Pink Pigmented Facultative Methylotrophic Bacteria(PPFMs): Introduction to Current Concepts (분홍색 색소를 형성하는 methylotrophic acteria(PPFMs): 최근 경향소개)

  • The non infecting, plant associated bacteria have attracted increased attention for stimulating plant growth and as environmental friendly plant protecting agents. Pink-pigmented facultatively methylotrophic bacteria (PPFMs), classified as Methylobacterium spp., are persistent colonizers of plant leaf surfaces. As the leaves of most or all plants harbor PPFMs that utilize leaf methanol as their sole source of carbon and energy, which is a specific attribute of the genus Methylobacterium. Although they are not well known, these bacteria are co-evolved, interacting partners in plant metabolism. This claim is supported, for example, by the following observations: (1) PPFMs are seed-transmitted, (2) PPFMs are frequently found in putatively axenic cell cultures, (3) Low numbers of seed-borne PPFMs correlate with low germinability, (4) Plants with reduced numbers of PPFM show elevated shoot/root ratios, (5) Foliar application of PPFMs to soybean during pod fill enhances seed set and yield, (6) Liverwort tissue in culture requires PPFM-produced vitamin B12 for growth, (7) treated plants to suppress or decrease disease incidence of sheath blight caused by Rhizoctonia solani in rice, and (8) the PPFM inoculation induced number of stomata, chlorophyll concentration and malic acid content, they led to increased photosynthetic activity. Methylobacterium spp. are bacterial symbionts of plants, shown previously to participate in plant metabolism by consuming plant waste products and producing metabolites useful to the plant. There are reports that inform about the beneficial interactions between this group of bacteria and plants. Screening of such kind of bacteria having immense plant growth promoting activities like nitrogen fixation, phytohormone production, alleviating water stress to the plants can be successfully isolated and characterized and integration of such kind of organism in crop production will lead to increased productivity.

Effects of Dispersivity of Clay on Thermal Stabilities of PP/Clay Nanocomposites (점토의 분산성이 PP/점토 나노복합재료의 열안정성에 미치는 영향)

  • The effect of ozone surface treatment of montmorillonite (MMT) was investigated in thermal stabilities of polypropylene (PP) nanocomposites. Sodium montmorillonite (Na$\^$+/-MMT) was organically modified with dodecylammonium chloride. The surface properties of MMT, including the specific surface area (S$\_$BET/), equilibrium spreading pressure ($\pi$$\_$e/), and London dispersive component (${\gamma}$s$\^$L/), were studied by the BET method with $N_2$ adsorption. Also, the thermal stabilities of the nanocomposites were investigated in DSC and TGA. As experimental results, $\pi$$\_$e/ and ${\gamma}$s$\^$L/ of the ozonized dodecylammonium chloride (DA-MK ( $O_3$)) were increased in about 1.7 and 3.5 mJ/ $m^2$, resulting from the increasing of the micropores. From the DSC results, it was found that the melting temperature and crystallization temperature of PP/DA-MK and PP/DA-MK ( $O_3$) were higher that those of pure PP. These results were explained that dodecylammonium chloride of nano-scale led to a nucleation effect for PP crystallization. Also, it was found that E$\_$t/ of the PP/DA-MK ( $O_3$) nanocomposies was increased within about 64 kJ/mol. These results were probably explained by the improvement of dispersivity of DA-MK ( $O_3$) in a PP matrix.

  • Numerical simulation for Resin Transfer Molding manufacturing process is attempted by using the eXtended Finite Element Method (XFEM) combined with the level set method. XFEM allows to obtaining a good numerical precision of the pressure near the resin flow front, where its gradient is discontinuous. The enriched shape functions of XFEM are derived by using the level set values so as to correctly describe the interpolation with the resin flow front. In addition, the level set method is used to transport the resin flow front at each time step during the mold filling. The level set values are calculated by an implicit characteristic Galerkin FEM. The multi-frontal solver of IPSAP is adopted to solve the system. This work is validated by comparing the obtained results with analytic solutions. Moreover, a localization method of XFEM and level set method is proposed to increase the computing efficiency. The computation domain is reduced to the small region near the resin flow front. Therefore, the total computing time is strongly reduced by it. The efficiency test is made with a simple channel flow model. Several application examples are analyzed to demonstrate ability of this method.

Characterization of Microtextures formed by Chemical Weathering in Crystalline Rocks and Implications for Rock Mechanics (화학적 풍화에 의한 결정질 암석내의 미세조직 발달특징과 암반공학적 의미)

  • Weathering can reduce rock strength and eventually affect the structural stability of a rock mass, which is important in the field of engineering geology. Several methods have been developed to evaluate the degree of weathering, including the chemical weathering index. In this study, we analyzed the weathering degree and characteristics of microtextures and pores in crystalline rocks (gneiss and granites) based on petrographic observations, the chemical weathering index, mineralogy by XRD, microtextural analysis by SEM/EDS, measurements of pore size and surface area by the BET method, and microporosity by X-ray CT. The formation of secondary minerals and microtexture in gneiss and granitic rocks are assumed to be affected by complex processes such as dissolution, precipitation, and fracturing. Hence, it is clear that some chemical weathering indices that are based solely on whole-rock chemistry (e.g., CIA and CWI) are unable to provide reliable assessments of the degree of weathering. Great care is needed to evaluate the degree of chemical weathering, including an understanding of the mineralogy and microtexture of the rock mass, as well as the characteristics of micropores.

Study on the Dextran and the Inner Structure of Jeung-Pyun (Korea Rice Cake) on Adding Oligosaccharide (올리고당 첨가 증편 발효 중 Dextran 형성과 증편의 내부구조에 관한 연구)

  • This study was carried out in order to investigate dextran formation and internal structure during fermentation of the oligosaccharide Jeung-Pyun. The dextran and sugar reducing contents of Jeung-Pyun batter and the specific volume and the internal structure of Jeung-Pyun were analyzed as a function of fermentation time. The specific volume of Jeung-Pyun peaked at the 7th hour of fermentation. The dextran content of Jeung-Pyun batters peaked at the 7~13th hour of fermentation, and Fructooligosaccharide Jeung-Pyun had the least peak value. Reducing sugar content of Jeung-Pyun batters slowly decreased as fermentation progressed. From the air pore size and distribution of Jeung-Pyun observed by SEM, the sucrose Jeung-Pyun fermented for 3~7 hours and oligosaccharide one fermented for 7 hours were judged as the best. It was concluded that dextran may be formed by fermentation of oligosaccharides as well as sucrose and dextran has a significant role on the volume expansion of Jeung-Pyun.

Evaluation of Antimicrobial Activity of Allyl Isothiocyanate (AITC) Adsorbed in Oyster Shell on Food-borne Bacteria

  • Oyster shells are a waste product from mariculture that creates a major disposal problem in coastal regions of southeast Korea. To make practical use of unused oyster shells, calcined oyster shell (COS) collected from a local company was allowed to adsorb AITC (allyl isothiocyanate), and then tested the powder's ability to inhibit the growth of some potential food borne disease-causing bacteria. COS powder showed bacteriostatic effect that inhibited cell growth of Escherichia coli, Staphylococcus aureus and Salmonella typhimurium from 3 to 5 log10 CFU/mL at concentrations around 1%. The MIC of pure AITC was found as 1 mg/mL, 0.8 mg/mL and 0.7 mg/mL for Escherichia coli, Staphylococcus aureus and Salmonella typhimurium, respectively. The calcined powder adsorbed about 225 mg of AITC per gram of shell, indicating porous material was created by calcination. FTIR data confirmed the adsorption of AITC by COS. Characterization of particle data showed very fine particle size and highly convoluted surface. AITC adsorbed calcined oyster shell (ACOS) completely inhibited bacterial cell at 1% concentration. ACOS showed better antibacterial effect than COS, indicating synergistic effect of AITC and calcined oyster shell powder on bacteria.

Development and Quality Evaluation of Hypo-Allergenic Bakery Products using Homegrown Wheat (항원성이 저감화 처리된 국내산 밀을 이용한 발효빵의 제조 및 품질 특성)

  • As an alternative of yeast, various concentrations to prepare hypo-allergenic bread (HAB). Addition of sodium bicarbonate increased $CO_2$ emission, but at higher than 2.0% caused bitter taste compared to other mixtures. Addition of sucrose significantly increased $CO_2$ emission. HABs 7 ($NaHCO_3$ 2.0g+sucrose 0.63 g) and 8 ($NaHCO_3$ 2.0g+sucrose 1.0g) showed higher grain and color scores. HAB 5 ($NaHCO_3$ 1.5g+sucrose 1.0g) and 6 ($NaHCO_3$ 1.5g+sucrose 2.0g) gave higher aroma and taste scores. HAB 5 showed significantly higher overall acceptability (p<0.05) score than others, whereas HABs 7 and 8, regardless of high external sensory qualities, showed lower overall acceptabilities than HAB 5 due to bitter taste. Prototypical HAB was obtained using wheat flour (100g) and water (62mL) incubated at $37^{\circ}C$ for 4 hr for treatment of b.p pretense (100mg) before adding sodium bicarbonate (1.5%), sucrose (1.0%), NaCl (1.0%), and citric acid (1.0%).

Degradation Characteristics of Cross-linked Hyaluronic Acid Membrane (가교된 히아루론산 막의 분해 특성)

  • The degradation characteristics of cross-linked lactide/hyaluronic acid (LA/HA) membranes were investigated for purpose of applying to tissue engineering. The lactide/hyaluronic acid cross-linked with 1,3-butadiene diepoxide (BD) and 1-ethyl-3-(3-dimethyl aminopropyl) carbodiimide (EDC) was degradated in deionized water in water bath at $37^{\circ}C$. As the LA/HA mole ratio or crosslinking agent concentration decreased, the degradation rate of the crosslinked membranes increased. In order to investigate the structure change of the membrane in the degradation process, the control sample and 3, 6, 9 days-degradated samples were analysed by the nuclear magnetic resonance spectroscopy. In case of the membranes crosslinked with EDC, the HA-EDC bonding structure was degradated slowly whereas the HA-LA bonding structure was degradated quickly and dissappeared completely after 6 days. In case of the membranes crosslinked with BD, all the crosslinked bonding structure degradated slowly. The HA-BD bonding structure maintained its original state about 89, 83% in case of 3, 6 days-degardated samples respectively whereas the HA-LA bonding structure maintained its original state about 83, 65%. The scanning electron microscopy of the degradated membranes showed that the pore density in the surface, and the structure in the surface and cross section, of the before and after-degradation membranes did not change greatly, so the membranes was shown to be applied to materials for tissue engineering.

The Effective Recovery of Gold from the Invisible Gold Concentrate Using Microwave-nitric Acid Leaching Method (마이크로웨이브-질산침출방법에 의한 비가시성 금의 회수율 향상)

  • This study aimed to liberate gold from invisible gold concentrate (Au = 1,840.00 g/t) through microwave nitric acid leaching experiments. For the purpose, this study conducted microwave-nitric acid leaching experiments and examined nitric acid concentration effect, microwave leaching time effect and sample addition effect. The results of the experiments were as follows: Au (gold) contents were not detected in all of the microwave leaching conditions. In the insoluble-residue, weight loss rate tended to decrease as the nitric acid concentration, microwave leaching time and sample addition increased. In an XRD analysis with solid-residue, it was suggested that gypsum and anglesite were formed due to dissolution of calcite and galena by nitric acid solution. When a fire assay was carried out with insoluble-residue, it was discovered that gold contents of the solid-residue were 1.3 (Au = 2,464.70 g/t) and 28.8 (52,952.80 g/t) times more than those of concentrate. But in the gold contents recovered, a severe gold nugget effect appeared. It is expected that the gold nugget effect will decrease if a sampling method of concentrate is improved in the microwave-nitric acid leaching experiments and filtering paper with smaller pore size is used for leaching solution and burned filter paper is used for sampling in lead-fire assay.

Application of Hierarchical ZnCo2O4 Hollow Nanofibers for Anode Materials in Lithium-ion Batteries (계층적 구조를 갖는 중공형 ZnCo2O4 나노 섬유의 리튬이온배터리 음극소재 적용)

  • Hierarchical $ZnCo_2O_4$ hollow nanofibers were prepared by electrospinning and subsequent heat-treatment process. The spinning solution containing polystyrene (PS) nanobeads was electrospun to nanofibers. During heat-treatment process, PS nanobeads in the composite were decomposed and therefore generated numerous pores uniformly in the structure, which facilitated the heat transfer and gas penetration into the structure. The resulting hierarchical $ZnCo_2O_4$ hollow nanofibers were applied as an anode material for lithium-ion batteries. The discharge capacity of the nanofibers was $815mA\;h\;g^{-1}$ ($646mA\;h\;cm^{-3}$) after the 300th cycle at a high current density of $1.0A\;g^{-1}$. However, $ZnCo_2O_4$ nanopowders showed the discharge capacity of $487mA\;h\;g^{-1}$ ($450mA\;h\;cm^{-3}$) after 300th cycle. The excellent lithium ion storage property of the hierarchical $ZnCo_2O_4$ hollow nanofibers was attributed to the synergetic effects of the hollow nanofiber structure and the $ZnCo_2O_4$ nanocrystals composing the shell. The hierarchical hollow nanofiber structure introduced in this study can be extended to various metal oxides for various applications, including energy storage.
