Effects of Cholic Acid/CDCA and FGF-19 on the Protein Levels of the Endogenous Small Heterodimer Partner (SHP) in the Mouse Liver and HepG2 Cells (생쥐의 간과 HepG2 세포에 있어서 내인성 small heterodimer partner (SHP)의 단백질 수준에 미치는 cholic acid/CDCA 및 FGF-19의 효과)

  • Recent studies determined that a chronic western-style diet increased the endogenous small heterodimer partner (SHP) protein levels in mice. In experiments with cell cultures, chenodeoxy cholic acid (CDCA) treatment increased endogenous SHP protein levels and reduced the degradation rate of exogenously expressed flag-SHP levels in the human hepatoma cell line, HepG2 cells. In addition, bile acid-induced intestinal fibroblast growth factor-19 (FGF-19) increased the half-life of the exogenously expressed SHP when HepG2 cells were transfected with ad-flag-SHP. However, both the expression level and the degradation rate of the endogenous SHP in response to cholic acid and FGF-19 have not been well understood, either in mice or in cultured HepG2 cells. This study examined the effects of cholic acid treatment on the endogenous SHP protein levels in mice and the effects of FGF-19 on the degradation rate of the endogenous SHP protein in HepG2 cells. Mice fed 0.5% cholic acid in normal chow showed an increase in endogenous SHP protein levels during both 12 hr and 24 hr treatment periods as compared to control mice fed only normal chow. In cultured HepG2 cells, treatment with CDCA did not noticeably change the rate of degradation in the endogenous SHP protein from cells not treated with CDCA. Although consistent with the previous studies on the exogenous ad-flag-SHP protein, treatment with FGF-19 significantly decreased the degradation rate of the endogenous SHP protein when HepG2 cells were treated with cyclohexamide. These results suggest that both bile acids and FGF-19 increase the endogenous SHP protein levels in mouse liver and HepG2 cells.

  • Human endogenous retroviruses (HERVs) are fossil viruses that began to be assimilated into the human genome some 30~40 million years ago, and now constitute nearly 8% of the human genome. These ancient retroviruses have since accumulated mutations that have rendered them defective; thus, they have been termed junk DNA. However, recent research indicates that not all HERVs remain silent passengers. Although they have not been shown to be causative of any human disease, endogenous retroviral sequences may become expressed under select pathological circumstances such as neurological disorders, including multiple sclerosis (MS), schizophrenia, and Amyotrophic Lateral Sclerosis (ALS); viral infections, including human immunodeficiency virus (HIV) and herpesvirus; and multiple types of cancers. This review focused on the possible interactions of HERVs and neurological diseases.

  • To investigate the relationship of endogenous retroviruses in peccaries and pigs, a set of degenerate primers was used in this study to amplify peccary retroviral sequences. The sequences of two putative retroviral clones showed close homology to mouse and pig retroviral sequences. The peccary endogenous retroviral sequences are significant in that they are the first such sequences reported in peccary species and repudiate old claims in the literature that peccaries do not have C-type retroviral sequences.

  • We experienced an extremely rare case of lipoid pneumonia combined with empyema. A 53-year-old patient was admitted because of chilling sensation and blood tinged sputum. Simple X-ray and computered tomography(CT) showed a huge homogeneous opacification in the left lower lung field. This patient was diagnosed as endogenous type lipoid pneumonia by a computed tomography guided needle biopsy; decortication and left lower lobectomy are performed. We diagnosed it as endogenous type lipoid pneumonia because it revealed a foreign body reaction and lipid laden macrophage on the pathologic examination.

  • 새 천년으로 들어선 2000년도부터 우리나라의 65세 이상 노령인구는 $7.1{\%}$를 차지하게 되어 본격적인 노령화사회 (UN에서 65세 이상인구가 총 인구의 $7{\%}$ 이상일 경우 노령화 사회로 정의)로 진입하였다. 평균수명의 계속적인 증가에 따라 노인 인구는 급격히 늘어날 전망이며, 따라서 노인성 질환의 발생도 늘어나게 되어 가정 또는 사회가 떠 맡아야할 경제적, 정신적 부담은 더욱 증가하게 된다. 건강하고 행복한 인생의 황혼기를 보내는 것은 인간의 권리이며 누구나 바라는 소망이다. 피부의 노화현상은 심장질환, 암 등에 못지 않게 중요하며, 노인의 경제적, 사회적 활동을 심각하게 위축시키고 있다. 피부노화를 예방하고 노인성 피부질환을 치료할 수 있는 방법을 개발함으로써 노인의 경제, 사회활동을 활동적으로 유지시킬 수 있다. 피부노화는 크게 두 종류로 나눌 수 있다. 그 한가지는 내인성노화(intrinsic aging)로서 세월이 흘러감에 따라 피할 수 없는 노화 현상을 말한다. 두번째는 광노화 (photoaging)로서 오랫동안 햇빛에 노출된 얼굴, 손등, 목뒤 등의 피부에서 관찰되는 노화현상을 말하는 것으로 내인성노화 현상과 자외선에 의한 영향이 합쳐진 결과로 발생한다. 광노화 현상은 자외선의 노출을 피하면 예방할 수 있는 피부노화 현상이다. 내인성 노화는 햇빛에 노출되지 않은 피부에서 주로 관찰된다. 임상적 특징은 비교적 경미하며, 잔주름, 피부건조증, 탄력감소 등을 들 수 있다. 그러나 광노화의 임상적 특징은 내인성 노화에 비하여 심하고, 일찍부터 관찰된다. 내인성 노화에 비하여 굵고 깊은 주름이 발생하며, 잔주름도 많이 발생한다. 햇빛에 노출된 피부에 불규칙한 색소침착이 발생하며 일광흑자 (solar lentigo) 등의 색소질환이 증가한다. 피부가 매우 거칠고, 건조해지며, 탄력성이 감소하여 심한경우 피부가 처지게 된다. 피부노화의 대표적인 증상은 주름살이며, 아직까지 그 발생기전에 대하여는 여러 가지 학설이 있으나 정확히 알려져 있지 않다. 피부에 존재하는 교원질, 탄력섬유등 기질단백질의 손상이 피부 주름살의 주 원인으로 알려져 있다. 또한 얼굴에 존재하는 근육의 분포와 움직임, 유전적 소인, 자외선, 흡연, 폐경, 산화적 손상, 열 등 여러 원인이 복합적으로 작용할 것으로 생각된다. 피부주름살의 원인을 밝히고, 원인인자가 피부주름살을 초래하는 분자생물학적 기전을 이해함으로써 주름살을 예방하고 치료할 수 있는 새로운 방법을 개발할 수 있다.

Effects of Electroacupuncture on Food Intake and mRNA Expressions of the Hypothalamic Cholecystokinin in Rats (전침이 흰쥐에서 먹이섭취와 시상하부의 콜레시스토키닌 mRNA 발현에 미치는 영향)

  • 목적 : CCK는 현재까지 가장 많이 연구된 식후의 포만신호 전달물질로, 음식섭취를 감소하고, 캡사이신 반응성의 미주신경에 의해 위장운동과 위내의 공복감을 억제시킨다 전침의 진통효과 발현기전에 영향을 미치는 항아편양 단백물질로서, 내인성 CCK와 그 수용체(CCK-A와 CCK-B)의 역할은 기존의 연구에서 이미 보고되어 왔다. 이에 착안하여, 본 연구에서는 포만감의 측면에서 전침자극이 내인성 CCK의 발현에 어떠한 영향을 미치는가를 살펴보고자 한다. 방법 : 48시간 절식 쥐 모델을 이용하여, 전침자극 후 30분과 60분 동안, 먹이 섭취량 변화를 측정하고, 먹이섭취량에 영향을 주는 신경전달경로에서 CCK가 관여하는지를 확인하기 위해 미주신경절제술을 시행한 쥐와 비교하였다. 한편 48시간 절식 쥐 모델을 대상으로하여 침자극 후 시상하부의 CCK mRNA 발현변화를 관찰하였다. 결과 : 전침군에서 30분과 60분 뒤의 먹이 섭취가 대조군에 비해 현저히 낮게 관찰되었는데, 포만감에 관련된 침의 이와 같은 효과는 CCK 수용체에 길항작용이 있는 lorglumide와 미주신경절제술에 의해 차단됨을 알 수 있었다. 시상하부의 CCK mRNA의 발현되는 대조군에 비하여 전침군에서 증가하는 경향이 관찰되었으나, 통계학적인 유의성은 확인 할 수 없었다. 결론 : 위의 결과에서, 전침은 포만감에 영향을 미치는 내인성 CCK 메카니즘을 활성화시키는 것을 알수 있었다.

Changes of Endogenous Steroids Profile After Oral Administration of Danazol (Danazol의 경구투여에 따른 내인성 스테로이드들의 변화)

  • Danazol, one of the IOC banned substances, is a synthetic steroid which stimulates the synthesis of cellular protein with multiple and diverse biologic effects. To confirm the androgenic effects of danazol, levels of eight endogenous steroids and its major metabolite in human urine were simultaneously analyzed by selected ion monitoring (SIM) of GC/MS after oral administration. The recovery range of this method was 71.59%~93.56% and the RSD values of precision and accuracy test were 1.87%~10.48% and 1.32%~11.25%, respectively. The limits of detection of most of the drugs were $0.01{\sim}0.05{\mu}g/mL$ and calibration was carried out using urine spiked with each endogenous steroid and ethisterone standard at a concentration of 0.1, 0.5, 1.0, 10, 20 and $50{\mu}g/mL$ and with $10{\mu}g/mL$ of internal standard. Correlation coefficients varied between 0.963 and 0.991. The endogenous steroid profiling is one of valuatle tool for decteation of anabolic steroids.

  • The endogenous steroids from human urine were simultaneously analyzed by selected ion monitoring method of GC/MS which is currently used for the doping procedure, together with anabolic steroids. The recovery range of this method was 72.33 %∼94.54% and the RSD values of precision and accuracy test were 1.43%∼10.86%, 0.96%∼9.98%, respectively. Using this method steroids profile was investigated in the urine of male volunteers after oral administration of nine anabolic steroids banned by IOC (International Olympic Committee). Urinary endogenous steroids level was varied specifically according to the excretion tendency of the metabolites of anabolic steroids.

Effect of Lidocaine on Utilization of Endogenous Substrates for Contractile Process of Isolated Rat Atria (심근 수축에 쓰여지는 내인성 기질 대사에 대한 Lidocaine의 영향)

  • The experiments were performed to determine whether the cardiac depressant action of lidocaine is directly associated with the utilization of endogenous substrates in isolated rat atria, by using citrate and bicarbonate-free medium known as potent inhibitors of phosphofructokinases (PFK) enzyme step. Citrate and bicarbonate-free medium produced negative inotropic action of isolated rat atria incubated in normal Krebs-Ringer bicarbonate glucose medium. Pyruvate and acetate increased the force of contraction of atria depressed by citrate or bicarbonate-free medium, whereas fructose was without effect indicating the inhibitory effect of citrate and bicarbonate-free medium at some point in the glycolytic pathway such as the PFK step in atria. In the absence of exogenous substrate, citrate and bicarbonate-free medium produced a marked depression of the force of substrate-depleted atria indicating that utilization of endogenous substrate above the PFK step, probably cardiac glycogen, is also impaired by citrate or bicarbonate-free medium. Lidocaine produced further depression of the contractile force of atria depressed by citrate. These results argue strongly for an additional mechanism of cardiac depression caused by lidocaine involving the sites below the PFK.

  • The pathophysiology of rotator cuff tears and the progression of asymptomatic tears to symptomatic tears are yet unclear and much controversy. It is likely to involve a number of factors such as a genetic predisposition, extrinsic impingement from structures surrounding the cuff and intrinsic degeneration from changes within the tendon itself. Degenerative changes in the rotator cuff with aging seem to be related to the anatomic and mechanical environment of the rotator cuff. The histopathologic appearance of rotator cuff tendon rupture specimens demonstrates a consequence of degenerative changes at the site of tendon insertion into bone. It weakens the tensile strength of the tendon. Is the process of degeneration intrinsic or extrinsic in nature? I suggest that degeneration is intrinsic and not caused by extrinsic factors. Even though, rotator cuff tear may be secondary to multiple factors, I believe that primary cause of rotator cuff tears is preexisting degenerative change.
