Effects of bulblet size, low temperature treatment and time of incubation on stem emergence during of Lilium oriental hybrids. (오리엔탈 백합의 순화재배시 자구크기, 저온처리, 배양기간이 경출현에 미치는 영향)

  • The experiment were carried out to establish the optimal condition of bulblet size, low temperature treatment and time of incubation in acclimatization on stem emergence of Lilium oriental hybrids. The large size bulblet was better than middle and small size bulblets in percentage of stem emergence and weight of bulblets. The highest percentage of stem emergence was 95% in large size bulblet 'Acapulco' but small size bulblet of 'Casablanca' was no stem emergence. Low temperature treatment(5℃)for breaking dormancy was needed at least more than 9weeks. Weight of bulblets and percentage of stem emergence was not good in 5℃ treatment for 6 weeks. Effects of Cultural Conditions on the In Vitro Propagation of Plantlets derived from Apical Meristem in Ipomoea batatas L. (고구마 정단분열조직 유래 식물체의 기내 증식에 미치는 배양조건의 영향)

  • Sweet potato is a crop vegetatively propagated by vine cuttings, an ineffective method for maintaining pathogene-free stock plants. As an alternative method, single-node cultures of virus-free plantlets derived from apical meristem in sweet potato (cv. Yulmi) was examined. Effective pH range, sugar concentration and nodal order were investigated to establish an in vitro mass propagation system with high quality virus-free stock plantlets to farmhouse. Although the plantlets grew at wide range of pH, the most effective pH of the medium was 4.8 in single-node cultures. High sugar concentration of 60∼80 g/L resulted in increased growth response in shoot length, root length, number of node, leaf area and fresh and dry weight of shoot and root, whereas reducing sugar contents below 6% was showed reduced growth response. The first node including meristem tip was the best for the rapid growth of plantlets and the other nodes also showed a very similar growth response. Uniform plantlet can be obtained massively at the same time by culture of single node except for the first node including meristem tip. In conclusion, the most effective pH range and sugar concentration of medium for the growth of plantlets via single-node cultures was 4.8, 60∼80 g/L respectively. The first node was the best for the rapid propagation of plantlets in nodal order.

Acclimatization of in vitro Plantlets of Wasabia japonica(Miq.) Matsum. Derived from the Apical Meristem Culture (고추냉이(Wasabia japonica (Miq.) Matsum.)의 정단분열조직유래 기내묘의 순화)

  • The repeated subcultures of in vitro plant materials in wasabi became highly vitrified and the capacity for multiple shoot formation from the vitrified plant materials was very low. In order to improve the quality of in vitro propagated planting materials, the experiments were carried out using culture vessels capped with membrane filter(MF). When vitrified shoots were cultured on MS medium with 0.2mg/L BA in the vessels with MF or without MF for 60 days, the shoots in the vessels with MF did not vitrified. In contrast, the shoots grown in the vessels without MF vitrified at 65%. The stomates of vitrified leaves were circular and inflated, whereas those of normal leaves acclimatizated in the vessels with MF were ovate in shape. The hardened shoots were also cultured on MS media without sucrose containing 0.01mg/L IBA in vessels with(photoautotrophic culture) or without(control) MF. Sucrose was necessary for survival of the in vitro plantlets in the vessels without MF. After 20 days of culture, the shoots in the vessels without MF on the sucrose-free media turned yellow and died. But the shoots in the vessels with MF in the sucrose-free media produced a lot of roots. When shoots were cultured on MS medium with 2% sucrose containing 0.01mg/L IBA in the vessels with(photomixotrophic culture) or without(heterotrophic culture) MF, best growth occured in photomixotrophic culture.

  • When cultured soybean immature seed on 15th days after flowering, suitable temperature in formation of callus were $24{\sim}27^{\circ}C$, and embryogenic callus(EC) were generated at medium containing NAA with growth regulators, and then, results were subcultured the EC, a plenty of shoots and roots were formed at medium supplemented BA 2mg/1 and IAA 2mg/1, respectively, however when used at medium the same time supplemented BA 2mg/1 and IAA 2mg/1, formation of cullus was energetic, and a symptom of organization was not showed , Total lipid contents include in each cultures were increased at low temperature of cultural conditions as much as possible, but glycolipid, phospholipid, free sterol contents were a little increased at $24{\sim}27^{\circ}C$, and free sterol content was increased at a case of embryogenic structure were generated. In fatty acid compositions in each cultures, the contents of unsaturated fatty acid were plenty in EC, and unsaturation rate was 0.837. Besides, in sterol compositions, cholesterol content was remarkably high in EC than that of other cultures.

Changes of Root Physiology of Tissue Cultured M.9 Apple Rootstock after Layering (기내 배양 사과 대목 M.9의 순화 후 휘묻이 번식 시 발근 관련 생리적 특성 변화)

  • This work was conducted to evaluate the effects of rooting on tissue cultured M.9 (Malus domestica Bork. cv, tcM.9) after layering in field. We investigated an appearance period of first root in shoot, rooting ratio, contents of indole-3-acetic acid (IAA), abscisic acid (ABA), inorganic matters, sugars, and lignin in rooting areas of stems by layering. First root in shoot of tcM.9 and natural M.9 appeared 25 and 30 days after layering (DAL), respectively. Rooting ratio was much higher in tcM.9 than in natural M.9. The content of IAA was higher in tcM.9 than in natural M.9 before layering, but it was reversed at 20 and 30 DAL. In contrast, the content of ABA was much higher in natural M.9 than in tcM.9 in case of both before and 10 and 20 DAL. The contents of N, B, Mn, and Zn were significantly higher in tcM.9 than in natural M.9 both before and 10 and 20 DAL. The contents of sugars in tcM.9 had the similar pattern of the contents of inorganic materials. There were statistically significant differences in the contents of sucrose and glucose at 30 DAL as well as the content of maltose at 20 and 30 DAL. The content of lignin was significantly higher in tcM.9 than in natural M.9 before layering and 10 and 30 DAL while there was no difference 20 DAL. Therefore, improvement of rooting ability in the tissue cultured root stock M.9 might be due to the changes of inorganic matters or lignin rather than that of sugars and hormones.

Expression of Organogenesis-related Genes and Analysis of Genetic Stability by ISSR Markers of Regenerants Derived from the Process of in vitro Organogenesis in Japanese Blood Grass (Imperata cylindrica 'Rubra') (기내배양 홍띠 단계별 재분화체의 기관분화 관련 유전자 발현과 ISSR에 기반한 유전적 안정성 분석)

  • The in vitro organogenesis is one of important issues in plant embryology, and somaclonal variations are existing in calli and/or regenerants induced from a process of the organogenesis with in vitro circumstances. In this study, expressions of organogenesis-related genes were evaluated and genetic stability of regenerants derived from the process of in vitro organogenesis were measured using ISSR markers in Imperata cylindrica 'Rubra', Poaceae. The expressions of organogenesis-related genes were detected all of regenerants at the process of the organogenesis. All ISSR markers produced with an average of 71 bands per in vitro-cultured regenerants, and the scorable bands were varied from two to eight with an average of 5.14 bands per a primer. The polymorphism rates of the in vitro regenerants were higher than that of mother plants (1.4%), showing 4.1% (pot-cultured regenerants), 4.3% (field-cultured regenerants), 4.2% (in vitro-cultured regenerants), 5.6% (calli with green shoots) and 1.4% (calli), respectively. The genetic similarity matrix (GSM) among all accessions ranged from 0.747 to 1.0 with a mean of 0.868. GSM of the regenerants showed differences (from 0.972 to 1.00) compared with that of mother plants (0.991). According to the clustering analysis, two independent groups were divided into; the one is mother plants and regenerants cultured at room and open field, the other is regenerants cultured in vitro. The results give a new insight for understanding the dynamics of organogenesis in monocot plant.

In Vitro Propagation of Alocasia cadieri Chantrier (Alocasia cadieri Chantrier의 기내번식)

  • In order to micropropagate uniform plantlets of Alocasia cadieri Chantrier in vitro, the shoot tips were cultured on media containing various concentrations of BA and thidiazuron (TDZ). Multiple shoot formation from shoot tips was very effective on medium containing 0.1mg/L TDZ. The formed shoots from shoot tips were separated into a shoot, and cultured on media with BA, TDZ, and NM combination for proliferation. The shoots were multiplied very vigorously on medium with 0.5mg/L TDZ and 0.5mg/L NAA. The rooting and growth of multiplied shoots were more effective on medium with 2.0g/L activated charcoal, rather than those with IBA and NAA. Rooted plantlets show high survival in soil mixed with perlite 1: vermiculite 1 or vermiculite alone.

Factors Affecting In Vitro Minimal Growth Conservation of Sedum sarmentosum (돌나물의 기내 활성보존에 영향하는 요인)

  • For in vitro minimal-growth conservation of S. sarmentosum, the in vitro shoots with 10 mm length were cultured on Murashige and Skoog's media (MS) containing different levels of agar (0.8, 1.2, 1.6, 2%), Gelrite (0.4, 0.6, 0.8, 1%), ABA (0, 5, 10, $20mg{\cdot}L^{-1}$), and sucrose (2, 3, 6, and 9%) without subculture at $4^{\circ}C$ and $25^{\circ}C$. All media were supplemented with $0.2mg{\cdot}L^{-1}$ BA, agar and Gelrite media, with 5% sucrose, sucrose media, with 1.2% agar, and ABA media, with 5% sucrose and 1.2% agar, respectively. In vitro minimal-growth conservation in room-temperature ($25^{\circ}C$) was effective in the media containing with $10mg{\cdot}L^{-1}$ ABA or 1.6% agar, and the healthy plantlets could be preserved for 10 months without subculture. After 12 months at $4^{\circ}C$, survival rate was 100% in all media. The in vitro minimal-growth conservation in low temperature ($4^{\circ}C$) was effective in the media containing with $10mg{\cdot}L^{-1}$ ABA or 6% sucrose, and the healthy plantlets could be preserved over 18 months without subculture. Especially, long-term conservation using minimal growth of S. sarmentosum was much more efficient in the medium containing high level sucrose at $4^{\circ}C$ compared to others.

Ultrastructural observation of Naegleyiu fowleri trophozoite in mouse brain and axonic culture (조직내 및 배야기내 자유생활아메바의 전자현미경적 비교연구)

  • Present study was undertaken to elucidate the changes of the ultrastructure of Naegleria fowleri trophozoite in brain tissue of mice and culture medium. Naegleria fowleri, 0359 strain, which used in this study was cultured in axonic liquid medium, CGVS medium. Each mouse was inoculated with amoebas intranasally under secobarbital anesthesia, and sacrificed on 7th day after the infection. Comparative observation of the ultrastructure of the amoebas in axonic culture and experimentally infected mice brain was done with transmission electron microscope. The results are summarized as follows: 1. The amoebas in mouse brain tissue were round in outline, whereas those of amoebas from axonic culture showed irregular appearance. 2. Mitochondria in the amoebas from axonic culture was oval, round and cylindrical shape and darkly stained, whereas those of the amoebas from mouse brain tissue showed dumbbell shape together with above forms. The stain was not unique, but light and/or dark. 3. Rough endoplasmic reticulum of amoebas in brain tissue was tubular, but from culture it was vesicular or tubular in shape. 4. Emity vacuoles were demonstrated in amoebas from culture, while food vacuoles with myelinated structures were abundant in those from tissue, suggesting a strong phagocytic activity. 5. Mouse brain tissue in ected were extensively destroyed, and Polymorphonuclear leukocytes were infiltrated predominantly with inflammatory lesion. Amoebas were observed in the vicinity of the capillary.

Micropropagation of Juvenile and Mature Tree of Corylopsis coreana by Axillary Bud Culture (액아배양에 의한 유묘 및 성숙 히어리나무의 기내번식)

  • We have developed an in vitro micropropagation system via shoot formation from axillary buds using nodal segments of Corylopsis coreana. Explants from both juvenile tree (one-year-old greenhouse stock seedlings) and mature tree (ten-years-old tree in nursery) were compared with regard to propagation efficiency. Combined treatment of both BA and zeatin were effective on shoot proliferation since the best result was obtained on MS medium supplemented with 0.5∼3.0 mg/L zeatin and 0.2 mg/L BA. Generally, juvenile explants were better in both shoot proliferation and growth than mature explants. However, as the duration of in vitro culture was proceed to 6 months, explants from mature tree also produced three shoots per explant. Distinctive differences in rooting and adaptability to soil of shoots obtained from mother trees. Whereas shoots originated from juvenile explants rooted as high as 97%, those from adult explants showed 62% rooting. Similar result was also observed in soil acclimatization. The plantlets derived from juvenile plants survived 67%, while only 48% of those from adult trees survived. The results showed a possibility of the micropropagation of Corylopsis coreana through shoot formation from axillary buds. In addition, the advance of the research still remain to enhance the frequency of acclimatization of plantlets from mature trees for practical application.
