  • Bagrid catfish hybrid between Korean bullhead (Pseudobagrus fulvidraco) and Ussurian bullhead (Leiocassis ussuriensis) was produced by artificial fertilization. Modal chromosome numbers of Korean bullhead, Ussurian bullhead and their hybrid were the same as 2n=52. The karyotypes were 22 metacentric (M), 22 submetacentacentric (SM), and 8 acrocentric (A) chromosomes for Korean bullhead, 22M+14SM+16A for Ussurian bullhead and 22M+18SM+12A for their hybrid. The hybrid bullhead karyotype consisted of each haploid set of parental species chromosomes. Erythrocytic sizes and DNA contents of hybrids were intermediate between two parental species.

  • Viability, early growth and sex ratio of induced gynogenetic diploid bagrid catfish (Leiocassis ussuriensis, Siluriformes) were examined. Survival rates of gynogenetic diploids and normal bi-parental diploids up to 180 days post hatching were not significantly different (P>0.05), and consequently resulted the mean percent survivals of 76% (gynogens) and 78% (bi-parental progeny) at the end of growth trial. Growth in both body weight and total length of gynogenetic diploids was also similar to those of normal diploid controls (P>0.05). Gynogenetic bagrid catfish displayed a similar level of ovarian development when compared to bi-parental diploid females, as judged at 10 weeks post hatching. Sex ratio of bi-parental progeny was normal 1:1 whereas all-femaleness was observed in gynogenetic progeny (P<0.05), suggesting that the genetic sex determination mechanism of this species should be XX-XY based female homogamety.

  • Treatment conditions for the induced diploid gynogenesis, a maternal-exclusive form of artificial parthenogenetic reproduction, were optimized in bagrid catfish (Leiocassis ussuriensis, Siluriformes). Optimal amounts of ultraviolet (UV) irradiation for the genetic inactivation of spermatozoa in bagrid catfish and Pseudobagrus fulvidraco were proven to be ranged from 3,600 to 4,800 $ergs/mm^2$ based on the examination of viability of embryos and haploid incidence. Haploid embryos were restored to diploidy by preventing the extrusion of the second polar body using cold shock treatment. Thermal treatments (4 or $6^{\circ}C$ for 30, 40 or 50 min) were carried out 3, 5 or 7 min post insemination. Best scores for embryo viability (38.6% of total eggs taken) and incidence of normal diploidy (87.9% of hatched larvae) were observed at the embryo group treated at $4^{\circ}C$ for 40 min, 5 min after insemination. Restoration of gynogenetic diploidy was confirmed based on the absence of haploid syndrome, cell size and/or nucleolar organizing region (NOR) counts.

  • 대농갱이는 메기목 동자개과에 속하는 종으로 전장이 50 cm에 이르는 대형 종이며 매운탕으로 기호도가 높은 중요 식용어이다(Lee and Kim 1990). 대농갱이는 암컷보다 수컷의 성장이 약2배 가량 빨라 실제 양식 현장에서는 전 수컷 종묘의 생산이 생산성을 크게 향상시킬 수 있다. 전 수컷집단을 생산할 수 있는 방법에는 자성호르몬 처리에 의해 생산된 phenotypic female (genetic male)을 친어로 사용하여 단순 교배에 의한 전 수컷 집단을 생산하는 방법인 유전학적 성전환 방법이 있으며, 호르몬 처리에 의해 성전환을 유도하여 전 수컷을 생산하는 생리학적 성전환 방법이 있다(Hunter and Donaldson, 1983).   • Backcross hybridization between Korean bullhead Pseudobagrus fulvidraco female and Korean bullhead P. fulvidraco $\times$ Ussurian bullhead Leiocassis ussuriensis hybrid male was performed, and early viability and karyotype of the backcross hybrids were examined along with their parental species. Mean fertilization rate (86.8%), hatching success (70.7%) and early survival rate (76.4%) of backcross hybrids were similar with those found in the maternal species, the Korean bullhead. From the chromosome analysis, modal chromosome numbers of Korean bullhead, Ussurian bullhead, their hybrid and backcross hybrid were the same as 2n = 52. However, their karyotypes were different among genotypes. The karyotype of backcross hybrid was 22 metacentric + 18 submetacentacentric + 12 acrocentric chromosomes.

대농갱이 인공종묘생산

  • 대농갱이는 체형이 동자개와 비슷하나 지금까지 인공부화는 물론 산란습성에 관해서도 잘 알려져 있지 않은 미개발 어종으로 양식품종의 다양화 및 경쟁력 있는 새로운 품종 개발을 위하여 인공종묘생산 시험을 시도하였다. 본 연구에 사용한 대농갱이 친어는 금강 중류역에서 서식하고 있는 자연산 친어로 암컷 210마리, 수컷 148마리를 사용하였다. 호르몬주사에 의한 인공 배란효과 조사를 위해 암컷 한 마리당 HCG 1,000 IU 주사군(75마리)과 2,000 IU 주사군(135마리)으로 나누어 배란율을 조사하였으며, 수컷은 주사하지 않고 정소를 적출하여 막자사발에 갈아서 등조액과 혼합하여 사용했다. 수정 방법은 난을 착출하면서 정자 등조액을 동시에 흘리는 방법, 난을 그릇에 받은 후 정자 등조액을 혼합 수정하는 방법, 채란용기에 정자 등조액을 미리 넣고 암컷의 난을 짜 넣어 수정시키는 방법 등 3가지 방법을 사용하였다. 부화된 자어는 초기먹이로 알테미아(3일 급이), 실지렁이(6일 급이), 배합사료 순으로 급이하였으며 수온 25~28$^{\circ}C$에 유지관리하면서 성장도 시험을 실시하였다. 호르몬 주사에 의한 배란효과는 HCG 2,000 IU 주사군(채란율 88.9%)이 1,000 IU 주사군(37.3%)에 비해 월등히 높았으며 난의 성숙상태도 좋았다. 수정방법에 있어서는 채란과 동시에 정자 등조액을 흘려 수정시키는 방법과 채란용기에 미리 정자 등조액을 넣고 암컷의 난을 한 마리씩 짜서 수정한 후 바로 부화지로 이동하여 고르게 뿌려 준 경우가 수정율 및 부화율이 좋았다. 부화수온은 23$\pm$1$^{\circ}C$에서 96시간, 26$\pm$1$^{\circ}C$에서는 72시간만에 부화되었으며 부화여건이 좋으면 단시간에 부화할 수 있도록 하는 것이 수생균 발생을 억제할 수 있어서 효과적이었다. 자치어의 성장도 실험에서는 사육수온 25~28$^{\circ}C$에서 직선식으로 나타낼 수 있어서 평균체장은 Y=0.9943X+0.7867, 평균체중은 Y=0.8800-0.6800로 각각 표시되었다.

  • The fine structure of spermatozoa of Leiocassis ussuriensis was examined with scanning and transmission electron microscopies. The spermatozoon of L. ussuriensis is approximately $68.8\;{\mu}m$ in length and a relatively simple cell with a spherical nucleus, a short midpiece and a tail. The ultrastructure of spermatozoa of L. ussuriensis is characterized by the following features. The nuclear fossa, the length of which is about two-thirds of the nuclear diameter, contains two centrioles. The centrioles are orientated approximately $180^{\circ}$ to each other. The mitochondria are arranged in two layers and their number is 12 or more. The axoneme is the 9+2microtubular pattern and has inner but no outer dynein arms as in other bagrids. The two axonemal fins are in the same plane with the two central microtubules, the doublets 3 and 8. The axonemal fins and the inner dynein arm are shared in Bagridae and the deep nuclear fossa is shared in Siluriformes. The axonemal finsobse observed in Bagridae and Amblycipitidae of Siluriformes might be the apomorphic character in Ostariophysi. They are not reported in Cyprinidae and Characiformes.

Change of Water Quality and Growth of Leiocassis ussuriensis Cultivated in a Biofloc System using Bacillus subtilis (Bacillus subtilis을 활용한 바이오플락 시스템에서 사육한 대농갱이(Leiocassis ussuriensis)의 성장 및 사육수 수질 변화)

  • This study observed changes in survival, growth performance and water quality for 90 days to confirm the possibility of Leiocassis ussuriensis farming using Biofloc technology (BFT) using Bacillus subtilis. Feed and molasses were added to the experimental tank to produce BFT water before planting the experiment, and B. subtilis was inoculated to stabilize the water quality for 40 days. The survival rate of the experimental fish was 92.7±3.2% in the control group and 95.8±3.3% in the BFT group. The Weight gain (WG) was 118.1±9.0% of the control and 197.7±15.6% of the BFT, and the Specific growth rate (SGR) was 0.87±0.5% of the control and 1.21±0.06% of the BFT. As for the feed efficiency, the control was 43.7±2.6% and the BFT was measured at 70.1±4.1%, indicating that the feed efficiency of the BFT was higher. As a result of measuring the water quality change during the experimental period, pH was reduced in both the control and the BFT, and Mixed Liquor Suspended Solids (MLSS) did not show any change in the control, but the BFT showed a significant increase at 90 days. NH4+-N and NO2--N showed a significant increase from the 30 days of the experiment in the control, but showed no change in the BFT. In conclusion, as a result of applying the BFT system using B. subtilis to the process of cultivating Leiocassis ussuriensis, the water quality tended to stabilize, and the growth rate and feed efficiency were found to be higher than those of the control, confirmiWng that it had a positive effect.
