Purification and Characterizationof Soluble Acid Invertase from the Hypocotyls of Mung Bean (Phaseolus radiatus L.) (녹두의 하배축에서 분리한 Soluble Acid Invertase의 정제와 특성)

  • The soluble acid invertase ($\beta$-D-fructofuranoside fructohydrolase, EC 3.2.1.26) was isolated and characterized from the hypocotyls of mung bean (Phaseolus radiatus L.). The enzyme was purified to apparent homogeneity by consecutive step using diethylaminoethyl (DEAE)-cellulose anion exchange, Concanavalin (Con) A affinity and Sephacryl S-300 chromatography. The overall purification was about 148-fold with a yield of about 15%. The finally purified enzyme exhibited a specific activity of about 139 $\mu$mol of glucose produced mg-1 protein min-1 at pH 5.0 and appeared to be a single protein by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and nondenaturing PAGE. The enzyme had the native molecular weight of 70 kD and subunit molecular weight of 70 kD as estimated by Sephadex G-200 chromatography and SDS-PAGE, respectively, suggesting that the enzyme was composed of a monomeric protein. On the other hand, the enzyme appeared to be a glycoprotein containing N-linked high mannose oligosaccharide chain on the basis of its ability to bind to the immobilized C on A. The enzyme had a Km for sucrose of 1.8 mM at pH 5.0 and maximum activity around pH 5.0. The enzyme showed highest enzyme activity with sucrose as substrate, but the activity was slightly measured with raffinose and cellobise. No activity was measured with maltose and lactose. These results indicate the soluble acid invertase is a $\beta$-fructofuranosidase.

  • Photostimulated luminescence (PSL) analysis was applied to determine whether some agricultural commodities of Korean and Chinese origins have been irradiated or not. Cereals (brown rice and buckwheat) and legumes (soybean, peanut, fed bean and mung bean) were exposed to gamma irradiation at 0.5 ∼4 kGy The Korean garlic was irradiated at 0.05 ∼0.5 kGy. Cereals and mung bean were tool applicable to the PSL measurement, showing the intermediate values between the lower (700 photon counts, negative) and the upper threshold values (5,000, positive), however PSL values determined for soybean, peanut, red bean and garlic were suitable for being discriminated between nonirradiated and irradiated samples. There was no significant difference in PSL properties accgrding to the sample origins.

  • The alkaline invertase ($\beta$-D-fructofuranoside fructohydrolase, EC 3.2.1.26) was isolated and characterized from the hypocotyls of mung bean (Phaseolus radiatus L.). The enzyme was purified by consecutive step using diethylaminoethyl (DEAE)-cellulose anion exchange, 1st Sephadex G-200, DEAE-Sephadex A50 and 2nd Sephadex G-200 chromatography. The overall purification was about 77-fold with a yield of about 6%. The finally purified enzyme exhibited a specific activity of about 48 $\mu$mol of glucose produced mg-1 protein min-1 at pH 7.0 and appeared to be a single protein by nondenaturing polyacrylamide gel electrophoresis (PAGE). The enzyme had the native molecular weight of 450 kD and subunits molecular weight of 63 kD and 38 kD as estimated by Sephadex G-200 chromatography and SDS-PAGE, respectively, suggesting that the enzyme is a heteromultimeric protein composed of two types of subunits. On the other hand, the enzyme appeared to be not a glycoprotein according to the results of Con A chromatography and glycoprotein staining. The enzyme had a Km for sucrose of 19.7 mM at pH 7.0 and maximum activity around pH 7.5. The enzyme was most active with sucrose as substrate, compared to raffinose, cellobiose, maltose and lactose. These results indicate the alkaline invertase is a $\beta$-fructofuranosidase.

Quality Characteristics of Cheongpomook Prepared with Different Levels of Mungbean Powder (녹두가루 첨가 비율에 따른 청포묵의 품질특성)

  • The objective of this study was to evaluate the quality characteristics of Cheongpomook prepared with five different levels of mungbean powder (0, 25, 50, 75, and 100%). We conducted the Hunter's color values, the rheological characteristics, sensory evaluations, and pasting properties of the Cheongpomook samples. The more mungbean powder was added, the more the luminance and Hunter's a values of Cheongpomook samples were decreased, but in Hunter's a values was reverse. With regard to the rheological properties of the Cheongpomook samples, the more mungbean powder was added, the values of hardness, springiness, gumminess, and chewiness were significantly decreased. In color, taste, and overall quality, the value of 25% mungbean powder added Cheongpomook (MP1) and 50% added Cheongpomook (MP2) were significantly higher than those of others. The resulting RVA viscogram, peak viscosity, hold viscosity, break down, setback, and final viscosity of Cheongpomook were decreased with an increase in mungbean powder, but the pasting temperature was increased slightly. Therefore, an addition of 25% mungbean powder appears to be an acceptable approach to enhance the quality of Cheongpomook without reducing acceptability.

  • The important legumes in Korea are mungbean, red bean, kidney bean and cowpea. Mungbean has traditionally been used for mook (jelly-like starch gel) preparation. Cowpea has recently been utillized for substitution of mungbean. The major use of sediment of red bean is for the sweet paste. The studies related to legume starches and flours (air-dried sediment) in Korea are concentrated on the understanding of the properties of mook. The structure of starch, gelatinization and gelling properties of legume starches and flours are reviewed with emphasis of Korean literatures.

Effects of Temperature on the Oviposition, Feeding and Emergence of the Azuki Bean Weevil (Callosobruchus chinensis L.) in the stored beans (저장두류(貯藏豆類)에 대(對)한 팥바구미의 산란(産卵).섭식(攝食) 및 우화(羽化)에 미치는 온도(溫度)의 영향(影響))

  • This study was conducted to study the effects of temperature on the oviposition, feeding and emergence of the azuki bean weevil(ABW) (Callesobruchus chinensis L.) in the stored beans. And ovipositional preference, hatching and emergence on the preference and non-preference hosts were also investigated. ABW has four generations a year and overwinters as larva stage in the bean grains. Optimum oviposition temperature of the ABW ranges from $25^{\circ}C\;to\;30^{\circ}C$. Oviposition preference in different hosts was in the order of azuki bean, mung bean, soy bean, kidney bean, black-soybean, pea. At $25^{\circ}C$, optimum development temperature emergence of period the ABW averaged 29 days on azuki bean, 31 days on mung bean, 49 days on soy bean, 46 days on black-soybean, 34 days on Pea. Percent hatch in different hosts was in the order of azuki bean, mung bean, soy bean, pea, black-soybean, kidney bean. But percent emergence was in the order of azuki bean, mung bean, pea, black-soybean, kidney bean. Especially, in azuki bean and mung bean, percentage of hatching and emergence was very high. And in soy bean and black-soybean, percentage of hatching was high but emergence was low. The kidney bean was hatching but not emergence at all. The longevity at the different temperatures appeared to be $4{\sim}5$ days at $35^{\circ}C,\;5{\sim}7$ days at $25^{\circ}C{\sim}30^{\circ}C$, $10{\sim}15$ days at $20^{\circ}C\;and\;19{\sim}24$ days at $15^{\circ}C$. Kidney bean of feeding non-preference host oviposited at Seed coat and Endosperm, but no larva developed. And cotyledon and embryo parts were not oviposited at all. The percentage of germination of egg laid bean grain and damaged bean grains was low. Especially, the percentage of germination of damaged bean grains was 0% on azuki bean, 5% on black-soy bean, 7% on Pea.

Effects of Selenium(Se) on Growth and Se Content of Mungbean (셀레늄 시용에 따른 녹두 생육 및 Se 함량 변화)

  • This study examined the effective treatment method of selenium and translocation characteristics of selenium in order to produce mungbean containing selenium. The foliar application of selenium at 3.5, 7, 14, and $28mg/{\ell}$ during the flowering period, yielded a relatively high record of seeds containing $0.41{\sim}3.96mg/kg$ and $0.27{\sim}2.38mg/kg$ of selenium, from the first and second harvesting. However, seeds from the first harvesting contained $52{\sim}71%$ more selenium than the seeds from the second harvesting. On the other hand, seeds from first and second harvesting of the non-treatment group had the same amount of selenium at 0.02mg/kg only. When the foliar application of selenium at $7mg/{\ell}$ was conducted two or three times, seeds from the first to third harvesting contained just the equal amount of selenium. However, when it was conducted only once, seeds from the first harvesting contained 56% and 67% more than seeds from the second and third harvesting, respectively. In seeds of mungbean containing 2.05mg/kg of selenium, cotyledon had 2.99mg/kg of selenium, which was 38% more than seed coat per unit weight. When mungbean sprout was produced, selenium content was 5.51mg/kg, but seed coats by-product of sprouts had 0.78mg/kg of selenium. The growth and quantity of mungbean was not significantly different according to the concentration and the frequency of foliar application of selenium used for in study.

Varietal Differences of Major Chemical Components and Fatty Acid Composition in Mungbean (녹두의 주요 성분함량과 지방산 조성의 품종간 차이)

  • These experiments were conducted to obtain basic information for breeding material on the utility characteristics in mungbean, The crude protein, fat and ash content were 24.3, 0.67, 3.6%, repectively, and the fat content showed varietal differences, whereas the protein and ash content was not significantly differences among the varieties. The negative correlation existed between protein and carbohydrate content, seed moisture and fat content, seed weight and fat content. The unsaturated fatty acid contain 60∼67% and there were oleic, linoleic, linolenic and archidonic acid, and the saturated fatty acid contain 33∼40% and there were stearic, palmitic, behenic and lignoceric acid. The major fatty acids in mungbean were linoleic, linolenic and palmitic acid, and have contained 33.46, 21.87, 21.72, respectively, and oleic, stearic and arachidonic acid contained 5.98, 5.88, 4.87, respectively, and the behenic and lignoceric acid left traces. Highly positive correlation existed between palmitic and linoleic acid, oleic and lignoceric, behenic. However; palmitic and arachidonic, lignoceric acid, oleic and linolenic acid, arachidonic and liniceric acid showed negative correlations with each other. Seed weight of tested varieties showed highly positive correlation with oleic, arachidonic and behenic acid.

Occurrence of Powdery Mildew on Mung bean(Vigna radiatus L.) Caused by Sphaerotheca phaseoli (Sphaerotheca phaseoli에 의한 녹두 흰가루병 발생)

  • Powdery mildew of mug bean was found in Suwon on September 2000. Many white and powdery mycelial colonies appeared on leaves of the plants. Conidia with fibrosin bodies were ellipsoid to elongate-ellipsoid, 25~38$\times$16~22 ${\mu}{\textrm}{m}$ in size and farmed in chains. Conidiophores were erect on superfical mycelium, 67~100 ${\mu}{\textrm}{m}$ in length. Foot cells were cylindric with 37~65$\times$10~11 ${\mu}{\textrm}{m}$ in size. Appressoria were indisdinct. Germ tubes from conidia were simple, not branched. Dark brown cleistothecia were observed on the leaves of mung bean. Cleisto-thecia were globose or subglobose, 85~1130 ${\mu}{\textrm}{m}$ in size. Wall cells were irregularly polygonal with 15~45$\times$7~26 ${\mu}{\textrm}{m}$. Appendages were mycelioid, 6-11 per ascocarp, coloured, throught when mature or paler upwards, 1~7 septa, 192~223$\times$5~7 ${\mu}{\textrm}{m}$ in size. A single sun had in a cleistothecium and was ovoid or subglobose with 62~95$\times$50~67 ${\mu}{\textrm}{m}$ in size. Ascusspore had 8 spores in an ascus and rarely developed, mostly imatuna ellipsoid-ovoid with 16~24$\times$12~16 ${\mu}{\textrm}{m}$. On the basis of morphological characters of the conidial stage and teleo-morph, the fungus was identified as Sphaerotheca phaseoli.
