Production and characterization of monoclonal antibodies against rabies virus (광견병바이러스에 대한 단크론항체 생산 및 특성)

  • Rabies virus which belongs to the genus Lyssavirus of the family Rhabdoviridae is known as a highly neurotropic virus and causes fatal encephalitis accompanied by severe neurological symptoms in almost all mammals, including humans. In this study, monoclonal antibodies (MAbs) against rabies virus were produced, characterized and applications of MAbs as diagnostic reagents were assessed Spleen and inguinal lymph node cells from Balb/c mouse immunized with purified rabies virus were fused with SP2/O myeloma cells using polyethylene glycol (PEG) and hybridoma cells producing rabies virus-specific MAbs were screened by an indirect fluorescent antibody test. A total of ten MAbs were produced against rabies virus. The protein specificity and neutralizing activity of MAbs were determined by Western blot analysis and fluorescent antibody virus neutralization test, respectively. As a result, two MAbs, 5G3 and 6H4 had specificity for nucleoprotein (N protein) and two other MAbs, 5B1 and 5C1 had neutralizing activity for rabies virus. Some MAbs recognized the rabies virus-infected bovine brain stem cells by immunohistochemistry (IHC) assay. In conclusion, it was confirmed that MAbs produced in this study were rabies virusspecific and could be used as reliable diagnostic reagents for the detection of rabies virus.

Natural Dyeing of Hanji with Alnus japonica Fruit Extractive (오리나무 열매 추출물을 이용한 한지의 천연염색)

  • We dyed traditional Korean hand-made papers (Hanji) with colorants extracted from fruits of Alnus japonica to investigate the effect of various dyeing factors (mordant, dye concentration and dyeing assistant) on colors, K/S values and light fastness of the dyed Hanji. The dyed hanji had brown color. The K/S value of dyed Hanji was increased by mordanting with alum and copper acetate. $a^*$ and $b^*$ value of dyed Hanji was decreased slightly by mordanting with alum and Iron(II) chloride. The K/S value and $b^*$ of dyed Hanji increased with increasing concentration of dye, but $L^*$ value of dyed Hanji decreased. The K/S value of dyed Hanji was also increased by fixing agent. When Hanji was mordanted with Iron(II) chloride and was not mordanted, use of fixing agent resulted in greater increase of K/S value. Use of fixing agent resulted in poor light fastness. When fixing agent was used, mordanting with copper acetate resulted in smaller color difference than mordanting with alum after aging test.

Dyeing Properties and Color of Silk Fabrics Dyed with Safflower Yellow Dye (홍화 황색소 견섬유에 대한 염색성과 색상)

  • The objective of this study is to investigate the dyeing properties of safflower yellow dye on silk for the standardization of dyeing process and color reproducibility. Yellow colorants were water-extracted from safflower petals, concentrated, and freeze-dried to obtain colorants powder. The effects of dye concentration, dyeing temperature, and pH of dye bath were studied in terms of dye uptake and shade. Fastness to dry cleaning and light was evaluated. Dye uptake increased with raising temperature and brighter and more vivid yellow shade was obtained when dyed at $30^{\circ}C$. As colorants concentration increased, dye uptake increased progressively and the shade got darker and deeper. Maximum color strength was obtained at pH 3.5. It was speculated that the adsorption of colorants seemed to occur mainly by hydrogen bonding and physical force at pH 5.5 and by ionic bonding as well as hydrogen bonding below isoelectric point(pH 3.8-4.0). The results of reproducibility test showed acceptable color difference in the range of $1.11{\sim}2.01$. Washing fastness was fairly good as 4/5 rating, while light fastness was 2/3 rating.

  • The painting group of Late Ming and Early Qing Dynasty is an era of confusion between the orthodox who faithfully cooperated with the Qing Dynasty and the individualist who had a hostile attitude. The country was ruined by these dynasties, and the family left the real world with the suffering of the breakdown and stayed in the mountains.Paldaesanin and Seokdo, the representative Yumin painters, did not form any a sect as a unique expression style for finding a true self.And, through his work, he expressed on madness and oddity through new ideas and composition that he feels misery and anger in his heart. The paintings of these paintings had semi-normative and impromptuous views of nature, which even the self burdened with grief was forgotten.And I expressed my own individuality and originality by forming a simple and free, on madness and oddity art aesthetic. The paintings of these paintings had semi-normative and impromptuous views of nature, which even the self burdened with grief was forgotten.And I expressed my own individuality and originality by forming a simple and free, on madness and oddity art aesthetic. Paldaesanin portrayed a strange figure with a spirit of resistance and a sad and angered emotion, through a unique technique of painting.The content is cynical, satirical, ironic, and on madness and oddity. On the other hand, Seokdo explained "ilhoeg" that the method of writing and the method of writing in "Hwaeolog" agree with each other.This has allowed us to achieve autonomy as a "rule without rules" that goes beyond the existing rules. And he delicately portrayed beauty, desire, and emotion with the use of sensuous brushes and the beauty of the colors.Their unique paintings were later conveyed to yangjupalgoe and led to the flow of paintings in the 18th century.
