Antioxidant and Anticancer Activities of Glycine Semen Germinatum Fermented with Germinated Black Soybean and Some Bacteria (발효콩 대두황권의 항산화 및 항암효과)

  • This study was conducted to evaluate the antioxidant (ABTS, DPPH radical scavenging and reducing power), nitric oxide (NO)) production and anticancer activities against human cancer cells (HeLa HepG2, HT-29 and MCF-7) for methanol extracts of Glycine Semen Germinatum (Daedoowhangkeun in Korean) fermented with germinated black soybean and some bacteria. Antioxidant activities were increased by increasing the concentration of the extract at dose-dependent manner, Their activities of black soybean were higher than those of yellow soybean. Non fermented sample was slightly higher than Glycine Semen Germinatum fermented with Bacillus subtilis and lactic acid bacteria. In 1 mg/mL of the extract NO production levels were $0.374\;{\mu}M$ for yellow soybean, $0.368\;{\mu}M$ for black soybean, and $0.367\;{\mu}M$ and $0.358\;{\mu}M$ for Glycine Semen Germinatum fermented with B. subtilis and lactic acid bacteria, respectively. Methanol extract of Glycine Semen Germinatum fermented with mixture broth of lactic acid bacteria was shown to be the highest activity for anticancer activities against human cancer cells tested and their activities were exhibited in the order of HeLa > HT-29 > HepG2 > MCF-7 cell.

Effect of the Extraction Method on the Soybean Embryo Factor 3 Activity (추출 방법에 따른 대두 배인자 3 역가)

  • Soybean nuclear extracts were prepared to detect SEF3(soybean embryo factor 3), which is presumed to be a trans-acting factor for the expression of the soybean ${\beta}-conglycinin\;{\alpha}'$ subunit gene. To increase the specific activity of DNA probe during labeling with $[{\alpha}-^{32}P]$dATP, dATP was added to a final concentration of 1.1 mM during the chase reaction. It results in approximately four-fold increase of specific activity of the DNA probe. Effects of several modifications in preparation of soybean nuclear extracts were examined. It was found that glycerol is effective to stabilize SEF3 during the preparation of nuclear extracts and polyethylenimine could be used to increase the specific activity of SEF3 in nuclear extracts.

  • Soybean nuclear extracts and S-100 were prepared to examine the soybean embryo factors which bind to the upstream region of soybean ${\beta}-conglycinin$ ${\alpha}'$ subunit gene. SEF3(soybean embryo factor 3), which is presumed to be a trans-acting factor for the expression of the gene, was detected in gel mobility shift assay using the DNA probe containing two AACCCA hexanucleotides. DNA probe containing CATGCAT or AACACA was used to find any other soybean embryo factor interacting with the upstream region of ${\beta}-Conglycinin$ ${\alpha}'$ subunit gene. It was found that there was no common DNA binding protein detected both in nuclear extracts and S-100. The relative levels of SEF3 binding activity both in nuclear extracts and S-100 of maturing soybean seeds were determined. SEF3 activity of nuclear extracts was first detected around 20 days after pollination and significantly increased around 32 days after pollination.

  • This study was performed to investigate the phenolic compounds content and antioxidative activity of methanol extract from Chinese soybean. The content of total phenolics and total flavonoids of Chinese soybean Heinong 48) extract were $9.91{\pm}0.57GAE\;mg/g$ and $4.15{\pm}0.39QE\;mg/g$, respectively. The antioxidative activities of Chinese soybean extract were significantly increased in a dose dependent manner on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS) radical scavenging($IC_{50}$ values were $2.04{\pm}0.16$ and $3.31{\pm}0.31mg/mL$, respectively). These results suggest that Chinese soybean may have great potential as a natural antioxidant source with health benefits.

The Antioxidative Effects of the Water-Soluble Extracts of Plants Used as Tea Materials (다류원료 식물류 물 추출물의 항산화 효과)

  • The water-soluble extracts of plants used as tea materials were investigated for their antioxidant activities and active components(total phenolics, ascorbate and selenium). Antioxidant activities of the plant extracts were determined by measuring the changes in both peroxide values(POV method) during storage(36 day, $55^{\circ}C$) and conductivity of soybean oil at $110^{\circ}C$(Rancimat method). Soybean oil without any additive was used as a control. Soybean oil treated with 0.02% BHT was used as a positive control. The test samples were prepared by mixing the plant extracts with soybean oil in 0.02% concentration by weight. The water-soluble extracts of lycii fructus(23 d), oolong tea(23 d), orange peel(23 d), citron(22 d), and apricot(22 d) showed longer induction periods, compared to control(21 d) and BHT(21 d) by POV method. Also water-soluble extracts of oolong tea(12 h), instant coffee(11 h), citron(10 h), cinnamomi cortex(10 h), schizandrae fructus(10 h), lycii fructus(10 h) and apricot(10 h) demonstrated longer induction periods, compared to control(8 h) and BHT(8 h) by Rancimat method. The contents of total phenolic compounds were observed to be high in water-soluble extracts of oolong tea, green tea, black tea, coffee, cinnamomi cortex, and cassiae semen. Ascorbate contents were found to be high in coffee, eucommiae cortex, black tea, ganoderma, cinnamomi cortex, persimmon leaf, chicory, green tea and oolong tea extracts. The selenium contents were not detected in all the extracts. The antioxidative effects of some plant extacts were seemed to be the combined effects of various antioxidant components such as phenolics and ascorbate.

Anti-cancer and Anti-inflammatory Effects of Mung Bean and Soybean Extracts (녹두 및 대두추출물의 항암 및 항염증 활성)

  • The quinone reductase (QR) inducing activities of mung bean and soybean solvent extracts were compared using murine hepatoma cells (Hepa 1c1c7). The mung bean extracts (ethylacetate and ethanol) showed higher chemoprevention index values (7.88-8.22) than those of soybean extracts (2.9-5.2) from four different cultivars. The mung bean extracts also had significantly higher inhibitory effects (47-62% at 100 ${\mu}g$/mL) than the soybean extracts (15-42% at 100 ${\mu}g$/mL) against the production of nitric oxide and prostaglandin E2 in lipopolysccharide stimulated macrophage RAW264.7 cells without cytotoxicity. Among seven recovered fractions of mung bean ethanol extract obtained by C 18 silica flash column chromatography, the most non-polar fraction exhibited the highest chemoprevention index of 10.4.

Biological Activities of Chungkugjang Prepared with Black Bean and Changes in Phytoestrogen Content during Fermentation (검정콩 청국장의 생리활성 및 발효중 Phytoestrogen 함량의 변화)

  • To assess functional properties of chungkugjang (CK), a traditional Korean soyfood, fermented with black beans at $42^{\circ}C$ for 72 hrs, some biological activities of methanol extract of CK and the changes in phytoesterogen (daidzein, genistein) contents during fermentation were investigated. The methanol extract of CK (MEC) prepared with soybean had no or a little antibacterial activity. MEC prepared with black bean inhibited the growth of all bacteria tested, and MEC of small black bean was higher in antibacterial activity than that of large black bean. Hydrogen-donating activities of MEC of large and small black bean were 76.4 and 75.5%, respectively, which were higher than that of soybean being 67.3%. Nitrite-scavenging activity was found to be above 90% in all MECs tested as compared with control group. MEC showed strong antioxidant activities against both peroxidation of linoleic acid and $H_{2}O_{2}-FeSO_{4}-induced$ peroxidation of rat liver homogenate. The antioxidant activities were high in the order of small black bean, large black bean and soybean. Contents of genistein and daidzein were gradually increased during fermentation of CK. The isoflavones were higher in black bean CK than in soybean CK and higher in large black bean CK than in small black bean CK. In black beans genistein content was about twice as much of daidzein.

  • These studies was conducted to investigate the influence of ALLELOPATHIC effects of upland weeds on crops and soil microorganisms. The aqueous extract of weed residues were used to identify physiological controll of plant and microorganisms. The results obtained are as follows : 1. The aqueous extract of weeds controlled germination and growth of plant, especially the 5%-extract of Potulaca oleracea suppressed the growth of soybean about 8~12%. 2. The 5%-extract of Potulaca oleracea inhibited effect of phytosynthesis of soybean about 60%. 3. The move the concetration of phenolic compounds in soil, the less a number of actinomycetes and bacteria. 4. The number of soil microorganism(fusarium oxysporum spp.) were decresed as to the concentration of vanillic acid was increased.

The Effects of Soybean Protopectinase on Melanin Biosynthesis (효소(Protopectinase) 처리한 대두가 세포내 멜라닌 생성에 미치는 영향)

  • This study was performed to assess the antioxidant activities and whitening effects of protopectinase enzymes and mechanical maceration from soybeans on melanin synthesis. The whitening effects of enzyme treatment and mechanical maceration were examined by an in vitro mushroom tyrosinase assay and by assessing markers in B16BL6 melanoma cells. We assessed inhibitory effects on the expression of melanogenic enzymes, including tyrosinase, tyrosinase-related protein 1 (TRP-1), and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effects on free radical generation were determined by measuring DPPH and hydroxyl radical scavenging activities. In DPPH radical scavenging activity, enzyme treatment and mechanical maceration had a potent anti-oxidant activity in a dose-dependent manner and significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. There was also an inhibition in the expression of tyrosinase, TRP-1, and TRP-2 in B16BL6 melanoma cells. Our results show that soybean protopectinase treatment inhibits melanogenesis, with the underlying mechanism possibly due to the inhibition of tyrosinase activity and tyrosinase, TRP-1, and TRP-2 expression. We suggest that soybean protopectinase should be contained as natural active ingredients for antioxidant and whitening cosmetics.

감초추출물을 첨가한 청국장의 품질 특성

  • 표준화된 청국장 제조법으로 대두를 정선, 수세 후 3배의 물을 가하여 24시간 침지하고 물빼기를 한 후 121$^{\circ}C$에서 1시간 동안 증자한 후 4$0^{\circ}C$까지 냉각한 후 재래청국장으로부터 분리한 B. lichenifomis 균주를 2% 접종, 4$0^{\circ}C$에서 54시간 발효시켜 청국장을 제조하였으며 발효 방법에 따라서는 종균을 접종한 경우가 가장 우수한 발효양상을 보였다. 감초추출물을 첨가하여 제조한 청국장의 품질특성을 알아본 결과 첨가농도가 증가할수록 pH가 증가하였으며 환원당 함량도 증가하였다. 색도는 첨가농도가 증가함에 따라 다소 어두워 졌으나 점질물의 생성량은 증가하는 경향이었다. 관능검사결과를 종합해 보면 5%의 첨가구에서 가장 높은 기호도를 보여 증자 대두에 감초추출물을 혼합하여 발효했을 경우 점질물의 생성의 증대는 물론 기호도의 향상에도 크게 기여하였다.
