Growth-promoting effect on Tricholoma matsutake mycelium by Terrabacteria isolated from pine mushroom habitats in Korea (국내 송이 자생지에서 분리된 Terrabacteria에 의한 송이균사체 생장촉진 효과)

  • To cultivate pine mushroom (Tricholoma matsutake) artificially, co-cultivation with microorganisms has been introduced. Here, experiments were performed to assess the growth-promoting effect of bacteria on T. matsutake mycelia. Bacteria were isolated from soil samples collected in Yangyang County, Korea. Four of the bacterial isolates (Y22_B06, Y22_B11, Y22_B18, and Y22_B22) exhibited a growth-promoting effect on T. matsutake mycelia (154.67%, 125.91%, 134.06%, and 158.28%, respectively). To analyze the characteristics of the bacteria, especially the antifungal activity, 𝛼-amylase and cellulase activity assays were performed. In comparison with the controls, the isolated bacteria exhibited low 𝛼-amylase and cellulase activity. 16S rRNA gene sequencing was performed to identify the four bacterial isolates. The isolates belonged to the Terrabacteria group and were identified as Microbacterium paraoxydans, Paenibacillus castaneae, Peribacillus frigoritolerans, and P. butanolivorans. These bacterial isolates are expected to have contributed to the growth promotion of T. matsutake mycelia and the artificial cultivation of T. matsutake.

Growth promoting effect on Tricholoma matsutake mycelium by bacteria from fairy Ring in Bonghwa-gun, Korea (국내 봉화 송이 자생지 내 균환 유래 토양세균의 송이균사체 생장촉진 효과)

  • As a member of ectomycorrhizal fungi, Tricholoma matsutake has a symbiotic relationship with its host, Pinus densiflora. To cultivate T. matsutake artificially, the co-cultivation of T. matsutake mycelia and bacteria from shiro was introduced. In this study, bacteria were isolated from soil samples in Bonghwa-gun, and seven bacterial isolates (B22_7_B05, B22_7_B06, B22_7_B07, B22_7_B08, B22_7_B10, B22_7_B13, and B22_7_B14) promoted the growth of T. matsutake mycelia (147.48, 232.11, 266.72, 211.43, 175.17, 154.62, and 177.92%, respectively). Sequencing of the 16S rRNA region of the isolated bacteria was performed. B22_7_B05 and B22_7_B10 were identified as Bacillus toyonensis, B22_7_B06 and B22_7_B08 as Paenibacillus taichungensis, B22_7_B07 and B22_7_B14 as P. gorilla, and B22_7_B13 as P. odorifer. These bacterial isolates were associated with the shiro community and are expected to contribute to the cultivation of T. matsutake.

The Genetic Characteristics of Strains and the Optimal Condition for Mycelial Growth of Naematoloma sublateritium on Sawdust Media (개암버섯 균주의 유전적 특성과 최적 배지조건에 관한 시험)

  • In order to compare collected strains of Naematoloma sublateritium, electrophoretic isozyme and RAPD band patterns of DNA from mycelia were studied. The two characteristics of the strain, ASI 11010 were much different from those of ASI 11005 and 11006. These patterns may be used as valuable criteria for identifying strains of N. sublateritium. Optimal conditions on the vegetative growth of N. sublateritium using sawdust media were favourable on alder sawdust media added with 30% beer waste. Optimal water content was 70%.

Isolation of Antagonistic Bacteria to Phytophthora capsici for Biological Control of Phytophthora blight of Red Pepper (고추역병의 생물학적 방제를 위한 길항세균의 분리)

  • To isolate of antagonistic bacteria to Phytophthora capsici, which cause Phytophthora blight in red pepper, 237 isolates of Pseudomonas spp. and 260 isolates of Bacillus spp. were screened in selective media from rhizosphere soils of red pepper at Kyongsan, Kyongju, Yongchon and Euisung in Kyongbuk. Among total 497 isolates, 8 isolates of Pseudomonas spp and 4 isolates of Bacillus spp. inhibited the mycelial growth of Phytophthora capsici above 50$\%$ . These antagonistic bacteria showed more inhibitory effect on TSA (tryptic soy agar) than V-8 juice agar. Four isolates, P0704, P1201, B1101 and B1901, showing the most prominent antagonistic activity were selected and identified as P. cepacia (P0704, P1201), B. polymyxa (B1101) and B. subtilis (B1901), respectively. Cell free filtrates of these isolates were shown to inhibit zoosporangia germination and mycelial growth of p. capsici indicating that these isolates turned out to be bacteria producing antifungal substances. As a result of antagonistic test to Phytophthora blight in green house p. cepacia (P0704) showed the highest antagonistic effect with 46.7$\%$ and the rest of them were in the range of 13.4$\%$ to 26.7$\%$ .

Comparison of Growth Characteristics of Tricholoma matsutake Mycelium Among the Types of Air Bubble Bioreactor (공기부양식 생물반응기의 형태별 송이균사의 생장특성 비교)

  • In order to select suitable bioreactor type inhibiting cell stress during submerged culture of Tricholoma matsutake mycelium, the growth characteristics and ergosterol contents were investigated using the external-loop type of air-lift bioreactor (ETAB), balloon type of air bubble bioreactor (BTBB) and column type of air bubble bioreactor (CTBB). Dry weights of the T. matsutake in the BTBB, ETAB and CTBB were 12 g, 11.4 g, and 9.5 g per 1 litter, respectively. BTBB, ETAB and CTBB reached stagnant phases 16, 20, and 24 days after cultivation, respectively, The BTBB was more suitable for liquid culture of T. matsutake mycelium compared to other bioreactors owing to much mycelia product and short culture period. The ergosterol contents produced by the mycelium in the bioreactors were in sequence of BTBB, CTBB, and ETAB at every growth phase. BTBB might affect the mycelium on producing the smallest size of pellets. BTBB and CTBB got the mycelium precipitated and coagulated under operation of bioreactor sparser, whereas ETAB shown no effect of above phenomenon. A renovated bioreactor combined between a balloon shape of BTBB and an external-loop of ETAB was developed to enhance the efficiency of culture technique.

  • Carpophores of ten Korean strains of Lentinus edodes (Berk.) Singer, an antitumor polysaccharide producing fungus, were extracted with 0.1N NaOH solution. The extracts were dialized for seven days in distilled water and lyophilized to produce crude polysaccharide powders. Thus obtained crude polysaccharide samples were assayed for sugar contents by colorimetric method with anthrone reagent. Among ten strains examined Lentinus edododes-DMC7 was found to be the richest strain in polysaccharide content of carpophores. By shake culture experiment for biomass production, L. edodes-DMC7 was found to be the second most productive strain among seven strains examined. Cultural characteristics of L. edodes-DMC7 were investigated by shake culture method. The best result was obtained when L. edodes-DMC7 was cultured in the medium containing glucose 8g, starch 80g, yeast extract 12g, $KH_2PO_4\;0.87g,\;MgSO_4{\cdot}7H_2O\;O.5g,\;CaCl_2\;0.3g,\;FeSO_4{\cdot}7H_2O\;10mg\;ZnSO_4{\cdot}7H_2O\;4mg,\;CuSO_4{\cdot}5H_2O\;lmg,\;MnCl_2{\cdot}4H_2O\;7mg\;per\;11\;at\;28^{\circ}C$, 180 rpm, for 12 days. Thus thirty-three grams of dry mycelia was obtained per one liter of medium.

Function of Dual Specificity Kinase, ScKns1, in Adhesive and Filamentous Growth of Saccharomyces cerevisiae (Saccharomyces cerevisiae의 균사형 생장에서 이중 특이성 인산화 효소, ScKns1p의 기능 분석)

  • In the previous study with the Saccharomyces cerevisiae S288c strains, no known function of the dual specificity kinase, ScKns1, was reported because its gene deletion did not show any noticeable phenotypic changes. Recent study with fission yeast, however, revealed the involvement of the LAMMER kinase in flocculation, filamentous growth, oxidative stress, and so on. Therefore we made Sckns1-deletion mutants with the ${\Sigma}1278b$-background, with which one can induce filamentous and adhesive growth in contrast to those of the S288c-background. The $Sckns1{\Delta}$ strains of both haploid and diploid showed defect in filamentous growth under conditions for inducing the filamentous growth such as nitrogen starvation and butanol treatment. Both kinds of the deletion mutants also showed decrease in adhesive growth on agar surface. Interestingly enough the defects of the $Sckns1{\Delta}$ strains were suppressed by the over-expression of each gene for the components of the MAPK signaling pathway such as STE11, STE12, and TEC1, respectively, but not by the upstream components, RAS2 and STE20, respectively. Although further investigations are required, these results indicate that the ScKns1 may act in place between the Ste20 and the Ste11 of the S. cerevisiae MAPK cascade.

Selection of suitable conditions of mycelial growth and materials of bag cultivation in Sparassis crispa (꽃송이버섯의 액체배양조건 및 봉지재배 적합배지 재료선발)

  • Sparassis crispa(Cauliflower mushroom) was an edible mushroom that shows remarkably high contents of ${\beta}$-glucan compared to other edible mushroom. In this study, we aimed to select suitable conditions and materials for mycelial growth and fruit body production. The longer saccharification time of barely result in higher sugar content for eight hours. The optimal sugar content of media for mycelial growth was showed at level of 6 $^{\circ}Brix$. Optimum temperature and pH for mycelial growth in liquid spawn were $25^{\circ}C$ and pH 5.0~6.0, respectively. In case of sawdust was used Larix kaempferi as main material, the fruit body yield and cultivation period of supplemented with 10% wheat flour and 20% corn flour were highest and fastest, respectively.

  • Among the organic sources of nitrogen tested, yeast extract and soytone were excellent for the mycelial growth of Tricholoma matsutake DGUM 26001. The mycelial growth was enhanced, when yeast extract at the concentration up to 1.0% was added to the starchpyridoxine medium. After 30-day cultivation of the mycelia at $24^{\circ}C$ in the medium supplemented with yeast extract, 518 mg/50 ml of dry mycelia could be harvested.
