Antioxidant and antidiabetic Activity of Jehotang according to Different Cooking Methods (제조방법에 따른 제호탕의 항산화 및 항당뇨 활성)

  • Jehotang is a cold traditional Korean drink made with honey and several ingredients used in traditional Korean medicine. The ingredients include Fructus mume, Fructus amomi, Fructus tsaoko, Santalum album and honey. In this study, Jehotang and its ingredients were determined through the analysis of antioxidant activity, total phenolic content, ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity. In addition, quality characteristics of Jehotang made by a traditional recipe(Yeonmil) and a modern recipe(double boiling and boiling) were also compared in terms of pH, color and sugar content. Total phenolic content of extract from Fructus ammomi was found to be 120.45 mg, and Yeonmil recipe was discovered 152.66 mg equivalent of gallic acid per g of extract. DPPH free radical scavenging activity were Feuctus amomi(93.13%) and Yeonmil recipe(56.44%). The Fructus amomi extract showed the highest ${\alpha}$-glucosidase inhibitory activity(89.51%) at the concentration of $100{\mu}g/mL$. ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity of boiled Jehotang were 52.38% and 72.52%, respectively. These results suggest that extract of Fructus amomi has an antioxidant activity and antidiabetic effects. Yeonmil recipe is useful for antioxidant effects more than the others. Also, the double boiling recipe has an excellent antidiabetic effect.

  • Experimental studies were carried out to develop an osmotolerant mutant of Candida magnoliae JH and to determine the optimum concentrations of carbon and nitrogen sources to improve erythritol yield and productivity. Mutants of C. magnoliae JH were prepared by treatment with ethylmethane sulfonate, and osmotolerant mutants were isolated from the agar plate colonies containing 2.5 M KCI. Among the mutants isolated, one mutant M26 was finally selected based on erythritol yield and productivity. In shake flask culture, glucose proved to be the best carbon source and the optimum yeast extract concentration was 5 g/L based on 100 g/L glucose. The erythritol yield and productivity of mutant M26 were greater than wild type in 100 g/L glucose medium. in the fermentation experiments, erythritol production increased with increased glucose concentration, up to a limit of 250 g/L. The maximum concentration of erythritol achieved 127.5 g/L, and the yield and productivity of erythritol production were 51.0% and 0.63 g/L-h, respectively.

  • To study the effect of nonwoven fabric materials on growth and yield of korea melon, Medium denier nonwoven fabrics (MDNF), Polyster nonwoven fabrics (PENF) and 12 ounce nonwoven fabric (control) were used. Mean temperature at night was 13.1$^{\circ}C$ at control, 14.7$^{\circ}C$ at MDNF and 13.3$^{\circ}C$ at PENF from Feb. 6${\sim}$7, 2009. Because of higher temperature, early growth of korea melon of MDNF was better than control at 30 days after transplanting. Days required to harvesting (DRH) of control was 89, those of PENF and MDNF were 88, 78, respectively. First harvesting of MDNF was 11 days earlier than control. Harvested fruit of MDNF had higher weight, soluble solid and marketable fruit ratio than control. Yield ratio of control and PENF was 4:3:3 (early:middle:late harvesting season), but MDNF was 5:3:2. Using of MDNF provided earlier harvesting. Total yield of PENF was similar to control, 1,844kg per lOa, MDNF was increased by 9%.

  • Torymus geranii was found to be a ectoparasitoid attacking the larvae of Dryocosmus kuriphilus. T. geranii has longevity of a $42.8{\pm}9.8$ days at $20^{\circ}C$ and $26.5{\pm}11.0$ day at $25^{\circ}C$ under conditions supplied with 100% honeydew. Oviposition numbers at 20 and $25^{\circ}C$ were $22.3{\pm}12.5$ and $42.2{\pm}18.4$, respectively. The eggs were oval in shape, measuring $0.56{\pm}0.33$ mm in length. The larvae, $2.94{\pm}0.18mm$ in length, was white and length of male and female pupae were $2.01{\pm}0.18mm$ and $2.73{\pm}0.09mm$, respectively. T. geranii had two generations with overwintering generation emerged on late May to early June at Chunchon, central part of Korea, however showed three generations with overwintering generation emerged on middle May to early June, the first one on late June to early July, and the second one on late July to earlyl August.

Effects of Deep Tillage before Planting on Physicochemical Properties of Soil, Growth and Fruit Characteristics in Cultivation of Watermelon under Plastic Film House (수박 시설 재배에서 정식 전 심경로타리 처리가 토양 이화학성, 생육 및 과실 특성에 미쳐는 영향)

  • To investigate effects of deep tillage before planting on physicochemical properties of soil, growth and fruit characteristics in cultivation of watermelon (Citrullus vulgaris L. cv. 'Uriggul') under plastic film house, this study was conducted in cultivating field of Gochang Junbuk. pH in soil after harvest compared with soil before planting of watermelon had almost no change, but EC lowed greatly in the two treatments. Available phosphate concentration in the soil with the deep tillage treatment was lower, K concentration of exchangeable cation decreased greatly than these with conventional tillage treatment. In the growth at 27th day after planting plant, stem length to 10th node from the first node, leaf width with deep tillage treatment were longer, bearing node of the first and second flower and wilting degree were lower than these with conventional tillage treatment. In the growth of harvesting time, the stem length to 30th node from the first node with the deep tillage treatment were longer, leaf discoloration degree was lower than these with conventional tillage treatment. Also, the harvested fruits length, diameter, peel hardness, and weight were significantly better than these with conventional tillage treatment.

Quality Properties of Honey in Korean Commercial Markets (국내 유통되는 벌꿀의 품질특성)

  • This study was carried out to examine the quality properties of honey in Korean commercial markets. The moisture content, stable carbon isotope ratio, invert sugar, cane sugar, and hydroxy-methylfurfural (HMF) contents of honey were measured according to the Korea Food Code and AOAC's (Association of Official Analytical Chemists) official methods. The stable carbon isotope ratio ranged from -25.18‰ to -12.60‰, which clearly differed between honey of $C_3$ origin (flower) and $C_4$ origin (artificial). Results of quality measurements revealed a moisture content of 18.12 to 19.70%, fructose content of 36.10 to 43.94%, glucose content of 22.61 to 31.91%, sucrose content of 1.56 to 4.75%, invert sugar content of 64.89 to 72.79%, and HMF content of 4.10 to 78.66 mg/kg. These values demonstrate that the quality of the tested honey meets the standard criteria of the Korean Food Code and Codex. However, it is necessary to reconsider the appropriate criteria for imported honey because it is circulated in the market through a long distribution process.

The Periodical Change and Regional Distribution Characteristics of Regional Speciality Products in Chungcheongbuk-do (충청북도 지역특산물의 시기별 변화와 지역적 분포 특성)

  • Regional specialty products should be representative of the regional characteristics for a long time, this was not done for geographical research. This study examines current the regional specialty products on 12 web-site of the cities counties in 2013 and the products registered on the Tosan(土産) category from old Jiriji(古地理誌) in Chungcheongbuk-do. Futhermore, the study analyzed periodical change and regional distribution characteristics of regional specialty products. Major findings are as Follows. First, current the regional specialty products types are 4 products(crops, marine products, forest products, livestock products) and 3 manufactures (processed food, handicraft, cosmetic products). Among the regional specialty products, various crops and processed food were distributed in all the regions. Second, the traditional products types are 5 products(crops, marine products, forest products, livestock products, mineral products) and 1 manufactures(handicraft) that registered on the old Jiriji in the Joseon Dynasty period. The jujube, ginseng and honey were cultivated in the most regions. Third, the traditional products types are 4 products except for marine products and 1 manufactures(handicraft) that registered on the old Jiriji in the period of Japanese colonial rule. Its total item numbers of the regional specialty products are greatly decreased, but items of handicraft relatively increased in the Danyang and Goesan regions. Finally, regional analyzing of current local items shows that there are only a few regional specialty products(18 items) that have historical traditionality.

Physicochemical and Microbiological Properties of Ginseng-Whey Beverages (인삼 유청음료의 이화학적 및 미생물학적 특성)

  • Ginseng-whey beverages were prepared with rennet whey, ginseng, sweetener, honey and Japanese apricot, inoculated with different strains of lactic acid bacteria or unfermented partly. The samples were stored at 4$^{\circ}C$ or 30$\pm$1$0^{\circ}C$ and then physicochemical and microbiological properties were investigated. The yield of whey was 78.8%. The pH-values reduced and acidities increased during the storage period. The contents of solid-substances, ash and lipid in ginseng-whey beverages were 7.90~8.20%, 0.62~0.66% and 0.16%, respectively. The protein contents of ginseng-whey beverages were 0.42~0.56% and the contents were not changed during the storage period. The lactose contents of fermented beverages were higher than those of unfermented beverages. During the storage period (1~5 weeks), the ranges of D(-) - and L(+)- lactic acid contents in fermented ginseng-whey beverages (17.3~156.1 mg/100g, 347.3~1894.2mg/100g) were higher than those of unfermented ginseng-whey beverages (6.2~82.8mg/100g, 7.1~885.5mg/100g). The contents of total saponin in unfermented sample and fermented sample (Lac. casei sub-sp. casei+Str. salivarius sub-sp. thermophilus) were increased during the storage period. But, those of the fermented sample(Lac. acidophilus+Lac. delbrueckii sub-sp. bulgaricus) were reduced. In the electrophoretic results of ginseng-whey beverages, an $\alpha$-lactalbumin and a $\beta$-lactoglobulin bands were shown apparently and there were no changes observed during the storage period. During the storage period (1~3 week) the coliform was not detected and total plate counts and psychrotrophs were increased according to the storage period.

  • Various insects live around us. Nowadays, insects are raised as a hobby at home or medicinal insects are bred in large quantities for commercial purposes. In addition, various studies have been conducted to understand the various growth characteristics of insects. In the laboratory, environmental factors were changed to do that, but only the growth characteristics could be analyzed. However, it is almost not easy to measure the skin temperature or body temperature basically needed to reveal the growth characteristics in the case of insects. In particular, the results of experiments on the skin temperature of winged insects are very insufficient. In this study, the skin temperature of insects was photographed and measured for bees and butterflies living in natural conditions under various biological activity conditions such as at rest and during flight using a thermal imaging camera, a non-contact temperature measurement method. Through quantitative analysis of the experimental results, the relationship between the biological activity characteristics of each insect and the skin temperature resulting from such biological activity was investigated. In addition, it was confirmed that honeybees maintained different skin temperatures for three important activities, such as flying, honey collection, and relocation, and that the butterflies performed a warm-up stage for flight similar to the characteristics of moths.

Morphometric Characterization of Honey Bee, Apis mellifera Linnaeus, Inbred Lines in Korea (국내 서양종꿀벌 순계의 형태적 특징)

  • The A, C, F colonies of Apis mellifera ligustica Spin. and D, V colony of Apis mellifera caucasia Gorb. bees were collected from 2005-2007. Consequently, inbred lines were derived from the bees of original colonies by matting in the isolated island with due regard for pure breeding. This project helps in the selection of colonies with higher production capacity, aiming to improve honey and royal jelly production and breeding programs. Twenty-three standard morphological traits of honeybee were evaluated, and samples were compared with the data of the two original subspecies. The result suggested that 8 traits partly preserved in bees of inbred lines, and the bees from A. m. ligustica preserved more traits than bees from A. m. caucasia. Among the studied inbred lines, the F line is distinguished by an increase in leg parameters, considered as a favorable phenotypic trait of inbred lines. Importantly, bred of beelines in the same area can be classified as remote and isolated areas. Therefore, we observed differences of inbred lines with the origin subspecies in description acquired with morphometric characteristics as a result of adaptation, breeding, purebred individual lines used as an important resource for breeding novel cross-breeding colonies.
