Changes of Oligosaccharide in Soymilk with Mixed Cultures of Lactobacillus acidophilus and Saccharomyces uvarum (Lactobacillus acidophilus와 Saccharomyces uvarum의 혼합배양에 의한 대두유의 당 변화)

  • The oligosaccharide metabolism in soymilk was investigated by mixed culture with Lactobacillus acidophilus and Saccharomyces uvarum. When Saccharomyces uvarum was cultured in soywhey, change of oligosaccharide could be shown apparently. However, Lactobacillus acidophilus could not utilize oligosaccharide in soywhey for growth and lactic acid production. During the fermentation of mixed culture with Lactobacillus acidophilus and Saccharomyces uvarum, Saccaharomyces uvarum was supposed to convert oligosaccharide to monosaccharide first and then Lactobacillus acidophilus to convert these produced monosaccharide to lactic acid.

O1igosaccharide Formation and Production of Transfructosylase and Transglucosylase by Aureobasidium pullulans (Aureobasidium pullulans가 생산하는 과당 및 포도당 전이효소에 의한 올리고당류의 생산)

  • Oligosaccharide formation and the production of transfructosylase and transglucosylase by Aureobasidium pullulans were studied in sucrose or maltose media, respectively. The initial uptake rates of substrate in sucrose-rich media were faster than that in maltose-rich media, also most parts of oligosaccharides formed and other monosaccharides released were utilized progressively as substrate during the cultivation periods. However, when the initial amount of sucrose was raised to $100g/\ell$, high concentration of monosaccharides were liberated, consequently high-level fructose was accumulated unused during fermentation. The biggest molecule of oligosaccharide synthesized was hexasaccharide in all cultivation media examined, of which the organism could not utilize isomalto-oligosaccharide of DP6 synthesized in a maltose-rich medium. The maximum amount of oligosaccharides produced was $58g/\ell$ when $100g/\ell$ of sucrose and $5g/\ell$ of maltose were used as initial substrate. From the early stage of growth both fructooligosaccharides and isomalto-oligosaccharides were synthesized and progressively utilized as substrates during the fermentation. Based on the experimental results, it was suggested that maltose could induce both transfructosylase and transg1ucosylase, whereas sucrose was unable to slimulate transglucosylase formation.

Antitumor and Immuno-potentiating Activities of Crude Polysaccharides from Fruiting Body of Agaricus brasiliensis (신령버섯(Agaricus brasiliensis) 자실체 추출 조다당류의 항암 및 면역증강 작용)

  • Agaricus brasiliensis, one of edible mushroom belonging to Basidiomycota, has been used for curing gastric ulcer and stomach cancer of human beings and also known to have good inhibitory effects on sarcoma 180 and Ehrlich carcinoma of mice. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were prepared from fruiting body of the mushroom. ${\beta}$-glucan and total protein contents were identify from fractions of edible mushrooms extract. The ${\beta}$-glucan and protein contents of all fractions of the mushrooms ranged from 21.54~32.31% and 0.16~9.34%, respectively. In vitro cytotoxicity tests, crude polysaccharides were not cytotoxic against cancer cell lines such as Sarcoma 180, HT-29, NIH3T3 and RAW 264.7 at the concentration of 10~2000 ${\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of 18.8~50.6% in mice previously inoculated with Sarcoma 180. Fr. HW increased the numbers of spleen cell by 1.2 fold at the concentration of 200 ${\mu}g/ml$ compared with control. Fr. MeOH and Na improved the immuno-potentiating activity of B lymphocyte by increasing the alkaline phosphatase activity by 1.6 fold compared with control at the concentration of 50~500 ${\mu}g/ml$. Fr. Na generated 15.9 ${\mu}M$ of nitric oxide (NO) when cultured with RAW 264.7 at the concentration of 200 ${\mu}g/ml$, while lipopolysaccharide, a positive control, produced 3.7 ${\mu}M$. The Fr. NaCl, Fr. HW and Fr. MeOH increased the secretion of TNF-${\alpha}$, IL-$1{\beta}$, Il-2 and IL-6 by 2.2 times compared with the control group. Fr. Na increased the numbers of peritoneal exudate cells by 4 folds at the concentration of 50mg/kg compared with control. Circulating leukocytes increased by 2.7 folds when Fr. HW from A. brasiliensis was inoculated at the concentration of 50 mg/kg body weight. The hematological and blood chemical analysis of the 3 fractions did not show any difference in blood compositions and enzyme activities compared with the control group (p<0.05). Therefore, the experimental results suggested that crude polysaccharides extracted from A. brasiliensis contain antitumor and immuno-potentiating activities against Sarcoma 180 in ICR mice.

  • 1) Conditions for the hydrolysis of starch by bacterial liquefying amylase (BLA), saccharifying amylase (BSA) and isoamylase were investigated. Out of four syrups prepared by different combinations of these enzymes, those made by BLA followed by BSA and/or isoamylase were comparable to sucrose syrup in canning of orange segments. 2) Two branched maltooligosaccharides were isolated from the hydrolyzate of starch by BLA and BSA, and their structures were tentatively identified as pentaose and hexaose having an ${\alpha}-1$, 6-linkage at the branching point.

  • Ordinary Korean Soy-sauce samples obtained from several homes in Seoul area were analysed by paper partition chromatography method on the free-sugars. The following results were obtained. 1) Galactose, glucose, arabinose and xylose were detected in ordinary soy sauce. 2) The abundance of the found sugars : galactose, arabinose, xylose and glucose in the order.

Antitumor and Immuno-modulatory Effect of Crude Polysaccharides from Fruiting Body of Tremella aurantialba Against Mouse Sarcoma 180 (생쥐의 Sarcoma 180에 대한 금목이(Tremella aurantialba) 자실체 추출 조다당류의 항암 및 면역조절 효과)

  • Tremella aurantialba, one of edible and medicinal mushrooms belonging to Tremellaceae of Basidiomycota, has been known to have outstanding curing effects on coughing, tracheitis and hypertension of humans and antitumor activity on the sarcoma 180 and Erhrlich carcinoma of mice. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from fruiting body of the mushroom. In vitro cytotoxicity tests showed that all crude polysaccharides extracted from the fruiting body were not cytotoxic against cancer cell lines such as RAW 309 CR.1 and Sarcoma 180 at the concentration of $2000\;{\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of $11.1{\sim}66.7%$ in mice inoculated with Sarcoma 180. Fr. MeOH improved the immunopotentiation activity of B lymphocyte by increasing the alkaline phosphatase activity by 1.16 folds at the concentration of $200\;{\mu}g/ml$. In case of Fr. HW, the numbers of peritoneal exudate cells and circulating leukocytes were increased by 1.42 and 2.87 folds, respectively.

Contents of Monosaccharides in the Hydrolysates of Some Forest Soil Horizons (삼림토양(森林土壤)의 층위별(層位別) 가수분해물중(加水分解物中) 단당류(單糖類)의 함량(含量))

  • Monosaccharide content of four forest soils were analyzed. Two soils under coniferous forest trees and another two under duciduous forest trees of Mts. Zeombong and Odae in Kangweon-Do were sampled from the surface horizon down into the subhorizons. 1. The largest amount of monosaccharide is found in the surface organic horizon of each soil and with increasing depth the amount decreases as might be expected considering total organic matter content. 2. Hexoses (galactose, glucose, mannose) predominate over pentoses (arabinose, ribose, xylose) and deoxyhexoses (fucose, rhamnose), the latter being in the smallest amount. Glucose is the most abundant monosaccharide in all samples regardless of vegetation of soil or depth. In general the content of each monosaccharide follows the order of glucose > manrtose > galactose > arabinose > xylose > rhamnose > fucose > ribose. 3. Very little amount of ribose is present even in organic horizons of coniferous forest soils. In samples taken from deciduous forest soils ribose is virtually absent. 4. The relative proportion of monosacchaiide to the total soil organic matter decreases with increasing depth, which may be resulted from the effect of prolonged humification. The total monosaccharide in the organic surface layer amounts to 27-50% of the total organic carbon or 15.7-29% of the total organic matter. Hexoses alone take the largest share of 20-38% of the carbon, or 12-22% of the organic matter.
