Prevalence of Toxin Genes and Antibiotic Resistance Profiles of Vibrio vulnificus strains isolated from Jeju Island (제주도에서 분리된 비브리오패혈증균의 독소 유전자 분포 및 항생제 내성)

  • Vibrio vulnificus, the most fatal waterborne and foodborne pathogens of 50% fatality rate in the world, is common in seawater and occurs particularly in warmer months. In this study, we investigated the toxin genes using reverse transcription-polymerase chain reaction (RT-PCR), antibiotic resistance status using Vitek, and genetic characteristics using pulsed-field gel electrophoresis (PFGE) of different V. vulnificus strains isolated from the Jeju Island seawater, distribution fishery products, and fish tanks. We examined a total of 487 samples and isolated a total of 46 strains (including overlapping strains) of V. vulnificus, 44 strains from seawater and 1 strain each from fishery products and fish tank. We detected toxin gene vvhA in all 46 strains and rtxA, viu in 8 strains (17.4%) and 9 strains (19.6%) strains, respectively. Antibiotic resistance tests indicated 100% resistance to cefoxitin antibiotics. The PFGE analysis of the 46 strains identified a total of 6 types showed 100% homology and the degree of similarity was 81.3-98.0%; however, there were no similarity between the regions and samples. These results indicate that V. vulnificus isolated from the seawater, fishery products, and fish tanks should be continuously monitored as cases of food poisoning caused by V. vulnificus with toxin genes have been reported in Jeju Island.

Cases and Significance of Inscriptions with Homophonic yet Misspelled Words on Buncheong Ware from the Early Joseon Dynasty: Buncheong Bowl with Inscription of "Naeja" ('내자(內子)'명(銘)으로 본 조선 전기 분청사기의 동음오자(同音誤字) 명문 사례와 의의)

  • The inscription found on buncheong ware (粉靑沙器, grayish-blue-powdered celadon) provides information on the bowl today, but in the past served as a device for controlling tributary payments (貢納) and the movement of government property (官物). The inscription had to be written or engraved clearly so that it could be recognizable to officials in charge of managing bowls. Such inscriptions offer important clues for contemporary research on buncheong ware. Buncheong Bowl with Stamped Design and Inscription of "Naeja" (內子) (bon 13808) in the collection of the National Museum of Korea bears a distinct inscription compared to other buncheong ware. Commonly, the inscription on buncheong indicated the name of the government office (官司名) to which the vessel belonged or the name of the place (地名) where the vessel was produced. However, the inscription "Naeja" on the buncheong bowl at the National Museum of Korea has no corresponding government office. "Naeja" is in fact a misspelling of Naeja (內資), meaning this bowl belonged to the Naejasi (內資寺, Royal Bureau of Procurement). Although "Naeja" (內子) was a misspelling of the intended Naeja (內資), it was still understood as a reference to the Naejasi since they were homophonic. Recently, buncheong ware with misspelled yet homophonous inscriptions have begun to be excavated in downtown Seoul. Examples including "Naeja" (內子) instead of Naeja (內資) and "Insu" (仁守) instead of Insu (仁壽) have been unearthed from historical sites in Seoul that functioned as consumption sites, meaning they were in fact circulated as government property despite the misspellings (whether accidental or the outcome of ignorance). Such misspelled yet homophonous inscriptions are characterized by the use of simple characters and a few sloppy strokes. Like other buncheong ware bearing the inscriptions of government office names, the bowls with the inscriptions of "Naeja" (內子) and "Insu" (仁守) were discovered at historical sites in Seoul. These misspelled homophonous inscriptions reveal that errors occurred during the process of engraving inscriptions on buncheong ware produced as a tributary payment during the early Joseon Dynasty and that buncheong ware with such errors were still distributed.

Influence of the Soluble Salt on the Exfoliation of the Stone Monument (수용성염이 석조문화재 표면 박리현상에 미치는 영향)

  • The mechanism of stone exfoliation and its cause in relation to chemical weathering by soluble salt were studied. Chemical, mineralogical and physical analysis was performed in exfoliation samples from stone monuments. The representative salt is gypsum in the exfoliation samples. In order to understand the salt reaction, stone samples(tuff and granite) were treated with two type of the salt, gypsum and sodium sulfate, which have different solubility. The capillary water uptakes are slight increased in impregnated with Na2SO4 and weathering simulation of two rock types. It means that the rock is deteriorated in the near of the surface by $Na_2SO_4$. $CaSO_4{\cdot}2H_2O$ bring out the thicker exfoliation than $Na_2SO_4$ because it is penetrated into the deeper zone and amount of accumulated salt is more abundant in the inner part than in the near of the surface. The exfoliation was formed in the tuff by salt treatment and 30cycle of weathering simulation, but there are not significant symptoms of exfoliation in the granite by same condition. This result was caused by the different capillary water uptakes and porosity of the rocks. In the tuff, salt solutions are penetrated into the inner part due to its high capillary water uptakes and porosity but the granite, which has low value relatively, can be formed thinner exfoliation.

  • At platform-reinforcing side stone plate having been exhibited in outdoor exhibition space of Gyeongju National Museum, split and exfoliation were proceeding due to weathering at joined·restored parts. Accordingly we checked imbibitional minerals in the stone through observation and analysis with polarized microscope, and studied capillary water absorption rate using Schmidt Hammer rebound hardness measurement and Karsten Tube.

  • The purpose of this is to make sure what the scholar of the Choseon thought the object of aesthetics, and which psychological element did they use to clear the center of the aesthetics. In addition, I consider their way to recognize the human's aesthetic point of view. A literary man in Choseon regarded the five senses as an obstacle which interrupted to see the nature of all things. So they set up the spiritual condition as their aesthetic subject, with suppressing the pleasure through five senses. The goal to make one's mind calm was to cause someone's spirit to activate freely. The condition of the calmness was considered as a process of recovering the human's natural spirit, which was connected to the poetic spirit and became the aesthetic subject. Simultaneously, a literary man at that time set up the condition of the calmness as a subject of objective consciousness, and estimated the common things as a objective existence, which led to the result that all the materials of the poem and the poet could be unified. This aesthetic centra put a premium on the senses caused by contacting with all natural things, and adopted the 'Xing' as a method of expression. This 'Xing' is not a sense based upon one's experience, but a sense which is created of itself. The expression method of 'Xing' caused the reader to feel and express, encourage, and sublimate what they feel through the poem, as well as purified reader's mind.

Molecular Characterization of Vibrio parahaemolyticus Isolates from Seawater, Fish Tanks, and Distributed Fishery Products in Jeju (제주지역 해수, 수족관물, 유통수산물에서 분리된 장염비브리오균의 분자생물학적 특성)

  • V. parahaemolyticus causes waterborne and foodborne disease such as acute diarrhea. In this study, V. parahaemolyticus isolates from seawater, fish tanks, and distributed fishery products in Jeju were investigated for potential toxin or species-specific genes (tdh, trh, tlh, and toxR) using RT-PCR and their genetic characteristics were analyzed using Pulsed-field gel electrophoresis (PFGE). Overall, V. parahaemolyticus of 90 strains (36.7%), including 33 strains from seawater, 8 strains from fish tanks, and 50 strains from fishery products, were isolated from 245 samples. All V. parahaemolyticus strains did not detect the tdh gene, whereas all strains detected tlh or toxR genes. In addition, trh genes were detected in 3 strains from seawater and 1 strain from fishery products. Monthly quantitative testing of seawater revealed that V. parahaemolyticus was positively correlated with water temperature. The 90 strains of V. parahemolyticus obtained in this study showed by gene homology between types, ranging from 64.0-97.3%. Among these, thirteen types showed 100% homology between genes. These results indicate that continuous monitoring is needed to facilitate food poisoning epidemiological investigations because some isolated V. parahaemolyticus strains harbored toxin genes and V. parahaemolyticus strains isolated from seawater, fish tanks, and distributed fishery products showed genetic similarity.

국내산 장석에 대한 산업광물로서의 광물특성 평가

  • 장석은 복잡다단한 광물학적 특성에도 불구하고 그 물질적 이해와 용도 측면에서의 단순성 때문에 관련 산업 부문에서 부가가치 향상이 잘 이루어지지 않고 있는 실정이다. 현재 전 세계적으로 10위권 내의 생산 수준을 유지하는 국내산 장석의 광석 유형은 그 관물상과 산출상태에 따라 페그마타이트상 장석, 반화강암질 및 우백질 화강암상의 장석으로 구분 될 수 있다. 현재 국내에서 개발되고 있는 장석들로는 일반적으로 반화강암질 유형의 광석이 가장 흔하다. 이 유형의 광체들은 흔히 알바이트화 작용이나 K-부화를 수반하는 열수변질 작용을 받은 양상을 보이는 것이 특징이다. 광물특성 상으로는 K-장석에 속하는 광체들이 Na-장석 유형들 보다 상대적으로 흔하고 대체적으로 그 부존 규모도 큰 것으로 나타난다. 장석의 부가가치 향상을 위해서는 그 품위를 화학조성에만 의존하지 말고 광물조성 단위로 이해하는 방식이 요구된다. 일반적으로 이 광물자원의 요업적 용도 특성상, 광물특성 평가에서 가장 주요한 기준이 되는 사항은 화학조성, 특히 알칼리 조성, $AI_{2}O_{3}$ 함량 및 철분의 함유도인 것으로 판단된다. 그렇지만 앞으로 그 수요가 확장될 전망이 있는 충진재 용도로 사용될 경우에는 이 같은 화학적 특징보다는 장석광물의 조성과 제반 물성적 특성에 그 품질이 의존될 것으로 여겨진다. 이에 따라 장석 광석의 화학조성과 물성은 기본적으로 장석의 광물상과 조성에 의존된다는 지식기반 하에서, 장석의 품위는 물론 품질 특성 평가에 있어서도 화학조성에만 의존하지 말고 그 광물상과 광물특성도 고려해서 평가하는 지혜가 요구된다.

A Study on the Application of SILRES BS OH 100 Consolidants for Shale (셰일에 대한 SILRES BS OH 100 강화제 적용연구)

  • The consolidation application of SILRES BS OH 100 was investigated, which has been used for consolidation of the weathered shale. The liquid SILRES BS OH 100 was polymerized by the sol-gel reaction with air moisture, and the XRD patterns showed that the gel was an amorphous solid. The drastic weight reduction of the sample was found by differential thermal analysis, which was followed to the formation of $Si(OH)_4$ particles. After consolidation, the polymerized gel was filled into the voids within the shale. The capillary water absorption of the consolidated shale was reduced to 48.7%, and the abrasive strength was improved.

Music, Language, and Life in Daoism and Confucianism (음악과 언어, 그리고 삶 - 도가와 유가를 중심으로 -)

  • This essay is an analysis on Daoist deconstructivism and Confucian constructivism about music and language. (1) Daoist criticizes that the Confucian constructive music and language fail to describe original sounds and original facts of doing nothing (wuwei, 無爲). According to Daoist, music and language can be an instrument to describe true facts in the world. So Daoists try to attain a state of 'seeing things as things themselves (yiwuguanwu, 以物觀物)' by 'forgetting oneself (wangwo, 忘我).' (2) However Confucian music and language is a part of one's life. Confucians try to get truth, goodness, and beauty by exercising one's music and language. Confucian music is associated with political and moral development in society. The Confucian genres of poetry (shi, 詩), appealing letter (shu, 疏), declaring writing (biao, 表), record (ji, 記), and written words (ci, 詞) are processes of developing one's life. Further, Confucian rhetoric of 'Xing (興)' in writing poem shows that one's language can be developed in contexts of one's life. (3) Although music and language is associated with human subjective narratives as if Confucians say, diverse narratives of different subjectivity cannot appear in one's lives if all kinds of narrative is absorbed in Confucian absolute ideological slogan to devide things into good and bad. Accordingly, the Confucian view of music and language can develop diverse narratives when it does not show an inclination toward moral dichotomy preunderstood by Confucian ideology.
