  • Sericulture is a main insect industry in Sangju (Kyungpook, Korea). This study reports an occurrence of a viral disease in the sericulture farms in 2020. More than 20% silkworm larvae (Bombyx mori) suffered diarrhea and melted tissues with pathogenic lethality at 4th or 5th instars. PCR diagnosis showed a positive response against B. mori nucleopolyhedrosis virus (BmNPV) infection. Tissue extract of the infected larvae was applied to healthy larvae by a leaf-dipping method and exhibited the same viral symptoms. The viral extract was used to be overlaid on Sf9 cells. The infected Sf9 cells showed polyhedra in the cytoplasm. These results indicate that the silkworm larvae reared in the sericulture farms in Sangju were infected with BmNPV.

  • The nuclear polyhedrosis viruses of Bombyx mori (BmNPV) and Hyphantria cunea (HcNPV) were characterized by electron microscopic observation, SDS-PAGE of polyhedral and virion proteins, and restriction endonuclease analysis of viral DNAs. Polyhedra of BmNPV were octadecahedral in shape with the diameter of $3 \mu\textrm{m}$, whereas those of HcNPV showed irregular appearances having the diameter of $1.5~2\mu\textrm{m}$. Under alkaline protease inactivated condition, polyhedral proteins of two NPVs were resolved into a major polypeptide, 30~31 KD, and several minor polypeptides by SDS-PAGE. Examination of virion proteins by silver staining after SDS-PAGE showed that BmNPV was composed of 47 polypeptides with M.W. range of 9.6~112 KD and HcNPV was composed of 48 polypeptides with M.W. range of 9.4~111 KD. The approximate genome size of two NPVs were determined by restriction endonuclease analysis: BmNPV and HcNPV were 114.6 Kb, respectively.

  • Practical application of silkworm artificial diet is very desirable to save labour in sericultural industry as the problem of labour shortage is becoming serious in Korea. However, silkworms reared on the artificial diet are more susceptible to viruses than those reared on mulberry leaves because of the lower anti-viral activity of gut juice of silkworms grown on artificial diet compared with that of silkworms grown on mulberry leaves. In this study, authors investigated the varietal difference of silkworm larvae, Bombyx mori L., reared on artificial diet which contained 20 percent of dried mulberry powder, in the susceptibility to nuclear polyhedrosis virus (NPV). The results showed that there is no difference in susceptibility to NPV among tested varieties when high concentration of NPV was admitted to silkworm larvae, but varietal difference appeared in lower concentration admitted. Among 7 hybrids tested, Hansaeng 1${\times}$Hansaeng 2 was most resistant to NPV with an $LC_{50}$ of 2.7${\times}$10$\^$6/ and Jam 111${\times}$Jam 112 was also more resistant comparatively than other hybrids.
