Anti-allergic Effect of Ethanolic Extract of Flos Sophora japonica L. on Ca++ Ionophore Stimulated Murine RBL-2H3 Cells (괴화(槐花) 에탄올 추출물이 RBL-2H3 비만세포에서 Ca++ Ionophore에 의한 알레르기 반응 조절에 미치는 효과)

  • Elevation of intracellular calcium ($Ca^{{+}{+}}$) triggers degranulation of mast cells by bypassing receptor activation. Flos Sophora japonica L. has been used as a natural dying source and has been reported to have biological activities such as anti-inflammatory and anti-allergic effects through $Fc{\varepsilon}RI$ and IgE crosslinking. In the present investigation, we report the regulatory effect of ethanolic extract of Flos Sophora japonica L. (S.F) on allergic mediators produced by $Ca^{{+}{+}}$ ionophore activation in mast cells. S.F significantly inhibited calcium ionophore (A23187)-induced interleukin (IL)-4 and tumor necrosis factor (TNF)-${\alpha}$ production as well as mast cell degranulation. Furthermore, administration of S.F suppressed allergic reactions in a 2,4-dinitrofluorobenzene (DNFB)-induced allergic dermatitis mouse model. Both oral administration and ear painting using 50 mg/kg of S.F significantly reduced levels of cytokines such as IL-4, TNF, and interferon-${\gamma}$ in ear tissues compared to the DNFB alone-treated group. Serum IgE level in the S.F-treated group also decreased compared to the DNFB alone-treated group. Weights of spleens and lymph nodes in the S.F-treated groups also decreased compared to the control group. Considering the data, we conclude that S.F mediates its anti-allergic effects not only through $Fc{\varepsilon}RI$ stimulation but also $Ca^{{+}{+}}$ influx in mast cells.

  • Sophora japonica has been used for treatment of liver and blood-related diseases in herbology. We evaluated whitening activities of Sophorae Flos and Fructus. Sophorae Flos and Fructus were extracted with methanol (MeOH) and divided into petroleum ether, ethyl acetate(EtOAC) and water fraction. For whitening effect by western blot in B16 F10 cells, we analyzed it by investigating the effect of tyrosinase, TRP-l (tyrosinase-related protein 1), TRP-2 (tyrosinase-related protein 2) and MITF (microphthalmia-associated transcription factor) protein expression. The protein expressions of tyrosinase, TRP-1, TRP-2 and MITF in B16 F10 cells treated with Sophorae Flos and Fructus extract were reduced in a dose-dependant manner. Therefore, these results suggest that Sophorae Flos and Fructus have useful plant resource to be developed as functional cosmetic.

쪽과 괴화를 이용한 양모직물의 복합염색

  • 천연염색의 색상다양화를 위하여 단백질 섬유인 양모직물에 쪽과 괴화를 이용하여 복합염색을 실시하였다. 복합염색시 쪽염액으로 먼저 염색한 후 괴화염액으로 복합염색한 경우가 괴화염액으로 염색한 후 쪽염액으로 복합염색한 경우에 비해 염착량이 크게 상승한 것으로 나타났다. 복합염색시, 양모직물의 경우 색상의 다양한 효과는 괴화로 먼저 염색하는 것이, 중간색상 발현의 경우에는 쪽으로 먼저 염색하는 것이 효과적이었다.

  • Flos Sophora japonica L. (Leguminosae), commonly called scholar tree, is a well-known traditional medicine used for the treatment of bleeding and as an antihemorrhagic agent. This research was conducted to determine biofunctional activities of Flos Sophora japonica extract. Methanolic extract from Flos Sophora japonica was partitioned by using organic solvents, including n-hexane, ethyl acetate, n-butanol, and water. Ethyl acetate soluble fraction showed the strongest antioxidant activity ($RC_{50}=3.13{\mu}g/mL$) among the fractions. In antimicrobial activity assays, ethyl acetate soluble fraction was effective to bacterial inhibition, such as Escherichia coli and Klebsiella pneumonia, with minimum inhibitory concentrations in $125{\mu}g/mL$. In anticomplementary activity assays, water soluble fraction was the most effective exhibiting 21% inhibitory activity.

  • The growth promoting effect of Clostridium hutyricum KCTC 1785 was investigated with natural products bearing antioxidative capacity, and combined two, three and four kinds of them. C. butyricum was showed a good growth by Lycii fructus, Sophorae flos, Chelidonium majus L., Atractylodis rhizoma alba, Paeonia japonica, alone, and two mixed com-binations were composed of Paeonia japonica and Epimedii herba, Paeonia japonica and Aurantii nobilis pericarpium, Paeonia japonica and Puerariae radix, Pneonia japonica and Angelicae gigantis radix, and three mixed combinations were organized with Epimedii herba, Sophorae flos and Nnelumbo nuclfera gaertner, and Epimedii herba, Sophorae flos and Scutellaria haicalensis george, and Epimedii herba, Sophorae flos and theae folium, and Epimedii herba, Paeonia japonica and Angelicae gigantis radix, and four mixed combinations were formed with Epimedii herba, Puerariae radix, Nelumbo nuclfera gaertner and Paeonia japonica, and Epimedii herba, Puerariae radix, Nelumbo nuclfera gaertner and Theae folium, and Epimedii berba, puerariae radix, Nelumbo nuclfera gaertner and Angelicae gigantis radix, and puerariae radix, Nelumbo nuclfera gaertner, paeonia japonica and Theae folium. As these combinations of natural products could activate some parts fo body, they might be applied to pharmaceuitcal applications, functional foods, antiaging tea, also expected to promote useful enterobacterial growth for multifunctional fermentative beverage.

The Effect of Chitosan Treatment of Fabrics on the Natural Dyeing using Japanese Pagoda Tree (I) (키토산 처리포의 괴화 천연염색에 관한 연구(I))

  • Cotton fabric md nylon fabric were chosen as base fabric specimens for dyeing using Japanese pagoda tree colorants through chitosan, treatment. With the chitosan treatment, the dye-uptake of the treated fabric increased. This treatment is also expected to be effective in terms of environment-friendliness. The effect of the dyeing methods, mordanting or non-mordanting, and chitosan treatment on the dye-uptake and air permeability of the treated fabrics was investigated. In case of cotton fabric, Al mordanted dyeing resulted in higher dye-uptake through the chitosan treatment. Therefore, the chitosan treatment is effective in this case. Japanese pagoda tree seems to have direct affinity for nylon fabric without the mordanting treatment. In case of cotton fabric, it seems that the cellulose molecules, colorants, and the chitosan make a complex, thereby reducing the air permeability. In case of nylon fabric, due to the fact the Japanese pagoda tree colorant molecules form direct physical bonding with the nylon molecules, it seems that there is not much of air permeability reduction.

Dyeing of Wool and Nylon Fabrics with Chinese Scholar Tree Extract (괴화 추출물에 의한 모와 나일론직물의 염색성)

  • This study was discussed the dyeing of wool and nylon fabrics with Chinese Scholar Tree extract. The extracts of Chinese Scholar Tree was prepared in the condition of heating at $95{\pm}5C$, for 1 hour and cooling to 40C. And then the extracts of color matter treate with vacuum concentration at $60{\pm}2^{\circ}C$, 30mmHg and dried with spray dryer. The dyeing of wool and nylon fabric in this experiment was also employed the mordant dyeing method such as pre-mordant, post-mordant and simultaneous mordant method. The mordanting agents used in this study were as followings ; aluminium potassium sulfate, copper(II) acetate monohydrate, chromium potassium sulfate$.$$7H_2O$, Tin(II) chloride dihydrate, iron(II)sulfate $7H_2O$. For an evaluation of the dyeing property of the mordanting agents, the pre-mordant method, the repeat dyeing and the fastness of the light, dry cleaning, washing and rubbing measured respectively. From the results of the dye absorption, the optimum dyeing condition of the wool and nylon fabrics with Chinese Scholar Tree extract is at 80C dyeing temperature for 60 minutes. The optimum concentration of mordanting agent is Al, Cr, Sn 1%, Fe, Cu 2% solution. In general, the fastness property of the dyed wool and nylon fabrics had a comparatively high grade.

  • In this study, the effects of chitosan pre-treatment on the mordanting effect and dyeability were investigated in the dyeing using Japanese pagoda tree. The chitosan treatment effects were evaluated mainly focused on the colors after dyeing of cotton and nylon fabrics which were treated with acidic aqueous solution of chitosan. In the case of chitosan untreated fabrics, the mordanting effects on the cotton and nylon were insignificant and the dyeability was very low. Regardless of the mordant types(Al, Sn, and Fe), the dyed fabrics exhibited almost identical yellowish colors. In the case of chitosan treated fabrics, dark colors were obtained compared to the case of chitosan untreated and ${\Delta}E$ values were increased significantly. In cotton fabrics, the most excellent result was obtained under the condition of unmordanted and chitosan treated fabrics. This verifies the fact that routin coloring matters establish direct linkages with the chitosan components, which exhibit cationic nature, easily. In nylon fabrics, the effects of mordanting treatment and chitosan treatment are so insignificant that the routin coloring matters are presumed to establish direct dyeing with the nylon molecular chains.

  • In this study, physical/chemical characteristics of the fabrics dyed using Japanese pagoda tree which is a yellowish dyestuff was investigated. We tried to estimate the dyeing mechanism among fiber macromolecules, mordants, chitosan, and dyestuffs by measuring the change of air-permeability according to the dyeing process. Also wash fastness and light fastness were measured in order to estimate the fastness according to the dyeing characteristics. When the chitosan pre-treatment was introduced, the wash fastness increased significantly in the cotton fabrics while the fastness did not change in the nylon fabrics significantly. The light fastness was not improved by the chitosan treatment. when the antibacterial activity was measured for the dyed cotton fabrics, Japanese pagoda tree itself did not exhibit antibacterial activity. However, chitosan treated dyed fabrics exhibited low antimicrobial activity.

A Study of Color Difference on Fabrics Dyed with Yellow Natural Material - by Natural Gardenia and Japanese Pagoda Tree - (황색계 천연염색물의 색차에 관한 연구 - 치자와 괴화를 중심으로 -)

  • Natural gardenia and Japanese pagoda tree have been known as them of the most widely used yellow vegetable dyes. Cotton and silk fabrics(KS K 0905) were chosen as specimens. Those were dyed with natural gardenia and Japanese pagoda tree. The conditions of dyeing were varied according to the kinds of mordants, using mordants or not and the concentrations of the natural dyes. Sn, Al, Fe and Cu were chosen for the mordants. 5%(owf), 10%(owf), 15%(owf) and 20%(owf) were used as for the concentration of the natural dyes. The color differences were measured by the Spectrometer(UV-VIS-NIR) before and after dyeing. The results are as follows: 1. Deeper colors were proportionately shown up to the concentration of the natural dye. 2. Silk was shown the higher dye affinity than the cotton at the same concentration of the dye. 3. The color difference between the Sn-mordanted specimens and the Al-mordanted specimens were significant. When the concentration of the dye increased, the colors got darker, specially in b* value. 4. The color difference between the Fe-mordanted specimens and the Cu-mordanted specimens were significant in L* value and b* value. 5. The deeper or lighter yellow were controllable under the concentration of the natural dye and the mordants. 6. The most effective and the economic concentration of the natural dye was 10% owf through the experiments.
