The Cytotoxic Effect of Oral Wet Wipes on Gingival Cells (시판 중인 구강청결티슈의 세포 독성 관찰)

  • Wet wipes are being increasingly used because of their convenience. Particularly, oral wet wipes are useful for regular cleaning of a baby's mouth after birth. Therefore, the consumption of oral wet wipes has increased over the past few years and a variety of products are commercially available. However, product information on safety is not sufficiently provided and still raises doubts regarding adverse effects. To confirm the safety of wet wipes as an oral hygiene item and provide information for their use, we investigated the cytotoxicity of oral wet wipes and verified the underlying mechanism. The anti-bacterial effect of oral wet wipes was analyzed using the disk diffusion method. The cytotoxic effects of oral wet wipes were observed based on morphological changes using microscopy and determined using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay in gingival epithelial cells and gingival fibroblasts. Evaluation of apoptosis by oral wet wipes was explored using propidium iodide flow cytometric analysis and a terminal deoxynucleotidyl transferase (TdT)-mediated deoxyuridine triphosphate (dUTP) nick-end labeling (TUNEL) assay. Apoptosis-related molecules were also analyzed using western blotting. Five types of oral wet wipes were tested, and two products from Fisher-Price and Dr. Kennedy revealed strong cytotoxic effects on gingiva epithelial cells and gingiva fibroblasts, although they also showed intense anti-bacterial effects on oral bacteria. Cell cycle arrest in the G2/M phase and apoptosis were observed based on treatment of extracts from Fisher-Price and Dr. KENNEDY. Relatively high TUNEL levels, reduction of proliferating cell nuclear antigen and cyclin-dependent kinase 4 expression, and fragmentation of poly (ADP-ribose) polymerase were also elucidated. These results suggest that commercial oral wet wipes could exert cytotoxic influences on oral tissue, although there are anti-bacterial effects, and careful attention is required, especially for infants and toddlers.

Influence of Some Commercially Available Mouthwashes on Teeth (일부 시판 구강양치액이 치아에 미치는 영향)

  • The purpose of this study was to investigate the chemical properties of some commercially available mouthwashes and to ascertain whether the mouthwashes accelerated mineral loss in dental enamel. Five commercially available mouthwashes were selected from the three largest malls in Korea: Perio Total 7 Aqua Cool Mint Strong $Fresh^{TM}$ (PS; LG Household & Health Care Ltd.), Garglin $Original^{TM}$ (Dong-A Pharmaceutical Co., Ltd.), Garglin $Zero^{TM}$ (Dong-A Pharmaceutical Co., Ltd.), Listerine Naturals $Citrus^{TM}$ (LC; IDS Manufacturing Ltd.), and Listerine Cool $Mint^{TM}$ (LM; IDS Manufacturing Ltd.). The composition, pH, and titratable acidity of the mouthwashes were investigated. Six bovine teeth specimens were prepared for each mouthwash group. Each of the six specimens was individually immersed in 30 ml aliquots of mouthwash for 1 minute, 30 minutes, 90 minutes, and 120 minutes, and the samples were placed in a $36.5^{\circ}C$ stirred incubator. The degree of mineral loss (${\Delta}F$) of the tooth surface area exposed to mouthwash, compared with normal teeth, was analyzed by quantitative light-induced fluorescence-digital. The difference in ${\Delta}F$ among mouthwash groups was examined by the Kruskal-Wallis H test (${\alpha}=0.05$). The contents of mouthwashes differed between Listerine and other products, and the pH ranged from 4.09 to 6.75. The titratable acidity of PS was the lowest at 0.63 ml and highest at 9.25 ml for LM. Minor mineral loss was observed when dental specimens were immersed in the Listerine products (LC and LM) for more than 90 minutes, but the degree of mineral loss for Listerine products was not statistically significantly different from that for groups without mineral loss. In conclusion, all five commercially available mouthwashes showed no harmful effects on tooth enamel.

Current status of fluoride concentration and information labeling of oral hygiene products on the Korean market (국내시판 불소함유 구강위생용품의 실태조사)

  • Objectives: This study was conducted to survey the currently available fluoride-containing oral hygiene products in Korea to provide consumers with information regarding the concentration and form of fluoride in each product, as well as to determine whether the information was easy to understand. Methods: A total of 64 types of domestic commercial oral hygiene products were purchased from an offline market and evaluated. Results: The domestic commercial toothpaste products contained fluoride in the form of sodium fluoride (NaF) and sodium monofluorophosphate (SMFP). In this study, toothpaste containing 1,000 ppm fluoride compounds accounted for the largest proportion (61.5%). Toothpastes containing below 1,000 ppm fluoride accounted for 34.6%, while toothpastes with fluoride above 1,000 ppm fluoride accounted for 3.9%. Toothpaste containing more than 1,000 ppm fluoride has not been popularized domestically. Mouthwash products contained fluoride compounds at less than 300 ppm concentration. Of the five types of mouthwash products, only two types had labels indicating fluoride concentration. In addition, the location of the labels indicating fluoride concentration differed between manufacturers and even within the same manufacturer. Conclusions: It is important to popularize toothpaste with fluoride levels above 1,000 ppm so that a broader selection of toothpaste can be offered to consumers in need. Standardized information needs to be provided for consumer convenience to aid in choosing appropriate oral hygiene products.

  • This study was intended to prepare the basic data required for the development of oral products for the patient with diabetes by analyzing the relation between diabetes and oral health and the actual condition of oral care from the senior citizens diagnosed with diabetes out of the senior citizens over 65. In this study, 8 senior citizens who visited the senior welfare center in D city, were organized into the group of 4 senior citizens and the interview approximately for 40 minutes per group was conducted. In the results of this study, 5 core topics of 'Diabetes and Oral Health", 'Oral Discomfort', 'Oral Care Method', 'Visit to Dentist' and 'Demand for Oral Care' were drawn. For the relation between diabetes and the oral health, the most of research subjects answered that they heard about it for the first time, They answered that for the oral health care, they brushed the teeth using toothpaste 3 ~ 4 times a day and for the visit to dentist, they visited the dentist only when they had oral problem. For what is necessary for oral care, they answered that they wish that the oral care education customized for patient with diabetes and diverse oral care products such as toothpaste, etc. would be developed. Considering the results of this study, the development of oral care management program and the development of oral products in future are deemed to be necessary.

The New in vitro Oral Irritation Test Method for Toothpaste using YD-38 Oral Mucosal Cell Line (치약에 대한 YD-38 세포주를 활용한 새로운 구강 점막 자극 시험방법)

  • Through our entire life, oral care products such as toothpaste are used. Thus the safety of oral care products used every day to our mouth is very important. As the previous study in animal tests or clinical trials, surfactant in toothpaste may cause the oral irritation. However, EU cosmetics legislation prohibits animal testing of cosmetics and its ingredient for animal welfare. Therefore the development of alternative in vitro test has been actively performed to replace or reduce using the animal in many areas. However, the way to evaluate oral mucosal toxicity has been done using animal models or clinical trials from now on. Even more, the experiment with human oral 3D tissue or human oral cell line is used recently. The aim of this study is the development of oral mucosal irritation method without using animal for the safety of the oral care product. We developed in vitro test method for oral irritation by using human oral cell line (YD-38 cell) acceptable to toothpaste which contains insoluble material. By the results of this assay, we could discriminate toothpaste with or without irritating substance as same manner in animal studies reported previously. In addition, we confirmed that toothpaste for babies and children toothpaste irritated oral musoca lower than the general adult toothpaste. The present study suggest that this new in vitro method by using human oral cell line (YD-38 cell) could be used for evaluation of oral irritation without using animal.

Variations of biological status and feeling for car conditioner with time (시간적 흐름에 따른 차량용 에어컨에 대한 쾌적감과 생체의 변화)

  • 차량용 에어컨은 열적 쾌적감을 고려해서 제품설계를 해야 하는 아주 민감한 제품이다. 본 연구에서는 차량용 에어컨의 성능 향상을 위한 기초연구의 하나로서 시간의 흐름에 따른 피실험자의 주관적 쾌적감과 피부온, 구강온, 맥박, 혈압의 변화를 비교 분석하고자 한다. 차량 운행중 에어컨을 사용한다면 바람을 직접적으로 받는 머리 부분은 10-20분 후에 급격히 피부온이 떨어지고, 등을 제외한 나머지 신체 부위는 40분 전후에서야 34$^{\circ}C$ 정도의 피부온을 보였다. 차량에 탑승한 후 50분과 70-80분 이후의 구강온이 초기의 30 분 이전의 구강온과는 유의한 차이를 보이고 있었다. 이것은 구강온이 떨어질 때 졸음의 욕구가 커지기 때문에 운행중 에어컨의 가동은 50 분 이하로 하는 것이 좋고 늦어도 70-80 분 이상을 하는 것은 안전 운행상 문제가 있는 것으로 생각된다.

Research on Natural Medicine for Wellness and Oral Health (웰니스 및 구강질환억제를 위한 천연물 유래물질 연구)

  • This study investigated the antioxidant and anti-inflammatory effects of Galla Rhois (Rhus verniciflua) on LPS (lipopolysaccharide)-stimulated human osteoblastic cells (MG-63). The aim of this study was to evaluate the LPS induced nitric oxide (NO) production and antioxidant radical in human osteoblast-like MG-63 cells. Therefore, the present work indicate that Galla Rhois extracts may be an ideal candidate for further research into their use for dental caries prevention component as well as, natural plant-based products. This suggested that 80% methanol and hexane extracts of Galla Rhois were inhibited NO generation and 1,1-diphenyl-2-pirylhydrazyl (DPPH) radical. Therefore, 80% methanol and hexane extracts of Galla Rhois may be utilized as a good source of protection against inflammation and oxidative stress. This study is intended to pursue wellness convergence in quality of life in the excavations in natural ingredients to create a variety of oral products with fewer side effects, cheap oral products for the dental treatment.

Assessment of Nutrient and Sugar Content and pH of Some Commercial Beverages (일부 시판음료의 영양성분, 당도 및 pH 평가)

  • The purpose of this study was to provide information on the labeling of nutritional components on beverages to aid in nutrition education and oral health promotion. The study was conducted to evaluate nutritional effects and risk factors associated with the consumption of different beverages with respect to oral health. A total of 52 products from seven different types of beverages were analyzed for their nutrient content, sugar content, and pH. The sugar content per serving size, based on the nutrition labeling of beverages, was highest for the milk beverages, at 26.6 g, and lowest for the teas, at 13.0 g. According to the recommendation of the World Health Organization (WHO), beverages should contain less than 10% (50 g) total sugars. Our assessment revealed that total sugars in and carbonated beverages were 53.2% and 50.0% of daily value, respectively. Therefore, the milk and carbonated beverages contained more than 50% sugars per serving size, exceeding the recommendation of WHO. The pH of the beverages, from the most acidic to the least acidic were: carbonated beverages, pH 3.0; fruit and vegetable beverages, pH 3.1; mixed beverages, pH 3.6; fruit and vegetable juices, pH 3.7; teas, pH 4.7; coffees, pH 6.6; and milk beverages, pH 6.8. The intake of acidic and sweetened beverages could potentially cause dental caries and erosion. Therefore, the results of this study could be used by oral health care professionals to counsel their patients by providing relevant information on the possibility of oral disease caused by consumption of commercial beverages.

  • 열적 쾌적감은 각 개인간의 대사작용의 차이, 실행중인 작업 의복의 종류나 계절의 영향, 환경순응, 습관이나 전통도 많은 영향을 미친다. 열적 쾌적감을 조절하는 기기중 하나인 에어컨은 특히 열적 쾌적감을 고려해서 제품설계를 해야 하는 아주 민감한 제품이다. 본 연구에서는 차량용 에어컨의 영향을 평가하기 위한 기초연구의 하나로서 차량용 에어컨이 작동 중일 때 피 실험자의 신체 부위별 주관적 쾌적감과 피부온, 구강온, 맥박, 혈압간의 관계를 분석하고자 한다. 실험은 1 인의 피실험자를 대상으로 9 회에 걸쳐 chamber 에서 1 회에 약 90 분간 실험했다. 차량용 에어컨에 대한 주관적 쾌적감에 가장 영향을 많이 미치는 요소로서는 머리 부위의 피부온과 구강온이 선택되었으며, 다른 부위의 피부온, 맥박, 혈압등은 큰 영향이 없는 것으로 나타났다.

A Research on the Life Span extension of Die Block in Cold Forging Die (냉간단조금형에서 다이블록의 수명연장에 관한 연구)

  • 냉간단조금형(Cold Forging Die)의 다이블록(Dieblock)을 제작하는 방법 중의 하나로, 다이블록 제작용 재료를 면가공 하여 다이블록 상면(上面)을 마스터펀치(Master Punch)인 호브(Hob)로 압입(Indentaion) 시켜 절삭가공((Cutting Work)이 아닌 다이호빙(Die Hobbing) 방법으로 임프레션(Impression)을 성형하여 제작하고 있다. 이 방법에 의하여 다이블록의 재료를 합금공구강(Alloy Tool Steel)인 SKD11을 사용하여 제작하고, 스테인리스판(Stainless Sheet Metal)을 제품 재료로 하여 냉간단조가공(Cold Forging Work)을 하였더니 6,000 스트로크(Stroke)에서 금형수명(Die Life)을 다 하였다. 본 논문에서는 다이블록 재료를 고속도공구강(High Speed Tool Steel)인 SKH51로 교체 제작하고, 탄소강(Carbon Steel)인 S45C를 제품 재료로 하여 냉간단조가공을 수행 하였더니 21,000 스트로크에서 금형수명을 다하고 종료 되어 종래의 방법과 비교 검토 하였을 때 350%의 금형수명 연장 효과를 얻게 되었다.
