Strain Development by Electrofusion between Phytase-producing Yeast and Salt-tolerant Yeast (전기융합법을 이용한 phytase 생성 내염성 효모 균주의 개발)

  • 내염성 조건에서 phytase를 생성하는 효모 균주를 개발하기 위하여 phytase를 생산하는 효모인 S. cerevisiae CY 균주와 내염성 효모인 Z. rouxii Y-80 균주 사이의 전기적 세포융합을 실시하였다. S. cerevisiae CY 균주의 Z. rouxii Y-80 균주의 배양된 세포를 1.0%의 ${\beta}$-mercaptoethanol로 전처리하고 Tunicase로 세포벽을 분해시킨 후 원형질체를 얻었다. 각 균주의 원형질체는 electrofusion 완충액에 1:1의 비율로 현탁한 후 AC 13V에서 30초간 처리하고 고전압(DC 500 V/100 ${\mu}sec$)을 가하여 전기적 세포융합을 실시하였다. 선별된 융합균주는 NaCl이 10% 첨가된 배지에서 친주들과 비교한 결과 Z. rouxii Y-80 균주보다는 생육면에서 25% 이상 증가하였으며, phytase 활성도는 친주인 S. cerevisiae CY 균주 대비 53%의 생성도를 확인하였다.

  • A halotolerant and extracellular pretense producing yeast was isolated from traditional Meju and identified as a strain of Hansenular sp. S-9 by investigation of its microbiological characteristics. The optimum pH, temperature and NaCl concentration for growth of Hansenular sp. S-9 were pH 7.5, $30^{\circ}C$ and 0.5M, respectively. The protease production from Hansenular sp. S-9 was maximized when it was grown on BD medium containing 1.0% beef extract, 1.0% glucose and 0.5M NaCl for 72 h at $30^{\circ}C$.

  • Four strains of Sporobolomyces genera were isolated from soy sauce mashes at the fermentating and aging periods and identified. Their salt-resistance and color formation were examined in order to improve the color density and nutrition of red pepper paste preparations. The results obtained were as follows; a) The strain L1, L2, L3 and L4 isolated were identified as Sporobolomyces gracilis, Sporobolomyces roseus, Sporobolomyes gracilis and Sporobolomyces roseus respectively. b) The yeasts isolated were grown on the media containing $0{\sim}10$ percent of sodium chloride but their growth were almost restrained on the media containing $15{\sim}18$ per cent of sodium chloride. And the strain L2 and L3 were more salt-resistant as compared with L1 and L4. c) The strain L3 were the best among the 4 strains of yeasts comparison with their salt-resistance, color and flavour formation.

Production of Yeast Cell Mass from Waste Brine and Cabbage Waste of Kimchi Factory (김치 공장의 절임 폐수 및 배추 쓰레기를 이용한 효모 배양)

  • The possibility of using waste brine and cabbage waste from kimchi industry as raw materials for the production of yeast cell mass was investigated. Among four strains of osmotolerant yeast, Candida guilliermondii ATCC 6260 showed the best growth in the waste brine containing about 1.0g/L of reducing sugar and 7% to 12% of NaCl. The growth of C. guilliermondii in waste brine was affected slightly between the temperature range of $25^{\circ}C$ to $35^{\circ}C$ and the initial pH of 3 to 6. The NaCl concentration up to 9% was not inhibitory to the growth of C. guilliermondii and the addition of 10mM of ammonium salts or 5mM of potassium phosphate had no effect on the growth. The growth of yeast reduced BOD of the waste brine by 85% within 24hours. When C. guilliermondii was cultured in waste brine added with cabbage juice extracted from waste cabbage, the cell mass was increased significantly.

  • The yeasts in the soy sauce mash were isolated and identified, and they were classified by coloring with the treatment of TTC(2, 3, 5, triphenyltetrazolium chloride) agar and counted in process of time. The results obtained were as follows: a) The number of ordinary and osmophilic yeasts in 1 ml. of the soy sauce mash showed a tendency to be increased from the mashing to the mature stages and to decrease in the aging stages: $127{\times}10^3$ immediately after mashing, $83{\times}10^3$ 1 month after, $356{\times}10^3$ 3 months after, $1250{\times}10^3$ 6 months after and $65{\times}10^3$ 2 years after mashing in the case of ordinary yeasts, and 0 after mashing, $40{\times}10^3$ 1 month after, $81{\times}10^3$ 3 months after, $358{\times}10^3$ 6 month after and $23{\times}10^3$ 2 years after mashing in the case of osmophilic yeasts. b) 50 strains of yeasts were isolated from the soy sauce mash optionally in process of fermentation period, and they were identified as 7 genera and 18 species: 10 strains of Saccharomyces rouxii, 1 strain of Saccharomyces marxianus, 3 strains of Saccharomyces rosei, 1 strain of Saccharomyces fermentati, 6 strains of Saccharomyces mellis, 1 strain of Saccharomyces acidifaciens, 1 strain of Saccharomyces pastori, 3 strains of Pichia polymorpha, 2 strains of Hansenula anomala, 1 strain of Hansenula saturnus, 2 strains of Hansenula suaveolens, 5 strains of Nadsonia fulvescens, 8 strains of Debaryomyces hasenii, 1 strain of Debaryomyces nicotianae, 1 strain of Debaryomyces kloeckeri, 2 strains of Torulopsis sake, 1 strain of Torulopsis holmii and 1 strain of Candida pelliculasa. c) Distribution of yeasts according to the fermentation period was as follows: i) Saccharomyces rouxii, Saccharomyces marxianus, Saccharoymces rosei, Pichia polymorpha, Debaryomyces hansenii, Torulopsis sake, Candida pelliculosa, Debaryomyces nicotianae, Nadsonia fulvescens, Hansenula suaveolens and Hansenula saturnus were found in the early stages of fermentation. ii) Saccharomyces rouxii, Saccharomyces rosei, Saccharomyces fermentati, Saccharomyces mellis, Saccharomyces pastori, Hansenula anomala, Saccharomyces acidifaciens and Debaryomyces hansenii appeared in the mature stages. iii) Saccharomyces rouxii, Saccharomyces mellis, Nadsonia fulvescenes, Dedaryomyces hansenii, Debaryomyces kloeckeri, Torulopsis sake and Torulopsis holmii were distributed in the aging stages. d) TTC white yeasts were found in abundance in the early stages of fermentation and TTC red yeasts appeared more than 50 per cent in the mature and aging stages. e) The yeasts belonging to Saccharomyces mellis and Saccharomyces pastori were classified as TTC red yeasts, Saccharomyces acidifaciens were reel pink, Hansenula saturnus Debaryomyces kloeckeri, and Torulopsis holmii were pink, Saccharomyces marxianus and Nadsonia fulvescens were white and the others were the same as the description in the previous report. Saccharomyces rouxii ware classified for the most part as TTC red yeasts, and while some of them were red pink. f) Species of yeasts in the soy sauce mash were similar to those in the soy sauce koji, but the latter were not osmophilic and in the former case, the osmophilic yeasts were increased in process of fermentation period.

Identification and Characterization of Osmotolerant Yeast Isolated from Soy Paste (된장에서 분리된 내염성 효모의 동정 및 특성조사)

  • Osmotolerant yeast isolated from soy paste could grow on media with 2 M NaCl. This strain was identified as Zygosaccharomyces rouxii by biological characteristics, RFLP of ribosomal DNA and mating with compatible haploid strain. Growing rate of the Z. rouxii YDJ was slower than Saccharomyces cerevisiae. Z. rouxii YDJ accumulated trehalose, which is known as one of the osmolytic protectants, in cells cultured on media with salt. Enzyme activity of trehalose phosphate synthase related to trehalose biosynthesis of the YDJ was lower than those of S. cerevisiae. Trehalase activity related trehalose degradation was also lower in Z. rouxii YDJ than S. cerevisiae. However, as Z. rouxii accumulated trehalose by salt treatment, salt tolerancy of Z. rouxii was assumed to be related to trehalose in additon to glycerol.

Characterization of Unrecorded Yeasts Isolated from Leaves of Trees of Oknyeobong Peak and Yeonjasan Mountain in Daejeon, Korea (대전광역시 옥녀봉과 연자산 나뭇잎으로부터 야생효모의 분리 및 국내 미기록 효모의 특성)

  • Twenty-two yeast strains of 15 species were isolated from the leaves of 20 trees on Oknyeobong Peak, and 24 yeast strains of 12 species were isolated from the leaves of 20 trees on Yeonjasan Mountain in Daejeon city, Korea. Cryptococcus bestiolae (5 strains) and Aureobasidium pullulans (8 strains) were the predominant isolates from Oknyeobong Peak and Yeonjasan Mountain, respectively. Of a total of 46 yeast strains, Cryptococcus kuetzingii JSL508, Cryptococcus vishniacii JSL509, and Dioszegia takashimae JSL510 from Okyeobong Peak, and Plowrightia periclymeni JSL514, Erythrobasidium hasegawanium JSL0193, and Rhodotorula nothofagi JSL0196 from Yeonjasan Mountain were determined to be yeast strains that were yet unrecorded in Korea. Morphological and cultural characteristics of these unrecorded yeasts were investigated. Erythrobasidium hasegawanium JSL0193 and Rhodotorula nothofagi JSL0196 did not form ascospores and pseudomycelia. All the strains, except Dioszegia takashimae JSL510, were halotolerant or halophilic, and Cryptococcus kuetzingii JSL508 and Dioszegia takashimae JSL510 were thermophilic, growing at $37^{\circ}C$.

  • Zygosaccharomyces rouxii is a salt-tolerant yeast which plays an important role during the ripening stage of soy sauce fermentation. Z. rouxii used in the experiment could grow in YPD (1 % yeast extract, 2% peptone and 2% glucose, pH5.0) medium with 18% (w/v) NaCl, whereas Saccharomyces cerevisiae could only grow in YPD medium with less than 8% NaCl. In the presence of 15% NaCl, Z. rouxii accumulates a large amount of glycerol as a compatible solute within the cells in the exponential phase. It is a characteristic of salt-tolerant yeasts. From the chemical analyses on membrane lipid fluidity, the membrane structure of the cells grown in 15% NaCl was suggested to become more rigid and its fluidity was decreased to keep glycerol within the cells in response to surrounding medium with high concentrations of salt.

  • Rhodosporidium fluviale 201-C-1 and Kluyveromyces thermotolerans 150-JE-2(1) were isolated from wild flowers in Geumsan-gun, Chungcheongnam-do and Wando-gun, Jeollanam-do, respectively. Rhodosporidium paludigenum 86-J-1 and Candida sp. 80-J-3 were also isolated from flowers in Jeju island, Korea. Characteristics for these yeasts were not reported before. Therefore, morphological and physiological characteristics for these yeasts were investigated in this study. R. fluviale 201-C-1 was halophilic asporogenous yeast and formed pseudomycelium. It also grew at $30^{\circ}C$ and pH 4.0-6.0. K. thermotolerans 150-JE-2(1) was sugar-tolerant and halotolerant asporogenous yeast. It formed pseudomycelium and showed positive urease activity. Candida sp. 80-J-3 and R. paludigenum 86-J-1 were asporogenous yeasts, and grew well in yeast extract-peptone dextrose(YPD) medium, potato-dextrose medium as well as vitamin-free medium and YPD medium containing 5% NaCl.

Effects of Blinds and Concentration of Salts on the Growth of Yeasts Isolated from Kimchi (소금 종류 및 농도에 따른 김치 효모균의 생육특성)

  • Effects of various types of salts (commercial low salt, sea salt, refined salt, and bamboo salt) and concentrations (0, 3.0 and 5.0% ) on the growth of yeasts isolated from Kimchi were investigated. The isolated yeasts used in the study are as follows : Saccharomyces cerevisia, Sporobolomyces albo-rubescens, Issatchenkia orientalis Cryptococcus luteolous, Ustilago maydis, Candida humilis, Pichia onychis, Cadida nitratophila, and Pichia jadinii. The growths of the yeasts were inhibited against each salt concentration. The growths of Candida sp. isolated from the later stage of fermentation was strongly inhibited against 5% concentration of salt. On the other hand the induction phase of Issatchenkia orientalis isolated at the early stage of fermentation was the quickest among all the tested yeasts. Among the salts, bamoo salt was found to be strongest inhibitor of the growth of yeasts.
