A Study on Sugars in Korean Sweet Rice Drink "Sikhye"(I) -Sugar Content and Its Composition- (식혜의 이소말토올리고당에 관한 연구(I) -정제 및 구조해석-)

  • A Korean traditional sweet rice drink "Sikye" was produced from the raw material of 20% of rice and 4% malt supplemented with 2l of tap water, by incubating the mixture at 6$0^{\circ}C$ for 7 hours. The product was found to contain 11.01% of maltose, 5.31% of isomaltooligosaccharides, 1.75% of maltotriose and 0.28% of glucose. Maltose, maltotriose and isomaltooligosaccharides in Sikye were seperated by ethanol (3 volume) precipitation repeated three times, followed by gel chromatography of Toyopearl HW-40S. 1H-NMR analysis revealed that the products of G2 and G3 size had only $\alpha$-1, 4-glucosidic linkage. but isomaltooligosaccharides showed both signal of $\alpha$-1, 4 and $\alpha$-1, 6-glucosidic linkage with its estimation ratio of 5:1. Isomaltooligosaccharides were hydrolyzed to produce maltooligosaccharide series from maltose to maltohexaose by pullulanase. These results, suggest that isomaltooligosaccharides were constructed by maltohexaose main chain with maltose or maltotriose and maltotetraose side chain.ide chain.

  • Limit dextrin of Korean beer(3 brands) and Japanese beer(21 brands) were separated by ethanol fractionation. Limit dextrin of Korean and Japanese beer was estimated to be 1.1%. 1H-NMR analysis revealed that the limit dextrin showed both signal of $\alpha$-1, 4- and $\alpha$-1, 6- glucosidic linkage with its estimation ratio of average 5.5:1. Limit dextrin was hydrolyzed to glucose with the yield of 57.22% by Aspergillus awamori $\alpha$-glucosidase(24.7 unit) plus human salivay $\alpha$-amylase(2.4 unit) in 100${mu}ell$ of 0.043M acetate buffer at 37$^{\circ}C$ for 5 hour. Among them, limit dextrin of Korean beer showed the highest hydrolysis rate of 76%. Small size sugars (64.8%) removed by ethanol fractionation and limit dextrin(21.4%) hydrolyzed by amylases that is digestable sugar. Non hydrolyzed limit dextrin(13.8%) by the amylases which can be a growth factor of Bifidobacterium in human intestine.

Structure of Hot-Water Soluble Rice Starch in Relation to the Structure of Rice Starch and Texture of Cooked Rice (열수 가용성 쌀 전분의 구조와 전분의 구조 및 밥의 텍스쳐와의 관련성)

  • The relationship between the molecular structure of rice starch and the texture of cooked rice was investigated using hot-water soluble rice starch. The structure of hot-water soluble starch bound amylose which was composed of small molecular weight and amylopectin which was composed of chain length of $\overline{DP}\;10{\sim}20$, and the average composition of amylose : amylopectin was 7 : 3. The molecular weight of amylose was the smaller and super long chain of amylopectin was the fewer, the extractable ratio of hot-water soluble rice starch was the higher. And hot-water solubility of rice starch be responsible for molecular structure of starch. On the texture of cooked rice, the extractable ratio of hot-water soluble rice starch was the higher, the hardness was the lower and the adhesiveness was the higher. The results suggest that the molecular structure of rice starch could be responsible for the texture of cooked rice.

Free Amino Acid and Free Sugar Contents of Liquid Koji Kochujang (액체국에 의한 숙성고추장의 유리아미노산과 유리당의 함량)

  • The three kinds of Kochujang were prepared with liquid koji equivalent to 30%, 50% and 70% of brewing water and the contents of free amino acids and free sugars during aging at $25^{\circ}C$ for 3 months were compared with those of the control group which was made from solid koji. All tested Kochujang were higher in glutamic acid, lysine and aspartic acid while lower in methionine, histidine and tryptophan. The content of total free amino acids was high in the order of control group, 70%, 50%, and 30% liquid koji group. Fructose and rhamnose were found in all groups, but glucose in only the control group and 70% liquid koji group. Fructose content was the highest of the free sugars and total free sugar content was higher in the control group and 70% liquid koji group than other groups.

  • Bio-based nylon 6-morpholinone random copolymers were prepared by the anionic ring opening polymerization of ${\varepsilon}$-caprolactam and morpholinone, both of which were prepared from lysine and glucose, respectively. From this work, a new biomass based nylon 6 with improved hydrophilicity was prepared. Optimizing the polymerization condition, copolymer with a viscosity-average molecular weight of 30000 g/mol was prepared, with a yield of 80%. It was possible to improve the hydrophilicity of nylon 6 by its copolymerization with morpholinone. The prepared nylon 6-morpholinone random copolymers were then characterized using several analytical methods such as DSC, TGA, XRD, viscosity measurement with U-shaped glass capillary viscometer and contact angle measurement.

  • This study is to improve the diagnosis of diabetes mellitus and the self-monitoring of blood glucose in people with diabetes by providing a non-invasive method of monitoring blood glucose. A near-infrared (NIR) spectrophotometer was used to measure absorption spectra of 80 glucose samples ranges from 1 mg/dL to 200 mg/dL, and shows the standard error of prediction 1.8 mg/dL. Also, to investigate the effect of interference in blood, NaCl and sand were added in glucose and found the standard error of prediction of 2.8 mg/dL and 3.8 mg/dL, respectively. A new and more accurate calibration system for the spectrophotometer was developed from systematic study of light scattering, which cause nonlinear spectrophotometer response.

  • Enzymatic cellulose hydrolysis depends on the several factors such as the structural features (CrI, particle size and surface area, etc.), the nature of cellulase enzyme system, the inhibitory effects of products, and enzyme deactivation. At the presence of products on the initial hydro- lysis rate of cellulose, cellobiose has more severe inhibitory effect than glucose. Othewise, the inhibition effect of products for adsorbed enzyme is related to the glucose and cellobiose conentration hyperbolically. Enzyme deactivation of FPA and ${\beta}-glucosidase$ were expressed by exponential decay profile.

  • Because the nanoporous electrodes has large electrochemical surface areas, extensive studies have been focused on their fabrication methods. In this paper, a method for introducing a nanoporous gold (NPG) structure on the surface of an ultramicroelectrode (UME) using potentiostatic anodization was investigated. A well-defined NPG structure was introduced on the surface of the UME when a potential of 1.3 V was applied in 0.1 M phosphate buffer solution (pH 8) containing 1 M KCl. The anodic oxidation efficiency was investigated by observing the effect of the applied potential, the reaction time, and the size of the electrode on the roughness factor (Rf) of the prepared NPG-UMEs. In a short time of about 10 minutes, NPG-UME with a large Rf value of about 2000 could be prepared, which could be effectively used for electrochemical glucose detection. The results shown in this work are expected to have great applicability when performing electrochemical analysis with a small sample volume.

  • To evaluate the applicability of cellular energy allocation (CEA) in the bivalves as a biomarker for the assessment of environmental contamination, the energy contents and energy consumption in several tissues of the Manila clam, Ruditapes philippinarum were analyzed. The contents of lipid, glucose, protein and electron transport system (ETS) activity in the foot, siphons, gills, and body of R. philippinarum exposed to crude oil-spiked sediments were measured at 1, 2, 4, 7, 10 days after exposure. The reserved energy (energy available, EA) in the lipid, glucose and protein decreased as contamination level and exposure time increased. In contrast, the ETS activity (energy consumed, EC) showed the reverse tendency. The order of available energy contents were foot > siphons > gill > body. Significant differences in both EA and EC were found only at the highest contamination level (58.3 mg TPAHs/kg DW). EA decreased significantly in the foot and gill at 1 day, in the body at 2 and 7 days after exposure. EC increased significantly in the body at 4 days after exposure. CEA showed higher sensitivity to the contamination than EA or EC. Especially, CEA in the foot and body decreased significantly at lower ranges of contamination level (as low as 6.5 mg TPAHs/kg DW) during 1 to 7 days after exposure. The CEA is more useful than EA or EC alone for the assessment of sediment contamination at lower level that acute toxicity could not be detected. CEA analyses in the body of R. philippinarum after 4 days' exposure to contaminated sediments seem to be the most sensitive and reliable.

Changes of Furfural and Levulinic Acid Yield from Small-diameter Quercus mongolica Depending on Dilute Acid Pretreatment Conditions (약산 전처리 조건에 따른 소경 신갈나무 유래 푸르푸랄 및 레불린산의 함량 변화)

  • In this study, dilute acid pretreatment was operated using small-diameter Quercus mongolica for evaluating the yield change of furfural and levulinic acid depending on pretreatment factors. The dilute acid pretreatment was conducted depending on reaction temperature ($140-180^{\circ}C$), reaction time (10-30 min), and sulfuric acid concentration (0-2%, w/w). Then, glucose, XMG (xylose + mannose + galactose), furfural, and levulinic acid contents in the liquid hydrolyzate were measured and analyzed after pretreatment. Glucose content increased to 16.02% as reaction temperature, reaction time, and sulfuric acid concentration increased, but it decreased at the sulfuric acid concentration of 2% (reaction temperature: > $170^{\circ}C$, reaction time: > 20 min). On the other hand, reaction temperature had a strong influenced on XMG content, and XMG content decreased to 1.63% through increasing of reaction temperature and sulfuric acid concentration, but XMG content was less affected by changes of reaction time. Furfural content increased with the increase of reaction temperature, reaction time, and sulfuric acid concentration, and maximum furfural content was 7.61% (reaction temperature: $180^{\circ}C$, reaction time: 20 min, sulfuric acid concentration: 1%) based on a weight of raw material, while furfural content was dropped in more severe condition than in maximum furfural content condition. Levulinic acid content also increased with higher reaction temperature, reaction time, and sulfuric acid concentration. Especially, the sharp increase of levulinic acid content was observed above $170^{\circ}C$, and maximum levulinic acid content was 10.98% (reaction temperature: $180^{\circ}C$, reaction time: 30 min, sulfuric acid concentration: 2%). However, less than 1% of furfural and levulinic acid content was obtained in non-acidic catalyst condition that in whole conditions of reaction temperature and reaction time.
