Effects of inoculation with selected Rhizobium japonicum on the yield, nodule formation and nitrogen fixing activity of soybean(Glycine max) (선발대두근류균(選拔大豆根瘤菌)의 접종(接種)의 대두근류형성(大豆根瘤形成) 질소고정호성(窒素固定浩性) 및 수량(收量)에 미치는 영향(影響))

  • To develop soybean inoculant the effects of inoculation with selected Rhizobium japonicum strains were tested in the field, and the results were as following. 1. With the inoculation of selected strains, increase of nodule number, nodule weight and nitrogen fixing activity were observed with or without applying N-fertilizer. 2. Chlorophyll contents was increased and senescence of plant was delayed with the inoculation of selected strains in case without applying N-fertilizer. But there was little difference of chlorophyll contents in case with applying N-fertilizer. 3. There were little differences of plant height and number of nodes, and some increase of plant stem dry matter and number of pods with inoculation of selected strains. 4. The mean yield of inoculation group (363-374kg/10a) was increased by 8-11% comparing to the uninoculated group (337kg/10a) in case without applyiag N-fertilizer. The mean yield of inoculated group (368-384kg/10a) was increased by 9-14% also in case with applying N-fertilizer.

Breeding a new white button mushroom cultivar 'Dodam' (백색 양송이 신품종 '도담' 육성)

  • The button mushroom, Agaricus bisporus, is one of the most widely cultivated mushrooms. The domestic production of cultivated mushrooms in 2016 was approximately 10,173 tons, with a total value of 59 billion won. Currently, 10 cultivars have been developed; however, Korean farmers continue to demand new cultivars of mushrooms with improved quality. To breed superior lines, KMCC00754 and KMCC00775 were selected as mother strains from the 170 collected genetic resources. The putative homokaryotic strains were 25 strains from the 120 SSIs of KMCC00754 and six strains from the 120 SSIs of KMCC00775 selected by using the AbSSR45 marker. These homokaryotic strains were crossed with each other and the crossing was confirmed by SSR analysis. Seventy-four lines were crossed into 150 lines for a 50% ratio of crossing. Abs2-2015-16 was selected as a superior line by three cultivations. A new cultivar, 'Dodam', was developed in 2017.

Analysis of the behavior of microorganisms isolated from the medium during cultivation of Agaricus bisporus (button mushroom) (양송이 재배 중 배지에서 분리한 미생물의 상호작용 분석)

  • This experiment investigates the characteristics of microorganisms isolated from a medium during cultivation process and reveals the relationship between these microorganisms and the growth of Agaricus bisporus. The domestically grown strains of Agaricus bisporus displayed a higher inhibition growth rate against microorganisms isolated from straw, chicken manure, and medium than imported strains. As for inhibition of mycelial growth among mushroom cultivars of the microorganisms separated by each fermentation step from the mushroom medium, the domestic cultivar, 'Saedo,' grew more vigorously among other cultivars. As the fermentation progressed, it was confirmed that inhibitation of microorganisms against Agaricus bisporus was weakened. A total of 21 strains of microorganisms that promote mushroom growth were isolated in the 4th turning process, and the microorganisms isolated from the mushroom medium affect the growth and as yield of the mushroom through secretory substances.

Physiological Functionalities and Enzyme Activities of Non-pathogenic Pigmented Wild Yeasts (비병원성 야생 색소 효모들의 생리활성과 효소활성)

  • This study aimed to produce novel bioactive compounds from non-pathogenic pigmented wild yeasts. Culture supernatants and cell-free extracts of non-pathogenic pigmented yeast strains were prepared, and their physiological functionalities and enzyme activities were measured. Cell-free extracts from Rhodosporidium paludigenum HHGG35-1 and culture supernatants from Rhodosporidium diobovatum NMD18-1 demonstrated very high antioxidant activity (76.6%) and anti-gout xanthin oxidase inhibitory activity (86.2%), respectively. Maximal production of the antioxidants (76.9%) was obtained when Rh. Paludigenum HHGG35-1 was cultured in a yeasts extract-peptone-dextrose (YPD) medium (pH 6.5) at 30℃ for 24 h. The xanthin oxidase inhibitor was also maximally produced (91.6%) when Rh. Diobovatum NMD18-1 was cultured at 30℃ for 96h in a YPD medium (pH 6.5). Rh. Paludigenum HHGG35-1 was oval in shape and formed ascospre. The Rh.diobovatum NMD18-1 specimen displayed dimensions of 1.6 × 1.6 ㎛ and produced ascospores; however, it did not form pseudomycelium. Both of Rh. Paludigenum HHGG35-1 and Rh. Diobovatum NMD18-1 grew well in a 40%-glucose-containing YPD medium and 10%-NaCl-containing YPD medium.

  • The useful fungi which secret extracellular enzymes was selected for deinking agent of old newsprint. Five fungal strains were isolated from a paper mill soil ground. The CMCase, FPase and xylanase activities of fungi on the liquid culture were investigated at optimal growth conditions. The results of this study were as follow: The optimal pH and temperature for culture growth were 4~8 and 27~$35^{\circ}C$, respectively. For screening of extracellular enzymes at optimal culture conditions the optimal culture period were less than 6-7 days. Fusarium pallidoroseum and Aspergiilus niger which shows relatively higher CMCase, FPase and xylanase activities than the other species were selected for further enzymatic deinking research.

Fungicide Selection for Control of Lycium chinense Anthracnose Caused by Colletotrichum spp. (구기자 탄저병 방제를 위한 살균제 선발)

  • Anthracnose caused by Colletotrichum spp. has been known as a significant disease which commonly infects to the fruits of Chinese matrimony (Lycium chinese) in the field conditions. To select effective fungicides for the control of Chinese matrimony anthracnose, the antifungal activity of 15 fungicides were evaluated with 13 different strains of Colletotrichum spp. in the laboratory condition. Six fungicides (Dithianon WG, Tebuconazole SC, Tebuconazole WG, Trifloxystrobin + Tebuconazole SC, Azoxystrobin SC and Polyoxin D zinc salt + Carbendazim WP) out of them, showed effective suppression with the mycelium growth of pathogenic fungus, and were selected to test in vivo of the field condition. Five fungicides, Dithianon WG, Tebuconazole WG, Trifloxystrobin + Tebuconazole SC, Tebuconazole SC and Azoxystrobin SC, were significantly effective to protect anthracnose of Chinese matrimony, the variety "Chungyang Jerae".

Control Efficacy of Fungicide Injection on Oak Wilt in the Field (살균제 나무주사를 이용한 참나무 시들음병 방제 효과)

  • Oak wilt caused by Raffaelea quercus-mongolicae was first noticed in South Korea in 2004 and, ever since, its distribution and damage have been increasing. To screen a fungicide effective for oak wilt control by tree injection, laboratory and field experiments were conducted. Ten fungicides and one antibiotic were examined in vivo for their effectiveness in restricting the growth of R. quercus-mongolicae and R. quercivora (Japanese oak wilt pathogen) isolates. To the Korean isolates of R. quercus-mongolicae, chlorothalonil showed the highest fungicidal effects, followed by benomyl and propiconazole. To the Japanese one, propiconazole was highest in the fungicidal effectiveness, followed by benomyl and bitertanol. Propiconazole was selected for field-testing of its control efficacy because it showed good fungicidal effects in vitro and systemic activity. The control efficacy in the field was 87.5% in the first year of injection and 66.7% in the second year, indicating the fungicidal effects last at least over one year.

  • For the study of human intestinal environments with the intestinal bacteria producing ${\beta}-glucuronidase\;and \;7{\alpha}-dehydroxylase$, genus Clostridium, known as the producer, were isolated from the fecal microflora. Through screening twice for one person, fecal microflora without major bacterial group seemed to be changed, which indicated the microflora would be changeable by the diet factors. With using Neomycin-Nagler selective medium during the screening, 14 Clostridium spp. were isolated and then the harmful enzyme activities were determined. Isolate-11 among them produced strongly ${\beta}-glucuronidase$ and its activity was 0.021 unit/mg Protein. However, the strain producing $7{\alpha}-dehydroxylase$ was not isolated. The Isolate-11 was tentatively identified as Clostridium scatologenes through cultural and physiological characteristic.

Molecular Diversity of Rhizobacteria in Ginseng Soil and Their Plant Benefiting Attributes (인삼토양 내 근권세균의 다양성 및 식물에의 유용 특성)

  • The purpose of this study was to investigate the molecular diversity of rhizobacteria associated with ginseng of varying age levels and their plant benefiting attributes. A total of 143 different isolates belonging to 15 different bacterial genera were recovered. Although variation was found in the rhizobacterial community due to age of the plant, majority of bacteria belong to Firmicutes (58%). In which, Bacillus was found to be the predominant genus irrespective of age of the ginseng. To assess the plant benefiting attributes, 30 representative isolates were selected. The results indicated that some of the isolates could exhibit multiple plant growth promoting traits like secretion of cell wall degrading enzymes, production of indole-3-acetic acid, synthesis of siderophores, solubilization of phosphates and soil pathogens inhibition. It can be suggested that strains of B. subtilis, B. amyloliquefaciens, B. velezensis, and B. licheniformis were positive for all the above traits, which have potential to be used as plant growth promoting inoculants to improve ginseng crop in the future.

Relationships Between Pathogenicty and Activities of Polygalacturonase, Laccase, and ${\beta}$-Glucosidase Produced by Botrytis cinerea (Botrytis cinerea 균주들이 생산하는 Polygalacturonase, Laccase, ${\beta}$-glucosidase의 균주 간 화성 및 병원성과의 상관관계)

  • Activities of polygalacturonase, laccase, and intra- and extra-cellular $\beta$-glucosidase produced by 20 Botrytis cinerea isolates in liquid culture media containing cucumber cell was as a carbon source were measured and their relationships to the pathogenicity were analyzed. No significant correlations between these enzyme activities and the pathogenicity of B. cinerea were found. Mycelial growth rate on Bayendamm media, however, was higthly correlated with the pathogenicity (r=0.522) anong these isolates. Immuno-blot analysis of the culture filtrate using antibody against against exo-polygalacturonase revealed that only one band with molecular weight of 66 kDa was detected amone 34 tested isolates. It appears that these enzymes may not be primary factors in dermining the pathogenicity of B. cinerea.
