Effect of Photosynthetic Bacterial Addition to Chlorella or ${\omega}-Yeast$ on Growth of Rotifer, Brachionus plicatilis, and its Dietary Value for Flounder, Paralichthys olivaceus, Larvae (Rotifer, Brachionus plicatilis, 성장을 위한 광합성세균의 첨가 효과와 넙치, Paralichthys oliraceus, 자어에 대한 먹이효율)

  • This study was carried out to investigate the effect of photosynthetic bacteria to chlorella or ${\omega}-yeast$ on Browth of the rotifer and its dietary value for flounder, Paralicbthys oliraceus. The rotifer fed the chlorella (200,000 cells/ind./day) with the addition of 20 times the photosynthetic bacteria of the chlorella concentration showed the highest growth. But the specific growth rate of 100,000 chlorella/ind./day with the addition of 30 times the photosynthetic bacteria was the most economical feeding regime for mass culture of the rotifer. The rotifer frd ${\omega}-yeast$ with 200,000 cells/ind./days with the addition of 20 times the photosynthetic bacteria of the chlorella conecentration showed the highest growth. Growth and survial rate of the larvae of Paralichithys oiivaceus fed the rotifer reared on both chlorella and ${\omega}-yeast$ with the addition of photosynthetic bacteria were higher than those without the bacteria, and the chlorella had better dietary value than the ${\omega}-yeast$ for the larvae. The larvae fed the rotifer which was cultured with the chlorella of 200,000 cells/ind./day and the photosynthetic bacteria of $4{\times}10^6$ cells/ind./day showed the highest survial rate and growth. The larvae reared with the addition of the photosynthetic bacteris had higher total lipid and the total content of EPA and DHA than those reared without the bacteria. The larvae fed the enriched artemia nauplius with the photosynthetic bacteria also showed higher suurval rate and growth than those fed the nauplius without the enrichment. The optimum enrichment concentration of the photosynthetic bacteria for artemia nauplius was $2{\times}1^7\;cells/ml$. The addition of the photosynthetic bacteria to the chlorella and the ${\omega}-yeast$ was effective to growth of the rotifer and dietary value for the flounder larvae. However, an excessive addition of the bacteria decreased both the growth of the rotifer and the dietary of the larvae.

Development and Application of the Photosynthesis Experimental Module Based on Scientist's Inquiry Processes (과학자의 탐구 과정을 재구성한 광합성 실험 모듈의 개발과 적용)

  • This study was intended to develop an experimental module based on inquiry processes conducted by photosynthesis scientists. It was aimed to enhance scientific inquiry ability of the middle school students by applying the developed module. Developed module included some experiments conducted by earlier photosynthesis scientists such as Helmont, Woodward, Priestly, Hales and Ingen-Hausz. Inquiry process was involved in the developed module for instructing the inquiry methods. Rapid-cycling Brassica rapa known as a Fast Plant was used for the experimental material. Developed module was applied to the experimental group consisting 27 eighth grader, while experiments suggested in the science textbook was applied to the control group consisting 30 eighth grader. Developed module was more effective in improving students' scientific inquiry ability, especially measuring, forecasting and hypothesizing ability as its subordinate elements. When the result of post-test was compared to one of pre-test in the experimental group, their observing, forecasting, and generalization ability were improved. Experimental group showed that students' conception in photosynthesis and conceptual development related with the role of plants in the ecosystem and plant's food and movement of the water and nutrients were also improved. Before application, students in the experimental group did not have enough understanding of the abstract concept such as the existence or the role of the materials like $CO_2$ or $O_2$ or the energy accumulation. Developed module could help students to achieve the comprehensive concept regarding the role of plants as producers of organic matter and oxygen and to enhance their scientific inquiry ability and concepts regarding photosynthesis.

Characterization of Photosynthetic Rates by Tomato Leaf Position (토마토 엽위별 광합성 특성 분석)

  • The photosynthetic rates according to leaf positions in tomato plants were investigated in relation to leaf age and flowering rate. In the experiment investigating the diurnal change of photosynthetic rates, three leaves below the 4th cluster was checked every hour from 2 hours before sunrise and 3 hours after sunset. It was checked twice with the replication of 3 plants. The photosynthetic rate increased sharply for 1 hour right after sunrise and remained steady until 2 hours before sunset. This trend can be applied to determine the irrigation schedule. In the experiment investigating the photosynthetic rates according to leaf positions, it three leaves below each clusters from 1st to 4th cluster were checked. Flowering rate was also investigated. The photosynthetic rates showed a decreasing tendency steadily after flowers bloomed fully, regardless of the leaf position. It seems to be because the leaves below the cluster with fully-bloom flowers lost their activities. This result suggests the flowering rate or the position of flower has deep relation with the photosynthetic rates of the concerned leaves. From the results the leaves under flowering cluster may be the good part to investigate the photosynthetic rate to evaluate the crop's activity, even the photosynthetic rates are different according to the position of clusters.

Modeling of Microalgal Photosynthetic Activity Depending on Light Intensity, Light Pathlength and Cell Density (빛의 세기, 투과거리 및 세포농도에 따른 미세조류의 광합성 활성 모델링)

  • The influenced of light intensity, cell density, and light pathlength on photosynthetic activity of Chlorella vulgaris were investigated. Since the light respon curve varied according to reaction conditions, the parameters estimated from nonlinear regression were proved to be apparent and could not be applied to various situations. The light response model incorporating the light penetration through the microalgal suspension was developed based upon the spatial distribution of the photosynthetic activity. This model showed a good agreement with experimental data at different cell densities and light intensities. Using the model the effects of cell density and light pathlenth were simulated and some dicussions about optimization of operation conditions of photobioreactors were carried out. Concludingly, the developed model can be useful for predicting microalgal photosynthesis and for determining the optimal operating conditions.

  • This report gives the results of studies on the photosynthetic kinetics. Futhermore, conformity and validity of the kinetic equation are elucidated by the experiment of sunflower leaves. The kinetic equations of photosynthesis are dirived from the equations of assimilation and dissimilation. The equations (6) and (7) are obtained by the result of this investigation. Conformity and validity of the above kinetic equations are elucidated by the fitted equation (8) on photosynthesis of sunflower leaves in relation to various environmental factors.

  • 식물체에서 일어나는 유전자 발현의 광조절 현상은 매우 복잡하다. 이는 각 유전자들이 조직에 따라 질적, 양적, 시간적 측면에서 빛에 대한 반응에 커다란 차이점을 보이고 있기 때문이다. 따라서 식물체를 대상으로 광조절 기작을 연구하는 데는 현상의 복합성과 실험과정의 기술적, 시간적, 경제적 제약이 많기 때문에 이에 관한 연구가 아직 초보적 수준에 머물고 있다. 광합성 세균 중에서 purple bacteria나 green bacteria와는 달리 식물성 광합성을 하며, 식물체보다 세포구조가 훨씬 간단한 cyanobacteria는 유전자 발현의 광조절 기작을 연구하는데 간단, 명료한 'model'로서 기대되는 바 크다. 따라서 이 글에서는 cyanobacteria의 광계와 광합성 색소인 chlorophyll의 생합성 과정을 중심으로 광조절 현상에 대한 최근 연구 동향과 전망을 살펴보고자 한다.

Highly Pure Culture of Photosynthetic Bacteria for Pilot-scale Production (광합성세균의 파이롯트 단위 생산을 위한 고순도 배양)

  • Lab-scale and pilot-scale productions of photosynthetic bacteria, that were able to efficiently treat wastewater from aquacultural farm, were experimented for their highly-pure culture. The results of experiments in a gas pack reactor, an anaerobic flask and a flask using $N_2$ gas as N-source showed that only photosynthetic bacteria formed red colonies on agar plate and their purity was over 90% in a colony, observed under a microscope. It was found that the basal medium could most promote the growth of photosynthetic bacteria, confirmed by experiments of serial cultures on various media. Under the culture conditions, the specific growth rate was found to be $0.18h^{-1}$ from the culture in 5L bioreactor and the same value could be obtained in pilot-scale production.

Correlation among Production Traits of Soybeans according to the Re-watering (재관수에 의한 대두의 물질생산 관련형질의 상호연관성)

  • This study was conducted to investigate the influence of soil moisture on the photosynthetic rate, transpiration rate, stomatal conductance, leaf water content and its any correlation in soybean. Followings were achieved as a conclusion. At the soil moisture was moved from drought condition to saturation condition, the photosynthetic rate, transpiration rate, stomatal conductance and leaf water content were gradually increased on the period of re-watering treatment. The recovery of photosynthetic rate was faster than others. There were positive correlation with the photosynthetic rate and leaf water content, the stomatal conductance and leaf water content, the photosynthetic rate and stomatal conductance respectively, but the correlation coefficient of photosynthetic rate and leaf water content was high.

  • Void fraction of air bubble in the seagrass bed by photosynthesis was estimated with sound speed dispersion. A field experiment was conducted at Seagrasss bed of which bottom type is sandy mud and 120 kHz CW waveform was transmitted to obtain backscattered signals from seagrass bed. The differences of the arrival time of received signal from seagrass bed were observed between day and night. The diurnal variation of arrival time was caused by sound speed dispersion of air bubble generated by photosynthesis of seagrass.

The Analysis of Elementary Pre-service Teachers' Reflective Thinking and Experiment Performance Ability on Photosynthesis Experiment (광합성 실험에서 나타난 초등 예비교사들의 반성적 사고와 실험 수행 능력 분석)

  • In order to find out Elementary pre-service teachers' reflective thinking and experiment performance ability related with Photosynthesis Experiment in the Korea Elementary School Science Textbook, the research is conducted targeting Elementary pre-service teachers. They are asked to carry out the experiment and write their own report about the difficulties and solutions of exploration process. This study aims to analyze Elementary pre-service teachers' reflection and experiment performance ability on Photosynthesis experiment based on 10 groups' reports and presentation materials. Reflective thinking extracts 108 statements which is associated with the four types of the sentence 'Knowledge, Procedure, Orientation, Attitude' in 10 reports. There are many sentences about reflective thinking acquired through analysis of the photosynthesis experiment. reflective thinking about the newly discovered type or changed concepts through experimentation in Knowledge is at the highest frequency. 56 sentences in relation to the ability to perform experiments are extracted by adding 4 different types of reflective thinking in 10 groups shown the highest frequency group and the lowest frequency group's report through analyzing 4 steps 'Experimental preparation and safety accident prevention', 'Experiments performance', 'Experimental results and generalization', and 'Experimental results and feedback.' Results of the analysis showed that there are the biggest difference between the two groups in 'experiment results supplement and feedback step.' In the lowest group's report, there's no contents related with 'Computer-assisted information processing' in the 'Experimental results summary and generalization stage', 'Alternative reagents and materials research', and 'Devising alternative experiment methods'.
