Characteristics of Pichia anomala K15 Producing Killer Toxin Isolated from Traditional Nuruk (전통 누룩으로부터 분리된 Killer Toxin 생산 균주 Pichia anomala K15의 특성)

  • In this study, killer yeasts were isolated from traditional Nuruk to improve storage and suppress contaminant in food industry. Among killer yeasts, yeast K15 showed strong killer toxin activity and inhibited growth of Salmonella Typhimurium and Vibrio parahaemolyticus. Killer yeast K15 was identified with Pichia anomala by the Microlog TM 4.0 identification system and homology of the ITS sequence. Killer toxin generated from P. anomala K15 was inactivated by pronase E and suggested to be a protein. Therefore killer toxin of P. anomala K15 was thought to be safe in human such as bacteriocin. P. anomala K15 was sufficient for growth in 50% glucose and could be used to prevent contaminant in initial stages of alcohol beverage fermentation.

  • Bacillus coagulans has been considered to be a prominent candidate for probiotics as well as a thermotolerant biomaterial producer. However, the species has not attracted the attention of Korean researchers, nor has it been commercialized in Korea. Therefore, isolates for functional studies are not readily available. To secure B. coagulans resources for future applications, we developed a rapid isolation method for the species from rice straw. Introduction of the enrichment culture at $50^{\circ}C$, the selection of acid producers with $CaCO_3$ supplemented medium, and the elimination of enterococci by selective medium, rendered the successful and rapid isolation of B. coagulans strains.

Cellulase and Xylanase Activity of Compost-promoting Bacteria Bacillus sp. SJ21 (부숙촉진 세균 Bacillus sp. SJ21 균주의 cellulase와 xylanase 활성)

  • In order to isolate thermophilic compost-promoting bacteria with high activity of cellulase and xylanase, spent mushroom substrates with sawdust were collected from mushroom cultivation farm, Jinju, Gyeongnam in Korea. Among of the isolates, one strain, designated SJ21 was selected by agar diffusion method. The strain SJ21 was identified as members of the Bacillus lincheniformis by biochemical characteristics using Bacillus ID kit and VITEK 2 system. Comparative 16S rDNA gene sequence analysis showed that strain SJ21 formed a distinct phylogenetic tree within the genus Bacillus and was most closely related to Bacillus subtilis with 16S rDNA gene sequence similarity of 99%. On the basis of its physiological properties, biochemical characteristics and phylogenetic distinctiveness, strain SJ21 was classified within the genus Bacillus, for which the name Bacillus sp. SJ21 is proposed. The cellulase and xylanase activity of Bacillus sp. SJ21 was slightly increased according to bacterial population from exponential phase to stationary phase in growth curve for Bacillus sp. SJ21.

Production and fermentation characteristics of seafood kimchi started with Leuconostoc mesenteriodes SK-1 isolated from octopus baechu kimchi (문어 배추김치에서 분리한 Leuconostoc mesenteroides SK-1을 이용한 수산물김치의 제조 및 발효 특성)

  • This study was carried out to investigate the effectiveness of Leuconostoc mesenteroides isolated from octopus baechu kimchi as a potential starter for seafood kimchi. L. mesenteroides is lactic acid bacterium currently used as a starter for kimchi production. We selected the most effective L. mesenteroides strain from the 7 strains isolated from octopus baechu kimchi and, based on biochemical properties and 16S rRNA sequencing, identified the selected strain as L. mesenteroides SK-1. The SK-1 strain exhibited acid-tolerance, good survival capacity, and excellent dextran productivity. We investigated the effects the SK-1 of starter on seafood kimchi fermentation. Octopus baechu kimchi was fermented with L. mesenteroides SK-1 at $4^{\circ}C$ for 35 d. The decrease in pH and increase in acidity in octopus baechu kimchi fermented with the SK-1 starter occurred more quickly than that in the control kimchi indicating that. Octopus baechu kimchi with SK-1 starter has a relatively slow rate of increase in lactic acid production. As a result, octopus baechu kimchi prepared with L. mesenteroides SK-1 can be maintained at a suitable ripening degree over an extended period of time compared to that of the control kimchi, Moreover, the octopus baechu kimchi started with L. mesenteroides SK-1 has excellent sensory properties, including a refreshing taste, and a weak sour odor.

  • The antagonistic fluorescent pseudomonas, which was isolated from continuous cropping rhizosphere of pepper and cucumber, was identified as Pseudomonas fluorescens (P.f.). For further study, transformant was derived from the isolated P.f. after spontaneous mutation to give antibiotic resistance to nalidixic acid and rifampicin as marked strain. Both P.f. and transformant strains were used for this study and the results obtained were summarized as follows. 1. One of the most effective antagonistic strain, KR164, was selected against F. solani, F. oxysporum, R. solani and this strain was identified and classified as Pseudomonas fluorescens biotype IV. 2. Transformant, KR1641, was derived from strain KR164 and both strains had the same biological and biochemical characteristics. 3, Mycelial lysis and abnormal mycelia of plant pathogenic fungi were microscopically observed after simultaneous culture of fungus and given bacterial strain. 4. The length of chinese cabbage to the autolyzed became longer with given bacterial strain in dark culture. 5. Percentage of germination, number of leaves, length of height, and length of root in chinese cabbage in pot experiment were improved by inoculation of given bacterial strain. 6. The number of given bacterial strain kept generally stable until 34 days after inoculation of itself in pot experiment. Inoculation of given bacterial strain did affect the number of plant disease fungi to be decreased but did not affect the number of other bacteria, Bacillus, in pot experiment.

Resistance of Chili Pepper Cultivars to Isolates of Phytophthora capsici (Phytophthora capsici 균주들에 대한 고추 품종들의 저항성)

  • Resistance of one hundred commercialized cultivars of chili pepper to four isolates of Phytophthora capsici was evaluated under controlled environmental conditions. The cultivars are commercialized as resistant (59%) and susceptible (41%) to Phytophthora blight in Korea. Mean disease severities of the cultivars on P. capsici MY-1, KPC-1, JHAI1-7, and KPC-7 isolates were 37, 55, 60, and 74%, respectively. In addition, 38 for MY-1, 48 for KPC-1, 56 for JHAI1-7, and 76 cultivars for KPC-7 showed susceptibility. To P. capsici MY-1, the weakest pathogenicity isolate among them, 59 cultivars represented high resistance. By contrast, only six cultivars showed high resistance to P. capsici KPC-7, the strongest isolate. Furthermore, resistance of most cultivars except for three cultivars was negatively correlated with the virulence of P. capsici isolates. And isolate-specific resistance of the chili pepper cultivars could not be found. Among them, six cultivars showing resistance to all the tested isolates were selected for further study. The development of Phytophthora blight on the six cultivars according to inoculum density ($5{\times}10^4$ to $1.5{\times}10^6$ sporangia/pot) and incubation temperature (25 to $30^{\circ}C$) after inoculation of P. capsici was tested. Resistance of the cultivars to P. capsici KPC-1 and JHAI1-7, moderately pathogenic isolates, was hardly affected. But to KPC-7 isolate, the highly resistant cultivars showed susceptiblility or moderate resistance when the seedlings were inoculated with inoculum density of $1.5{\times}10^6$ sporangia/pot and incubated at 28 to $30^{\circ}C$. From these results, it is likely that resistance of chili pepper cultivars to Phytophthora blight is affected by the virulence of P. capsici isolate.

Isolation and Identification of Hydrolytic Enzyme-producing Bacteria from Spent Mushroom Substrate (버섯부산물유래 가수분해효소분비 박테리아의 분리 및 동정)

  • This study was conducted to isolate and identify xylanase- and cellulase-producing thermophilic bacteria from stacked spent mushroom substrates and to determine the optimal medium conditions for their growth. Bacteria with the highest xylanase and CMCase activities were strain 3 and 201-7. Both of them were identified as Bacillus spp. and named Bacillus subtilis KU3 and Bacillus subtilis KU201-7. The optimal medium condition of Bacillus subtilis KU3 was obtained when 3%(w/v) of yeast extract and 1%(w/v) of maltose were used as nitrogen and carbon sources, respectively. That of Bacillus subtilis KU201-7 was obtained when 0.5%(w/v) of yeast extract and 0.5%(w/v) of CMC were used as nitrogen and carbon sources, respectively.

  • Fish flesh of Alaska pollack containing 5% of corn starch were fermented with 8 strains of mold and monitoring of their flavor characteristics, acceptability, nucleotides and their related compounds, amino acid compositions were conducted. All strains were grown vigorously on fish flesh media and formed their characteristic spores with unique flavor by strains. Amino type nitrogen $(NH_2-N)$ content of fermented fish flesh (FFF) were 25-26 times higher than that of raw flesh and 6-15 times higher in extractable nitrogen (Ex-N) content . The strains which produce more ADP (Adenosine 5'-diphosphate) in FFF also showed much higher level of IMP (Inosine 5'-monophosphate) and GMP (Guanosine 5'-monophosphate) content than that of raw flesh. Amino acid composition were differ by strain but lysine was generally highest and arginine, glutamic acid, leucine and alanine in order In review of sensory evaluation , total content of nucleotides, $NH_2-N$, Ex-N and amino acid compositions, suitable strains for fish flesh fermentation were Aspergillus oryzae KFCC 11371, Asp. oryzae KFCC 32343, Penicillium roqueforti KFCC 11269 and Asp. quercinus.

Cloning Genes Involved in Aniline Degradation from Delftia acidovorans. (Delftia acidovorans로부터 Aniline 분해관련 유전자의 분리)

  • Delftia acidovorans 51-A isolated from river water degrades aniline. In order to clone genes involved in aniline degradation, transposon Tn5-B20 was inserted into the strain 51-A to generate a mutant strain 10-4-2 that cannot utilize aniline as a carbon source. The mutant strain was not an auxotroph but could not degrade aniline. Southern hybridization analysis indicated that the transposon was inserted into the mutant bacterial DNA as a single copy. Flanking DNA fragment of Tn5-B2O insertion was cloned and sequenced. DNA sequence analysis revealed three ORFs encoding TdnQ, TdnT, and TdnA 1 that arc responsible for catechol formation from aniline through oxidative deamination. The analysis also confirmed that Tn5-B2O was inserted at the immediate downstream of tdnA1. The result suggests that the transposon insertion behind tdirA1 disrupted the pathway of the catechol formation from aniline, resulting in the mutant phenotype, which cannot degrade aniline. A large plasmid over 100-kb in size was detected from D. acidovorans 51-A and Southern hybridization analysis with Tn5-B2O probe showed that the transposon was inserted on the plasmid named pTDN51. Our results indicated that the tdn genes on pTDN51 of D. acidovorans 51-A are involved in aniline degradation.

Control of Ragweed (Ambrosia artemisiifolia) of Mutant N-29 Broth Filtrate of Streptomyces scopuliridis KR-001 (토양 방선균 N-29 배양 여액의 생태계교란 식물 돼지풀 방제효과)

  • We isolated Streptomyces scopuliridis KR-001 that produced herbicidal active metabolite(s) against several grass and broad leaf weeds. In this study, potential as natural herbicide of mutant N-29 broth filtrate of S. scopuliridis KR-001 was investigated to Ambrosia artemisiifilia in a greenhouse and field condition. The broth filtrate of mutant N-29 by foliar application showed a strong herbicidal activity to A. artemisiifilia with leaf stage in a greenhouse condition. Also, field trial of foliar application within treatment range had effectively controlled with early and middle stage of A. artemisiifilia at the natural habitats. Phytotoxic symptoms of mutant N-29 broth filtrate by foliar application were wilting and discoloration, and burn-down of leaves and finally plant death. These results suggest that mutant N-29 broth filtrate is considered possible as a natural herbicide for controlling environmentally friend to invasive alien plant such as A. artemisiifilia and may provide a new lead molecule for a more efficient herbicide.
