Laboratory culture of the native kelp Kjelimaniella crassifolia and the introduced species Laminaria japonica in east coast of Korea were compared at each stage of their life cycles. In the zoospore stage, L. japonica grows optimally at a water temperature of $15{\~}20^{\circ}C$ achieving $95\%$ spore release in 24 hours, whereas K. crassifolia requires 48 hours to achieve $90\%$ spore release in these conditions. Good growth of gametophytes occurred at $10^{\circ}C$ and $15^{\circ}C$ in both species. L. japonica grows optimally under high light intensity ($80{\~}120{\mu}mol{\cdot}m^{-2}s^{-1}$) while K. crassifolia grows best under low light intensity ($40{\~}60{\mu}mol{\cdot}m^{-2}s^{-1}$). Growth of juvenile sporophytes of L. japonica was good in various water temperatures ($10{\~}20^{\circ}C$) and light levels ($40{\~}120{\mu}mol{\cdot}m^{-2}s^{-1}$) while K. crassifolia grew to optimal blade length only under specific conditions ($10{\~}40{\mu}mol{\cdot}m^{-2}s^{-1}$). While the optimal culture conditions for K. crassifolia were more constrained than those of L. japonica which tolerated a wide range of water temperatures and light intensities, the laboratory culture conditions for both of these species reflect the natural environment in which these species are found.

Growth tests on the Wando and Baengnyeongdo cultivars of Saccharina japonica were performed at the Myeongcheon and Gyedo aquafarms, Goheung in Jeollanamdo, from February to July in 2003. Five environmental conditions and 2 traits were measured monthly. The data were used to analyze the growth patterns, relationships between traits and principal component. Box plots were used to display the growth patterns. Scatter plots and regression and correlation coefficients were used to determine the strength of relationships between the traits. A principal component analysis revealed that the first principal component explained more than 91.4% and 90.5% of the total sample variance in the Myeongcheon and Gyedo aquafarms. From the viewpoint of the economic traits (blade length, blade weight), the growth of populations from the Gyedo aquafarm was stronger than that of those from the Myeongcheon aquafarm, and the growth of the Baengnyeongdo cultivar was superior to that of the Wando one.

Effects of the various dietary additives on growth and tolerance of abalone Haliotis discus hannai to the stresses were determined in the 16-week feeding trial. Seventy juvenile (an initial body weight of 4.2 g) abalone per container were randomly distributed into 21, 50 L plastic rectangular containers each. The six kinds of experimental diets were prepared: control (CON) with no additive, by-product of green tea (BPG), extract of figs (EF), extract of green tea (EG), commercially available product of Hearok (PH), and Haematococcus (HC). In addition, dry sea tangle (ST) was prepared to compare the efficiency of the experimental diets. Fishmeal, soybean meal and shrimp head meal were used as the protein source, and dextrin, sea tangle powder and wheat flour, and soybean oil and fish oil were used as the carbohydrate and lipid sources, respectively in the experimental diets. The experimental diets were fed to abalone once a day at a satiation level with a little leftover. The feeding trial lasted for 16 weeks. At the end of the 16-week feeding trial, abalone was exposed to the different types of stresses (air exposure, and sudden changes of rearing temperature and salinity). Survival of abalone fed the sea tangle was highest. However, weight gain of abalone fed the EF, EG and PH diets was significantly (P<0.05) higher than that of abalone fed the BPG diet or dry sea tangle. Shell length of abalone fed the all experimental diets was significantly (P<0.05) higher than that of abalone fed the dry sea tangle. Accumulated mortality of abalone fed the sea tangle was low when exposed to the different types of stresses. Also, relatively low mortality was achieved in abalone fed the HC and EF diets. In considering these results, it can be concluded that the various sources of additives is effective to improve production of abalone, and Haematococcus and extract of figs can be considered as dietary additives to improve resistance of abalone against the different types of stresses.
A 20-week growth trial was conducted in flow-through aquarum system to investigate the practical dietary carbohydrate sources for juvenile abalone (Haliotis discus hannai). Four replicate grops of the abalone averaging 0.125g were fed one of eight diets containing 24.2% wheat flour (WF), 20% dextrin (DEX), 20% sucorse (SUC), 10% $^{\alpha}$-potato starch+10% $^{\beta}$-potato starch (ab-S), 15% $^{\alpha}$-potato starch (a-S15), 20% $^{\alpha}$-potato starch (a-S20), 25% $^{\alpha}$-potato starch (a-S25), or mixture (MIX) with practical ingredients such as soybean meal, corn gluten meal, cotton seed meal and heat flour. In addition, these formulated diets were compare with macroalgae such as dried sea mustard Undaria (D-SM) or dried sea tangle Laminaria(D-ST). Survival rate, weight gain, shell growth and soft body weight of abalone were not significantly affected by the different dietary carbohydrate sources (P>0.05), whereas those fed a-S15 diet were slightly low. These values of abalone fed D-ST were lowest (P<0.05), followed by those fed D-SM. Lipid contents of soft body from abalones fed a-S25, D-ST or D-SM were significantly lower than those of abalone fed other diets (P<0.05). These data indicate that abalone can equally utilize any carbohydrate sources used in this study.

Investigation of optimal temperature, irradiance and photoperiod conditions for free-living gametophytes of Saccharina sculpera for natural resources conservation and mass cultivation of endangered species in the eastern coast of Korea. Induction of regeneration and maturation of the free-living gametophytes of S. sculpera were cultured at temperatures (5, 10, 15, 20 and $25^{\circ}C$), irradiance (10, 20, 40, 60 and $80{\mu}mol\;photons\;m^{-2}\;s^{-1}$ and photoperiods (14:10, 12:12 and 10:14 h L:D). The female gametophyte were actively regenerated without reaching sexual maturity under $10^{\circ}C$, $20{\mu}mol\;photons\;m^{-2}\;s^{-1}$ and 12:12 h (L:D) conditions. In contrast, the conditions for male gametophytes were slightly different at $15^{\circ}C$, $10{\mu}mol\;photons\;m^{-2}\;s^{-1}$ and 12:12 h (L:D). The sexual maturation of female and male gametophytes took place under $15^{\circ}C$, $40{\mu}mol\;photons\;m^{-2}\;s^{-1}$, 14:10 h (L:D) and $10^{\circ}C$, $20{\mu}mol\;photons\;m^{-2}\;s^{-1}$, 10:14 h (L:D), respectively. These results provide basic information for controlling the regeneration and maturation of free-living gametophytes for conservation and utilization of S. sculpera.

To investigate the contribution of macroalgae to biogeochemical nutrients and carbon cycles, we measured the uptake rates of nutrients and $CO_2$ and characteristics of fluorescence of Saccharina japonica (Laminaria japonica Areschoug) using an incubation method in an acrylic chamber. From January to May 2011, S.japonica was sampled at Ilkwang, one of well-known macroalgae culture sites around Korea and ranged 46~288 cm long and 4.8~22.0 cm wide of whole thallus. The production rate of dissolved oxygen by S. japonica (n=25) was about $6.9{\pm}5.8{\mu}mol\;g^{-1}$ fresh weight(FW) $h^{-1}$. The uptake rate of total dissolved inorganic carbon ($TCO_2$), calculated by total alkalinity and pH, was $8.9{\pm}7.9{\mu}mol\;g^{-1}\;FW\;h^{-1}$. Mean nutrients uptake were $175.6{\pm}161.1\;nmol\;N\;g^{-1}\;FW\;h^{-1}$ and $12.7{\pm}10.1\;nmol\;P\;g^{-1}\;FW\;h^{-1}$. There were logarithmic relationships between thallus length and uptake rates of nutrients and $CO_2$, which suggested that younger specimens (<100-150 cm) were much more efficient at nutrients and $CO_2$ uptake than old specimens > 150 cm. There was a positive linear correlation ($r^2$=9.4) existed between the dissolved oxygen production rate and the $TCO_2$ uptake rate, suggesting that these two factors may serve as good indicators of S. japonica photosynthesis. There was also positive linear relationship between maximal quantum yield ($F_v/F_m$) and production/uptake rates of dissolved oxygen, $TCO_2$ and phosphate, suggested that $F_v/F_m$ could be used as a good indicator of photosynthetic ability and $TCO_2$ consumption of macroalgae. Maximum relative electron transport rate ($rETR_{max}$) of S. japonica increased as thallus grew and was high in distal part of thallus which may be resulted from the increase of photosynthetic cell density per area. The annual $TCO_2$ uptake by S. japonica in Gijang area was estimated about $1.0\sim1.7{\times}10^3C$ ton, which was about 0.02-0.03% of carbon dioxide emission in Busan City. Thus, more research should be focused on macroalgae-based biogeochemical cycles to evaluate the roles and contributions of macroalgae to the global carbon cycle.
