexia, and loss of intestinal sound. At necropsy, several liters of turbid yellowish green fluid were seen in the abdominal cavity. Yellowish creamy and fibrinous or bloody materials were presented on the serosa of various abdominal organs that were intensively adhered with each other. Spleen and mesenteric lymph node were remarkably enlarged. Affected lobes of the lung showed severe congestion, hemorrhage and doughy consistency. Histologically, the lung showed hemorrhagic pneumonia with diffuse congestion and edema. Severe diffuse fibrinopurulent peritonitis with Gram-positive bacterial cocci and adjacent fibrosis were showed in the serosa of various abdominal organs such as liver, spleen, stomach, and intestine. And multifocal abscess pouches were presented in the granulation tissue of abdominal viscera. S. equi subsp. zooepidemicus was isolated from the peritoneal swab, abdominal organs, and lung. Hematogenous dissemination of bacteria from hemorrhagic pneumonia is proposed as the route of infection in this case.
