Direct Conversion of Raw Starch to Maltose in an Agitated Bead Enzyme Reactor using Fungal $\alpha$-Amylase (분쇄마찰 효소반응계에서 Fungal $\alpha$-Amylase를 이용한 생전분의 직접전환에 의한 Maltose 생산)

  • Direct conversion of raw starch without liquefaction to maltose using maltose-forming fungal a-amylase (Fungamyl) was carried out in an agitated bead enzyme reactor (bioattritor). The reaction rate in bioattritor was comparable with conventional method which utilized liquefied soluble starch. Moreover the extent of maltose formation increased substantially compared with conventional method; from 150 g / I of raw starch, around 95 g/l of maltose was formed and 72% of maltose content in sugar mixture was achieved. Especially, pH influenced greatly not only on total sugar formation from raw starch in bioattritor but also on maltose content in sugar mixture. The optimal pH for maltose formation from raw starch was shifted into the weak alkaline pH, the optimal pH of 8.0~9.0 in bioattritor contrast to pH of 5.0~5.5 for liquefied starch. The maltose formation and content were also affected by the amounts of Fungamyl added and raw starch concentration. Consumption of maltose-forming Fungamyl can be substantially reduced by supplementary addition of starch liquefying a-amylase (Termamyl).

  • The organic/organic core-shell composite polymer for nonwomen binder were synthesized by stage polymerization of methyl methacrylate and styrene with ammonium persulfate after preparing monomer pre-emulsion in the presence of anionic surfactant. We study the effect of initiator concentration, $0.79{\times}10^{-3}{\sim}3.16{\times}10^{-3}mol/L$ for core polymer, $2.0{\times}10^{-4}{\sim}8.0{\times}10^{-4}mol/L$ for shell polymer, sulfactant concentration, $1.45{\times}10^{-5}{\sim}4.15{\times}10^{-5}mol/L$ for core polymer, $0.73{\times}10^{-5}{\sim}2.91{\times}10^{-5}mol/L$ for shell polymer on core-shell structure of polymethyl methacrylate/polystyrene and polystyrene/polymethyl methacrylate. Emulsion stability was major test method, particle size and particle size distribution were measured using particle size analyzer and the morphology of the core-shell composite polymer was determined using transmission electron microscope, glass temperature was also measured using differential scanning calorimeter.

A Study on Whitening and Anti-inflammatory Effects of Eriobotrya Japonica Leaf Extracts with Different Extraction Methods (추출 방법에 따른 비파엽 추출물의 미백 및 항염활성에 관한 연구)

  • In this study, we prepared Eriobotrya japonica leaf extracts by several extraction processes and then evaluated their biological activities for their potential application as a new raw material of functional cosmetic. Their whitening effects were measured by tyrosinase inhibitory activities, and anti-inflammatory effects were determined by inhibitory activities of nitric oxide (NO) production. Among the several extracts obtained from E. japonica leaf, supercritical fluid extract showed tyrosinase inhibitory activities at the concentration of 10%. Inhibitory activity on NO production effect related to anti-inflammatory efficacy was in the order: supercritical fluid extract > ethanol extract > hot water extract. According to the results of MTT assay, cell cytotoxicity was not observed at all concentrations except for a 5% concentration of the 70% ethanol extract. For whitening effects, 30% ethanol and 70% ethanol extract showed mushroom tyrosinase inhibitory activity at the concentration of 5%. These results indicated that E. japonica leaf extracts could have the functional effects when they are added as ingredients in cosmetics.

  • This is the research about the concentration of trace citrus flavor components in water by pervaporation. We have investigated the permeation characteristics depending on the material and formation of membranes using four siloxane-based polymer composite membranes. We have also chosen the optimal membrane and investigated the permeation characteristics depending on the feed temperature, concentration and flow rate. And then it has been analyzed by using resistance-in series model. In the permeation experiment of citrus essence aroma model solution through the four siloxane-based polymer composite membranes, PVDF/POMS membranes have showed the best flavor flux and enrichment factor. As a result of the permeation experiment depending on the feed temperature, concentration and flow rate, we can find that as the feed temperature and concentration increase, the flavor flux increases while the enrichment factor decreases. And the flavor flux and enrichment factor increased as the flow rate increases.

Analysis of the Effects of CO Poisoning and Air Bleeding on the Performance of a PEM Fuel Cell Stack using First-Order System Model (일차계 모델을 이용한 고분자전해질 연료전지 스택의 CO Poisoning 및 Air Bleeding 효과 분석)

  • We analyze the effects of CO poisoning and air bleeding on the performance of a PEM (polymer electrolyte membrane) fuel cell stack fabricated using commercial MEA (membrane electrode assembly). Dynamic response data from the experiments on the performance of a stack are identified by obtaining steady-state gains and time-constants of the first-order system model expressed as a first-order differential equation. It is found that the cell voltage of the stack decreases by 1.3-1.6 mV as the CO concentration rises by 1 ppm. The time elapsed to reach a new steady state after a change in the CO concentration is shortened as the magnitude of the change in the CO concentration increases. In general, the steady-state gain becomes bigger and the time-constant gets smaller with increasing the air concentration (air-bleeding level) in the reformate gas to restore the cell voltage. However, it is possible to recover 87%-96% of the original cell voltages, which are measured with free of CO, within 1-30 min by introducing the bleed air as much as 1% of the reformate gas into the stack.

  • 본 연구에서는 자연하천에서 Rhodamine-WT를 이용하여 추적자 실험을 수행하고 이를 바탕으로 오염사고대응예측모델에 대한 검증을 실시하고자 하였다. 최근 국내에서는 다양한 형태와 규모의 수질오염사고가 매년 수십건 이상 발생하고 있으며, 따라서 수치모형 기반의 수질오염 사고 대응 예측시스템에 대한 높은 신뢰성이 요구되고 있다. 수질사고에 노출되어 있는 지표수를 각종 용수로서 안전하게 공급하기 위해서는 정확한 수질예측이 필수적이며, 이를 위해서 수십 년간 연구되어 온 수질모델을 오염사고 대응예측시스템에 적합하도록 정확성과 신뢰성을 갖추기 위한 연구가 진행되어야 한다. 수치 모형을 이용한 물질의 이송 및 확산 모의에서는 오염물질 도달시간과 확산 농도를 결정하는 것이 가장 중요한 요소이므로 이송 및 확산 모의에 대한 검증 기법 개발 및 적용이 필요하다. 본 연구에서는 낙동강수계 지류하천인 감천에서 추적자 실험을 4회 실시하여 측정한 수리량과 농도 실측치를 이용하여 분산계수를 종 횡분산계수 산정이 가능한 2차원 유관추적법을 적용하여 산정하였다. 각 단면에 유속은 ADV-3차원 유속계인 Flow-Tracker를 사용하여 도섭으로 측정하였으며 하천의 흐름 방향의 직각으로 측선을 설치하고 펌프를 이용하여 채수를 한 다음 Rhodamie WT의 농도를 측정하였으며 측선의 위치 보정은 GPS를 통하여 보정하였다. 측정 자료를 이용하여 2차원 유관추적법으로 분산계수를 산정한 결과 각각의 측선에 따라서 다소 차이가 발생하였으며, 일부 구간에서는 이론식으로 계산한 분산계수와 근사한 값이 나타났다. 이는 사주가 매우 발달하고 만곡이 많은 실험구간의 특성상 Elder와 Fischer 식으로 계산한 값과 차이가 발생할 가능성이 높은 구간이기 때문인 것으로 판단된다. 또한 하폭에 대한 수심비가 커질수록 분산계수도 증가하고 평균유속에 대한 전단유속에 비에 비례하는 것으로 나타났다.

Nutrient Variations in the Jindong Bay during Summer by Ecosystem Modeling (해양생태계모델에 의한 하계 진동만의 영양염변동)

  • During summer, the DIN (dissolved inorganic nitrogen) and DIP (dissolved inorganic phosphate) observed in the Jindong Bay in the southern sea of Korea show much higher values in the inner area of the bay. In general, they have high values in the upper (0-1 m) and lower layers (8 m-bottom), but are relatively lower in the middle layer (1-8 m). These features in their distribution are examined using an ecosystem model with considering the wind, tidal current, horizontal gradient of water density and residual flow. The experiments were focused on how to influence nutrients associated with these conditions. In the experiment with tide-induced residual flow, the values of nutrients appeared lower than the observation, and were well corresponded to it when the effects of wind, tide-induced residual current and horizontal gradient of water density were additionally imposed. A statistical analysis identifies these results. This paper suggests that variation of nutrient in the Jindong Bay during summer should be seriously a(footed wind-driven current by the wind and density-driven current is induced by the horizontal gradient of water density as well as tidal current.

A Study on Foam Mixing Characteristics in Steady State to Enhance the Performance of Proportioner for Foam System (포 소화설비용 소화약제 혼합장치의 성능향상을 위한 정량 혼합특성에 관한 연구)

  • This study describes to analyze foam mixing characteristics in steady state to enhance the performance of proportioner for foam system designed to accurately proportion a foam liquid concentrate into a water stream up to constant concentration. The proportioner developed is experimentally evaluated in performance evaluation system consisted of a pump, tanks, pressure gauges, flow meters, a nozzle. As a result, the foam mixing performance of the line proportioner is found to increase with increased the water flow rate due to the venturi effect and with increased the cross-sectional area of the orifice and is analysed with 3 % in the error rate of $\pm4%$. For the pressure proportioner, the foam mixing performance is analyzed to increase with increased the water flow rate and with increased the inlet pressure and is analysed with 3% in the error rate of $\pm2%$.

Performance of Soil Flushing for Contaminated Soil Using Surfactant (계면활성제를 이용한 오염 토양 세정 성능 평가)

  • In this study, a series of experiments were carried out to remove total petroleum hydrocarbon(TPH) and toluene by soil flushing. In batch experiments, Triton X-100 and SWA 1503 showed TPH removal efficiency of 79.0% and 69.0%, respectively. Although the TPH removal efficiency increased as the surfactant was increased in the concentration range 1-11mmol/L, the optimum concentration was 1mmol/L, considering the ratio of the removal efficiency to the amount of surfactant injected. In column experiment, the optimal velocity was 0.3mL/min. The physical aquifer model(PAM) result revealed that the soil flushing removed as much as 5.5% of the toluene under 3 pore volume(PV) conditions. To improve the soil flushing efficiency, it is necessary to find optimal condition through recirculation or reuse of surfactant.

Separation of $CO_2$ from Syngas Using Gas Hydrate Formation (가스 하이드레이트 동공점유특성을 이용한 합성가스로부터의 $CO_2$ 분리 공정)

  • 석탄가스화복합발전과 연계하여 사용할 수 있는 $CO_2$분리법으로 연소 전 탈탄소화는 연료가 연소되기 전에 $CO_2$를 회수하는 방법으로 현재 여러가지 분리법이 사용되고 있다. 본 연구에서는 가스 하이드레이트의 다양한 응용 분야 중 이산화탄소 격리분야에서 합성가스로부터 $CO_2$롤 효과적으로 분리/회수하기 위하여 가스 고형화법에 관한 연구를 진행하였다. 가스 하이드레이트 형성과정에서의 반응 특성을 살펴보기 위하여 순수계와 촉진제 첨가계(TBAB, TBAF, THF)에 대하여 반응시간에 따른 가스소모량 및 기상의 $CO_2$ 조성 변화를 측정하였다. 그 결과 하이드레이트 상에 고농도의 $CO_2$가 포집되는 것을 확인 할 수 있었다. 순수계와 THF 첨가계의 경우 가스 소모량이 다른 계에 비하여 높게 나타났다. 이는 순수계의 경우 구조-I의 큰 동공과 작은 동공에 모두 기체가 점유되기 때문이며, THF 첨가계의 경우 구조-II의 큰 동공에만 기체가 점유되지만 THF의 첨가로 인해 전환율이 증가되기 때문이다. 반면, TBAF와 TBAB 첨가계의 경우에는 상재적으로 낮은 가스 소모량을 보였다. 기체 소모량이 큰 경우 최종 기상의 $CO_2$ 조성이 낮게 나타났다. 그리고 모든 실험조건에서 1시간 이내에 하이드레이트 형성반응이 종결되는 것을 확인할 수 있었다. 또한, 촉진제 첨가에 의한 하이드레이트의 구조적인 변화를 확인하기 위하여 Raman 분광법과 $^1H$-NMR을 이용하여 혼합가스 하이드레이트를 분석하였다. 본 실험으로 얻어진 결과는 가스 고형화법을 이용한 합성가스 분리 공정 설계 및 개발에 중요한 기초자료가 될 것으로 사료된다.
