A Study on the Nitrogen Sources for the Enhancement of the Nitrogen Bioavailability in Rats with Peptic Ulcer -The Ratio of Casein and Casein Hydrolysate- (소화성 궤양 흰쥐에서 체내 질소이용율 증진을 위한 체내 질소원에 관한 연구 - 단백질과 단백질 가수분해물의 비율을 중심으로 -)

  • This study aimed to verify the nutritional and curative effects of protein hydroysate and optimal ratio between protein and protein hydroysate as nitrogen source in rats with cysteamine-induced duodenal ulcer. Duodenal ulcer rat model was established by intraperitoneal injections of cysteamine. Sprague-Dawley, female rats weighing approximately 200g were intrapertionealy injected twice cysteamine(13mg/100g BW) at intervals of 3hours per day. This procedure was repeated 3 times at intervals of 3 days. Animals fed on 10% casein diet for injection periods. After last injection, 5 kinds of kiets (the ratio of casein and casein hydrolysate was 100 : 0(C100), 75 : 25(CH 25), 50 : 50(CH 50), 25 : 75(CH 75), 0 : 100(CH 100)) were given. The rate were sacrificed after feeding diet, 1, 3, 5 days. Ulcer index, hexosamine content of stomach and duodeum, gastric motility, trypsin activity, blood glutathione, plasma total protein, albumin, amino-N, urinalry urea nitrogen, creatinine, hydroxyproline and retention rate of nitrogen were analyzed for nutritional effects of diet treatments. There were no differences among diet groups in the view of the growth and diet treatments. There difference of ulcer curation by diet was appeared after 3 days. The ulcer indexes of C100 and CH 25 of 3, 5 days were significantly higher than those of CH 50, CH 75 and CH 100. This result was the same as hexosamine content of stomach, plasma protein, albumin concentration and nitrogen retention rate. The more casein hydrolysate diet had, the lower trypsin activity was. The more casein gydroysate diet had, the higher excretion of hydroxyproline was. These results show that protein hydrolysate can be applied in diet therapy for the patients with gastronitestinal ulcer. It suggests that it has curative effect of diet when nitogen sources include at least over than 50% of protein hydrolysate.

Effects of Buckwheat on the Activities of Pancreatic Digestive Enzymes in Streptozotocin-Induced Diabetic Rats (메밀급여가 Streptozotocin 유발 당뇨쥐의 췌장 소화효소 활성에 미치는 영향)

  • This study was undertaken in order to elucidate the effects of raw, roast and steamed buckwheat on fecal protein, Pancreas weight, the activities of $\alpha-amylase,$ chymotrypsin and lipase 91 the pancreas, and $\alpha-amylase,$ chymotrypsin and trypsin activities of the feces in streptozotocin-induced diabetic rats. Fecal proteins of raw, roast and steamed buckwheat diabetic groups were increased up to 99%, 91%, 103%, respectively compared to those of the diabetic control group. Feeding of buckwheat diet increased pancreas weight, especially raw buckwheat diabetic group(p<0.05). Pancreatic chymo-trypsin activity was decreased in buckwheat diabetic groups compared to diabetic control group, wheres any significant difference was observed in $\alpha-amylase$ and lipase activities. Fecal chymotrypsin activi-ty was significantly increased in all buckwheat diabetic groups. Fecal trypsin activity was increased in roast buckwheat diabetic groups compared to diabetic control group and fecal $\alpha-amylase$ activity in buckwheat diabetic group was not significantly different. These results suggest that feeding of buckwheat diet enhances the impaired exocrine pancreatic function of diabetic rat.

Nutritional Evaluation of Tofu Containing Dried Soymilk Residue(DSR) 1. Evaluation of Protein Quality (건조비지 첨가 두부의 영양적 품질평가 1. 단백질의 영양가)

  • The effect of dried soymilk residue (DSR) on protein quality of tofu was studied. The amount of added DSR into soybean water extract was corresponding 10% (dry basis) of soybean used in tofu manufacturing. Proximate composition and in vitro protein qualities of soybeans at different stages of the conversion into tofu have also been investigated. Partially substituted tofu with DSR (TDSR) had higher moisture content (80.6%) than that of tofu prepared in traditional manner (TT). TDSR contained lower content of protein (38.9%) and total lipid (26.9%) compared to 45.8% of protein and 34.3% of total lipid for TT. A large amount of trypsin inhibitor (TI) in raw soybeans was diminished and extracted through tofu processing, and only 10~13% of TI in raw soybean remained in both tofu products (TDSR and TT). There was not a considerable difference in amino acid profiles between TT and TDSR, but TDSR had a higher content of lysine than that in TT. in vitro studies showed that TDSR and TT were comparable in terms of both in vitro digestibilities (90% over for four-enzyme digestibility and predicted digestibility) and discriminant computed protein efficiency ratio (2.07~2.14, DC-PER). Unlike those in vitro indices for protein quality, computed protein efficiency ratio (C-PER) of TDSR was much lower (1.4) than that of TT (1.95). It was revealed that C-PERs of tofu products were not in agreement with rat-PERs (1.7~1.9) in previous reports except for TT. However, DC-PER assay was more recommendable for protein quality of tofu products than C-PER assay.

성장율 및 사료효율에 있어서 키토산의 첨가급여가 육용계 두 품종에 나타나는 상관반응에 관한 연구

  • Two experiments were conducted to investigate the response of chitosan supplementation in diet on the major economic traits of broiler in two different breeds. In the both experiments, the Arbor Acres and Ross breeds were used as experimental stocks and two groups were assigned in each breed. The control group birds(CON) were fed with basal diet only and the experimental group birds(EXP) were fed with basal diet added with 10.5mg chitosan/bird/day. The chitosan was supplied to birds from day-old in experiment 1 and from 15-day-old in experiment 2. In experiment 1, the mean body weight at 35-day-old were significantly(P〈0.05) heavier by 121.2 g and 243.7 g in the EXP groups than in the CON groups of Arbor Acres and Ross, respectively. Whereas, the mean body weights at 35-day-old in experiment 2 were lighter by 91.7 g and 70.2 g in the EXP groups than in the CON groups of Arbor Acres and Ross, respectively : however, the comparisons between breeds in the mean body weight at 35-day-old did not show significant difference in each other in both breeds. In the mean feed conversion ratio of Arbor Acres from 14 to 35-day old in experiment 1, it did not show significant difference between EXP and CON groups although the feed conversion ratio of the EXP group of Ross was significantly higher(P〈0.05) than the CON group. In experiment 2, the feed conversion ratios from 14 to 35-day-old did not show significant differences between the two breeds. The percentage of mean abdominal fat depositions of EXP groups in both breeds In experiment 1 were significantly(P〈0.05) higher than those of CON groups. And the percentage of mean abdominal fat deposition of Ross was significantly(P〈0.05) lower than that of Arbor Acres. In experiment 2, the percentage of mean abdominal fat depositions did not show significant difference between EXP and CON groups in both breeds. Whereas, the interaction effects between breed and experimental groups on the above economic traits did not show significant in both experiments.

Antioxidative Effect and Digestive Enzyme Inhibition of Grape Seed Extract (GSE) (포도씨 추출물의 항산화 효과 및 소화효소 저해 효과)

  • The purpose of this study is to investigate the antioxidative activity and digestive enzyme inhibition of grape seed extract (GSE). The GSE was tested for its effect on various antioxidative potentials (scavenging activities of DPPH radical, superoxide anion radical and hydroxyl radical) and inhibitory effect of various digestive enzymes (trypsin, $\alpha$-chymotrypsin, $\alpha$-amylase, $\beta$-glucosidase and lipase). DPPH radical scavenging activity ($SC_{50}$, 50% scavenging concentration) of GSE was 4.76${\pm}$0.27 ppm while those of positive controls (EGCG and vitamin C) were 2.22${\pm}$0.12 ppm and 9.50${\pm}$0.72 ppm, respectively. $SC_{50}$ value of GSE against superoxide anion radical and hydroxyl radical were 3.82${\pm}$0.07 ppm and 803.23${\pm}$27.16 ppm, respectively. In addition, $IC_{50}$ values of GSE against trypsin, $\alpha$-chymotrypsin, $\alpha$-amylase, $\alpha$-glucosidase and lipase were 2.17${\pm}$0.59 ppm, 7.46${\pm}$1.25 ppm, 18.25${\pm}$3.54 ppm, 12.30${\pm}$1.12 ppm, and 653.23${\pm}$79.34 ppm, respectively. These results suggest that GSE may be useful for the prevention or treatment of obesity.

  • This experiment was planned to investigate the effects of delayed sealing on fermentation and digestibility of grass silage. The results are summarized as follows: 1. Internal temperature of roll bale silage during storage was $26-28^{\circ}C$ in control, but in delayed sealing it was up to $55^{\circ}C$ by heating with delaying seal. 2. The lactic acid contents was markedly decreased in delayed sealing. $NH_3-N/T-N$ was tended to increase. In the chemical composition of silage, ADIN/T-N, fiber and lignin contents were increased in delayed sealing. 3. Crude protein digestibility was lowered in delayed sealing as compared with control. Fiber digestibility was higher in delayed sealing than in control. 4. The ruminal pH was lowered in delayed sealing. Difference of $NH_3-N$ concentration between control and delayed sealing was not observed. Concentration of total volatile fatty acids (total VFAs) was lower and proportion of acetic acid of total VFAs was higher in delayed sealing than in control.

Nutrient Utilization of Broiler Litter and Bakery By-product Ration in Sheep (육계분-제과부산물 발효 완전혼합사료(TMR)의 면양 체내에서의 영양소 이용성 평가)

  • This study was conducted to determine the effect of feeding a total mixed ration(TMR) of broiler litter(BL) and bakery by-product(BB) with additional BL or rice straw incorporated at 10% of dietary DM as a roughage source on behavior pattern, nutrient intake, digestibility, digestible nutrient intake, ruminal and blood parameters, and N balance of sheep. All the treatment diets were formulated to be isoenergetic[total digestible nutrients(TDN) 66.9%]. Compared with the conventional formulated feed - rice straw feeding system(control), feeding TMR with BL(T1) or rice straw(T2) at 10% of dietary DM resulted in reduced eating, ruminating and total chewing time(P<0.05), similar DM intake, low(P<0.05) digestible DM, OM, fiber and total nutrients intake, low(P<0.05) nutrients digestibilities except EE, similar ruminal characteristics(pH, VFA concentrations and ratios, efficiency of carbohydrate fermentation, NH3-N), and favorable N digestion and retention. There were no differences in the above parameters between T1 and T2 with the exception of increased(P<0.05) eating, ruminating and total chewing time for T2. These results suggested that a TMR of BL and BB with or without rice straw may replace the conventional formulated feed and rice straw in ruminant diets successfully and furthermore feeding the TMR with rice straw made sheep behavior pattern more favorable.

Comparison of the Chicken Large Intestine to the Large Intestine of Pigs and the Rumen of Cows (닭의 대장을 돼지의 대장이나 소의 제 1위 (Rumen)와 비교 연구)

  • The large intestine of the chicken differs both anatomically and physiologically from the pig's large intestine and the men of the cow. The chicken's large intestine is less developed than the pig's large intestine or the cow's lumen. This paper summaries these differences. The chicken's large intestine contains a microbiological population similar to that found in the rumen. The chicken's caeca especially contains a large number of microorganisms, but this population varies according to age, fred, maturity, antibiotic use and etc.. Protein is an essential nutrient for the formation of intestinal microvilli. A study showed that the length of the small intestine was 63 % of the total gastrointestinal tract (GIT) length, while caecum was 8.1 %, and the colon and rectum were 4.6 %. The establishment of the microbial population of the small intestine occurs earlier than that of the caeca, but the identity of approximately 90 % of microbial population of the chicken GIT is hon. Recent studies have shown that energy, volatile fatty acid (VFA) and electrolytes that are found in the large intestine may be absorbed to a certain degree. The chicken small intestine is the primary location for digestion with a variety of enzymes being secreted here. Much research is being conducted into the digestion of sucrose thermal oligosaccharide caramel (STOP), fructooligosaccharides (FOS), mannanoligosaccharide (MOS), galactooligosaccharides (GOS) and isomalto-oligosaccharides (IMO) in the chicken caeca and large intestine. Excessive fibre content in the feed has detrimental effects, but proper fibre supplementation to chicken diets can improve the length and capacity of the small intestine.

Effect of Recombinant CagL Immunization on the Gastric Diseases Induced by Helicobacter pylori in Mongolian gerbils (CagL 재조합 단백질 접종후에 Mongolian gerbil에서 나타나는 Helicobacter pylori 감염에 대한 반응)

  • Helicobacter pylori is an important factor of chronic gastritis, digestive ulcer, and stomach cancer. CagL, a virulence factor of H. pylori, is well-known as a pilus protein which acts as adhesion to host cell and a component of Type 4 secretion system. In this study, we evaluated the protective response of recombinant CagL protein (rCagL) using Mongolian gerbil animal model for H. pylori infection. The cagL gene was cloned from 26695 H. pylori followed by over-expression and purification of the protein in E. coli. Mongolian gerbils were immunized with rCagL protein mixed with aluminum adjuvant via intramuscular injections once a week during 4 weeks. At a week after the last immunization, the Mongolian gerbils were administrated with H. pylori 7.13 strain into the stomach and sacrificed to measure antibody titer on rCagL by ELISA and bacterial colonization in the stomach, and to examine the histopathological changes and cytokine expression at 6 week after challenge. Antibody titers on recombinant protein were significantly increased from a week after the first immunization. There was no significant change of the number of bacterial colony between control group and immunized group. The relative stomach weight was significantly decreased in immunized group, but the significant change of histopathological assessment was not observed in the stomach. Cytokine expression such as IL-$1{\beta}$ and KC also was not significantly different between control and immunized groups. These results indicate that rCagL could effectively induce the formation of the specific IgG antibodies. However, bacterial colonization and histopathological lesions could not be inhibited by the immunization in the stomach, indicating not enough protection against H. pylori infection. We consider that along with CagL other adequate antigens could be needed stimulating immune response and inducing protective effects against gastric disease, and also a better adjuvant could be considered.

Effects of Different Dietary Protein Sources on Apparent Digestibility and Growth in Juvenile River Puffer Takifugu obscurus (단백질원료의 종류에 따른 황복(Takifugu obscurus) 치어의 소화율 및 성장 평가)

  • An 8-week feeding trial was conducted to determine the apparent dry matter and protein digestibility of seven different dietary protein sources by juvenile river puffer Takifugu obscurus. Eight diets containing white fish meal (WFM), brown fish meal (BFM), squid liver powder (SLP), krill meal (KM), leather meal (LM), soybean meal (SM), or fermented soybean meal (FSM) were prepared by mixing a basal diet (BD) with one of the seven test ingredients at a ratio of 7 to 3. Fish averaging $10.8{\pm}0.04g$ were fed the experimental diets in triplicate groups. The apparent dry matter digestibilities of BFM, WFM, FSM, SLP, LM, SM, and KM were 80, 78, 72, 67, 56, 55, and 54%, respectively, while the corresponding apparent protein digestibilities were 96, 96, 93, 92, 89, 88, and 86. The weight gain of fish fed BFM was significantly greater than that of fish fed SM or LM (P<0.05), while it was not significantly different from that of fish fed the BD, WFM, FSM, BFM, SLP, or KM. These results indicated that WFM, BFM, FSM, and SLP are potentially good protein sources for replacing fish meal in river puffer feed.
